물과 주정을 이용한 삼채 뿌리와 잎 추출물의 항산화 활성 및 항염증 효과 비교 (Comparison of Effect of Water and Ethanolic Extract from Roots and Leaves of Allium hookeri)

  • 본 연구는 삼채의 뿌리와 잎을 물과 주정을 이용하여 추출하여 항산화 활성 및 항염증 효과를 비교 평가하였다. 삼채의 추출물에 포함된 총 폴리페놀 함량과 전자스핀공명기기를 이용한 DPPH, alkyl, hydroxyl 라디칼 소거능, ABTS를 이용한 라디칼 소거 활성 및 FRAP 및 ORAC을 이용한 항산화 활성을 평가하였는데, 삼채뿌리 추출물보다는 삼채잎 추출물의 항산화 활성이 높았으며 이는 잎에 포함된 총 폴리페놀 함량과 관련이 있을 것으로 판단된다. 물 추출의 경우가 뿌리와 잎 모두 수율이 높았으며 폴리페놀 함량과 ORAC value와 alkyl 라디칼 소거 활성은 비슷한 경향을 나타내었으나 DPPH와 hydroxyl 라디칼 소거 활성과는 다른 결과를 나타내었다. 또한 간세포를 이용한 세포 독성 평가 실험 수행 결과, 0.25 mg/mL의 농도까지는 삼채의 뿌리와 잎의 물 추출물 및 주정 추출물 모두 독성을 나타내지 않았다. RAW264.7 세포를 이용하여 독성을 나타내지 않는 농도인 0.2 mg/mL 이하에서 항염증 활성 평가를 수행한 결과 삼채뿌리 및 삼채잎의 물과 주정 추출물은 모두 NO의 생성을 농도 의존적으로 감소시키는 경향을 나타내었다. 뿐만 아니라 삼채 추출물은 염증성 사이토카인인 IL-6와 TNF-${\alpha}$의 생성을 감소시켰으며, 특히 삼채뿌리 물 추출물은 가장 높은 IL-6와 TNF-${\alpha}$의 생성 억제를 나타내었다. 또한 삼채 추출물이 LPS로 유도한 세포내 활성산소종을 농도 의존적으로 억제하는 것을 확인하였으며, 위의 결과로 삼채잎 추출물은 항산화 활성이 우수하고 삼채뿌리 추출물은 항염증 활성이 높음을 확인하였다. 따라서 삼채 추출물이 우수한 항산화 및 항염증 활성을 가지고 있는 것으로 판단되며, 이와 관련된 항산화 및 항염증 생리활성 물질에 관한 추가적인 연구가 필요할 것으로 사료된다.

Anti-Inflammatory Activity of the Solvent-Partitioned Fractions from Spergularia marina in LPS-Stimulated RAW 264.7 Cells

  • As a part of ongoing research to elucidate and characterize antioxidant and anti-inflammatory nutraceuticals, solvent-partitioned fractions from Spergularia marina were tested for their ability to scavenge radicals and suppress inflammation. The results of the 2',7'-dichlorofluorescein diacetate assay indicate that solvent-partitioned fractions from S. marina scavenged intracellular radicals in $H_2O_2$-stimulated mouse macrophages. The tested fractions decreased the generation of nitric oxide (NO) and the expression of inflammation mediators, namely, inducible nitric oxide synthase (iNOS) and interleukin (IL)-6, by lipopolysaccharide (LPS)-induced mouse macrophages, indicating that S. marina decreases inflammation. Among all tested fractions [i.e., $H_2O$, n-buthanol (n-BuOH), 85% aqueous methanol (aq. MeOH), and n-hexane], the 85% aq. MeOH fraction showed the strongest antioxidant and anti-inflammatory response. The 85% aq. MeOH fraction scavenged 80% of the free radicals produced by $H_2O_2$-induced control cells. In addition, NO production was 98% lower in 85% aq. MeOH fraction-treated cells compared to LPS-induced control cells. The mRNA expression of iNOS and IL-6 was also suppressed in 85% aq. MeOH fraction-treated cells. The results of the current study suggest that the phenolic compound components of S. marina are responsible for its antioxidant and anti-inflammatory effects.

