Application of Probiotics for the Production of Safe and High-quality Poultry Meat

  • Poultry industry has always been a dynamic and integral part of national economies in many countries. Economic losses incur especially in large-scale rearing facilities, often attributed to the deterioration of environmental conditions, poultry exposure to stressors and development of diseases. While antibiotics have been commonly used for prophylactic purposes and as growth stimulants, extensive documentation of antimicrobial resistance among pathogenic bacteria due to indiscriminate utilization of antibiotic in the industry has led to public and governmental outcries. Elimination of antibiotics from poultry production has thus encouraged intensive search for alternatives. In this review, we discuss the immense potential of probiotics to fill the gap as alternative growth promoters and evidences of beneficial effects of probiotic application in poultry production.

Fungichromin Production by Streptomyces padanus PMS-702 for Controlling Cucumber Downy Mildew

  • Streptomyces padanus PMS-702 strain produces a polyene macrolide antibiotic fungichromin and displays antagonistic activities against many phytopathogenic fungi. In the present study, experimental formulations were assessed to improve the production of fungichromin, the efficacy of PMS-702 on the suppression of sporangial germination, and the reduction of cucumber downy mildew caused by Pseudoperonospora cubensis. PMS-702 strain cultured in a soybean meal-glucose (SMG) medium led to low levels of fungichromin accumulation and sporangial germination suppression. Increasing medium compositions and adding plant oils (noticeably coconut oil) in SMG significantly increased fungichromin production from 68 to $1,999.6{\mu}g/ml$. Microscopic examination reveals that the resultant suspensions significantly reduced sporangial germination and caused cytoplasmic aggregation. Greenhouse trials reveal that the application of PMS-702 cultural suspensions reduced downy mildew severity considerably. The addition of Tween 80 into the synthetic medium while culturing PMS-702 further increased the suppressive efficacy of downy mildew severity, particularly when applied at 24 h before inoculation or co-applied with inoculum. Fungichromin at $50{\mu}g/ml$ induced phytotoxicity showing minor necrosis surrounded with light yellowish halos on cucumber leaves. The concentration that leads to 90% inhibition (IC90) of sporangial germination was estimated to be around $10{\mu}g/ml$. The results provide a strong possibility of using the S. padanus PMS-702 strain as a biocontrol agent to control other plant pathogens.

Comparison of the Effect of Green Tea By-product and Green Tea Probiotics on the Growth Performance, Meat Quality, and Immune Response of Finishing Pigs

  • The objective of this experiment was to compare the effects of green tea by-product and green tea probiotics on the growth performance, meat quality and immune response of finishing pigs. A total of 72 crossbred "Landrace$\times$Yorkshire" finishing pigs with an average of 76 kg body weight were assigned to 4 dietary treatments in a completely randomized design. Each treatment had 3 replications with 6 pigs per replication. The four dietary treatments were control, antibiotics (control diet with 0.003% chlortetracycline added), and diets containing 0.5% green tea by-product or 0.5% green tea probiotic supplementation. Weight gain was increased in 0.5% green tea probiotics treatment compared to others, but there was no significant difference (p>0.05). The incorporation of 0.5% green tea probiotics to diets reduced the feed conversion ratio in finishing pigs (p>0.05). The incorporation of 0.5% green tea by-product into the pig diet reduced the crude protein and fat contents of the meat (p>0.05). Pigs fed diets containing 0.5% green tea probiotic supplementation had lowered meat TBA values compared to those fed 0.5% green tea by-product (p<0.05). The proliferation of spleen cells stimulated with Con A (concanavalin: 0.1, 0.3, and $1.0{\mu}g/ml$) significantly increased with 0.5% green tea by-product treatment compared to antibiotic treatment (p<0.05), but was significantly decreased in 0.5% green tea probiotics treatment compared to the antibiotic treatment (p<0.05). When stimulated with $1.0{\mu}g/ml$ Con A, splenocyte production of IL-6 from pigs treated with 0.5% green tea by-product or green tea probiotics was significantly increased compared to the antibiotic treatment group (p<0.05). Splenocyte production of TNF-${\alpha}$ after treatment with $1.0{\mu}g/ml$ Con A was significantly higher following 0.5% green tea probiotics treatment (p<0.05), while TNF-${\alpha}$ production after $10.0{\mu}g/ml$ LPS (lipopolysaccharide) was significantly higher in the 0.5% antibiotic treatment group (p<0.05).

