The Anti-tumor Effect of Soonkiwhajungtang with Doxorubicin in MKN-45 Conclusion (순기화중탕과 Doxorubicin의 병용이 MKN-45의 항암효과에 미치는 영향)

  • Objectives : To evaluate the anti-tumor and synergic effect of Soonkiwhajungtang with doxorubicin. Methods : The inhibitory concentration (IC), $IC_{50}$ and $IC_{90}$ of single use of doxorubicin and Soonkiwhajungtang with their concomitant treatment against MKN-45 (human stomach carcinoma) cell line were observed using MTT (microculture tetrazolium test) assay. In addition, their anti-tumor effects were also observed in xenograft nude mice models against the MKN-45 cell line. Results : Soonkiwhajungtang has only minimal direct anti-tumor effect against MKN-45 cell line but it reduced general depressed signs induced by implantation of the tumor cell lines and increased the total WBC and lymphocyte numbers. Conclusions : It is considered or expected that Soonkiwhajungtang extract reduces the critical toxicity of doxorubicin and has favorable synergic anti-tumor effect when administered concomitantly with doxorubicin.

  • Objectives : To evaluate the anti-tumor and synergic effect of Bojungikkeehapdaechilki-tang (BJDC) with doxorubicin. Membrane-bound p35 Subunit of IL-12 on Tumor Cells is Functionally Equivalent to Membrane-bound Heterodimeric Single Chain IL-12 for Induction of Anti-tumor Immunity

  • In this study, we compared two different tumor cell vaccines for their induction of anti-tumor immunity; one was a tumor cell clone expressing a membrane-bound form of IL-12 p35 subunit (mbIL-12 p35 tumor clone), and the other was a tumor clone expressing heterodimeric IL-12 as a single chain (mb-scIL-12 tumor clone). The stimulatory effect of mb-scIL-12 on the proliferation of ConA-activated splenocytes was higher than that of mbIL-12 p35 in vitro. However, the stimulatory effect of mbIL-12 p35 was equivalent to that of recombinant soluble IL-12 (3 ng/ml). Interestingly, both tumor clones (mbIL-12 p35 and mb-scIL-12) showed similar tumorigenicity and induction of systemic anti-tumor immunity in vivo, suggesting that tumor cell expression of the membrane-bound p35 subunit is sufficient to induce anti-tumor immunity in our tumor vaccine model.

Anti-tumor Effect of Carrot(Docus carota L.) Extracts in the Human Lung Cancer Cell Line NCI-H1299 (인체 페암세포주 NCI-H1299에 대한 당근 추출물의 항암효과)

  • This study was designed to investigate the anti-tumor effects of fresh carrot juice, methanol-extracts, and $\beta$-carotene on the human lung cancer cell line NCI-H1299. The anti-tumor effect was evaluated by the MTT assay in vitro. The anti-tumor effect of fresh carrot juice against NCI-H1299 lasted up to 96 hours after exposure; the viability rate of lung cancer cells decreased below 50% after 48 hours, and further after 72 hours. The strongest propagation inhibition effect of fresh carrot juice was shown at the concentration of 2000 $\mu\textrm{g}$/$m\ell$ after 72 hours and the viability rates was 45.98% even at the concentration of 25 $\mu\textrm{g}$/$m\ell$. The value of $IC_{50}$/ was 23.1$\mu\textrm{g}$/$m\ell$ when the elapsed time was 72 hours. The viability rate of methanol-extract was 52.4% under the concentration of 2000 $\mu\textrm{g}$/$m\ell$ and the elapsed time of 72 hours. Under the concentration of 1000 $\mu\textrm{g}$/$m\ell$ and the elapsed time of 48 hours, $\beta$ -carotene decreased the viability rate to 29.99%. The $IC_{50}$/ value of $\beta$-carotene was 691.2$\mu\textrm{g}$/$m\ell$ after 72 hours. According to the above results, the anti-tumor effect arose in NCI-H1299 when the concentration of the fresh carrot juice or the $\beta$-carotene was more than 25 $\mu\textrm{g}$/$m\ell$ or 1000 $\mu\textrm{g}$/$m\ell$, respectively. On the other hand, the methanol-extracts showed a weak anti-tumor effect even at a concentration as high as 2000 $\mu\textrm{g}$/$m\ell$.

