Synthesis and Antimicrobial Activity of Novel Tetrahydrobenzothienopyrimidines

  • Due to the rapidly growing number of resistant strains of bacteria, the search for antibacterial agents with new modes of action will always remain an important and challenging task. Thus, the reaction of 2-substituted or. unsubstituted-4-(4-acetylanilino)-5,6,7,8-terahydrobenzo[b] thieno[2,3-d]pyrimidine derivatives 1-3 with the hydrazine derivatives, semi and / or thiosemi-carbazides, provided the corresponding hydrazones 4-6 and semi and/or thiosemicarbazones 7-9. Claisen-Schmidt condensation of compounds 1 or 2 with the appropriate aldehyde yielded the chalcones 10, 11 which, when treated with hydroxylamine hydrochloride gave rise to the isoxazoline-containing compounds 12, 13. Furthermore, reacting the respective chalcones 10, 11 with different hydrazines, urea and/or thiourea, furnished compounds 14, 15, 16, and 17 respectively. Representative compounds were tested for their antimicrobial activity against Candida Albicans and certain gram-positive and gram-negative bacteria. Their MICs were then determined. Compound 15e, showed a broad spectrum of activity while most of the other com-pounds showed varying antimicrobial activity.

  • In the present study, various extracts of Clerodendrum infortunatum leaves have been studied for its anthelmintic, antimicrobial and antipyretic activities. In the anthelmintic and antipyretic study it was observed that the benzene extract was more potent than the other two extracts (ethanol and aqueous) even though all the three extracts were endowed with both the properties. The study reveals antimicrobial activity of the extracts against the tested strains of microorganisms between concentration ranges of $75\;{\mu}g/ml$ and $350\;{\mu}g/ml$ and shows effectiveness more against gram-positive bacteria than the gram-negative bacteria.

해조류 추출색소의 기능성을 활용한 립밤 제조 (Preparation of Lip Balm Utilizing Functionalities of Colorants Extracted from Marine Algae)

  • The aim of this study is to utilize the algae extracts with functionalities such as antioxidant and antimicrobial properties for preparing lip balm. Functional properties of extracts from Dictyota coriacea and Ulva pertusa were evaluated in terms of DPPH radical scavenging activity and antimicrobial activity. Lip balms contained the algae extracts were tested its efficacy by measuring the moisture retention of lip skin after applying human subjects. According to the results of skin moisture retention measurement, lip balm was effective to increase the moisture retention of lip skin. It is considered that the utilization of colorants extracted from algae which are verified of antioxidant effect and antimicrobial activity is valuable and useful in developing the cosmetic products.

글루코사민-글루타르알데히드 가교결합체의 항균 효과 (Effect of Antimicrobial Activity of the Glutaraldehyde Cross-linked Glucosamine)

  • This study was investigated the antimicrobial activity of glutaraldehyde cross-linked glucosamine. Glutaraldehyde was used as a cross-linker which specifically combines an amine-group of molecules. To optimize the mixing ratio of glutaraldehyde and glucosamine, mixing ratio was set up 1:1, 2:1, 3:1 and 0.5:1 in molarity, respectively. The optimum mixing ratio of glucosamine and glutaraldehyde was found to be 3:1 using thin layer chromatography based on the production of complex. Glucosamine-glutaraldehyde cross-linked complex (Ggcc) revealed significant antimicrobial activity toward PWG than F1, both microbial strains were isolated from porcine semen as antibiotics resistance bacteria (ARB). These results clearly demonstrate that Ggcc has potential bactericidal activity toward ARB in porcine semen.

Synergistic Antimicrobial Effect of Achyranthes japonica Nakai Extracts and Bifidobacterium Supernatants Against Clostridium difficile

