Effect of capsaicin on melanoma growth and metastasis

  • Capsaicin (8-methyl-N-vanillyl-6-nonenamide), a pungent ingredient of hot chili peppers, has been reported to possess substantial anticarcinogenic and antimutagenic activities. In our previous study, we found that capsaicin (100 ${\mu}$M) induced significant inhibition of matrix metalloproteinase-2 activity by gelatin zymography, and that capsaicin (i.p., 2.5mg/kg) inhibited development of lung colonization (58%) in experimental lung metastasis assay.(omitted)

Effect of Quercetin on Melanogenesis in Melan-a Melanocyte Cells (Quercetin이 Melan-a 멜라닌세포의 멜라닌합성에 미치는 영향)

  • Flavonoid seems to have various biological effects. Quercetin is a kind of natural plant flavonoids and has multiple biological effects such as antioxidant, antimutagenic and anticarcinogenec agent. Melanogenesis is a physiological process resulting in the synthesis of melanin pigments, which play a crucial protective role against skin photocarcinogenesis. This present study was designed to investigate effect of quercetin on proliferation and melanogenesis in Melan-a melanocyte cells. After 48h treatment of cells with quercetin, the cells exhibited a dose-dependent inhibition in their proliferation without apoptosis. Therefore, thε growth retardation by the extract may be due to the cell arrest or cell differentiation. We also investigated the effect of quercetin on melanogenesis of this cells. Melan-a melanocyte cells were grown for 48h in the presence of $0.01-60\;{\mu}g/ml$ quercetin and the total melanin content and activity of tyrosinase were measured. Quercetin stimulated melanization of the cells in low concentrations $(0.01-1.0\;{\mu}g/ml)$, whereas it inhibited melanization in high concentrations $(5.0-30\;{\mu}g/ml)$. It was observed that quercetin differently regulates melanogenesis of Melan-a melanocyte cells dependent on Its concentrations.

Effect of Antioxidant, Antimutagenicity and Anticancer of Root Extract from Ixeris dentata Nakai (씀바귀 뿌리 추출물의 항산화성, 항돌연변이원성 및 항암활성 효과)

  • Ixeris dentata root were extracted with methanol and then fractionated with n-hexane, EtOAc and BuOH to get active fractions. and their antioxidant and antimicrobial activities in each fraction were determined. Ethyl acetate fraction of Ixeris dentata root showed strong antioxidant activities, but aqueous fraction did not show any activities. But in the antimicrobial test, aqueous fraction showed strong antimicrobial activities except to Escherichia coli. especially, aqueous fraction showed the strongest activities against Hypocrea nigricans. and butanol fraction showed the strongest activities against Cladosporium herbarum. This study was performed to determine the antimutagenic and cytotoxic effect of Ixeris dentata root methanol extract on Salmonella typhimurium TA98, TA100 and cancer cell lines using ames test and cytotoxicity assay, respectively. Cancer cell lines include human lung carcinoma(A549), human breast adenocarcinoma(MCF-7) and human hepatocellular carcinoma (Hep 3B). Futher fractionations with hexane, ethyl acetate, butanol and water from methanol extract of Ixeris dentata root were performed to obtain effective fraction, methanol extracts showed 79.94% inhibitory effect on the mutagenesis induced by N' -methyl- N' -nitro-N-nitrosoguanidine(MNNG) against TA100, while 89.99% inhibition was observed on the mutagenesis induced by 4-nitroquinoline-1-oxide(4NQO) against TA98. In the meanwhile, butanol fraction showed 89.92% and 71.01% inhibitory effect on the mutagenesis induced by benzo(a)pyrene(B(a)P) against TA98 and TA100, respectively. Ethyl acetate fraction showed the strongest effect against A549, MCF-7 and Hep3B at the same concentration compared to those of other fration.

  • Antitumor activities of tannin extract and chloroform fraction extract from the persimmon leaves, and 2, 4-decadienal identified as an antimutagenic compound in persimmon leaves were examined in sarcoma 180 implanted tumor in mice by using both light and transmission electron microscopes. Among them, tannin extracted from the persimmon leaves delayed the progression of malignant tumor but the other two did not show any noticeable effect. The antitumorigenic activity of tannin extract might not come from the selective cytotoxicity against tumor cells, but might be, in an anaerobic environment, from the inhibitory action against oncogenic protein synthesis or from the proteolysis of the preformed oncogenic proteins by autophagocytic granules. therefore, the tannin from persimmon leaves might protect cells from fast progression of malignant tumorigensis.

