Ginseng saponins and the treatment of osteoporosis: mini literature review

  • The ginseng plant (Panax ginseng Meyer) has a large number of active ingredients including steroidal saponins with a dammarane skeleton as well as protopanaxadiol and protopanaxatriol, commonly known as ginsenosides, which have antioxidant, anticancer, antidiabetic, anti-adipocyte, and sexual enhancing effects. Though several discoveries have demonstrated that ginseng saponins (ginsenosides) as the most important therapeutic agent for the treatment of osteoporosis, yet the molecular mechanism of its active metabolites is unknown. In this review, we summarize the evidence supporting the therapeutic properties of ginsenosides both in vivo and in vitro, with an emphasis on the different molecular agents comprising receptor activator of nuclear factor kappa-B ligand, receptor activator of nuclear factor kappa-B, and matrix metallopeptidase-9, as well as the bone morphogenetic protein-2 and Smad signaling pathways.

한국, 중국, 일본에서 자생하는 '마' 속 식물의 화학 성분과 활성 (Phytochemical and pharmacological profiles of Dioscorea species in Korea, China and Japan)

  • Plants of genus Dioscorea have long been served as important carbohydrate-stuffed foods in the tropical and subtropical regions, and utilized as traditional herb medicines to enhance digestive function, improve anorexia, and treat diarrhea in oriental countries. It is known that around 600 species of Dioscorea are distributed in the world including 107 species in Asia, but actually utilized Dioscorea species are restricted to small numbers. Phytochemical investigations for Dioscorea species have revealed a number of chemical components such as sapogenins, saponins, phenanthrenes, stilbenes, diterpenes and purine derivatives. According to recent pharmacological studies, Dioscorea species possess significant antioxidant, antibacterial and anti-inflammatory activities as well as anticancer, antidiabetic, cholesterol-lowering and hypolipidemic effects. Here, seven Dioscorea species (D. batatas, D. japonica, D. bulbifera, D. opposita, D. tokoro, D. nipponica and D. alata), mainly distributed and used in Korea, China, and Japan, are reviewed to provide their botanical, phytochemical and pharmacological properties were described.

장뇌삼 에탄올 엑스의 대사성증후군 개선 활성 (Wild Ginseng Improves the High-Fat Diet Induced Metabolic Syndrome In ICR Mice)

  • The ginseng root has been used as a tonic remedy, and its antidiabetic activity has been demonstrated as early as 1920s. Although wild ginseng was anecdotally thought to be superior to cultivated ginseng in terms of pharmacological properties, there have been no prior reports on its improvement of metabolic syndrome. In this study, we figured out whether wild ginseng ethanol extract (WGEE) exerted the preventive effects on high fat diet-induced metabolic syndrome as well as treatment effect in ICR mice. In the preventive mode experiment, WGEE at 500 mg/kg significantly inhibited body weight gain $(16\%)$, fasting blood glucose $(37\%)$ and insulin $(37\%)$, triglyceride $(15\%)$, and free fatty acid levels $(32\%)$ when compared to those in high fat diet (HFD) fed control group. WGEE-treated mice at doses of 250 and 500mg/kg improved the insulin resistance index by $55\%\;and\;61\%$ compared to the HFD control group, respectively. In the treatment mode experiment, WGEE also markedly reduced the blood glucose levels (210 mg/dl in control group was lowered to 167 mg/dl).Taken together, WGEE has potential as a preventive and treatment agent for metabolic syndrome and deserves clinical trial in the near future.

  • Impaired insulin action in Type 2 diabetes is thought to lead to hyperglycemia, with both environmental and complex genetic factors playing key roles. Although the primary lesion in Type 2 diabetes is unknown, a number of studies suggest that metabolic defects in the liver, skeletal muscle and fat, and pancreatic ${\beta}$-cells contribute to the disease. These metabolic abnormalities are characterized by the overproduction of hepatic glucose, impaired insulin secretion, and peripheral insulin resistance. In current pharmacological treatment of Type 2 diabetes, sulfonylurea (SU) drugs have mainly been used as oral hypoglycemic drugs to stimulate endogenous insulin secretion from ${\beta}$ cells. SU drugs, however, sometimes aggravate the disease by causing fatigue of the pancreatic ${\beta}$ cells, which leads to reduced drug efficacy after long-term treatment. This class of drugs also leads to enhanced obesity arising from the stimulation of endogenous insulin secretion in obese Type 2 diabetic patients, plus an increased incidence of SU-induced hypoglycemia. Since 1980, a major challenge has been made by us to develop a potential pharmacological therapy for the treatment of insulin resistance in peripheral tissues and/or suppression of abnormal hepatic glucose production in Type 2 diabetic patients. Such a drug would be expected to have fewer side effects and retain long-term efficacy.

