유통 축·수산물 중 잔류벤질페니실린의 검출 및 모니터링 (Detection and Monitoring of Benzylpenicillin Residues in Livestock and Marine Products)

  • 국내 유통 중인 돼지고기, 소고기, 닭고기, 넙치 232건의 시료에 대해 벤질페니실린의 잔류량을 조사하기 위해 현행 식품공전상 축 수산물 중 벤질페니실린 분석법에 대한 검증을 실시하고 모니터링을 수행하였다. 그 결과 벤질페니실린에 대한 분석법은 CODEX에서 요구하는 기준에 적합한 정밀성과 정확성을 보였다. 벤질페니실린의 잔류량 모니터링 결과 축산물 193건 중 11건, 수산물 39건 중 2건 총 13건이 검출되었다. 검출된 수준은 축 수산물 12건에서 0.001-0.004 mg/kg으로 잔류허용기준 이하로 검출되었으나, 축산물 중 국내산 돼지고기 1건에서 잔류허용기준을 초과한 0.134 mg/kg 수준으로 검출되어 해당 유관기관에 부적합 통보 및 사후관리를 요청하였다. 따라서 벤질페니실린 등 항생제는 다른 동물용 의약품에 비해 사용빈도가 많은 만큼 지속적으로 잔류모니터링과 관리가 필요할 것으로 판단된다.

Biophysical Studies Reveal Key Interactions between Papiliocin-Derived PapN and Lipopolysaccharide in Gram-Negative Bacteria

  • Papiliocin, isolated from the swallowtail butterfly (Papilio xuthus), is an antimicrobial peptide with high selectivity against gram-negative bacteria. We previously showed that the N-terminal helix of papiliocin (PapN) plays a key role in the antibacterial and anti-inflammatory activity of papiliocin. In this study, we measured the selectivity of PapN against multidrug-resistant gram-negative bacteria, as well as its anti-inflammatory activity. Interactions between Trp2 of PapN and lipopolysaccharide (LPS), which is a major component of the outer membrane of gram-negative bacteria, were studied using the Trp fluorescence blue shift and quenching in LPS micelles. Furthermore, using circular dichroism, we investigated the interactions between PapN and LPS, showing that LPS plays critical roles in peptide folding. Our results demonstrated that Trp2 in PapN was buried deep in the negatively charged LPS, and Trp2 induced the ${\alpha}$-helical structure of PapN. Importantly, docking studies determined that predominant electrostatic interactions of positively charged arginine residues in PapN with phosphate head groups of LPS were key factors for binding. Similarly, hydrophobic interactions by aromatic residues of PapN with fatty acid chains in LPS were also significant for binding. These results may facilitate the development of peptide antibiotics with anti-inflammatory activity.

전국에서 도축된 가축의 잔류검사에서 검출된 잔류물질 비교조사 (Comparison of antibiotic residues detected by the residue tests of slaughtered livestock in Korea)

  • It was carried out to investigate the antibiotic residues detected by the residue tests for edible tissues of slaughtered livestock in Korea from January to November in 2019. Positive cases of qualitative test and analytical test for 11 months were 689 and 341, respectively. Positive cases of analytical test for cattle, pigs, horses and poultry were 164, 168, 2 and 7, respectively. Antibiotics were detected from 472 materials from 341 cases and classified as β-lactams 162 (37.9%), aminoglycosides 141 (33%), quinolones 69 (16.2%), sulfonamides 23 (5.4%), amphenicols 10 (2.3%), tetracyclines 9 (2.1%), macrolides 6 (1.4%) and the rest 7 (1.4%). Other materials were benzyl penicillin 119 (27.9%), (dihydro)streptomycin 105 (24.6%) and enrofloxacin (including ciprofloxacin) 39 (9.1%). In conclusion, strict management of benzyl penicillin and streptomycin in cattle and pigs should be implemented to reduce the positive cases of slaughtered livestock in Korea.

Application of a solid-phase fluorescence immunoassay to determine ampicillin residues in muscle tissue of olive flounder (Paralichthys olivaceus)

  • $Parallux^{TM}$, a solid-phase fluorescence immunoassay(SPFIA) developed for antibiotics residue detection in milk, was applied for analysis of fish muscle. The recommended therapeutic dose of ampicillin(100 mg/kg body weight, withdrawal period 7 days) was orally administered to a group of 25 olive flounders(Paralichthys olivaceus) for consecutive five days. Muscle was sampled after drug treatment 1st, 2nd, 3rd, 4th and 5th day. The concentration of ampicillin in muscle, determined by SPFIA, was compared with that of internal standard(10 ppb as ampicillin). The absorbance ratio of sample to internal standard(Bs/Bo) was employed as an index to determine the muscle residue in olive flounder. To investigate the recovery rate, the standard solutions were added to muscle samples to give final concentrations in muscle of 10 and 50 ng/ml. The recovery rates of all spiked samples were > 89% of the spiked value. Ampicillin was detected in muscle of fishes treated until the 3rd day of withdrawal period. The present study showed that the SPFIA can be easily adopted in predicting tissue residues for ampicillin in farmed fishes.