Screening of the Mutagenicity and Antimutagenicity of the Hot Water Extracts from Medicinal Plants (한약재 열수 추출물의 변이원 및 항변이원 활성의 검색)

  • The mutagenicity, antimutagenicity, and cytotoxicity of the hot water extracts of 130 medicinal plants were examined. The results obtained using authentic alkaline phosphatase activity as the growth representative of the indicator cell, E. coli PQ37, demonstrated that the extracts of 14 medicinal plants including, Forsythia koreana Nakai, showed strong toxic effect on the cell growth, however, the extracts of 22 medicinal plants, including Phlomis umbrosa Turcz., were shown to have stimulatory effect on the cell growth. The mutagenicity and antimutagenicity of the medicinal plants were screened using SOS chromotest. The mutagenic activity was detected from 36 kinds of the samples, including Forsythia koreana Nakai and Coptis japonica Nakai. On the contrary, 12 medicinal plants, including Crataegus pinnatifida Bunge, were shown to have a significant antimutagenic activity.

  • The alginate extracted from Hizikia fusiform by the acid alkali soluble alginate (AASA) extraction method was reacted with Na$_2$SO$_4$. The amount of sulfate absorbed in the 3% alginate solution was determined about 18,435 ppm by inductively coupled plasma-atomic emission spectrometry. Both alginate and S-alginate stimulated the growth of Lactobacillus acidophilus in MRS broth and peptone water medium. The addition of S-alginate to the media showed the higher bacterial numbers than alginate supplement. There was no mutagenic activity of S-alginate in the Ames test using the Salmonella Typhimurium TA98 and TA100 strains. The S-alginate showed suppressive effect against 2-NF and MMS of mutagens.

Quality Characteristics of Steamed Bread Containing Pomegranate (Punica granatum L.) Peel Powder (석류 껍질 분말을 첨가한 찐빵의 품질특성)

  • Purpose: Pomegranate has been reported to exhibit medical properties such as antimicrobial, antimutagenic, anticancer, and anti-inflammatory effects. Especially the pomegranate peel as a by-product of the juice industry possesses more polyphenols with an antioxidant effect than the pomegranate flesh does. This study aimed to investigate the effect of the pomegranate peel on the quality characteristics of steamed bread. Methods: Steamed bread containing pomegranate peel was prepared with 0%, 1%, 3%, 5%, and 7% replacements of wheat flour with pomegranate peel powder, respectively. Results: The pH of dough significantly decreased from 5.66 to 4.42 with the amounts of pomegranate peel powder rising. After a 1st fermentation, the pH of all groups decreased. With an increase of the pomegranate peel powder level, the fermentation tension decreased from 42.83 mL to 29.03 mL. In addition, the volume, the specific volume, and moisture decreased as the pomegranate peel powder content level heightened. For color, steamed bread added with 7% pomegranate peel powder displayed low lightness but high redness and yellowness. In texture analysis, hardness and chewiness of the steamed bread rose as the pomegranate peel powder content increased, while the springiness and cohesiveness declined. Both total polyphenol and DPPH radical scavenging activity significantly increased with growing levels of pomegranate peel powder. In a sensory evaluation, the group with 1% pomegranate peel powder added showed the highest sensory preference scores for color, flavor, chewiness, taste, and overall acceptability. Conclusion: The results of this study suggest that steamed bread prepared with the addition of 1% pomegranate peel powder is the most optimal product for appropriate quality characteristics.