Role of Curcuma longa, a traditional ayurvedic medicinal plant, in diabetes

  • Curcuma longa belongs to the family Zingiberaceae and can be found in the tropical and subtropical regions of the world. It is widely used in Asiatic countries, especially India and South East Asia where it is cultivated commercially as a condiment. Its rhizomes exhibit anti-inflammatory, anti-human immunodeficiency virus, anti-bacterial, antioxidant effects, nematocidal activities, antiproliferative and antiangiogenic activities and are of pharmaceutical importance. Another relevant medicinal property exhibited by it is antidiabetic property which is reviewed here. Studies on the efficacy of crude C.longa extracts against type 2 diabetes in murine models reveal that it demonstrates a hypoglycemic effect by lowering the blood glucose levels under in vivo conditions. Clinical studies have revealed the safety of curucmin (major principle component exhibiting pharmaceutical properties from C.longa) on humans but with very low bioavailability. In view of its effective hypoglycemic effect and its low bioavailability, further studies are needed for the characterization of the bioactive principles and formulating the development of C.longa extracts as a novel anti-diabetic therapeutic agent.

Effect of gemigliptin on cardiac ischemia/reperfusion and spontaneous hypertensive rat models

  • Diabetes is associated with an increased risk of cardiovascular complications. Dipeptidyl peptidase-4 (DPP-IV) inhibitors are used clinically to reduce high blood glucose levels as an antidiabetic agent. However, the effect of the DPP-IV inhibitor gemigliptin on ischemia/reperfusion (I/R)-induced myocardial injury and hypertension is unknown. In this study, we assessed the effects and mechanisms of gemigliptin in rat models of myocardial I/R injury and spontaneous hypertension. Gemigliptin (20 and 100 mg/kg/d) or vehicle was administered intragastrically to Sprague-Dawley rats for 4 weeks before induction of I/R injury. Gemigliptin exerted a preventive effect on I/R injury by improving hemodynamic function and reducing infarct size compared to the vehicle control group. Moreover, administration of gemigliptin (0.03% and 0.15%) powder in food for 4 weeks reversed hypertrophy and improved diastolic function in spontaneously hypertensive rats. We report here a novel effect of the gemigliptin on I/R injury and hypertension.

UPLC를 이용한 지역별 헛개나무(Hovenia dulcis) 플라보노이드의 정량분석 (Quantitative Analys is of Flavonoids in Hovenia dulcis by Region Using UPLC)

  • Hovenia dulcis is a herbal plant, which belongs to the Rhamnaceae family and is a native of Japan, China and Korea. Its fruit stalk is called 'Jiguja' in Korea. It has been traditionally used as a medicinal plant in East Asia. It was reported to have detoxification effects on alcohol poisoning, and antioxidant, antidiabetic etc. Sample of 5 g was extracted with 50 mL of 70% EtOH. The supernatant was filtered by 0.45 ㎛ membrane filter before analysis. The UPLC system was performed on Waters alliance UPLC HSS T3 column (2.1 × 100 mm, 1.7 ㎛) with a UV detector. The gradient system was a binary eluent of 0.1% formic acid in water(A) and 0.1% formic acid in acetonitrile(B) with gradient conditions as follows: Initial, 10% B; 1 min, 10% B; 4 min, 20% B; 10 min, 25% B; 12 min, 30% B; 14 min, 90% B; 17 min, 90% B; flow rate of 0.2 mL/min. The samples were injected by 2 µL and were detected at UV 355 nm. As a result of analysis, chromatographic patterns appeared in two cases: samples analyzed for ampelopsin and myricetin, and samples analyzed for taxifolin and quercetin. Among the four compounds, the largest regional difference was found to be taxifolin.

커피 볶음 정도에 따른 생화학적 성분 및 생리활성의 변화 (Changes of biochemical components and physiological activities of coffee beans according to different roasting conditions)

  • 4종류의 커피 원두(CS, Colombia superimo; EY, Ethiopia yirgacheffee; IM, Indonesia mandheling; IMM, India monsooned malabar AA)를 생두(raw coffee bean)와 각각 다른 온도(200, $250^{\circ}C$)와 시간(10, 15, 20 min)으로 로스팅하였으며, 최적의 로스팅 조건을 찾기 위하여 로스팅 중에 변화되는 pyrazine을 비롯한 몇 가지 성분과 항산화, 항당뇨 효과를 조사하였다. Caffeine 함량은 로스팅 정도에 따라 크게 차이를 보이지 않았으나, 5-caffeoylquinic acid와 total phenolic compounds는 대체로 로스팅이 강해질수록 크게 감소하였다. Pyrazine 화합물은 IMM에서 가장 많이 생성되었으며, $250^{\circ}C$에서는 시간이 경과할수록 급격히 줄어들었다. DPPH 라디칼 소거활성은 같은 처리농도에서의 BHT와 ${\alpha}$-tocopherol의 활성에 비하여 대부분 80% 이상의 효과를 나타내었다. 한편 FRAP assay에서는 로스팅 정도가 강해질수록 활성이 약간 감소하였다. ${\alpha}$-Glucosidase 저해활성은 유의할 만한 수준이었으나, $250^{\circ}C$에서 20분 roasting할 경우에는 활성이 80% 이상 줄어들었으며, ${\alpha}$-amylase 저해활성도 유사한 경향이었다. 이러한 결과로 로스팅 정도에 따른 생리활성과 커피의 고소한 향과 초콜릿 향을 내는 pyrazine 화합물의 생성을 종합적으로 보았을 때, $200^{\circ}C$에서 15분정도 로스팅하는 것이 가장 최적의 조건인 것으로 판단하였다. 따라서 이러한 결과는 커피산업에서 로스팅 조건을 달리한 원두커피의 향과 생리활성이 높은 커피를 생산하는데 활용될 수 있을 것이라 기대된다.

