Study on skin anti-inflammatory activity of fig (Ficus carica L.) fruit extract fractions (무화과(Ficus carica L.) 열매 추출 분획의 피부 항염증 활성 연구)

  • Figs has known to have antioxidant, whitening, anti-inflammatory, and antibacterial effects in their leaves, roots, stems, latex, and fruits. In order to develop cosmetic materials based on natural products, we have studied on the skin activity of the ficin in latex as well as the whitening function of the fruit extract with 70% ethanol, and used it as a raw material for released cosmetic product. However, there is little research on the demand for the development of new eutectic solvent extraction methods and its ability to control skin inflammation and psoriasis regulation. Thus, in this study, we evaluated the effectiveness of fig fruit extracts and fractions using eutectic solvent extraction for skin inflammation control and psoriasis. First, fig fruits were extracted under optimal eutectic solvent conditions and fractionated with n-hexane, dichloromethane, ethyl acetate, and butanol. First, the antioxidant activity and inhibition of nitric oxide (NO) production were confirmed in mouse macrophage RAW264.7 cells. In addition, as a result of observing the mRNA expression through RT-PCR, pro-inflammatory cytokines such as TNF-α, IL1α, and IL-1β were suppressed significantly in the hexane, dichloromethane, and ethyl acetate fractions. In addition, it was confirmed in TNF-α stimulated HaCaT keratinocyte model. Finally, chemokine CC motif ligand 20 (CCL20), marker gene of human psoriasis skin disease, was significantly suppressed in the hexane, dichloromethane, and ethyl acetate fractions. These results suggested its anti-inflammatory and skin soothing effect and the possibility of development as an excellent skin soothing natural cosmetic material in the future through future clinical trials.

The Anticarcinogenic and Antioxidative Activity of Hemicentrotus pulacherrimus Fractions in Various Cancer Cells. (말똥성게 분획물에 의한 항 발암 및 항산화 효과)

  • This study was carried out to investigate the anticarcinogenic and antioxidative activities of Hemicentrotus pulacherrimus (HP). HP was extracted with methanol (HPM), which was then further fractionated into four sub-fractions by using the solvent partition method, affording methanol (HPMM), hexane (HPMH), butanol (HPMB) and aqueous (HPMA) soluble fractions. We determined the anticarcinogenic activities of these four fractions in four kinds of cancer cell lines, such as HepG2, HT29, MCF-7 and B16-F10, by MTT assay. Among various fractions from HPM, the HPMH showed the strongest growth inhibition effect. We also determined the inductive effect on quinone reductase (QR) of HP fractions. HPMB fraction exhibited strong inductive effects in HepG2 cells at a level of 90 ${\mu}g/ml$, showing inductive indexes of 2.26 compared to the control value of 1.0. The antioxidant activities of fractions from HP were also investigated by measuring the scavenging activities of HP against reactive oxygen speicies (ROS), peroxynitrite (ONOO-) and NO. Among the various solvent fractions, HPMH fractions displayed marked antioxidative activities.

Study on Cosmeceutical Activities and Anti-inflammatory Activities of Magnolia biondii Extracts (신이화(辛夷花, Magnoliae Flos)추출물의 화장품약리활성 및 항염증효과에 관한 연구)

  • Existing pharmaceutical studies show that Magnolia biondii is effective in treating rhinitis and in reducing cholesterol, given its endogenous, volatile ingredients. The study herein seeks to assess the cosmeceutical activities and anti-inflammatory activities of Magnolia biondii extracts for possible application as cosmetic ingredients. The cosmeceutical and anti-inflammatory activities were investigated using hydroxyl radical scavenging, superoxide dismutase (SOD)-like activity, xanthine oxidase (XO) inhibition, cell viability, nitric oxide (NO) inhibition, and inducible nitric oxide synthase (iNOS) expression by Western blotting. Magnolia biondii extracts were identified to have antioxidant activities in hydroxyl free radical scavenging, SOD-like activity, and XO inhibition. In testing the anti-inflammatory activities of the extracts, NO production was inhibited in a dose-dependent manner. Additionally, in a dose-dependent manner, the Magnolia biondii extracts were able to suppress iNOS expression in LPS-stimulated RAW 264.7 macrophage cells. From these results, Magnolia biondii showed adequate potential for application in cosmetic production and related industries as well as a functional material.

A Study on the Development of Baby Powder Using Silk gland Powder of Silkworm (누에생실샘 미세분말을 이용한 베이비파우더 소재 개발에 대한 연구)

  • In this study, silk gland powder of silkworm were investigated to see the possibility for baby powder cosmetics materials. To test possibility as a baby powder cosmetics, total content rate of amino acids, DPPH free radical scavenging assay, MTT assay, and clinical trial were done. According to the result of the analysis of the amino acids of silk gland powder, serin (26.77%) content was the highest and asparatic acid (15.47%), and glycine (9.62%) were followed. DPPH free radical scavenging activity of silk gland powder was lower than vitamin C by 82.3% and 97%, respectively, which is relatively good. Moisture effect were increased in silk gland powder compared to control cosmetics by 50%. Also, silk gland powder was classified as a practically non-irritating material based on the score 0.05 of primary irritation index. Thus, these results suggest that silk gland powder of silkworm may have beneficial properties as a material for baby powder cosmetics.