  • The synergistic antimicrobial effect of Achyranthes japonica Nakai (AJN) and Bifidobacterium extracellular factors against Clostridium difficile were measured using a turbidity method. Each broth supernatant of Bifidobacterium infantis ($68.8{\pm}0.02%$) and Bifidobacterium adolescentis ($33.2{\pm}0.2%$) obtained by adding ethyl acetate soluble fractionate from A. japonica Nakai ethanolic extracts (AJNEA, 100 ppm, no inhibition) showed high synergistic antimicrobial activity against C. difficile. In addition, the antimicrobial activity in a laboratory medium and yogurt products against C. difficile were evaluated. In yogurt prepared with a starter 5 (Lactobacillus acidophilus: Streptococcus thermophilus: B. adolescentis =1 : 1 : 1) and a starter 4 (L. acidophilus: S. thermophilus: B. infantis=1 : 1 : 1) and 0.5% AJNEA powder, high antimicrobial effects were recorded that measured 79.0 and 65.2%, respectively. The results indicated the potential of AJN extract for use as an antimicrobial agent. In addition, the efficiency of the antimicrobial activity of the extracts was further improved in combination with lactic acid bacteria, which suggests that they have the potential to be used as a highly effective antibiotic-tolerant microorganism prevention system. Such a strategy can be used for alternative drugs or functional food additives for treatment of antibiotic-associated diarrhea.

무좀균과 비듬균에 대한 대나무 기름의 항균효과 (Antimicrobial Effect of Bamboo (Phyllosrachys Bambusoides) Essential Oil on Trichophyton and Pityrosporum)

  • 1. 무좀균에 대한 각 시료들이 항균력으로 terbinafine > 100%-bamboo oil> 50%-bamboo oil순으로 나타내었다. 무좀균에 대한 Bamboo oil에 대한 영향으로는 T.M > E.F > T.R의 순서대로 항균력을 나타내었으며, 특히 T.M에 대하여 가장 우수하게 나타내었다. 2. 비듬균에 대한 각 시료들이 항균력으로는 terbinafine > 100%-bamboo oil > 50%-bamboo oil순으로 나타내었다. 비듬균에 대한 Bamboo oil에 대한 영향으로는 P.O-75 > P.O-77의 순서대로 항균력을 나타내었으며, 특히 P.O-75에 대하여 가장 우수 하게 나타내었다. 3. 50%-bamboo oil은 무좀균과 비듬균에 대하여 항균력이 있는 겋으로 나타내었으au, 100%-Bmboo oilㅇ의 항균력과 비슷하게 나타내었으며, 경비 절감 측면에서 기대를 걸 수 있을 것으로 사료된다.

구강미생물에 대한 고삼의 항균효과 (Antimicrobial Effects of Sophorae Radix Extracts against Oral Microorganisms)

  • Objectives : Sophorae Radix (SR), the dried root of Sophorae Flavescens Aiton, has been used to treat atherosclerosis, arrhythma and skin diseases including scabies and eczema. The present study was examined to evaluate antimicrobial activities of SR extracts against oral microorganism. Methods : Antimicrobial properties of SR extracts were determined by agar diffusion method and minimum inhibitory concentration (MIC) against Streptococcus mutans, Streptococcus sobrinus and Actinomyces viscosus. Analysis of kurarinone from SR extracts was conducted using UPLC (Ultra Performance Liquid Chromatography). Results : The ethanolic extracts of SR showed stronger antimicrobial effect than methanolic extracts, while the aqueous extracts of SR had no activity. In addition, the higher content of kurarinone was found in ethanolic extracts than methanolic extracts. The purified kurarinone from ethanolic extracts showed potent antimicrobial activity with the MIC value of $3.9{\sim}7.8{\mu}g/m{\ell}$. Conclusion : An ethanolic extract of SR showed antimicrobial properties against several oral microorganisms, and kuranrinone contributed to antimicrobial action of SR. Thus, ethanolic extracts of SR or purified kurarinone should be beneficial for the preparation of the useful agent for treating oral disease including anticaries.

Burkholderia sp. OS17의 항균활성 증진을 위한 배양최적화 (Antimicrobial activities of Burkholderia sp. strains and optimization of culture conditions)