Alloxan에 의한 HIT-T15 세포 손상에 대한 쑥갓주정추출물의 세포보호효과 (The Protective Effects of Chrysanthemum cornarium L. var. spatiosum Extract on HIT-T15 Pancreatic β-Cells against Alloxan-induced Oxidative Stress)

  • The objective of the present study was to evaluate the potential antidiabetic and antioxidant effect of the ethanol extract from Chrysanthemum cornarium L. var. spatiosum(CSE) against alloxan-induced oxidative stress in pancreatic ${\beta}$-cells, HIT-T15. In this study, the antidiabetic effect of CSE was examined using the 3-(4,5-dimethylthiazolyl-2)-2,5-diphenyltetrazoliu bromide(MTT) cell proliferation assay, lactate dehydrogenase(LDH) release assay, $NAD^+$/NADH ratio and insulin secretion. To further investigate whether CSE is involved in the antioxidant activity of alloxan-damaged HIT-T15 cells, its antioxidant effect against alloxan-induced oxidative stress was measured in HIT-T15 cells by determining the levels of antioxidant enzymes including superoxide dismutase(SOD), glutathione S-transferase(GST), glutathione reductase(GR) and glutathione peroxidase(GPx). The results of this analysis showed that alloxan significantly decreased cell viability, increased LDH leakage, and lowered $NAD^+$/NADH ratio and insulin secretion in HIT-T15 cells. However, CSE significantly increased the viability of alloxan-treated cells and lowered LDH leakage. The intracellular NAD+/NADH ratio and insulin secretion were also significantly increased by 1.7-fold and 1.3-fold, respectively, after treatment with 100 ${\mu}g/m{\ell}$ CSE. The HIT-T15 cells treated with alloxan showed significant decreases in the activities of antioxidant enzymes, while CSE significantly elevated the levels of antioxidant enzymes. These findings suggest that CSE could have a protective effect against cytotoxicity and dysfunction of pancreatic cells in the presence of alloxan-induced oxidative stress.

천연 암반 심해수의 항산화 활성 및 면역반응에 대한 연구 (Study of Deep Ground Sea-Like Water on Antioxidant Activity and the Immune Response in RAW264.7 Macrophages.)

  • 해양성 심층수(KDSW)는 해양심층수(DSW)와 유사한 mineral 조성을 가지고 있어 식품 및 의약분야에서 그 유용성에 대한 관심이 증대되고 있다. 본 연구에서는 KDSW 및 탈염한 Danasoo의 항산화력, 면역활성, 함암 및 당뇨에 대한 효과를 연구하였다. KDSW와 Danasoo의 항산화 활성은 DPPH radical scavenging 활성, SOD-like 활성 및 PCL 법에 의하여 항산화 활성을 조사하였다. KDSW와 Danasoo의 항산화 활성은 첨가된 농도에 의존적으로 증가하였으며, 특히 PCL법에 의한 KDSW와 Danasoo의 항산화능은 85.32과 14.02(nmol of ascorbic acid equivalent/ml)로 높은 항산화 활성을 나타내었다. Macrophage RAW 264.7 cell에서 LPS에 의하여 유도된 NO 활성은 Danasoo (20%)를 첨가에 의하여 약 30%정도의 NO 합성이 저해되었으며, 이 농도에서 세포 독성이 없는 것으로 MTT assay에 의하여 확인하였다. NO는 nitric oxide synthase에 의하여 합성되며, tumor성장 및 angiogenesis에서 중요한 역할을 하는 것으로 밝혀져 있으며, Danasoon (20%)는 위암세포와 폐암세포의 iNOS 발현을 현저히 감소시켰다. STZ에 의하여 유도된 당뇨 쥐의 혈당량은 Danasoo (20%) 식이에 의하여 대조구와 유사한 당뇨효과를 나타내었다. 이러한 결과들은 KDSW가 우수한 biological 활성을 가지고 있으며, 또한 천연 기능성 제품생산을 위한 소재로 사용될 수 있는 높은 가능성올 시사하고 있다.
