Application of a solid-phase fluorescence immunoassay to determine neomycin residues in muscle tissue of olive flounder (Paralichthys olivaceus), rockfish (Sebastes schlegeli), and red sea bream (Pagrus major)

  • Parallux, a solid-phase fluorescence immunoassay (SPFIA) developed for detection antibiotics residue in milk, was applied for analysis of antibiotics in muscle tissue of olive flounder (Paralichthys olivaceus), rockfish (Sebastes schlegeli), and red sea bream (Pagrus major). Fishes were dipped in neomycin 140 mg/ton water, the recommended therapeutic dose, for 24 h. Muscle samples were obtained on 1st, 2nd, 3rd, 4th and 5th day after drug treatment. The concentration of neomycin in muscle was determined using an internal standard (100 ppb as neomycin). The absorbance ratio of sample to internal standard (S/C) was employed as an index to determine the muscle residues in fishes. To investigate the recovery rate, the standard solutions were added to muscle samples to give final concentrations in muscle of 0.2 and 0.5 mg/ml. The recovery rates of all spiked samples were > 85% of the spiked value. Neomycin was detected in muscles of fishes treated after the 1st day of withdrawal period. On the 2nd day after drug treatment, all muscle samples showed negative reaction (S/C ration ${\leq}$ 1.0). The present study showed that the SPFIA can be applied for predicting residues of neomycin in muscle tissues of farmed fishes.

  • The steady growth in population has led to an enhanced water demand and immense pressure on water resources. Pharmaceutical residues (PRs) are unused or non-assimilated medicines found in water supplies that originate from the human and animal consumption of antibiotics, antipyretics, analgesics etc. These have been detected recently in sewage effluents, surface water, ground water and even in drinking water. Due to their toxicity and potential hazard to the environment, humans and aquatic life, PRs are now categorized as the emerging contaminants (ECs). India figures in the top five manufacturers of medicines in the world and every third pill consumed in the world is produced in India. Present day conventional wastewater treatment methods are ineffective and don't eliminate them completely. The use of nanotechnology via advanced oxidation processes (AOP) is one of the most effective methods for the removal of these PRs. Present study is aimed at reviewing the presence of various PRs in water supplies and also to describe the process of AOP to overcome their threat. This study is also very important in view of World Health Organization report confirming more than 30 million cases of COVID-19 worldwide. This will lead to an alleviated use of antibiotics, antipyretics etc. and their subsequent occurrence in water bodies. Need of the hour is to devise a proper treatment strategy and a decision thereof by the policymakers to overcome the possible threat to the environment and health of humans and aquatic life.

  • Fish pathology is one of the main scientific bases upon which this expansion in aquaculture has been dependent and requires a wide knowledge of the environmental constraints, the physiology and characteristic of the various pathogens, the responses of the host, and the methods by which they may be controlled. The primary disease and parasite problems in aquaculture animals related to viral, bacteria, fungal and protozoan epizootics. Parasitic nematodes, trematodes and cestodes are commonly found in aquaculture animals, but seldom are they present in concentrations sufficient to cause significant problems, When an epizootic does occur and chemical treatment is indicated, the appropriate chemical must be selected an properly applied. We have antibiotics, sulfa, nitrofuran and other chemicals for treatment of fish diseases, Some may be mixed with the feed during formulation, added to the pellets of feed as a surface coating, given in the form of an injection or used as a bath. Even though a drug or chemical has been officially approved for use in aquaculture, the substance should never be used unless there is a clear need, Some of the reasions for this view are as follows: (1) the constant use of antibiotics can leak to the development of resistant strains of bacteria, (2) biofilter efficiency may be impaired or destroyed by chemicals added to closed recirculating water systems, and(3) the injudicious use of chemical can have a damaging effect on the environment as well as on human.

양식산 무지개송어에서의 항생제와 말라카이트 그린 잔류량 조사 (The Residues of Antibiotics (Tetracycline, Oxolinic Acid and Ciplofloxacin) and Malachite Green in Cultured Rainbow Trout)

  • Products from aquaculture have sometimes been focused on the problems caused by the contamination of chemical agents as the use of chemical agents in aquaculture has been annually increased. The risk of contamination of products by chemical agents is greater in freshwater than in seawater. In order to evaluate the food safety of a fish grown in freshwater, we investigated the residues of antibiotics (tetracycline, oxolinic acid and ciplofloxacin) and malachite green in cultured rainbow trout, Oncorhynchus mykiss. Malachite green, which was prohibited in the application of aquaculture, was not detected in samples tested in this study. The residual content of tetracycline was determined to be less than the permissible amount, <0.2 mg/kg. The contents of ciplofloxacin was also less than the permissible amount, <0.1 mg/kg. However, in case of oxolinic acid, one of samples was only exhibited higher content than the permissible amount (<0.1 mg/kg). The results obtained in this study suggested that the control and regulation of chemical agents such as antibiotics was important to maintain a safe and worry-free seafood supply.