The Effects of Paeonia Lactiflora Pallas on Inhibition of Oxygen Free Radical, Anti-inflammation and MMP-1 Inhibitory Activity (적작약 꽃 추출물의 활성산소 억제와 항염증 및 MMP-1 발현 억제능 효과에 관한 연구)

  • This study attempted to investigate the effects of Paeonia Lactiflora Pallas (P. lactiflora) on the inhibition of oxygen free radical, anti-inflammation and MMP-1 inhibitory activity and examine its possibility as a functional cosmetic material. For test methods, the inhibition of oxygen free radical after measuring reactive oxygen species (ROS) in the cell, cytotoxicity assessment and anti-inflammation were measured, and MMP-1 inhibitory effects in the HDF cell were measured. According to the test, the inhibition of ROS was confirmed in RAW 264.7 and HDF cells. In terms of cytotoxicity assessment, 90% or higher cell viability was detected at $5/10{\mu}g/mL$ Paeonia Lactiflora Pallas extract while it was 80% or higher at other concentration levels in both RAW 264.7 and HDF cells. In addition, NO production was inhibited in the RAW 264.7 cell while MMP-1 was significantly inhibited in the HDF cell. The above results reveal a possibility of Paeonia Lactiflora Pallas extract as a functional cosmetic material after confirming the inhibition of ROS synthesis in the cell, antioxidant and anti-inflammatory effects by inhibiting NO synthesis, low toxicity on skin cells and anti-aging effect through MMP-1 inhibition.

Increased Genotoxicity of N'-methyl-N'-nitroguanidine by Oxidative Stress (산화적 스트레스에 의한 N'-methyl-N'-nitroguanidine의 유전독성증가)

  • To investigate the possible enhancement of genotoxicity in stress environment, we examined the of effect of genotoxic material in oxidative stress-induced condition using human tell line. Human lymphoblast cell line, TK6 was treated with hydrogen peroxide ($H_2O_2$) for induction of oxidative stress, and treated with N'-methyl-N'-nitroguanidine (MNNG), af a genetoxic material. We carried out MTS assay to set treatment doses. TK6 was treated with $H_2O_2$ at 6.75 (low dote) or $13.5\;{\mu}M$ (high dose) for 2 h, and treated with MNNG af 0.117 (low dose), 0.234 (middle dose), $0.468\;{\mu}M$ (high dose) for 2 h. As results, a treatment of MNNG induced DNA dam age as dose dependently. And TK6 treated with $H_2O_2$ at low as well as high dose followed by MNNG treatment showed higher DNA damage compared to MNNG alone treated groups. Malondialdehyde, as a marker of lipid peroxidation was increased in $H_2O_2$ and MNNG treated groups. Real-time RT-PCR analyses for expression of several antioxidative enzymes showed that catalase mRNA and glutathione peroxidase 1 mRNA expression were decreased in $H_2O_2$ and MNNG treated groups. Taken together, we conclude that genotoxicity induced by MNNG is enhanced in a condition of oxidative stress induced by $H_2O_2$ and it suggests that it should be associated with induction of lipid peroxidation and decrease of antioxidant enzymes.

Morphological Characteristics of 'Ilbon' (Ziziphus jujuba) with Higher Seed Productivity and Functional Component (종자형성능과 기능성 성분이 높은 대추 유전자원 '일본'의 형태적 특성)

  • Morphological characteristics of Ziziphus jujuba accessions, including 'Ilbon', were investigated to verify the possibility of 'Ilbon' as a breeding material. 'Ilbon' has smaller leaves and fruits and a lower total soluble solids content compared to the major Z. jujuba accessions. However, total polyphenol content and antioxidant capacity were highest in 'Ilbon' among the Z. jujuba accessions. In addition, 'Ilbon' could produce intact seeds and seed productivity of 'Ilbon' was highest compared with major Z. jujuba accessions. Principal component analysis and clustering analysis using the morphological characteristics of Z. jujuba accessions have been classified them to geographical origin. In particular, 'Ilbon' was clearly distinguished from the major Z. jujuba accessions. Therefore, 'Ilbon', which has the highest seed productivity and functional component, will be used as a material for cross breeding and will promote genetic study of Z. jujuba.

