Antioxidative activities, total phenolic compounds and the sulforaphane contents of cooked broccoli extracts were studied. Total phenolic compounds were determined to be 0.96 mg/g(flower) and 0.76 mg/g(stem) in fresh broccoli extracts. The total phenolic compounds of cooked broccoli extracts showed 0.92 (1 min), 0.79 (3 min), 0.67 (10 min) mg/g when a boiling process was used and 1.27 (1 min) mg/g when a steaming process was used. In the DPPH assay, the steam process showed the highest free radical scavenging capacities. Sulforaphane has been of increasing interest in the nutraceutical and phamaceutical industries due to its anti-cancer effect. Sulforaphane was isolated from fresh and boiled, steamed broccoli using dichloromethane as an extract solvent. The sulforaphane contents of fresh broccoli were higher in the flower (14.78 mg/kg) than in the stem (6.16 mg/kg). The sulforaphane content dramatically decreased after the boiling ($100{\pm}2^{\circ}C$) or steaming ($100{\pm}2^{\circ}C$) processes were used.
Cetrizine dihydrochloride (CTZH) is widely used as an anti-allergic drug. Sodium N-bromo-p-toluenesulfonamide or bromamine-T (BAT) is the bromine analogue of chloramine-T (CAT) and is found to be a better oxidizing agent than CAT. In the present research, the kinetics of oxidation of CTZH with BAT in acid and alkaline media was studied at 313 K. The experimental rate laws obtained are: -d[BAT]/dt=$k[BAT][CTZH]^{0.80}[H^+]^{-0.48}$ in acid medium and -d[BAT]/dt=$k[BAT][CTZH]^{0.48}[OH^-]^{0.52}[PTS]^{-0.40}$ in alkaline medium where PTS is p-toluenesulfonamide. Activation parameters and reaction constants were evaluated. The solvent isotope effect was studied using $D_2O$. The dielectric effect is positive. The stoichiometry of the reaction was found to be 1:1 and the oxidation products were identified as 4-chlorobenzophenone and (2-piperazin-1-yl-ethoxy)-acetic acid in both media. The rate of oxidation of CTZH is faster in acid medium. Suitable mechanisms and related rate laws have been worked out.
The present study aimed to prepare a novel efficient flame retardant additive for polypropylene. The new flame retardant was prepared by chemical grafting of melamine to graphene oxide with the aid of thionyl chloride. Fourier-transform infrared spectroscopy and thermogravimetric analysis proved that melamine had been successfully grafted to the graphene oxide. The modified graphene oxide was incorporated into polypropylene via solution mixing followed by anti-solvent precipitatio. Homogeneous distribution as well as exfoliation of the nanoplatelets in the polymer matrix was observed using transmission electron microscopy. Thermogravimetric analysis showed a significant improvement in the thermo-oxidative stability of the polymer after incorporating 2 wt% of the modified graphene oxide. The modified graphene oxide also enhanced the limiting oxygen index of the polymer. However, the amount of improvement was not enough for the polymer to be ranked as a self-extinguishing material. Cone calorimetry showed that incorporating 2 wt% of the modified graphene oxide lowered total heat release and the average production rate of carbon monoxide during burning of the polymer by as much as 40 and 35%, respectively. Hence, it was concluded that the new flame retardant can retard burning of the polymer efficiently and profoundly reduce suffocation risk of exposure to burning polymer byproducts.
Twenty-four different natural food materials extracted with 80% methanol were used to investigate the inhibition of cell-mediated immediate type allergic reactions induced by compound 48/80 in rat peritoneal mast cells (RPMCs). Nine 80% methanol extracts screened at a concentration of $10\;{\mu}g/mL$ inhibited histamine release from RPMCs induced by compound 48/80. Of these, two materials (Ulmus parvifolia and Liriope platyphylla Wang et Tang) were extracted and fractionated into four different solvent types (chloroform, ethylacetate, butanol, and water), and the fractions with major anti-allergic effects were assessed. The chloroform fraction of U. parvifolia (UP) at $5\;{\mu}g/mL$ and the ethylacetate fraction of L. platyphylla Wang et Tang (LPWT) at $1\;{\mu}g/mL$ showed the greatest inhibition of histamine release induced by compound 48/80. The chloroform fraction of UP and the ethylacetate fraction of LPWT in combination showed a greater inhibition of histamine release than either fraction alone. The cAMP levels in RPMCs treated with UP and LPWT were significantly greater than in cells treated with compound 48/80 alone. Our studies suggest that extracts from UP and LPWT may alleviate immediate type hypersensitivity reactions through the increase of cAMP levels in the mast cells.
