육계.부자의 약침과 경구투여가 2형 당뇨 모델 생쥐의 혈당강하에 미치는 영향 (Effects of Cinnamomum Cassia and Aconitum Carmichaeli's Pharmacopuncture and Oral Administration on Blood Sugar in Type II Diabetic Mice)

  • Objectives : This study was designed to evaluate the antidiabetes effects of Cinnamomum cassia and Aconitum carmichaeli in type II diabetes mellitus model mice. Methods : The experimental animals were divided into 3 groups and treated for 50 days as follows:control group, pharmacopuncture group and extract administration group. The effects of Cinnamomum cassia and Aconitum carmichaeli were observed by measuring body weight, blood glucose, insulin, insulin immunoreactivity of the pancreas and subpopulation of splenocytes. Results : Cinnamomum cassia sample groups were reduced a little body weight than control group. Glucose level of extract administration group lowered than control and pharmacopuncture group. Insulin immunoreactivity of Aconitum carmichaeli extract group appeared more effective than control and pharmacopuncture group. Conclusions : This study shows that Cinnamomum cassia and Aconitum carmichaeli have a distinct antidiabetes effect in type II diabetes mellitus model.

곶감, 생감 및 감잎 추출물의 생리활성 효과 (Physiological Activities of Dried Persimmon, Fresh Persimmon and Persimmon Leaves)

  • 본 연구에서는 곶감, 생감 및 감잎의 메탄올 추출물을 제조한 후 이들의 생리활성을 검색하였다. 총 폴리페놀 함량은 곶감, 생감 및 감잎 각각 147.79, 301.45 및 $315.90\;{\mu}g/mg$으로 생감 및 감잎에 폴리페놀 함량이 곶감에 비해 유의적으로 많았다. 감잎의 경우 폴리페놀 함량에 비례하여 DPPH radical 소거활성, 지질과산화 억제효과 및 salivary $\alpha$-amylase 저해활성이 증가되었다. 항암활성도 폴리페놀 함량이 높은 생감 및 감잎추출물이 위암 세포인 AGS에 대해 $65{\sim}70%$의 높은 저해율을 나타내었다. 그러나 폴리페놀 함량에 차이가 있는 곶감과 생감의 경우 free radical 소거능 및 salivary $\alpha}$-amylase 저해활성은 유의적인 차이가 없었다. 또한, 항고 혈압 활성은 곶감, 생감 및 감잎 추출물 모두 80% 이상의 높은 저해활성을 나타내었다. 항균활성은 감잎 추출물의 항균력은 나타나지 않았으나, 곶감 및 생감의 경우 E. coli O157:H7에 대해서만 약한 항균력을 볼 수 있었다. 이와 같이 같은 감류임에도 건조 상태 및 부위에 따라 다양한 차이를 보이는 것으로 나타났다. 이상의 결과를 종합하면 감잎의 항산화, 항당뇨 및 항암 활성은 폴리페놀 함량에 기인하나, 곶감은 생감 및 감잎에 비하여 폴리페놀 함량은 상대적으로 적으나 항산화, 항당뇨, 항고혈압 및 항균효과가 있는 물질을 함유한 것으로 판단되며, 이들 효과를 나타내는 원인물질에 대한 연구가 진행되어야 할 것이다.

어성초의 항혈전, 항당뇨 및 항산화 활성 (Antithrombosis, Antidiabetes, and Antioxidant Activities of Houttuynia cordata)

  • 어성초(Houttuynia cordata)는 삼백초과(Saururacease)에 속하는 다년생 초본으로 잎, 줄기 및 뿌리의 전초를 약제로 사용하고 있으며, 민간에서는 혈행개선, 폐렴, 급만성기관지염, 장염, 요로감염증, 종기 치료에 주로 이용되고 있다. 최근에는 항산화, 항염증, 항균 및 암세포 성장억제, 간손상 및 신경세포 사멸 억제, 카드뮴 독성 감소효과 등이 보고되고 있다. 본 연구에서는 어성초 지상부의 에탄올 추출물 및 이의 헥센 분획물, 에틸아세테이트 분획물, 부탄올 분획물 및 물 잔류물을 조제하여 항혈전, 항당뇨, 항산화 및 적혈구용혈활성을 평가하였다. 어성초의 총 폴리페놀 및 총 플라보노이드 함량은 에틸아세테이트 분획물에서 가장 높았으며, 상기 분획물은 5 mg/ml 농도에서 용매 대조구보다 2.09배, 2.19배 및 1.69배 연장된 TT, PT, aPTT를 보여 강력한 항혈전 활성을 나타내었으며, DPPH, ABTS 및 nitrite radical에 대해 각각 30.9, 19.9 및 49.6 ㎍/ml의 RC50을 보여, 9.8, 5.2 및 11,8 ㎍/ml의 RC50를 나타낸 vitamin C의 항산화력에 비교되었다. 트롬빈 저해활성이 강력한 헥센 분획물이 강력한 적혈구 용혈활성을 나타내는 바와 달리, 상기 에틸아세테이트 분획물은 1 mg/ml 농도까지 전혀 용혈활성이 나타나지 않았다. 본 연구결과는 어성초의 항혈전 활성에 대한 최초의 보고로, 어성초의 quercitirin, hyperoside, orientin 및 isoquercitrin이 항혈전 및 항산화 활성에 관련되어 제시하고 있으며, 향후 어성초를 이용한 기능성 항혈전 식품소재 개발 가능함을 제시하고 있다.