Clinical Studies on the Anti-Irritation Effects of Mung Bean (Phaseolus aureus) Extract in Cosmetics (녹두추출물의 자극완화 효과에 관한 임상 연구)

  • The aim of this study is to assess the anti-irritation activities of mung bean (Phaseolus aureus) extract against various irritants used in cosmetics. For its antidotal activity, mung bean has been used as a medicinal or cosmetic material since ancient times. However, there have been few reports describing the biological activities of these beans and no comprehensive surveys of the constituents. We obtained an ethanolic extract of mung bean and isolated the major constituents, such as vitexin and isovitexin. And we previously reported that the mung bean extract containing vitexin and isovitexin had excellent antioxidant and anti-inflammatory activities. To investigate the mechanisms of anti-inflammatory activity of mung bean extract, we examined the inhibitory effects on histamine release from rat peritoneal mast cells and lipoxygenase activity. Mung bean extract inhibited histamine release in a concentration dependent manner but showed no inhibitory activity in the 5-lipoxygenase assay. And, clinical studies were conducted to evaluate the anti-irritation effects of mung bean extract against various irritants used in cosmetics such as lactic acid, retinol, and preservatives. When 2.0% of mung bean extract was applied to cosmetic formulae containing each of irritants, it revealed considerable anti-irritation efficacy. Our results of the human patch test with 20 volunteers showed that this extract reduced skin irritations caused by 5.0% lactic acid, 4000 IU retinol, and 1.0% preservative mixture by about 60%, 30%, and 50% respectively. The stinging potential test for assessing subjective irritation also showed that the extract reduced the unpleasant sensations by about 50∼30%. Finally, we performed a double-blind usage test with 30 subjects to compare formulae containing mung bean extract with placebo. From the results of questionnaires for 4 weeks of use, we confirmed the excellent anti-irritation effect of mung bean extract. Conclusively, we could discover new material that had anti-irritation effects and apply this mung bean extract to the final cosmetic products successfully.

Optimization of extraction conditions of flavonoid compounds from Thyme (Thymus vulgaris Libiatae) (타임으로부터의 플라보노이드 화합물의 추출 조건 최적화)

  • Total polyphenol contents, flavonoid contents, 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activities of ethanol extracts of 20 herbal plants (sage, turmeric, mace, bayleaf, fenugreek, oregano, blackpepper, whitepepper, clove, marjoram, cinnamon, coriander, basil, dillseed, mustard, cadamon, thyme, celery, rosemary, cumin) were analyzed for the screening of high flavonoid-containing plant resource. Thyme extract, showing the highest flavonoid content and a high degree of antioxidant activity, was selected as a bioactive cosmetic material. The total polyphenols and flavonoids contents of thyme extracts were measured as 6.90 mg chlorogenic acid equivalent (CAE)/100 g and 1.71 mg naringin equivalent (NE)/100 g, respectively, and DPPH radical scavenging activities was 90%. Among the tested organic solvents, hexane gave the highest extraction yield. Thus hexane was selected as the most suitable solvent for the extraction of thyme. Response surface method was used to obtain optimized extraction conditions for thyme: reaction temperature of 35.9 ℃, raw material to hexane ratio of 1.63:25 (w/v), and reaction time of 192 min. These predicted extraction conditions was validated by a total flavonoid extraction experiment showing a value equivalent to 96.3% of the predicted total flavonoid content. It is expected that the optimized solvent extraction conditions could be used for the production of flavonoid using thyme.

Genotoxicological Safety of Octadeca-9,12-dienyl-3,4,5-trihydroxybenzoate from Gallic and Linoleic Acids and Its Biological Functions in Cream-based Emulsion (Gallic Acid와 Linoleic Acid로부터 합성한 Octadeca-9,12-dienyl-3,4,5-trihydroxybenzoate의 유전독성학적 안전성 및 화장품 제형을 통한 생리 기능성 평가)

  • The objective of this study was to investigate the genotoxicological safety and biological functions of octadeca-9,12-dienyl-3,4,5-trihydroxybenzoate (GA-LA) in cream-based emulsion for future application as a functional cosmetic material as well as food. GA-LA was synthesized chemically from gallic acid and linoleic acid. The Ames test showed that GA-LA did not have mutagenical toxicity. The control cream-based emulsion containing GA-LA was prepared by commercial method and tested for 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity. Ferric reducing antioxidant power (FRAP) and inhibition effect against tyrosinase of the emulsions were tested for the evaluation of antioxidative and skin-whitening activities. The results showed that DPPH radical scavenging activity in the cream-based emulsion containing GA-LA was higher (52.65%) than that of the control (4.30%). The FRAP value of the sample was 12.85%, however, no activity was found in control. The inhibition effect of tyrosinase showed also a higher value (26.29%) when compared to the control. The results indicate that GA-LA, which showed superior antioxidative and skin-whitening activities in cream-based emulsion, is a useful functional material applicable in cosmetic products as well as food.
