Quality Changes in 'Hayward' Kiwifruit Wine Fermented by Different Yeast Strains (효모에 따른 참다래 'Hayward' 와인의 품질 변화)

  • The yeast strains used for fermentation are known to influence the qualities of wine. We investigated the effects of fermentation using different yeast strains on the properties of wine produced from 'Hayward' kiwifruit (Actinidia deliciosa). The physicochemical characteristics of wine produced using various yeast strains for fermentation were also analyzed. Saccharomyces cerevisiae Gervin No.5 strain (GVN), S. bayanus Lavin strain EC1118 (EC1118), and S. cerevisiae Red star Davis No. 796 (No.796) are commercial dry yeast strains selected for optimization of fermentation. Although the soluble solid contents of samples fermented by all three yeast strains decreased by a similar extent, the levels of alcohol production differed, particularly during the first week of fermentation. Use of the GVN strain resulted in the highest alcohol concentration (13.8%, v/v), whereas fermentation with No.796 and EC1118 strains yielded alcohol contents of 13.0% and 12.5% (both v/v), respectively. Upon sensory evaluation, GVN-fermented wine had a strong taste and bitterness, with high acid and alcohol contents. Wine fermented using No.796 had a chemical profile similar to that of GVN-fermented product, but the taste remained sweet, consistent with the lower alcohol content. EC1118-fermented wine was soft and sweet in taste, high in flavor, and had a low alcohol content. Total phenolic levels and antioxidant activities in wine fermented by EC1118 were significantly higher than in wines prepared using No.796 or GVN. When previously described characteristics were additionally considered, EC1118 was selected as an optimum strain for further study. In conclusion, fermentation of kiwifruit using different yeast strains yielded wines with distinct characteristics. The yeast strain EC1118 had the most desirable properties, and is considered suitable for kiwifruit fermentation. Valuable attributes of wine fermented by this yeast include overall sensory acceptance, an appropriate level of total phenolics, and good antioxidant activity.

Effect of Salt Fermentation on the Physicochemical Properties and Antioxidant Activities of Sea Urchin Roe from Anthocidaris crassispina and Pseudocentrotus depressus (염장처리가 성게 알의 이화학 품질 특성과 산화방지 활성에 미치는 영향)

  • Sea urchin roe obtained from Anthocidaris crassispina and Pseudocentrotus depressus was briefly salt-fermented (5%), followed by ethanol treatment (1%) and the physicochemical properties as well as antioxidant activity were investigated. Compared to raw sea urchin roes, the salted one showed a significantly low amount of water (p<0.001) high salinity (p<0.05), ash content (p<0.001) and Na content (p<0.001). With salt-fermentation, the redness (p<0.05) and yellowness (p<0.001) of roe decreased noticeably, indicating the decomposition of endogenous carotenoids. Accordingly, the salted roe showed a lower DPPH radical scavenging activity than its unsalted counterpart. Additionally, it showed a significantly lower metal-chelating activity (p<0.05) and metal chelator content (e.g. ortho-phenolics) displayed by a negligible difference in titratable acidity. The salted roe showed significantly increased hardness (p<0.05) and total reducing capacity (p<0.001), which were attributed to the protein coagulation and the release of antioxidants bound to macromolecules after the ethanol treatment, respectively.

Physiological activitive of Grifola frondosa by log cultivation and bottle cultivation (원목재배와 병재배 잎새버섯(Grifola frondosa)의 생리활성)

  • This research was carried out to determine the differences of physiological activites between Grifola frondosa of log cultivation(LC) and Grifola frondosa of bottle cultivation(BC). Total flavonoids content, total phenolics content, electron donating ability(EDA), nitrite-scavenging ability(NSA), SOD-like activity and inhibitory effect of Xanthine oxidase were examined. The highest value of total flavonoid content is $5.96{\pm}0.81mg/g$ in water extract from Grifola frondosa of log cultivation at $40^{\circ}C$ (LC-W40) but, one of total phenolics compound is $44.53{\pm}0.89mg/g$ in water extract from Grifola frondosa of bottle cultivation at $40^{\circ}{\cdots}$ (BC-W40). The EDA using DPPH of BC-W40 extract showed the highest value of $97.14{\pm}0.71%$. Nitrite-scavenging ability was $62.55^{\circ}{\ae}0.36%$ in extract from Grifola frondosa of BC-W40 at pH 1.2. The value was SOD-like activity showed the highest value of $18.95^{\circ}{\ae}1.39%$ in extract from LC-W40. Xanthine oxidase inhibitory activity was the highest value of $54.31{\pm}0.40%$ in extract from Grifola frondosa BC-W40, and dependent on concentrations. These results showed that a the antioxidant effects of Grifola frondosa is excellent. However, physiological activities of Grifola frondosa were not depend on caltivation method regulary, and were different according to kind of solvents, concentraitions and physialogical factors examined such as EDA, SOD-like activity and NSA.