쥐참외뿌리 추출물의 항산화, 미백, 항염증 활성 연구 (A Study on the Antioxidant, Whitening and Anti-Inflammatory Activities of Trichosanthis Cucumeroidis Radix Extract)

  • 본 연구에서는 쥐참외뿌리 추출물의 항산화 활성 및 미백, 항염증 활성에 대한 생리활성 효과와 기능성 소재로써의 활용 가능성을 확인해 보고자 하였다. 쥐참외뿌리 추출물의 DPPH 라디칼 소거 활성을 통한 항산화 활성, 멜라닌 세포인 B16F10 melanoma 세포에 대한 멜라닌 생성 억제능을 통한 미백 활성 효과, 대식 세포인 RAW 264.7 세포에 대한 NO 생성 억제능을 통한 항염증 활성 효과를 확인해 보고자 하였다. 연구결과, 쥐참외뿌리 추출물은 농도 의존적으로 DPPH 라디칼 소거활성이 확인되었으며, 양성 대조군인 Ascorbic acid와 비슷한 DPPH 라디컬 소거활성이 확인되었다. 100 nM 𝛼-MSH로 유도된 멜라닌 생성 억제 활성과 LPS 1 ㎍/mL로 유도된 NO 생성 억제능을 유의하게 억제 하는 것을 확인되었다. 이에 따라 쥐참외뿌리 추출물은 항산화 및 미백, 항염증 효과를 가진 기능성 소재로써의 활용 가능성이 있음이 사료되어 진다.

Antioxidant and Anti-Inflammatory Effects of NCW Peptide from Clam Worm (Marphysa sanguinea)

  • Clam worms (Marphysa sanguinea) are a rich source of bioactive components such as the antibacterial peptide, perinerin. In the present study, we explored the physiological activities of a novel NCWPFQGVPLGFQAPP peptide (NCW peptide), which was purified from clam worm extract through high-performance liquid chromatography. Tandem mass spectrometry (MS/MS) revealed that NCW was a new peptide with a molecular weight of 1757.86 kDa. Moreover, NCW peptide exhibited significant antioxidant effects, causing a 50% inhibition of DPPH radical at a concentration of 20 μM without showing any cytotoxicity. These were associated with a reduction in the activity of catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and malondialdehyde (MDA) in LPS-stimulated RAW264. 7 cells. Furthermore, NCW peptide exhibited anti-inflammatory effects in LPS-stimulated RAW264.7 macrophages via inhibition of the abnormal production of pro-inflammatory cytokines including nitric oxide (NO), inducible nitric oxide synthase (iNOS), and cyclooxygenase-2 (COX-2). These anti-inflammatory effects of NCW peptide were associated with the inhibition of interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α). Our results therefore suggest that this novel NCW peptide with antioxidant and anti-inflammatory effects could be a good therapeutic agent against inflammation-related diseases.

Effect of Astaxanthin on Anti-Inflammatory and Anti-Oxidative Effects of Astaxanthin Treatment for Atopic Dermatitis-induced Mice

  • Background: This study sought to determine whether the antioxidant effects of astaxanthin (AST) could have an anti-inflammatory effect to reduce inflammation caused by atopic dermatitis (AD). Methods: Using a mouse model of AD induced by phtalic acid (PA), the levels of inflammation, inflammatory agents, and evidence of antioxidant activity were examined in PA treated mice (n = 3), PA-AST treated mice (n = 3), and a control group of mice (n = 3). This included measurements of ear thickness, levels of mast cells, IgE, inflammatory cytokine, malondialdehyde (MDA), hydrogen peroxide, HO-1, and GPx-1. Results: AST treatment significantly prevented inflammation as measured by ear thickness (p < 0.05), mast cell count (p < 0.001), and IgE concentration in the blood (p < 0.001). Levels of TNF-α (p < 0.001), IL-1β (p < 0.001), IL-6 (p < 0.001), and MDA (p < 0.05) were also significantly lower. In addition, GSH levels increased significantly (p < 0.001), and the level of hydrogen peroxide significantly reduced (p < 0.01). The expression of HO-1, GPx-1 increased. Conclusion: In this small experimental study, AST acted on inflammatory mechanisms that induced AD, through anti-inflammatory and antioxidant mechanisms, and is a candidate of interest in the clinical treatment of AD.

흑마늘박 추출물의 항산화활성 및 항염효과 (Anti-oxidative and Anti-inflammatory Effects of Black Garlic Pomace Extract)

  • 본 연구에서는 흑마늘 제조 시 발생하는 부산물을 이용하여 기능성 사료 첨가제룰 개발하기 위한 사전 실험으로 수행되었다. 흑마늘박의 항산화 활성과 면역증강 활성을 측정하였으며 흑마늘박의 항산화 활성을 측정한 결과 흑마늘박이 항산화 활성이 있는 것으로 나타났다. 실험에 사용된 시료중 흑마늘박을 포함한 식물성 시료의 면역증강 활성을 실험하기 위해 Nitric oxide(NO) assay 실험을 한 결과 식물성 추출물인 흑마늘박, 섬쑥부쟁이, MIX가 최고 농도에서 LPS(100%)대비 각각 흑마늘박(69.4%), 섬쑥부쟁이(35.9%), MIX(45.3%)의 NO를 생성하였다. 따라서, 흑마늘박이 항염증 효과를 함유하고 있고 흑마늘박과 섬쑥부쟁이의 최적의 혼합비를 선정한다면 항염증 효과를 함유하고 있는 사료 첨가제로써 충분한 가치가 있을 것으로 사료된다.