경북지방 도축장의 지육에 대한 잔류물질 실태조사 (Survey on residual antibiotics for beef, pork and chicken at slaughter house in Gyeongbuk province)

  • This survey was carried out to detect the residual antibiotics in beef (n = 1,071), pork (n=7,837) and chicken (n=1,536) from slaughter houses in Gyeongbuk province by EEC-4 plate method, Charm II and HPLC during 2005. Residues of antibiotic were detected from 9 beef (0.8%) and 119pork (1.52%) by EEC-4 plate method, and total positive rates were 1.23% (128). 126 samples of the 128 positive samples by the EEC-4 plate method were detected by charm II test. 128 samples were classified as tetracyclines 110 (95.5%), ${\beta}$-lactam 2 (1.6%), sulfonamide 22 (17.2%), quinolone 1 (0.8%). The highest residual concentration of oxytetracycline, tetracycline, chlor-tetracycline, penicillin, sulfamethazine, sulfadimethoxine, sulfaquinoxaline, sulfamerazine, sulfamonomethoxine and enrofloxacin were 7.57, 0.27, 0.40, 0.24, 14.24, 4.33, 8.59, 0.12, 0.09 and 1.98 ppm, respectively and 49 samples were exceeded legal admitted levels.

Risk Assessment of Growth Hormones and Antimicrobial Residues in Meat

  • Growth promoters including hormonal substances and antibiotics are used legally and illegally in food producing animals for the growth promotion of livestock animals. Hormonal substances still under debate in terms of their human health impacts are estradiol-$17\beta$, progesterone, testosterone, zeranol, trenbolone, and melengestrol acetate (MGA). Many of the risk assessment results of natural steroid hormones have presented negligible impacts when they are used under good veterinary practices. For synthetic hormonelike substances, ADIs and MRLs have been established for food safety along with the approval of animal treatment. Small amounts of antibiotics added to feedstuff present growth promotion effects via the prevention of infectious diseases at doses lower than therapeutic dose. The induction of antimicrobial resistant bacteria and the disruption of normal human intestinal flora are major concerns in terms of human health impact. Regulatory guidance such as ADIs and MRLs fully reflect the impact on human gastrointestinal microflora. However, before deciding on any risk management options, risk assessments of antimicrobial resistance require large-scale evidence regarding the relationship between antimicrobial use in food-producing animals and the occurrence of antimicrobial resistance in human pathogens. In this article, the risk profiles of hormonal and antibacterial growth promoters are provided based on recent toxicity and human exposure information, and recommendations for risk management to prevent human health impacts by the use of growth promoters are also presented.

Use of veterinary medicinal products in the Philippines: regulations, impact, challenges, and recommendations

  • Agricultural production is a major driver of the Philippine economy. Mass production of animal products, such as livestock and poultry farming, is one of the most prominent players in the field. Filipino farmers use veterinary medicinal products (VMPs) when raising agricultural animals to improve animal growth and prevent diseases. Unfortunately, the extensive use of VMPs, particularly antibiotics, has been linked to drug resistance in animals, particularly antibiotics. Antimicrobial gene products produced in animals due to the prolonged use of VMPs can passed on to humans when they consume animal products. This paper reviews information on the use of VMPs in the Philippines, including the regulations, their impact, challenges, and potential recommendations. The Philippines has existing legislation regulating VMP use. Several agencies were tasked to regulate the use of VMPs, such as the Department of Agriculture, the Department of Health, and the Philippine National Action Plan. Unfortunately, there is a challenge to implementing these regulations, which affects consumers. The unregulated use of VMPs influences the transmission of antibiotic residues from animals to crops to humans. This challenge should be addressed, with more focus on stricter regulation.