Anti-inflammatory Activities of Methanolic Extracts from Different Rose Cultivars (품종별 장미꽃 메탄올 추출물의 항염증 효과)

  • The genus Rosa (Rosaceae) is an abundant source of phenolics and is traditionally used as a food supplement and as herbal medicine. Various plant phenolics are known to have anticancer, antioxidant, and anti-inflammatory properties. In this study, we investigated the anti-inflammatory effects of rose methanolic extracts (RMEs) from four different rose cultivars (Macarena, Onnuri, Oklahoma, and Colorado) in lipopolysaccharide (LPS)-activated RAW 264.7 cells. Our results demonstrated that pretreatment of REMs ($500{\mu}g/mL$) significantly reduced NO production by suppressing iNOS protein expression in LPS-stimulated cells. Anti-inflammatory effects by RMEs were observed in the following order: Oklahoma > Colorado > Onnuri > Macarena. Consistent with this finding, RMEs inhibited the translocation of $NF-{\kappa}B$ from the cytosol to the nucleus via the suppression of $I{\kappa}B{\alpha}$ phosphorylation and also inhibited LPS-stimulated $NF-{\kappa}B$ transcriptional activity. These findings suggest that RMEs exert anti-inflammatory actions and help to elucidate the mechanisms underlying the potential therapeutic values of RMEs. Therefore, RMEs could be regarded as a potential source of natural anti-inflammatory agents.

The Effects of Anti-Inflammatory Activities and Active Fractions Analysis of Ethanol Extract from Red Rose Petals (붉은 장미꽃잎 에탄올 추출물의 활성 분획물 분석 및 항염증 활성 효과)

  • Red rose petals are usually disposed but they are an abundant source of phenolics and traditionally used as food supplement and as herbal medicine. Of the Various phenolics, they are known to have anticancer, antioxidant, and anti-inflammatory properties. In this study, we investigated the anti-inflammatory effects of red rose ethanolic extracts(GRP) on lipopolysaccharide (LPS)-activated RAW 264.7 cells. The results demonstrated that pretreatment of GRP(500㎍/mL) significantly reduced NO production by suppressing iNOS protein expression in LPS-stimulated cells. Anti-inflammatory effects byred rose petal were observed in the following. Anti-melanogenesis in B16F0 Melanoma Cells by Extract of Fermented Cordyceps militaris Containing High Cordycepin (Cordycepin-고함유 동충하초(Cordyceps militaris) 발효 추출물의 미백효과)

  • To find a novel skin whitening agent, the effect of cordycepin-enriched Cordyceps militaris (CM${\alpha}$) extract fermented by fungi on anti-melanogenesis in B16F0 mouse melanoma cells was investigated. Fermented CM${\alpha}$ was prepared with fungi, including Monascus purpureus (Mp), Aspergillus oryzae (Ao), Aspergillus kawachii (Ak), and Rhizopus oryzae (Ro), respectively. When the content of the phenolics and the flavonoids and the activities of the antioxidant and the mushroom tyrosinase inhibition were measured in the CM fermented by Ak (AkF-CM), the highest content of the phenolics was 46 mg/g dry weight and the highest content of the flavonoids was 0.93 mg/g; the highest activity of the DPPH radical scavenging was 62.74% and the highest activity of the mushroom tyrosinase inhibition was 79.97% CM${\alpha}$CM${\alpha}$. From this result, AkF-CM${\alpha}$ exhibited the highest mushroom tyrosinase inhibitory activity and so it was used in subsequent anti-melanogenesis. B16F0 melanoma cells were treated with 1-10 mg/ml concentrations of AkF-CM${\alpha}$ and 200 ${\mu}M$ arbutin as the positive control. The melanin content and cell viability of the melanoma cells by arbutin treatment decreased to 43% and 92% of the control, respectively. AkF-CM${\alpha}$ treatment at 1, 3, and 5 mg/ml concentrations decreased the extracellular melanin release induced by IBMX treatment by 35%, 45%, and 53%, respectively. AkF-CM${\alpha}$ showed inhibitory activity against both intracellular tyrosinase in melanoma cells and mushroom tyrosinase. AkF-CM${\alpha}$ reduced the protein level of tyrosinase in the IBMX-stimulated cells. These results indicate that AkF-CM${\alpha}$ suppressed the activity and protein content of cellular tyrosinase and decreased the total melanin content in cultured B16F0 melanoma cells.

