The effects of cell lytic enzyme treatment on total phenolic content, antioxidant and antityrosinase activities of lotus leaf were investigated. The dried lotus leaves were hydroyzed by cell lytic enzymes such as Promozyme, Ceremix, Pectinex, Ultraflo, Celluclast, Pentopan, Tunicase, Viscozyme at their optimum pHs (pH 5-8) at $50^{\circ}C$ for 4 hrs. Depending on the enzymes used, total phenolic compounds content was measured as $1,079-1,476{\mu}g$/mL, and antioxidant activities and whitening activities were increased by 5~10% and 20%, respectively Among the tested hydrolytic enzymes, Promozyme (pullulanase) was selected as the most suitable enzyme for the extraction of total polyphenol from lotus leaf. The optimal dosage of Promozyme were found to be 1-2% (w/w). By Promozyme treatment, total phenolic compounds content of the lotus extract significantly increased compared to the extraction without enzyme treatment.
BACKGROUND/OBJECTIVES: Isoflavones are widely believed to be beneficial to human health, in relation to their antioxidant potentials. Exercise can cause an imbalance between reactive oxygen species (ROS) and antioxidants. This study was conducted in order to investigate the ability of isoflavones in amelioration of oxidative stress induced by exercise. MATERIALS/METHODS: Male Sprague-Dawley rats were assigned to one of four groups: isoflavone-free with no exercise (CON-sd), isoflavone-free with exercise (CON-ex), isoflavone-supplemented with no exercise (ISF-sd), and isoflavone-supplemented with exercise (ISF-ex). Animals exercised on the treadmill for 30 minutes per day, five days per week. TBARS as a marker of oxidative stress and antioxidant enzyme activity, including SOD, GSH-px, and catalase were determined in liver tissue. Serum lipid profile was also examined. RESULTS: A significant effect of isoflavone alone was observed on abdominal fat pad mass. ISF-ex had significantly less abdominal fat pad than CON-ex. Both exercise and isoflavone treatment had significant effects on lowering plasma triglyceride (TG), thus, the ISF-ex group had a significantly lower TG level than the CON-sd group, by 30.9%. However, no differences were observed in plasma cholesterol, HDL-C, and cholesterol/HDL-C ratio. Exercise, isoflavone, and exercise-isoflavone interaction effects were significant on thiobarbituric acid reactive substances (TBARS) (P = 0.001, 0.002, and 0.005, respectively). The CON-ex group showed a higher TBARS level than the other three groups. By contrast, in the ISF-ex group, TBARS was restored to the level of the ISF-sd or CON-sd group. Isoflavone had a significant effect on superoxide dismutase (SOD) (P = 0.022) and catalase activities (P = 0.049). Significantly higher SOD and catalase activities were observed in ISF-ex than CON-ex. SOD and catalase activities showed an inverse pattern of TBARS. Taken together, isoflavones increased the activities of SOD and catalase with concomitant decreases in TBARS, indicative of decreased oxidative stress. CONCLUSIONS: Isoflavone supplementation enhances antioxidant action with attenuation of exercise-induced oxidative stress, as measured by decreases in TBARS, and inhibits body fat accumulation and plasma TG increase. Antioxidative effects ascribed to isoflavones may be partially exerted via enhancement of antioxidant enzyme activities.
Antioxidant and redox enzyme activities are known to be involved in the cellular responses to various stresses. Their variations were observed according to the growth cycle of the fission yeast Schizosaccharomyces pombe. Peroxidase activity appeared to be notably higher in the early exponential phase than in the mid-exponential and stationary phases. However, catalase activity showed a variation pattern resembling the growth curve. Glutathione S-transferase activity was higher in the early exponential and late stationary phases. Activities of the two redox enzymes, thioredoxin and thioltransferase (glutaredoxin), were high in the stationary phase. However, their activities appeared to increase from the early exponential to mid-exponential phase. Total glutathione content had a varying pattern similar to that of thioredoxin and thioltransferase. However, its content in the early exponential phase was high. These results propose that antioxidant and redox enzymes tested are also involved in the mechanism of cell growth.