A novel method to depurate β-lactam antibiotic residues by administration of a broad-spectrum β-lactamase enzyme in fish tissues

  • As a novel strategy to remove ${\beta}$-lactam antibiotic residues from fish tissues, utilization of ${\beta}$-lactamase, enzyme that normally degrades ${\beta}$-lactam structure-containing drugs, was explored. The enzyme (TEM-52) selectively degraded ${\beta}$-lactam antibiotics but was completely inactive against tetracycline-, quinolone-, macrolide-, or aminoglycoside-structured antibacterials. After simultaneous administration of the enzyme with cefazolin (a ${\beta}$-lactam antibiotic) to the carp, significantly lowered tissue cefazolin levels were observed. It was confirmed that the enzyme successfully reached the general circulation after intraperitoneal administration, as the carp serum obtained after enzyme injection could also degrade cefazolin ex vivo. These results suggest that antibiotics-degrading enzymes can be good candidates for antibiotic residue depuration.

육류중 잔류 항생물질 및 테트라싸이클린 조사 (Detection of Residual antibiotics and tetracyclines in beef, pork and chicken)

  • The purpose of the survey were determined the residual antibiotics and tetracyclines in beef(n=1,364), pork(n=2,817) and chickens(n=1,921) by the EEC 4-plate method, Charm IIand HPLC. The results were summarized as follows ; 1. The recovery rates in spiked samples were 98.8-107.2% for oxytetracycline(OTC), 33.2-48.6% for tetracycline(TC) and 64.1-72.3% for chlortetracycline(CTC) at 0.05-0.1ppm by HPLC using MSPD. 2. Residues of antibiotic were detected from 10 beef(0.75%) and 36 pork(1.31%) by EEC 4-plate method. In case of chickens were not detected. 3. Twenty-eight from 46 positive samples by the EEC 4-plate method were classified as TCs(60.9 %) by Charm II and TC of 12 samples were detected by HPLC. 4. Ten samples were detected levels of OTC ranging from 0.035 to 0.635 ppm and 3 samples were levels of CTC ranging from 0.066 to 0.150 ppm. OTC and CTC levels in 3 beef and 4 pork samples were exceeded the current tolerance level of 0.1 ppm.

Antibacterial Activity and Synergism of the Hybrid Antimicrobial Peptide, CAMA-syn

  • A 20-residue hybrid peptide CA(1-8)-MA(1-12) (CAMA) incorporating residues 1-8 of cecropin A (CA) and residues 1-12 of magainin 2 (MA) has high antimicrobial activity without toxicity. To investigate the effects of the total positive charges of CAMA on the antibacterial activity and toxicity, a hybrid peptide analogue (CAMA-syn) was designed with substitutions of $Ile^{10}\;and\;Ser^{16}$ with Lys. According to CD spectra, structure of CAMA-syn with increase of cationicity was very similar to that of CAMA in DPC micelle. CAMA-syn showed antimicrobial activity similar with CAMA while CAMA-syn has no hemolytic activity and much lower cytotoxicity against RAW 264.7 macrophage cells than CAMA. Also, CAMA and CAMA-syn significantly inhibited NO production by LPSstimulated RAW264.7 macrophage at 10.0∼20.0 $\mu$M. CAMA-syn displayed salt resistance on antimicrobial activity against Escherichia coli at the physiological concentrations of $CaCl_2\;and\;MgCl_2$. The combination studies of peptides and antibiotics showed that CAMA-syn has synergistic effects with synthetic compound and flavonoid against Enterococcus faecalis and VREF. CAMA-syn can be a good candidate for the development of new antibiotics with potent antibacterial and synergistic activity but without cytotoxicity.

Application of ELISA for the Detection of Oxytetracycline Residue in Live Animals

  • Oxytetracycline has been widely used in the cattle industry to control pneumonia, shipping fever, foot rot, bacterial enteritis, and uterine infections. Extensive use of antibiotics in veterinary clinics has resulted in residues in tissue and bacterial resistance to antibiotics. To prevent unwanted drug residues from entering the human food chain, extensive control measures have been established by both government authorities and industries. The demands for reliable, simple, sensitive, rapid and low-cost methods for residue analysis of foods are increasing. In this study, we established a rapid test for tissue residues of oxytetracycline in cattle. The recommended therapeutic dose of oxytetracycline (withdrawal time, 14 days) was administered to 10 cattle. Blood samples were collected from each cow before drug administration and during the withdrawal period. The concentration of oxytetracycline in plasma, determined by a semi-quantitative ELISA, was compared to that of the internal standard, 10 ppb. The absorbance ratio of internal standard to sample (B/Bs) was employed as an index to determine whether the residues in cattle tissues were negative or positive. That is, a B/Bs ratio less than 1 was considered as residue positive and that greater than 1 as negative. Based on this criterion, all plasma samples from cattle were negative to oxytetracycline at pre-treatment. Oxytetracycline could be detected in the plasma treated cattle until day 14 post-treatment. The present study showed that the semi-quantitative ELISA could be easily adapted in predicting tissue residues for oxytetracycline in live cattle.