  • 본 연구는 담수환경에서 항균활성을 보유한 미생물을 발굴하고, 활성 증진을 위해 배양조건을 최적화하는 것이다. 상주시 중동면 오상저수지에서 시료를 채취하여 38종의 미생물을 순수분리하였다. 16S rRNA 염기서열 분석에 근거하여 Proteobacteria강(22종), Actinobacteria강(7종), Bacteroidetes강(6종), Firmicutes강(3종)으로 구성되어있는 것을 확인하였다. 메티실린내성 황색포도상구군 등 10종의 유해미생물에 대한 항균활성을 보유한 Burkholderia sp. OS17 균주를 선발하였다. 항균활성 증진을 위한 상용배지, 온도, 초기 pH별 생육 및 항균활성 비교실험을 수행하였다. OS17 균주는 YPD 배지, $35^{\circ}C$, pH 6.5로 배양했을 때 가장 활성이 높았다. LB, NB, TSB, R2A 배지와 $20^{\circ}C$, $25^{\circ}C$ 배양했을 때는 생장은 가능하나 항균활성이 전혀 없었다. 이전결과를 바탕으로 YPD 배지, $35^{\circ}C$에서 배양하면서 5 L fermenter를 이용하여 생육, 항균활성, pH 확인을 통해 배양 48시간을 최적 배양시간으로 선정하였다. 항균활성을 보유한 미생물의 배양 최적화는 항균물질 생산에 영향을 미치고, 이는 상업적 응용에 이점으로 작용할 수 있다.

Probiotic Properties of Lactobacillus brevis KU200019 and Synergistic Activity with Fructooligosaccharides in Antagonistic Activity against Foodborne Pathogens

  • This study aims to evaluate the probiotic properties of Lactobacillus brevis (L. brevis) KU200019 and the synergistic activity with prebiotics on antimicrobial activity, and the potential application as an adjunct culture in fermented dairy products. The commercial strain, L. brevis ATCC 14869 was used as reference strain. L. brevis KU200019 was showed higher viability in simulated gastric (99.38±0.21%) and bile (115.10±0.13%) conditions compared to reference strain. L. brevis KU200019 exhibited antimicrobial activity against various foodborne pathogens. The supplementation of fructooligosaccharides (FOS) enhanced viability of lactic acid bacteria (>8 Log CFU/mL) and antioxidant activity [2,2-diphenyl-2-picrylhydrazyl radical assay (DPPH) assay, 31.23±1.14%; 2,2-azinobis (3-ethylbenzothiazoline-6-sulfonic acid (ABTS) assay, 38.82±1.46%] in fermented skim milk during refrigerated storage. L. brevis KU200019 was distinguished from the reference strain by its higher probiotic potential, antimicrobial activity, and higher antioxidant activity in fermented milk. Therefore, L. brevis KU200019 with FOS was demonstrated promising properties for further application in fermented dairy products with enhanced safety and quality.

Antimicrobial Effect of Acanthopanax sessiliflorum Fruit Extracts against Selected Oral Bacteria

  • This study aimed to evaluate the antimicrobial effects of Acanthopanax sessiliflorum fruit (ASF; Ogaza) extracts on Streptococcus mutans and Streptococcus sobrinus, which are agents that cause dental caries, and on Streptococcus mitis and Streptococcus salivarius, the microbial flora of the oral cavity. The ASF extracts obtained using 70% ethanol were fractionated in the order of ethyl acetate and n-Butanol, concentrated under reduced pressure, and lyophilized to give powdery solvent extracts. The antimicrobial activity of ASF extracts from each solvent was examined using the disk diffusion method. As a result, only those extracts obtained using an ethyl acetate solvent showed antimicrobial activity. These extracts were selected, and the minimum inhibitory concentration was measured by disk diffusion method at various extract concentrations. Results showed a minimum inhibitory concentration of 32 mg/ml. The viable cell count was measured to confirm the minimum bactericidal concentration. Results showed a minimum bactericidal concentration of 64 mg/ml. In the cytotoxicity test using normal human dermal fibroblast cells, the absorbance value of the test group was similar to that of the control group at 0.64, 1.28, and 6.4 mg/ml. The bacteria and their colonies were examined using a scanning electron microscope. Boundaries between the antimicrobial activity region and non-antimicrobial activity region were observed around the paper disk, which was immersed in the extract with 32 mg/ml concentration. Bacterial colonization was not observed in the area with antimicrobial activity. This finding suggests that ASF extracts can inhibit the growth of some microorganisms in the oral cavity, in addition to the effects of these extracts known to date. In particular, ASF extracts may be used as a preparation for preventing dental caries by adding the extract to the toothpaste or oral mouthwash.