Antioxidant activities of mulberry leaf extracted with different concentrations of EtOH were investigated. Total phenolic content and electron donating abilities of extract from 70% EtOH were the highest. Extracts obtained from EtOH-water mixture were shown to be significantly higher superoxide dismutase (SOD)-like activities than other treatment (p<0.05). Angiotensin-converting enzyme (ACE) inhibition was the greatest at 50% EtOH concentration (p<0.05). The extracts from 30-70% EtOH exhibited higher ferric reducing ability of plasma (FRAP) value than rest of the concentration (p<0.05). In case of nitrite scavenging activity, much higher scavenging activities were observed when the extraction was performed with EtOH or EtOH-water mixture (p<0.05). The results indicate that concentration of EtOH as extraction solvents can affect the antioxidant activity of mulberry leaf, which may provide useful information on the optimal solvent conditions for the extraction.
Objectives : This study has been carried out to understand the effect of Uncooked Korean Food(F-DM) on blood glucose and antioxidant enzyme activities in streptozotocin-induced Diabetic Rats. Methods : SD rats were separated into four groups(each with 20 rats). Except normal two group, the other two groups were injected into intra-peritoneal with streptozotocin 60 mg/kg. Experimental group was eated Feed with 25% F-DM for 4 weeks. The change of plasma glucose level, body weight were observed. After 4 weeks, liver and kidney weight, antioxidant enzyme activities, survival rate were observed with histological changes on liver, kidney and pancreas. Results : In experimental group, body weight and survival rate increased, plasma glucose level were decreased significantly. Liver and kidney weight, XOD activity were decreased in experimental group compared to control group. GSH-px and CAT activities, insulin- immunoreactive granules in p-cells were increased significantly in experimental group compared to control group. Conclusions : This study shows that the F-DM might be effective for treatment of diabetes and its complications, as well as reduction of the oxidative stress.
A laboratory experiment was conducted to determine the content of phenolics and flavonoids, antioxidant activity and antioxidant enzyme activity for the extracts from cowpea seed and sprouts. Plant length and weight of cowpea sprouts were significantly increased until 7 days after seeding. Total phenolics level [mg chlorogenic acid equivalents (CAE) $kg^{-1}$ DW] was highest in dry seed (DS) extracts of cowpea ($63.9mg\;kg^{-1}$), followed by imbibed seed (IS) ($56.8mg\;kg^{-1}$) and 1-day-old sprout (1DOS) extracts ($46.4mg\;kg^{-1}$), and significantly reduced with increase of sprout age (p < 0.05). The antioxidant activity of the methanol extracts from all the samples showed same tendency to the results of total phenolics level, and dose-dependently increased. DPPH (1,1-diphenyl-2-picryl hydrazyl radical) free radical scavenging activity was higher in DS (87.3%) and IS (41.2%) than in cowpea sprouts from 1DOS to 7DOS, ranging from 17.1 to 30.4%. Antioxidant enzymes, APX, POX, and POX activities were highest in 7DOS and lowest in DS. SOD activity showed much higher activity in sprouts and in seeds. Correlation coefficient between physiological-active substance and the activity was highest between APX and CAT activities ($r^2$= 0.9574). Especially, total phenolics content was more highly correlated with antioxidant or with antioxidant enzyme activities than was total flavonoid level.
To elucidate the relationship between blood parameters related bone metabolism and antioxidant enzyme activity in postmenopausal period 60 women residing in Iksan area were recruited. Food and nutrient intake of each individual subject were estimated by 24-hour recalls of 3 non-consecutive days. The biochemical markers including total protein, albumin, osteocalcin (intact bone gla protein; BOP), calcium, phosphorus and hemoglobin were measured in fasting blood. In addition, parameters of antioxidative capacity including the activities of superoxide dismutase (SOD), glutathione peroxidase (GPx) , catalase (CAT) and total antioxidant capacity (TA) were monitored in blood, also. The mean age, height, weight, and BM! of subjects were 64.8 years, 151.1 em, 59.5 kg $26.0\;kg/m^2$, respectively. The mean SOD, GPx, and CAT activities were 138.5 U/ml, 1,273.8 U/ml and 314.3 kU/l respectively, and TA was 1.16 mmol/l without significant difference among different age groups. BMI was positively correlated with SOD activity (p < 0.01). SOD activity and CAT activity showed positive correlation with serum albumin (p < 0.05) and hemoglobin (p < 0.01). In conclusion, this study revealed that antioxidant enzyme activity holds a significant relationship with the blood parameters like as serum albumin and hemoglobin in postmenopausal women and further systematic research is needed to investigate the their relation mechanism.