어류 중 4계열 잔류 항생물질 검출을 위한 Lateral Flow Immunoassay Kit 개발 (A Lateral Flow Immunoassay Kit for Detecting Residues of Four Groups of Antibiotics in Farmed Fish)

  • A lateral flow immunoassay kit based on antigen-antibody interactions was developed to detect residues of beta-lactams, quinolones, tetracyclines, and sulfonamides in farmed fish. Group-specific antibodies showing cross-reactivity with other antibiotics in the same group were produced in rabbits. The rabbits were immunized eight times to obtain the maximum titers. Antibodies were extracted from the antisera collected from the immunized rabbits and produced group-specific reactions with antibiotics from the four groups. A kit was prepared that optimize conditions for the antigen-antibody reaction, using colloidal gold conjugated antibodies, and was designed to detect the four groups of antibiotics simultaneously. The kit enabled the detection of antibiotics in the four groups at below maximum residue limits (MRLs), which were $200{\mu}g/kg$ for tetracyclines, $100{\mu}g/kg$ for sulfonamides, $50{\mu}g/kg$ for beta-lactams, and $100{\mu}g/kg$ for quinolones. The cross-reactivity of the antibodies ranged from 10-80% for the sulfonamides, 20-100% for tetracyclines, 38-100% for quinolones, and 20-100% for the beta-lactams, confirming that the antibodies were group specific. The test kit was used 30 times to examine spiked antibiotics at the limits of detection (LODs) and all produced positive results, indicating high sensitivity. The LODs for the assay ranged from 4-20 ng/mL for beta-lactams, 25-50 ng/mL for sulfonamides, 20-100 ng/mL for tetracyclines, and 30-80 ng/mL for quinolones, and there were no false negative reactions at above these LODs. In addition, all of the LODs of the developed kit were correlated with high-performance liquid chromatography (HPLC) data. Our lateral flow immunoassay kit can simultaneously detect antibiotic residues from a large number of fish samples rapidly, strengthening the safety of domestic farmed and imported fish.

LC-MS/MS를 이용한 육류 중 페니실린계 항생제 8종의 동시분석 및 적용성 검증 (Simultaneous Determination of Penicillin Antibiotics in Meat using Liquid Chromatography Tandem Mass Spectrometry)

  • 본 연구에서는 LC-MS/MS를 이용하여 페니실린계 동물용의약품 8종, 암피실린, 아목시실린, 클록사실린, 디클록사실린, 나프실린, 옥사실린, 페니실린 G, 페니실린 V의 동시분석법을 확립하고 잔류실태조사를 수행하였다. 피페라실린을 내부표준물질으로 적용하였으며, 이동상으로는 0.05% 포름산 용액와 0.05% 포름산 아세토니트릴 용액을 gradient mode로 사용하였고 $C_{18}$ 칼럼을 이용하여 분석하였다. 각 분석물질은 MRL의 0.25, 0.5, 1, 2, 4, 8배 농도범위에서 검량선을 작성하여 $R^2{\geq}0.99$의 직선성 및 재현성을 확인하였다. 소고기, 돼지고기, 닭고기에 대하여 MRL의 0.5, 1, 2배인 세 가지 농도에서 6회 반복으로 분석을 실시한 결과, 평균 회수율은 71.0~106%, 상대표준편차는 4.0~11.2%, LOD는 0.003~0.008 mg/kg, LOQ는 0.01~0.03 mg/kg이었다. 국내 유통 소고기, 돼지고기, 닭고기에서 페니실린계 항생제 8종의 분석결과, 총 193건 중 경기도 지역의 국내산 소고기 1건에 대해서 클록사실린이 MRL 이하인 0.08 mg/kg이 검출되었으며, 나머지 192건의 검체에서는 불검출로 분석되었다. 본 연구를 통하여 확립된 페니실린계 동물용의약품 8종 동시분석법은 식육에 대한 페니실린계 동물용의약품의 안전관리 체계 방안을 제시하기 위한 기초자료로 활용 가능한 것으로 사료된다.