인동초(Lonicerae Japonica Thunb) 추출물의 혈당강하 효과 (Antidiabetic Effect of Lonicerae Japonica Thunb Extract)

  • 본 실험은 문헌상으로 혈당강하효과가 알려져 있는 인동초를 에탄올로 추출하여 그 추출물을 streptozotocin(STZ)으로 당뇨를 유발시킨 흰쥐에게 투여 후 혈당강하작용을 확인하였다. 또한, 그 유효 성분의 추적을 위하여 여러 용매(hexane, chloroform, ethylacetate, butanol, water)로 계통분획하여 그 추출물의 항 당뇨효능을 분석한 결과 hexane, water층의 분획물이 당뇨유발 흰쥐에서 glycogen 함량증가와 glucose-6-phosphatase(G-6-Pase), glucose-6-phosphate dedehydrogenase (G-6-PDH)등의 당대사효소 활성에 효과가 있었다.

큰방가지똥 추출물의 항당뇨 및 항고혈압효과 (Antidiabetes and Angiotensin Converting Enzyme Inhibitory Activity of Sonchus asper (L) Hill Extract)

  • In this study, we evaluated the bioactivities of methanol extract and its solvent fractions of Sonchus asper (L.) Hill. The EtOAc fraction of S. asper exhibited more strong antioxidant activity than other extracts as evidenced by the strongest 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity with a $EC_{50}$ value at $33.55\;{\mu}g/mL$ and reducing power, the total polyphenol (180.71 mg GAE/g) and flavonoid contents (145.86 mg QE/g) of S. asper extract were higher than other extracts. The EtOAc fraction of the S. asper also showed 47.38% mushroom tyrosinase inhibition activity, 56.22% ${\alpha}$-glucosidase inhibition and 46.58% ${\alpha}$-amylase inhibition ratio at 1 mg/mL. Both methylene chloride and EtOAc fractions of methanol extract of S. asper effectively reduced of the 86.34% and 62.03% angiotensin I converting enzyme (ACE) activity at 2 mg/mL, respectively. These findings suggest that the EtOAc fraction of the S. asper could be a potential antioxidant in food additive, medicinal, and industry product.

  • This study examined the antidiabetic and antihyperlipidemic activities of an ascidian (Halocynthia roretzi) in rats in vivo. Rats were fed on experimental diet including dried ascidian powder (200 mg/kg body weight) for 4 weeks, and then the triglyceride and total cholesterol levels in blood were analyzed. On the ascidian tunic powder diet, the triglyceride level decreased by more than 20.9% and the total cholesterol content decreased by more than 24.4%. In comparison, the triglyceride and total cholesterol level in the blood of rats fed ascidian meat powder decreased only slightly. Therefore, the ascidian tunic powder might be a healthy food with antidiabetic and antihyperlipidemic effects.

  • Carotenoids being most important pigments among those occurring in nature, have received increased interest owing to their beneficial effects on human health. An effort is made to review marine carotenoids as important bioactive compounds with reference to their presence, chemical, and biofunctional benefits they afford. The potential beneficial effects of marine carotenoids were particularly focused on astaxanthin and fucoxanthin, major marine carotenoids found in marine animals and aquatic plants, respectively. Both carotenoids show strong antioxidant activity which is attributed to quenching singlet oxygen and scavenging free radicals. The potential role of the carotenoids as dietary antioxidants has been suggested as being one of the main mechanism by which they afford their beneficial health effects such as anticancer activity and anti-inflammatory effect. Only recently, antiobesity effect and antidiabetic effect have been noted as specific and novel bio-functions of fucoxanthin. Nutrigenomic study reveals that fucoxanthin induces uncoupling protein 1 (UCP1) expression in white adipose tissue (WAT) mitochondria to lead to oxidation of fatty acids and heat production in WAT. Fucoxanthin improves insulin resistance and decreases blood glucose level, at least in part, through the down-regulation of tumor necrosis factor $\alpha$ ($TNF{\alpha}$) in WAT of animals.

Inhibitory Effect of Hexane Fraction from Myagropsis myagroides on Pancreatic α-Amylase In Vitro

  • A Myagropsis myagroides (Mm) methanol extract showed α-amylase inhibitory activity of 13% at a concentration of 5 mg/ml. Results showed that the hexane fraction from the Mm methanol extract exhibited α-amylase inhibitory activity with an IC50 value of 4.24 mg/ml. The hexane fraction was separated using silica-gel column chromatography, and six subfractions were obtained. The fraction eluted with CHCl3:MeOH = 50:1 showed the highest α-amylase inhibitory activity with an IC50 value of 0.72 mg/ml. This fraction was purified using Sephadex LH-20 column chromatography and an octadecyl silica (ODS) Sepak cartridge, obtaining seven subfractions. Fraction (Fr.) 4 also showed a strong α-amylase inhibitory activity with an IC50 value of 0.75 mg/ml. Fr. 4 was purified by Sephadex LH-20 column chromatography and ODS Sepak cartridge, obtaining six subfractions. Fr. 4-2 was identified as sargachromanol I with an IC50 value of 0.40 mg/ml, and the inhibition pattern analyzed from Lineweaver-Burk plots revealed it to be an uncompetitive inhibitor. These results suggest that Mm has potential as a natural antidiabetes agent.

  • Antidiabetic and beta cell-protection activities of purple corn anthocyanins (PCA) were examined in pancreatic beta cell culture and db/db mice. Only PCA among several plant anthocyanins and polyphenols showed insulin secretion activity in culture of HIT-T15 cells. PCA had excellent antihyperglycemic activity (in terms of blood glucose level and OGTT) and HbA1c-decreasing activity when compared with glimepiride, a sulfonylurea in db/db mice. In addition, PCA showed efficient protection activity of pancreatic beta cell from cell death in HIT-T15 cell culture and db/db mice. The result showed that PCA had antidiabetic and beta cell-protection activities in pancreatic beta cell culture and db/db mice.
