Antioxidant, anticoagulant, and α-glucosidase inhibitory effects of mistletoe (Viscum album var. coloratum) extract (겨우살이(Viscum album var. coloratum) 추출물의 항산화, 항응고 및 α-glucosidase 저해 효과)

  • Antioxidant, α-glucosidase inhibition, and anticoagulant effects of 80% ethanolic extract from mistletoe (Viscum album var. coloratum) were investigated. The yield and polyphenol content of the mistletoe extract were 30.9±0.4% and 57.6±1.5 mg gallic acid equivalents/g, respectively. The antioxidant effects of the mistletoe extract such as free and cationic radical scavenging ability, nitrite scavenging ability, and reducing power increased in proportion to its concentration. Also the mistletoe extract inhibited the activity of α-glucosidase, and delayed plasma coagulation mainly by inhibiting the extrinsic and common pathways in blood coagulation system.

  • Rhus verniciflua Stokes (RVS) is a widely used herbal plant with various biological properties. Our previous study using in vitro platelet aggregation in whole blood showed that the fractions of RVS had strong anti-aggregatory activity. In this study, using in vitro platelet aggregation in PRP and coagulation parameters, to investigate the anti-platelet activity and anticagulant effects of RVS ethyl acetate layer, the layer was subsequently fractionated by ODS columm chromatograph (50% MeOH).   • The anticoagulant activities of fucoidan fractions extracted from Sporophylls of Undaria pinnatifida, Laminaria religiosa, Hizikia fusiforme and Sargassum fulvellum were studied to assess the relationship between chemical characteristics and the activities. Crude fucoidans extracted with diluted HCl solution (pH 2.0) at $65^{\circ}C$ were precipitated with cetylpyridinum chloride and then fractionated by dissolving the precipitated complex with increasing $CaCl_2$, concentrations (1.0 M, 1.5 M, 3.0 M). The anticoagulant activities of the fractions with respect to activated partial thromboplastin (APTT) increased with increase in their sulfate content and Undaria finnatifida Fr-3.0 fraction, prepared by dissolving with 3.0 M $CaCl_2$ solution, exhibited the highest activity. The Undaria finnatifida Fr-3.0 fraction was further modified with pronase and laminase. The pronase and laminase treatment decreased protein and glucose content and the APTT activity was higher than that or parent Undaria finnatifida Fr-3.0 fraction. The pronase and laminase modified Undaria finnatifida Fr-3.0 was composed of fucose, galactose, mannose, sulfate, uronic acid in the approximately molar ratio of 1.00 : 1.30: 0.03 : 2.70 : 0.08.

  • Persimmon has been considered to have a therapeutic value diseases in Korea, Dried, persimmon was applied to a wounded part for an anti-inflammatory and analgesic activities, Anti-coagulating activity has been detected in persimmon by a thrombin-induced clotting assay using human plasma. The anti-coagulating activity was detectible in stem, pulp, core, leaf, and seed of persimmon The stem part of persimmon contains maximal anti-coagulating activity after-size-fractionation. Both of an anticoagulant and its denaturating factor were present in persimmon pulp. The concentration of anti-coagulating activity was determined and compared in different harvesting time, species, and available foods. The level of anti-coagulating activity was highest in persimmon(Fuyu)harvested in November. The anti-coagulating activity was decreased significantly in its processed foods. Persimmon could be expected to be effective in prevention of diseases induced by excess coagulation.

  • Artemisia princeps Pampanini (AP) has been used as a traditional medicine in Korea, China and Japan and reported to exhibit various beneficial biological effects including anti-inflammatory, antioxidant, anti-atherogenic and lipid lowering activities; however, its antiplatelet and anticoagulant properties have not been studied. In the present study, we evaluated the effects of an ethanol extract of Artemisia princeps Pampanini (EAP) and its major flavonoids, eupatilin and jaceosidin, on platelet aggregation and coagulation. To determine the antiplatelet activity, arachidonic acid (AA)-, collagen- and ADP (adenosine diphosphate)-induced platelet aggregation were examined along with serotonin and thromboxane A2 ($TXA_2$) generation in vitro. The anticoagulant activity was determined by monitoring the activated partial thromboplastin time (aPTT) and prothrombin time (PT) in vitro. The data showed that EAP and its major flavonoids, eupatilin and jaceosidin, significantly reduced AA-induced platelet aggregation and the generation of serotonin and $TXA_2$, although no significant change in platelet aggregation induced by collagen and ADP was observed. Moreover, EAP significantly prolonged the PT and aPTT. The PT and/or aPTT were significantly increased in the presence of eupatilin and jaceosidin. Thus, these results suggest that EAP may have the potential to prevent or improve thrombosis by inhibiting platelet activation and blood coagulation.

A Novel Anticoagulant Protein with High Affinity to Blood Coagulation Factor Va from Tegillarca granosa

  • A novel inhibitory protein against blood coagulation factor Va (FVa) was purified from muscle protein of granulated ark (Tegillarca granosa, order Arcoida, marine bivalvia) by consecutive FPLC method using anion exchange and gel permeation chromatography. In the results of ESI-QTOF tandem mass analysis and database research, it was revealed that the purified T. granosa anticoagulant protein (TGAP) has 7.7 kDa of molecular mass and its partial sequence, HTHLQRAPHPNALGYHGK, has a high identity (64%) with serine/threonine kinase derived from Rhodopirellula baltica (order Planctomycetales, marine bacteria). TGAP could potently prolong thrombin time (TT), corresponding to inhibition of thrombin (FIIa) formation. Specific factor inhibitory assay showed that TGAP inhibits FVa among the major components of prothrombinase complex. In vitro assay for direct-binding affinity using surface plasmon resonance (SPR) spectrometer indicated that TGAP could be directly bound with FVa. In addition, the binding affinity of FVa to FII was decreased by addition of TGAP in dose-dependant manner ($IC_{50}$ value = 77.9 nM). These results illustrated that TGAP might interact with a heavy chain of FVa ($FVa_H$) bound to FII in prothrombin complex. The present study elucidated that non-cytotoxic T. granosa anticoagulant protein (TGAP) bound to FVa can prolong blood coagulation time by inhibiting conversion of FII to FIIa in blood coagulation cascade. In addition, TGAP did not significantly (P < 0.05) show fibrinolytic activity and cytotoxicity on venous endothelial cell line (ECV 304).

Purification of Antithrombotic Material from Auricularia auricular-judae Extracts and Its Antithrombotic Activity (목이버섯 추출물로부터 항혈전물질의 정제와 항혈전효과)

  • Blood coagulation and aggregation of platelet are crucial events in the pathogenesis of various ischemic diseases. The substance which can prevent blood coagulation and platelet aggregation was extracted from wood ear mushroom (Auricularia auricular-judae) and its anticoagulation activity was investigated. The dried A. auricular-judae was extracted with 0.1 N NaOH and its supernatant was further extracted with methanol and ethanol followed by $H_{2}O$. The resulting methanol soluble fraction showed significant antithrombotic activity in activated partial thromboplastin time, thrombin time, and prothrombin time assays with values of 100, 124, and 54 sec, respectively. This active substance was purified with DEAE-Sepharose CL6B and Sephacryl 400-HR and was found to be polysaccharide with the average molecular weight of over 150 kDa. This polysaccharide was xyloglucomannan of which the main component was mannose, and its anticoagulant activity was mostly mediated by inhibition of thrombin activity.