Antioxidant and Anti-Inflammatory Effect of Probiotic Lactobacillus plantarum KU15149 Derived from Korean Homemade Diced-Radish Kimchi

  • Lactobacillus plantarum KU15149 was demonstrated to have probiotic behavior and functions, including antioxidant and anti-inflammatory activity. L. plantarum KU15149 obtained from homemade diced-radish kimchi has a high survival rate under artificial gastric acid (pH 2.5, 0.3% pepsin) and bile salt (0.3% oxgall) conditions. However, L. plantarum KU15149 did not produce β-glucuronidase, which is known to be a carcinogenic enzyme with resistance to several antibiotics, such as gentamycin, kanamycin, streptomycin, tetracycline, and ciprofloxacin. L. plantarum KU15149 strongly adhered to HT-29 cells and had high antioxidant activity in terms of 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical-scavenging and β-carotene bleaching assays. L. plantarum KU15149 also exhibited a pronounced inhibition of nitric oxide (NO) production, along with expression of nitric oxide synthase (iNOS) and cyclooxygenase -2 (COX-2) as well as pro-inflammatory cytokines, such as TNF-α, IL-1β, and IL-6, when RAW 264.7 cells were stimulated with LPS. Therefore, L. plantarum KU15149 exhibited pharmaceutical functionality as a potential probiotic.

Antioxidant and NO-scavenging Activities of Acanthopanax senticosus var. subinermis Leaf Extracts Prepared Using Ethanol and Extrusion Processing

  • Acanthopanax senticosus has been used in East Asia as a traditional medicine. The purpose of this study is the investigation of an extraction method for A. senticosus var subinermis (ASvS) leaves to improve their antioxidant and anti-inflammatory activities. Six extracts were prepared: extracted with water (W), ethanol (Eth), water or ethanol after ultra high pressure (WP, EthP), and water or ethanol after an extrusion process (WEx, EthEx). Diphenylpicrylhydrazyl (DPPH) radical scavenging activity and ferric reducing ability showed that all extracts had a significant antioxidant activity. In anti-inflammatory activities, Eth and EthEx significantly inhibited the release of lipopolysaccharide (LPS)-induced nitric oxide (NO), tumor necrosis factor (TNF)-${\alpha}$, and interleukin (IL)-$1{\beta}$ independent of cell viability in RAW 264.7 macrophage cells. Inducible NO synthase (iNOS) protein was significantly decreased by EthEx. These findings present that Eth and EthEx extracts of ASvS leaves have anti-inflammatory activities, and EthEx extract suppresses LPS-induced NO through the down-regulation of iNOS and pro-inflammatory cytokines.

Effect of Extraction Ethanol Concentration on Antioxidant and Anti-Inflammatory Activity of 30-Year-Old and 120-Year-Old Dangyuja (Citrus maxima (Burm.) Merr.)

  • Dangyuja (Citrus maxima (Burm.) Merr.) is a native fruit of great economic importance in Jeju island in Korea. To provide experimental evidence for the antioxidant and anti-inflammation properties on extraction ethanol concentration of Dangyuja, 2 cultivars, including 30-year-old and 120-year-old were evaluated. 30-year-old Dangyuja 50%, 70% ethanol extracts had the highest polyphenol and flavonoid content, and the strongest 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity. To investigate the anti-inflammatory activity of Dangyuja ethanol extracts, we used BV-2 microglia cells and induced inflammation using lipopolysaccharide (LPS). Then, we measured levels of inflammatory mediators as nitric oxide (NO). Among the 6 extracts, 30-year-old Dangyuja 50% ethanol extracts show the highest anti-inflammatory activity. The results suggest that 30-year-old Dangyuja 50% ethanol extracts provides significant health benefits and may be used for developing new functional materials.

시호 추출물의 화장품 생리활성에 관한 연구 (A Study on the Physiological Activity of Cosmetics in Bupleurum falcatum L. Extract)

  • 시호는 약용작물의 하나로 해열, 진정, 간 장애 억제, 항염, 항암 등 우수한 약리 활성을 지니고 있다. 현재 식품 또는 약용으로 사용되고 있지만 화장품으로서의 사용은 미비하여 화장품 소재로서의 적합성 및 이용 가치를 검토하였다. 시호 추출물을 이용하여 폴리페놀, 플라보노이드, 항산화, 항염 실험을 진행하였다. 폴리페놀 함량은 14.71±0.16 mg·TAE/g, 플라보노이드 함량은 1.42±0.05 mg·QE/g를 나타내었다. 항산화 활성은 농도 의존적으로 우수한 활성을 보였으며, NO 발현 억제능은 1,000 ㎍/mL에서 61.21%의 저해율을 나타내어 항염 활성이 뛰어남을 확인하였다. 본 연구를 통해 시호 추출물의 항산화, 항염 효능이 뛰어나기에 화장품의 천연 소재로서 충분한 가치가 있다고 본다.