남해안 양식어류의 fluoroquinolone계 항균제 잔류량 (Residual Concentrations of Fluoroquinolones in Farmed Fish in the Southern Coast of Korea)

  • Fluoroquinoles have a wide range of antimicrobial properties and are effective in the treatment of bacterial diseases in fish. The use of fluoroquinoles continues to grow steadily. Fluoroquinolone antibiotics are probably the most important class used among synthetic antibiotics in human and veterinary medicines because of their broad activity spectrum and good oral absorption. This study was conducted to estimate the residue of antibiotics in four species of farmed fishes, including olive flounder (Paralichthys olivaceus), black rock fish (Sebastes schlegeli), red sea bream (Pagrus major), and sea bass (Lateolabrax japonicus), collected from fish farms located in the southern coastal area of Korea. The residues of fluoroquinolones were determined using high performance liquid chromatography (HPLC) with a fluorescence detector. Residuals of five fluoroquinolones in muscle tissue of farmed fish were analyzed. We found that enrofloxacin was the most common agent in fish muscle, and that ciprofloxacin was the next most common. The range of detected concentrations of fluoroquinolones in olive flounder muscle was 0-0.859 mg/kg in 32.6% of all samples. Enrofloxacin was commonly detected in sea bass muscle at a range of 0-0.143 mg/kg in 38.9% of all samples. Fluoroquinolones were detected in 6.9% of black rock fish muscle and in 16.6% of sea bream, although the detected concentration was below 0.01 mg/kg. The maximum detection value of enrofloxacin and ofloxacin in olive flounder at the time of shipping was 0.102 mg/kg and 0.09 mg/kg, respectively; no other antimicrobials were detected. We detected no antimicrobial substances in red sea bream.

식품 중 잔류동물용의약품의 안전관리 (Management of Veterinary Drug Residues in Food)

  • 동물용의약품은 가축의 질병예방 및 치료, 성장 촉진 등 생산성을 높이는 목적으로 사용되는 의약품임에도 불구하고 무분별하게 과용 또는 남용되면 축 수산물에 잔류하게 되고 장기간 노출될 경우 인체에 유해할 수 있기 때문에 관리가 필요한 물질이다. 국내 동물용의약품의 관리는 국립수의과학 검역원에서 국내 사용되는 동물용의약품의 허가를 담당하고 있으며, 식품의약품안전청은 해당 동물용의약품의 잔류자료, 일일섭취허용량, 식품섭취량 및 국민평균체중 등을 이용하여 위해평가를 실시하고 그에 알맞은 잔류허용기준을 고시하고 있다. 현재 국내의 동물용의약품에 대한 잔류허용기준은 116 개가 고시되어 있으며, 2009년 말까지 총 142종에 대하여 기준이 설정될 예정이다. 또한 2010년 1월 1일부터 축 수산물 (유, 알 포함) 및 벌꿀에서 기준 미설정 항생제 및 합성항균제가 검출될 경우 0.03 mg/kg의 일률기준을 적용함으로써 축 수산물에 대한 안전관리가 강화하게 된다. 현재 동물용의 약품의 기준관리는 이미 선진국 수준으로 운영되고 있으며, 잔류분석법도 또한 국제적 수준에 맞게 개선할 예정이다.

HPLC-PDA를 이용한 닭고기 중 Nitroxoline 분석법 개발 (Development and Validation of Analytical Method for Nitroxoline in Chicken Using HPLC-PDA)

  • 우리나라는 2007년부터 원료축산물에 대한 동물용의약품 잔류허용기준 설정을 강화하면서 관련 시험법을 식품공전에 등재하여 활용하고 있다. 더불어 국외에서는 국제식품규격위원회와 유럽연합의 규제 강화 추이에 따라 불검출기준 물질에 대한 MRL 설정 등 낮은 농도의 정량한계를 가지는 검증된 분석법 개발 등이 활발히 진행되고 있다. 따라서 본 연구는 가금류 중 잔류허용기준은 아직 설정되어 있지 않으나, 식품안전성 조사 및 연구 등을 위해서는 시험법의 확립이 필요한 nitroxoline의 닭고기 근육 중 잔류시험법을 개발하고자 하였다. 분석에 사용된 검체는 닭고기의 근육을 이용하였다. 검체에 아세트로니트릴을 가하여 추출한 후, 이를 감압 농축하여 메탄올로 녹인 다음 MCX 카트리지로 정제한 후 HPLC-PDA에 주입하였다. 기기분석은 C18 컬럼을 사용하였고, 0.1 % TBAOH-인산용액과 메탄올을 기울기 조건으로 하여 360 nm에서 측정하였다. 또한, 액체크로마토그래프-질량분석기를 통해 확인시험을 수행하였으며, 모든 검증은 CAC 가이드라인(CAC/GL 16, 1993; CAC/GL 71, 2009) 규정에 따라 실시하였다. 그 결과, nitroxoline의 LOQ는 0.02 mg/kg 수준이었고, 회수율은 72.9~88.1 %로 나타났다. 또한, 변동계수는 2.5~11.7 %로 CAC 가이드라인(CAC/GL 16, 1993; CAC/GL 71, 2009) 규정에 만족하는 수준이었다. 따라서 개발된 분석법은 잔류동물용의약품의 분석에 있어 보다 신속하고 경제적인 분석 및 모니터링에 적용 가능할 것으로 기대된다.