The non-enzymatic antioxidants and antioxidant enzyme from the extracts of Parthenocissus tricuspidata PLANCH. were examined in order to utilize natural product for cerchemopreventive agents. The antioxidant potential and enzyme activities on plant positions in the extracts of Parthenocisuss tricuspidata PLANCH. showed considerable differences. The antioxidant activity of the leaf extracts by Ethyl acetate fractions of Parthenocisuss tricuspidata PLANCH. was the highest among three positions ($7.57\mu\textrm{g}/m\ell$). The highest activities showed in S-5 (in leaf), S-4 (in stem) and S-3 (in root) fraction by Silicagel column chromatography and the antioxidant activity showed, in purified extract of each positions, $7.06\mu\textrm{g}/m\ell$ (in leaf), $6.99\mu\textrm{g}/m\ell$ (in stem) and $12.39\mu\textrm{g}/m\ell$ (in root) respectively. The activities of DPPH by LH-20 column chromatography revealed much higher than those by silica-gel column chromatography. These were identified as the phenolic compounds known as antioxidant compounds such as Benzoic acid(Gallic acid), 1-methyl-3-(2-phenylethen) benzene, phloroglucinol and 1,2-dihydroxy-4-(1-propyl)benzene by GC/MS. POD activities in the stem and root were higher than in the leaf. SOD activity was highest in the leaf, stem and root activity was comparatively low. Especially, SOD activity in leaf was over 2 times higher than root.
Ultraviolet (UV) A penetrates deeply into the skin and induces the generation of reactive oxygen species (ROS) causing damage to fibroblasts, which leads to aging of the skin. However, the body has developed an antioxidant defence system against the harmful effects of ROS. Enzymes such as superoxide dismutase (SOD) and catalase (CAT) play critical roles on the removal of excess ROS in living organisms. In this study, the antioxidant activities of anthocyanins (cyanidin 3-galactoside and cyanidin 3-lathyroside) from Acanthopanax divaricatus var. albeofructus (ADA) fruits were investigated by xylenol orange, thiobarbituric acid reactive substances (TBARS), and antioxidant enzyme assay. As a result, generation of $H_2O_2$ and lipid peroxide induced by UVA-irradiation in human dermal fibroblast (HDF-N) cells was reduced by treatment of anthocyanins. Also, augmented enzyme (SOD and CAT) activities were observed in UVA-irradiated cells when treated with anthocyanin. In conclusion, the results obtained show that anthocyanins from ADA fruits are potential candidates for the protection of fibroblast against the damaging effects of UVA irradiation. Furthermore, anthocyanin may be a good candidate for antioxidant agent development.
This study investigated concentrations of the several air pollutants and compared antioxidative enzyme activities on Platanus occidentalis because this tree species is one of the widespread street trees in Korea. This species has been emerging the ambient air pollutants during its growing periods. The purpose of this study was to identify the relationship between air pollution on the tree species and antioxidant enzyme activities on the trees. $O_3$, $NO_2$, CO and $SO_2$ concentrations of several cities in Korea were compared for last decades. Among the air pollutants, $O_3$ and $NO_2$ concentrations in six big cities in Korea showed similar increasing trends during this period. In contrast, $SO_2$ and CO concentrations in the same cities dramatically decreased between 1994 and 2005. Platanus occidentalis trees were controlled to investigate, ascorbate peroxidase (APX) and glutathione reductase (GR) activity. Ozone exposure generally increased APX and GR activities of tree seedlings. It is a typical compensatory strategy of stressed trees.