Antimutagenic and Cytotoxic Effects of Ethanol Extracts from Five Kinds of Seaweeds (다섯 가지 해조류 에탄올 추출물의 항돌연변이 활성 및 암세포 성장억제 효과)

  • The protective effects of ethanol extracts from 5 seaweeds on the mutagenic and cytotoxic damage were evaluated. They were separately extracted using ethanol from dried samples at room temperature, and freeze-dried. The inhibition effects on the mutagenicity in Salmonella assay by Ames test and cancer cell inhibitory effect in HeLa cell, MCF-7 cell and SNU -638 cell by MTT assay were assayed. Seaweed fusiforme, sea tangle and green laver showed strong inhibitory effect against 2-nitrofluorene, sodium azide- or 2-anthramine-induced mutagenicities in Salmonella Typhimurium TA 98 and TA 100 at the level of 2.5 mg ethanol extract per plate. Cancer cell inhibitory effect was shown with all of the seaweed extracts. Green laver, sea mustard, sea tangle and seaweed fusiforme showed strong cytotoxicity against HeLa and MCF-7 cells, with inhibiting by $92\~93\%$ and $89\~92\%$, respectively. These data show that 5 seaweeds tested in this study might be potent functional foods for cancer prevention, and consumption of these seaweeds in adequate amount is recommended.

  • Cirsium japonicum var. ussuriense were extracted with methanol and then fractionated with nhexane, EtOAc and BuOH to get active fractions. And their antioxidant and antimicrobial activities in each fraction were determined. Ethyl acetate and butanol fraction of Cirsium japonicum var. ussuriense showed strong antioxidant activities, but hexane fraction did not show any activities. But in the antimicrobial test, Ethyl acetate fraction showed strong antimicrobial activities except to Aspergillus awamori, Asperigillus niger. Especially, Ethyl acetate fraction showed the strongest activities against Bacillus subtilis. And aqueous fraction showed the strongest activities against Cladosporium herbarum, Hypocrea nigricans. This study was performed to determine the antimutagenic and cytotoxic effect of Cirsium japonicum var. ussuriense methanol extract on Salmonella typhimurium TA98, TA100 and cancer cell lines using Ames test and cytotoxicity assay, respectively. Cancer cell lines include human lung carcinoma(A549), human breast adenocarcinoma(MCF-7) and human hepatocellular carcinoma (Hep3B). Futher fractionations with hexane, ethyl acetate, butanol and water from methanol extract of Cirsium japonicum var. ussuriense were performed to obtain effective fraction, methanol extract showed 60.14% inhibition effect on the mutagenesis induced by MNNG against TA100, while 77% and 72.5% inhibition was observed on the mutagenesis induced by 4NQO against TA98 and TA100, respectively. and methanol extract showed 82.25% and 73.7% inhibitory effect on the mutagenesis induced by Trp-P-1 against TA98 and TA100, respectively. methanol extract showed the strongest effect against A549, MCF-7 and Hep3B at the same concentration compared to those of other fration.

Desmutagenic Effect of Water Extract from Artemisia capillaris THUNB on the Mutagenicity of Benzo[a]pyrene (Benzo[a]pyrene의 변이원성에대한 인진쑥 물 추출물의 항돌연변이 효과)

  • The antimutagenic activity of the extract of Artemisia capillaris THUNB on the mutagenicity induced by benzo(a)pyrene [B(a)P] in the presense of S9 mixture was studied using bacterial mutagenic assay system. Samples harvested in summer and autumn were extracted using ethanol and hot water. Among these extracts the water extract of summer sample had the strongest inhibitory effect against the mutagenenicity of B(a)P, The water extract of Artemisia capillaris THUNB was separated again into ethanol soluble and insoluble parts. The ethanol insoluble part(El) of water extract exhibited higher inhibition effects than the ethanol soluble part against the mutagenic activity of B(a)P. El showed dose-dependent activity on the mutagenicity of B(a)P in SOS Chromotest and Ames test. The 50% inbibition concentraction $(IC_{50}$ of El were $200{\mu}g/assay$ $600{\mu}g/plate$ and $800{\mu}b/plate$ in E. coil PQ37 S. typhimurium TA100 and TA98 respectively. El were showed desmutagenic effect but had no effect on the DNA repair system for B(a)P-induced mutagenesis. HPLC analysis showed that the formation of aflatoxin M1 by cytochrome P-450 1A1 known as playing an impotant role on B(a) P-induced mutagenicity was highly inhibited by El. Therefore we encluded that B(a)P-induced mutagencity can be reduced possible due to the interference of el with cytochrome P-450 1A1-dependent bioactivation.