사료내 Cu-methionine Chelate와 Cu-soy Proteinate가 산란계의 생산성, 소장내 미생물 균총 및 면역체계에 미치는 영향 (Effect of Dietary Supplementation of Cu-methionine Chelate and Cu-soy Proteinate on the Performance, Small Intestinal Microflora and Immune Response in Laying Hens)

  • 본 실험은 Cu-methionine chelate(Cu-Met)와 Cu-soy proteinate(Cu-SP)가 산란계의 생산성과 소장내 미생물 균총 및 면역 체계에 미치는 영향을 조사하기 위해 실시하였다. 사양시험은 39주령의 산란계(Hy-line $Brown^{(R)}$) 960수를 선별하여 A형 2단 4열 케이지에 대조구 포함 총 6처리구로 구성하여 처리당 4반복, 반복당 40수씩(2수 수용 케이지 20개) 난괴법으로 임의 배치하였다. 시험 기간 동안 물과 사료는 자유 섭취하게 하였으며, 일반적인 점등 관리(자연 일조+조명; 16 hr)를 실시하였다. 시험구는 대조구, 항생제구(avilamycin 6 ppm), Cu- Met 50(Cu-methionine으로 Cu 50 ppm), Cu-Met 100(Cu- methionine으로 Cu 100 ppm), Cu-SP 50(Cu-soy proteinate로 Cu 50 ppm) 그리고 Cu-SP 100(Cu-soy proteinate로 Cu 100 ppm) 등 총 6처리였다. 일계 산란율(Hen-day egg production)과 산란지수(Hen-house egg production)는 Cu 첨가구들과 항생제 첨가구들이 대조구에 비해 높은 경향을 보였는데, 특히 Cu-Met 100 구는 대조구에 비하여 유의적으로(P<0.05) 높았으며 Cu 처리구들 간에 종류나 첨가 수준에 따른 차이는 유의하지 않았다. 난중에서는 항생제구와 Cu-SP 첨가구들이 대조구보다 무거운 경향을 보였고, Cu-Met 첨가구들보다는 유의하게(P<0.05) 무거웠다. 난각 강도, 난각 두께, 난각색, 난황색, Haugh unit 등 계란 품질에 있어서 처리구간에 유의적인 차이는 나타나지 않았다. Leukocytes와 erythrocytes 관련 모수들은 평균 적혈구 혈색소량(MCH)을 제외한 모든 항목에서 처리간에 유의적인 차이는 나타나지 않았다. MCH는 Cu-SP 100이 대조구와 비교하여 유의적으로 높았다(P<0.05). 혈청 IgG와 IgA의 농도는 처리구간에 유의적인 차이는 나타나지 않았다. 간 내 구리의 함량은 처리간에 유의한(P<0.05) 차이가 있었는데, 구리의 첨가 수준이 높을수록 높아지는 경향이 있었으며 같은 첨가 수준에서는 Cu-SP 첨가구들이 Cu-Met 첨가구들보다 높은 경향이 있었다. 소장 내용물 중 Cl. perfringens의 수는 항생제와 Cu 첨가구들이 대조구에 비해 적었고 E. coli 수는 첨가구들에서 감소하는 경향이 있었으며, Lactobacilli 수는 Cu 첨가구들이 대조구나 항생제구와 비교하였을 때 많았다. 결론적으로 Cu-Met와 Cu-SP는 산란계의 산란율 개선에 있어서 항생제와 유사한 효과를 나타내었다. Cu-SP는 Cu-Met보다 난중을 유의하게 증가시켰으나 다른 항목들에 있어서는 Cu 공급원(Cu-Met vs Cu-SP)과 첨가수준(50 vs 100 ppm)에 따른 유의적 차이는 없었다.

Nutritional Studies on Production of Antibacterial Activity by the Zebra Mussel Antagonist, Pseudomonas fluorescens CL0145A

  • Pseudomonas fluorescens strain CL0145A was discovered at the New York State Museum Field Research Laboratory as an effective agent against the environmentally destructive zebra mussel, which has contaminated US waters. Dried cells of the microbe are being commercialized as an environmentally friendly solution to the problem. We found that antibiotic activity against the Gram-positive bacterium Bacillus subtilis is produced and excreted by this strain. We have carried out studies to optimize production of the antibiotic. Studies were begun in a complex corn meal medium. Activity was found in both cells and culture supernates and was maximal after one day of fermentation. Static fermentation conditions were found to be superior to shaken culture. Production of extracellular antibiotic in complex medium was found to be dependent on the content of sucrose and enzyme-hydrolyzed casein. Indeed, production was greater in sucrose plus enzyme-hydrolyzed casein than in the complex medium. Of a large number of carbon sources studied as improvements over sucrose, the best was glycerol. An examination of nitrogen sources showed that production was improved by replacement of enzyme-hydrolyzed casein with soy hydrolysates. Production in the simple glycerol-Hy-Soy medium was not improved by addition of an inorganic salt mixture or by complex nitrogen sources, with the exception of malt extract. In an attempt to keep the medium more defined, we studied the effect of amino acids and vitamins as replacements for malt extract. Of 21 amino acids and 7 vitamins, we found tryptophan, glutamine, biotin, and riboflavin to be stimulatory. The final medium contained glycerol, Hy-Soy, tryptophan, glutamine, biotin, and riboflavin.