  • Gekko has been used for several diseases including cancer in Oriental medicine and fork herbalogy. Nevertheless, its origin as herbal medicine and its efficacy and mechanism as anti-tumor drug have not yet been thoroughly reported in Korea. This study aimed to investigate anti-tumor effect of Gekko through selected articles from cqvip database in China. In vitro and In vivo, Gekko could obviously inhibit tumor growth, induce tumor cells apoptosis, reduce micro-vessel density in tumor tissue through down regulating VEGF & bFGF protein expression, promote cytotoxicity of lymphocyte. Gekko could improve survival rate, relive clinical symptoms, improve quality of life, and relieve anti-tumor treatment reaction, suggesting that Gekko might be a effective anti-tumor drug.

Dendritic Cell (DC) Vaccine in Mouse Lung Cancer Minimal Residual Model: Comparison of Monocyte-derived DC vs. Hematopoietic Stem Cell Derived-DC

  • The anti-tumor effect of monocyte-derived DC (MoDC) vaccine was studied in lung cancer model with feasible but weak Ag-specific immune response and incomplete blocking of tumor growth. To overcome this limitation, the hematopoietic stem cell-derived DC (SDC) was cultured and the anti-tumor effect of MoDC & SDC was compared in mouse lung cancer minimal residual model (MRD). Therapeutic DCs were cultured from either $CD34^+$ hematopoietic stem cells with GM-CSF, SCF and IL-4 for 14 days (SDC) or monocytes with GM-CSF and IL-4 for 7 days (MoDC). DCs were injected twice by one week interval into the peritoneum of mice that are inoculated with Lewis Lung Carcinoma cells (LLC) one day before the DC injection. Anti-tumor responses and the immune modulation were observed 3 weeks after the final DC injection. CD11c expression, IL-12 and TGF-${\beta}$ secretion were higher in SDC but CCR7 expression, IFN-${\gamma}$ and IL-10 secretion were higher in MoDC. The proportion of $CD11c^+CD8a^+$ cells was similar in both DC cultures. Although both DC reduced the tumor burden, histological anti-tumor effect and the frequencies of IFN-${\gamma}$ secreting $CD8^+$ T cells were higher in SDC treated group than in MoDC. Conclusively, although both MoDC and SDC can induce the anti-tumor immunity, SDC may be better module as anti-tumor vaccine than MoDC in mouse lung cancer.

The Anti-tumor Activity of Vitamin C via the Increase of Fas (CD95) and MHC I expression on Human Stomach Cancer Cell Line, SNU1

  • It is already known that high concentration of vitamin C induces apoptosis on tumor cells. However, there is no report regarding the function of vitamin C on the modulation of immune susceptibility of cancer. Therefore, we investigated whether vitamin C can modulate immune susceptibility of tumor cells, especially on the induction of Fas-mediated apoptosis. First, the optimal concentration of vitamin C, which cannot induce damages on tumor cells for 36 hrs. We found that 2 mM of vitamin C did not show harmful effect. In addition, the optimal concentration of agonistic anti-Fas Abs for 18 hrs was examined. As a result, 400 ng/ml of agonistic anti-Fas Abs did not induce apoptosis on tumor cells. Next, we tried to find the effect of 2 mM of vitamin C on the modulation of the susceptibility to agonistic anti-Fas Abs. When tumor cells were cultured with 400 ng/ml of agonistic anti-Fas Abs for 18 hrs, after pre-treatment with 2 mM of vitamin C for 24 hrs, viability of cells was decreased. Interestingly, we found that the expression of Fas (CD95) and MHC class I was increased by the treatment of vitamin C. Taken together, vitamin C increases the susceptibility of tumor cells to anti-Fas Abs and the expression of Fas (CD95) and MHC class I on tumor cells.