Studies on Quality Characteristics and Biological Activities of Macaroons supplemented with GABA(γ-Aminobutyric Acid) Rice Powder and Xylose (마카롱 제조 시 가바쌀 가루와 자일로스의 첨가에 따른 품질 특성과 생리 활성에 대한 연구)

  • The purpose of this study was to evaluate the biological activities and quality properties of macaroons made with GABA rice powder and xylose. GABA rice powder was added to macaroons at weight percentages of 0, 2.5, and 3.5%. Hunter's color values ($L^*$, $a^*$, $b^*$), total sugar contents, total phenolics, ABTS and DPPH radical scavenging activities, textures, and sensory characteristics supplemented with different xylose and GABA rice powder contents were measured. As the ratio of GABA rice powder in macaroons increased, total phenol contents and ABTS and DPPH radical scavenging activities increased. In the texture profile analysis, fracturability and brittleness were lower as GABA rice powder content increased. In the sensory evaluation, significant differences (p<0.05) were observed in color, sweetness, texture, and overall acceptability depending on the addition of GABA rice powder to macaroons. Overall qualities of macaroons were different from GABA rice powder and xylose added ones. Therefore, xylose and GABA rice powder may be used to make healthy macaroons.

Antioxidative Activities of Various Solvent Fractions from Fruit and Leaf of Pinkpop Borisu (핑크팝 보리수 열매와 잎 용매 분획물의 항산화 활성)

  • To get basic data for the utilization of Pinkpop Borisu fruit and leaf as a raw material in functional food, antioxidative activities were investigated. The radical scavenging activity of various solvent fraction from fruit and leaf against the DPPH and ABTS radicals increased with the increasing amount of various solvent fractions and butanol fraction of leaf showed the highest DPPH radical scavenging activities. Among the various solvent fractions from fruit and leaf of Pinkpop Borisu, butanol fraction of leaf showed the strongest scavenging effect on ABTS radicals. Reducing power and ferric reducing/antioxidant power (FRAP) of the various solvent fractions from fruit and leaf increased with the increasing concentration of fractions and butanol fraction of leaf showed the highest reducing power. The butanol fraction of fruit and leaf showed strong antioxidative activity on linoleic acid system. Total phenolics contents of butanol fraction from fruit and leaf were 106.34 and 252.46 mg/g, respectively. The antioxidative activities of the butanol fraction from fruit and leaf are attributed to phenolic compounds. The present results suggest that butanol fraction of Pinkpop Borisu leaf would have the protective potential from oxidative stress induced by free radicals.

Identification and Antioxidative Effects of Phenolic Compounds from Chinese Soybean (Heinong 48) (중국산 대두 (Heinong 48)의 페놀성 화합물 검출 및 항산화 효과)

  • This study was performed to investigate the phenolic compounds content and antioxidative activity of methanol extract from Chinese soybean. The content of total phenolics and total flavonoids of Chinese soybean Heinong 48) extract were $9.91{\pm}0.57GAE\;mg/g$ and $4.15{\pm}0.39QE\;mg/g$, respectively. The antioxidative activities of Chinese soybean extract were significantly increased in a dose dependent manner on 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical scavenging and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS) radical scavenging($IC_{50}$ values were $2.04{\pm}0.16$ and $3.31{\pm}0.31mg/mL$, respectively). These results suggest that Chinese soybean may have great potential as a natural antioxidant source with health benefits.

Phenolic Compounds and Antioxidant Activity in Cell Wall Materials from Deodeok (Codonopsis lanceolata) (더덕 부위별 세포벽 물질의 페놀성화합물과 항산화 활성)

  • This study measured the amounts of uronic acid, total sugars, non-cellulosic neutral sugars, phenolic compounds as well as antioxidants activity in cell wall materials (CWM) derived from different parts of deodeok (Codonopsis lanceolata). The values of the uronic acid (UA): neutral sugars (NS) ratio in polymers extracted from the CWM of the flesh and skin were 4 and 6, respectively. The total sugar contents of the flesh and skin were 788.6 and 824.9 ${mu}g/mg$ of CWM, respectively. Galactose and arabinose were the main noncellulosic neutral sugars. The chemical structure of five phenolic compounds from the CWM were analyzed and identified as vanillic acid, p-OH-benzaldehyde, vanillin, ferulic acid, and 8-O-4' diferulic acid by HPLC spectral data. Among them, p-OH-benzaldehyde, vanillin, and 8-O-4' diferulic acid were the first compounds identified from the deodeok. The content of 8-O-4' diferulic acid in the skin CWM was 56.1 ${mu}g/g$ AIR (alcohol insoluble residue). The ethanol-NaOH fractions from CWM had the highest oxygen radical absorbance capacity (ORAC) activities, followed by the AIR fractions and ethanol fractions.