Detection, Characterization and Antibiotic Susceptibility of Clostridioides (Clostridium) difficile in Meat Products

  • Clostridioides (Clostridium) difficile is a Gram (+), anaerobic, spore forming, rod shaped bacterium that can produce toxin. The objective of this study is to reveal the presence of C. difficile in meat products, to analyze the ribotype diversity by PCR and to evaluate the antibiotic susceptibility of isolated strains. The organism was isolated in 22 out of 319 (6.9%) examined meat product samples and 9 out of 22 (40.9%) isolates were identified as RT027 and all isolates had the ability of toxin production. In terms of antibiotic susceptibility, all isolates were susceptive to amoxicillin-clavulanic acid, tetracycline and vancomycin and 21 (95.4%) isolates to metronidazole. On the other hand, imipenem and cefotaxim resistance was observed in all. In conclusion, the results of this comprehensive study conducted in Turkey deduced the presence of C. difficile in different meat products. Therefore, these products can be evaluated as a potential contamination source of C. difficile from animals to humans especially for elders, youngsters, long terms wide spectrum antibiotic used and immuno-suppressed individuals.

다제내성 Acinetobacter baumannii에 유효한 방선균 B-51의 탐색 및 이 균주가 생산하는 항생물질 발효 최적 배양 조건 (Screening and Optimal Culture Conditions of Antibiotic-Producing Actinomycetes B-51 for Multidrug Resistant Acinetobacter baumannii)

  • 다제내성균 A. baumannii에 유용한 항생물질을 생산하는 방선균을 선별하기 위하여 토양에서 분리한 방선균 300여 균주를 대상으로 항균 활성을 실험하여 그 활성이 가장 뛰어난 B-51 균주를 최종적으로 선별하였다. 이 균으로부터 항생물질 생산을 위한 최적 조건을 조사하기 위하여 발효 배지를 기본배지로 하여 동일조건 하에서 탄소원, 질소원, 무기염 및 인삼염만을 다르게 하여 항균 활성을 실험한 결과 glycerol 2%, soybean meal 0.5%, $CaCl_2$ 0.01%, $MgSO_4{\cdot}7H_2O$ 0.01%, $KH_2PO_4$ 0.01%에서 항생물질 생산이 가장 높았다. 또한 이렇게 결정된 배지 조성에서 초기 pH 6.0, 배양온도 $30^{\circ}C$, 배양시간 76시간 조건 하에서 항생물질 생산이 가장 많았다. 항생물질을 생산하는 대부분의 방선균이 항생물질 생산 시 배양액의 final pH가 중성 혹은 약 알칼리인 점을 비교해 볼 때 본 균주인 B-51의 경우 항생물질 생산능이 높은 경우 final pH 4.0 부근인 산성 pH를 보여 그간의 논문보고와는 상반되는 결과를 보였다.

청국장 발효 균주인 Bacillus subtilis 028-1의 항생물질 생산과 특성 (Characterization and Production of Antibiotic by Bacillus subtilis 028-1, a Chungkookjang Fermenting Strain)

  • Bacillus subtilis 028-1 균주는 청국장 발효에 사용하는 균주로 Staphylococcus sp. LS2 뿐만 아니라 여러 yeast 균주의 생장을 억제하는 항생물질을 생산하며, soybean meal 2%와 maltose와 같은 이당류를 1% 첨가하여 15~18시간 진탕 배양하였을 때 최대의 항생물질 생산을 보였으며 배지의 pH는 6.5 이하였다. 항생물질의 활성은 약염기성 조건에서 극대화되었으며, $100^{\circ}C$에서 20분간 가열하여도 활성은 크게 감소하지 않았고, 실온 보관 시 한 달 이상 효과가 지속되며 chymotrypsin과 papain 같은 단백질 분해효소 처리에 의해 서서히 활성이 줄어들었다. 투석에 의해 항생물질의 분자량을 측정한 결과 1,000에서 500 dalton 사이인 것으로 나타났으며 항미생물 효과는 있으나 fibrin 분해 능력이 없으므로 surfactin이 아닌 iturin 계열의 peptide성 항생물질로 보인다.

Effect of Salvia miltiorrhiza aerial parts on growth performance, nutrient digestibility, and digestive enzymes in rabbits

  • Objective: This study aimed to investigate the effect of Salvia miltiorrhiza (S.m.) aerial parts as an alternative ingredient on growth performance, nutrient digestibility, and digestive enzymes in growing rabbits. Methods: Treatments included five tested diets: a control (basal diet), antibiotic (basal diet+enramycin at 5 mg/kg), and S.m. aerial parts powder added at 3.0%, 6.0%, and 9.0% of feed using 300 growing rabbits. Results: The diets with S.m. aerial parts addition at 9.0% decreased (p<0.05) feed/gain compared to the control, but there were no differences in feed intake and body weight gain. In contrast with the control, the addition of antibiotic increased (p<0.05) digestibility of dry matter, crude protein, energy, fiber, and ash. The herb addition did not cause differences in the digestibility of most nutrients compared to the antibiotic, but fiber digestibility of the herb at 6.0% and 9.0% was lower (p<0.05) than that of the antibiotic. Moreover, the antibiotic and the herb also similarly increased (p<0.05) the activities of duodenal α-amylase, maltase, lipase, and trypsin, compared to the control, and the herb at 6.0% and 9.0% showed a greater (p<0.05) activity of elastase than the dose 3.0%. Conclusion: The obtained data indicate that S.m. aerial parts can be a potential forage in rabbit's diet at 9.0% with a beneficial regulation on nutrition and digestion.