Comparison Study of the Anti-tumor Effects of Hangamjedoktang(Kangaizhidu-tang) with Holotrkhia and Hangamyagjaebang(Kangaiyaocai-fang) (항암제독탕가제조와 항암양제방에 대한 비교연구)

  • Objective : Though modern medicine has made various studies in cancer treatment, the results of the treatments are not satisfactory. Considering this, Oriental medicine can be a breakthrough in treatment of cancer, and therefore, its constant research eagerly needed. According to preceding studies, Hangamjedoktang (Kangaizhidu-tang) with Hagocho appeared to be statistically significant against cancer, and therefore to seek a better medication for cancer, Holotrichia which seemed to be effective against cancer was added to the formula, and herbs which showed an anti-tumor effect in preceding studies composed Hangamyagjaebang. The efficacy of both Hangamjedoktang with Holotrichia (HJJ) and Hangamyagjaebang (Kangaiyaocaijang) (HM) was compared. Methods : To examine the anti-cancer effect of HJJ and HM, inhibitory effect on solid tumor growth in mice induced by Sarcoma-180 (s-180), change of body and organ weight in tumor bearing mice and the activity of machrophages and lymphocytes in the spleen were examined. Results : 1. In the HJJ and HM treated groups, tumor growth was markedly decreased. 2. HJJ and HM increased the activity of ALP which is produced from the splenocytes transplanted with S-180. 3. HJJ and HM increased the ACP activity of the macrophages of the mice transplanted with S-180. Conclusion : These results suggest that HJJ and HM are good candidates for new drugs for cancer therapy.

Combined Treatment of Herbal Mixture Extract H9 with Trastuzumab Enhances Anti-tumor Growth Effect

  • Extracts from Asian medicinal herbs are known to be successful therapeutic agents against cancer. In this study, the effects of three types of herbal extracts on anti-tumor growth were examined. Among the three types of herbal extracts, H9 showed stronger anti-tumor growth effects than H5 and H11 in vivo. To find the molecular mechanism by which H9 inhibited the proliferation of breast cancer cell lines, the levels of apoptotic markers were examined. Proapoptotic markers, including cleaved PARP and cleaved caspases 3 and 9, were increased, whereas the anti-apoptotic marker Bcl-2 was decreased by H9 treatment. Next, the combined effect of H9 with the chemotherapeutic drugs doxorubicin/cyclophosphamide (AC) on tumor growth was examined using 4T1-tumor-bearing mice. The combined treatment of H9 with AC did not show additive or synergetic anti-tumor growth effects. However, when tumor-bearing mice were co-treated with H9 and the targeted anti-tumor drug trastuzumab, a delay in tumor growth was observed. The combined treatment of H9 and trastuzumab caused an increase of natural killer (NK) cells and a decrease of myeloid-derived suppressor cells (MDSC). Taken together, H9 induces the apoptotic death of tumor cells while increasing anti-tumor immune activity through the enhancement of NK activity and diminishment of MDSC.

Anti-tumor Metastatic Effect and Activation of Innate Immunity by Extract of Mori Radicis Cortex (상백피(桑白皮)의 선천면역 활성화에 의한 항암 효과)

  • Purpose: This study was carried out to investigate the anti-tumor metastasis effect and activation of innate immunity by extracts of Mori radicis cortex. Methods: Anti-tumor metastatic experiment was conducted in vitro and in vivo by using colon 26-M3.1 carcinoma cell, L5178Y-R lymphoma cell and HeLa cell. To observe the activation of innate immunity by extracts of Mori radicis cortex, we estimated IL-6, IL-10, IL-12, TNF-${\alpha}$ from peritoneal macrophages. And we evaluated the activation of NK cell by using anti-asialo-GM1 serum. Results: We found that the administration of Mori radicis cortex extracts significantly inhibited tumor metastasis. In an in vitro cytotoxicity analysis, Mori radicis cortex affected tumor cell growth above specific concentration. Mori radicis cortex also stimulated peritoneal macrophage, which was followed by the production of various cytokines such as IL-6, IL-10, IL-12, TNF-${\alpha}$. The depletion of NK cells by anti-asialo GM1 serum partly abolished the inhibitory effect of Mori radicis cortex on tumor metastasis. Conclusion: Mori radicis cortex appears to have considerable activity on the anti-metastasis by activation of innate immunity.
