Antioxidant and Antiproliferative Activities of Methanol Extracts from Leafy Vegetables Consumed in Korea

  • There has been growing interest in the antioxidative and anticarcinogenic effects of vegetables. This study was aimed at evaluating the antioxidative and antiproliferative activities of 13 leafy vegetables consumed in Korea. New-beet and ornamental kale contained >1,000 mg of polyphenolics/100g fresh weight (FW), which was the greatest amount among the test vegetable extracts. Ornamental kale also contained the greatest amount (232.84 mg/100g FW) of flavonoids. With the exception of chicory, values of 1.1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging activities showed similar trends. New-beet was found to be the greatest antioxidant among the test vegetable extracts. New-beet exhibited the highest antiproliferative activity (>60%) against all tumor cells. No relationship was found between antiproliferative activity and antioxidant contents or antioxidant activities among samples.

  • Capsaicin (8-methyl-N-vanillyl-6-nonenamide), a pungent ingredient of hot chili peppers, has been reported to possess substantial anticarcinogenic and antimutagenic activities. In the present study. we investigated the effects of capsaicin on pulmonary metastasis of B 16-F10 murine melanoma cells. as well as its mechanism of action. Capsaicin (i.p.. 2.5mg/kg) suppressed the number of lung colonies (58%) in experimental lung metastasis assay. (omitted)

  • Arginine deiminase (ADI, E.C. 3.5.3.6), one of the arginine deprivation enzymes, exhibits anticarcinogenic activities. The present study investigated the anti-inflammatory activities of the purified recombinant ADI originating from Lactococcus lactis ssp. lactis ATCC7962 (LADI). LADI dose-dependently inhibited lipopolysaccharide (LPS)-induced upregulation of inducible nitric oxide synthase and the production of nitric oxide in RAW 264.7 murine macrophages. The induction of cyclooxygenase-2 expression and subsequent production of prostaglandin $E_2$ by LPS was also attenuated by LADI treatment. Moreover, LADI inhibited the production of interleukin-6 in LPS-stimulated RAW 264.7 macrophages. These results indicate that LADI exerts anti-inflammatory effects, which may in part explain its chemopreventive potential.

  • We have researched quantitative structure activity relationships between molecular structure of medicines and physiological activity. Since they are non-linear, neural networks are useful tool to research them. There are many ranks for the non-linearity; therefore, the neuron function must be selected carefully. As the results of some trial calculations, Ire find the sigmoid-linear functions pair. We call the neural network constructed of the pair as ANN. The inter- or extrapolation abilities of the ANN are excellent; therefere, ANN is a superior predictor for the relationships. We evaluated the anticarcinogenic medicines, Carboquinone derivatives, by the developed ANN and leave-one-out method.

감자를 첨가한 김치의 발효 특성 및 항암효과 (Fermentation Properties and In vitro Anticancer Effect of Kimchi Prepared with Potato)

  • Potato kimchi, fermentation was carried out at $10^{\circ}$C for 15 days using various ratios of potato to kimchi (2.5%, 5%, 10%). The samples were determined according to the fermentation time, pH, acidity and growth of lactic acid bacteria in potato kimchi. The addition ratio of potato to kimchi had little effect on the pH, acidity or growth of lactic acid bacteria in potato kimchi. In comparison to baechu kimchi and mul-kimchi, the pH, acidity and growth of lactic acid bacteria was better in potato kimchi than in the other kimchi samples. The in vitro anticancer effect of potato kimchi was investigated using human cancer cells, AGS human gastric adenocarcinoma cells and HT-29 human colon adenocarcinoma cells. MTT assay revealed that the methanol extract of potato kimchi showed the highest anticarcinogenic effects.

  • A wide array of phenolic substances, particularly those present in dietary and medicinal plants, have been reported to possess substantial anticarcinogenic and antimutagenic activities. The majority of naturally occurring phenolics retain antioxidative and anti-inflammatory properties which appear to contribute to their chemopreventive or chemoprotective activity. Inducible cyclooxygenase (COX-2) and nitric oxide synthase (iNOS) are important enzymes that mediate inflammatory processes.(omitted)

  • Silymarin, a polyphenolic flavonoid antiodant, has been shown to have anti-inflammatory, hepatoprotective, and anticarcinogenic effects. In the present study, we report the inhibitory effect of silymarin on nitric oxide (NO) production and inducible nitric oxide synthase (iNOS) mRNA expression in macrophages. In vivo administration of silymarin attenuated NO production of peritoneal macrophages in lipopolysaccharide (LPS)-treated mice.(omitted)

  • Induction of NAD(P)H : (quinone-acceptor) oxidoreductase (QR) which obligatory two electron reduction of quinones and prevents their participation in oxidative cycling and thereby the depletion of intracellular glutathione, has been used as a marker for chemopreventive agents. We postulated that vitamin E, an antioxidant, which induces QR as the gene of QR was reported to contain antioxidant reponsive element in the 5'-flanking region. Vitamin E resulted in significant induction of QR in both hepalclc7 cells and mouse tissues. QR induction was observed; to be maximal at 25uM vitamin E for hepalclc7 cells while it was maximal in the level of 2.5∼5 μmoles vitamin E/㎏ BW for mouse tissues. Thus the cancer-preventive effect of vitamin E may be exerted by it induction of intracellular detoxifying enzymes.

  • The first original inducer of interferon had been synthesized in Russia in 1965-1967. It was 2-car-boxiethyl-germanysesquioxan (Lx-13) and other-1 hydroxigermanytrans-monohydral (Lx-5), These compounds have very high induct$\circledcirc$on of interferon. The strain of Panax ginseng C. A. Meyer was recultivated on medium with different concentiation of (Lx-13) and (Lx-5) many times. The strain was stabilized when biomass of cells contain of the Ge form 10${\times}$10-3mg% to 2.3x10-3mg% to ashes, Biomass are hard physical work (swimming test) and also had stress-protective, immunomodulating action and anticarcinogenic effect hampered the sarcoma-180 by 33-80% and suppressed quantity of metanes on 43-46%.