Previously, the methanolic extract of Paeonia lactiflora seeds was shown to have strong ingibitory activity against soybean liposygenase (SLO). Four phenolic compounds were isolated from the seeds by solvent fractionation Sephadex LJ-20 column chromatography and preparative HPLC, and three of them showed strong SLO inhibitio and were characterized as trans-resveratrol, $\varepsilon$-viniferin and luteolin by UV, IR, 1H-NMR, 13C-NMR and MS spectrometry. trans-Resveratrol (IC50=1.02$\mu$M), $\varepsilon$-viniferin (IC50=0.81$\mu$M) and luteolin (IC50=10.01$\mu$M), first found in the above seeds, exhibited a potent SLO inhibitory activity although their activity was lower than that of a well-known lipoxygenase inhibitor, nordihydroguaiaretic acid (NDGA) (IC50=0.57$\mu$M). These results suggest that Paenia lactiflora seeds, now an unused plant seed, may be developed into useful sources of anti-inflammatory drugs.
𝛽-Patchoulene (𝛽-PAE) is a tricyclic sesquiterpene which performed many potential bioactivities and can be found in patchouli oil but in very low concentration. This study aimed to obtained 𝛽-PAE in high concentration by conversion of patchouli alcohol (PA) in patchouli oil under acid catalyzed reaction. Patchouli oil was fractinated by vacuum distillation at 96 kPa to get the fraction with the highest PA content. H2SO4 and ZnCl2 were used respectively as homogeneous and heterogeneous acid catalysts in the conversion reaction of the selected fraction. Patchouli oil, the fractions and the products were analysed by using GC-MS and FTIR instruments. Moreover, the interaction of 𝛽-PAE to COX-2 protein was studied to understand the antiinflammation activity of 𝛽-PAE. The results showed that patchouli oil contains 25.3% of PA. The selected fraction which has the highest PA content (70.3%) was distilled at 151 - 152 ℃. The application of ZnCl2 catalyst in conversion reaction did not succeed. In contrast, H2SO4 as a catalyst in acetic acid solvent succeeded in converting the overall fraction of PA to 𝛽-PAE. Furthermore, the molecular docking study of 𝛽-PAE against COX-2 enzyme showed van der Waals and alkyl-alkyl stacking interactions on ten amino acid residues.

Lactic acid bacteria are well-known probiotics, conferring several health benefits. In this study, we isolated lactobacilli from human breast milk and identified Lactobacillus reuteri LM1071 (RR-LM1071) using 16S rDNA sequencing. We tested the hemolytic activity, biogenic amine production, and antibiotic susceptibility of this strain to assess its safety. RR-LM1071 was found to be negative for hemolytic activity and biogenic amine production, as well as was measured in susceptible level for antibiotics in the minimal inhibitory concentration (MIC) test. The adhesive properties of RR-LM1071 were higher than those of LGG in HT-29 cells, and showed a greater hydrophobicity than LGG in hexadecane solvent. Under inflammatory conditions, RR-LM1071 suppressed the mRNA expression of IL-6, TNF-α, and IL-4 produced in IL-1β-induced HT-29 cells. Our results suggest that RR-LM1071 is a safe and valuable probiotic that can be used for the treatment of inflammatory bowel disease.

Single crystal of organic lead halide ($CH_3NH_3PbX_3$; $CH_3NH^+_3$ = methylammonium (MA), $X=Cl^-$, $Br^-$, $I^-$) is the best candidate for material intrinsic property studies due to no grain boundary and high crystal quality than the film having a lot of grain boundary and surface defects. The representative crystallization methods are inverse temperature crystallization (ITC) and anti-solvent vapor assisted crystallization (AVC). Herein, we report bandgap modulated organic lead halide single crystals having a bandgap ranging from ~ 2.1 eV to ~ 3 eV with ITC and AVC methods. The bandgap modulation was achieved by controlling the solvents and chloride-to-bromide ratio. Structural, optical and compositional properties of prepared crystals were characterized. The results show that the crystals synthesized by the two crystallization methods have similar properties, but the halide ratios in the crystals synthesized by the AVC method are controlled more quantitatively than the crystals synthesized by ITC.
Objectives : This study was performed to investigate the antioxidant activities of isolated compounds from the shoot of Aralia elata. Methods : The methanol extract from the shoot of Aralia elata was fractionated into ethyl acetate, n-BuOH and $H_2O$ layers through solvent fractionation. Repeated silica gel, ODS column chromatography of n-BuOH layer afforded four flavonol glycosides. Their antioxidant activity was determined by measuring free radical scavenging activity by DPPH, ABTS and superoxide dismutase (SOD) like activity assay. Results : They were identified as quercetin 3,7-di-O-$\alpha$-rhamnopyranoside (1), quercetin 3-O-$\beta$-D-galactoside-7-O-$\alpha$-L-rhamnoside (2), kaempferol 3-O-$\beta$-glucosyl($1{\rightarrow}2$)-$\alpha$-rhamnoside-7-O-$\alpha$-rhamnoside (3) and quercetin 3-O-$\beta$-glucosyl($1{\rightarrow}2$)-$\alpha$-rhamnoside-7-O-$\beta$-rhamnoside (4) on the basis of spectroscopic data. The result showed that 1 is the most active compound in the DPPH and ABTS radical scavenging test. Conclusions : Isolated Compounds from the shoot of Aralia elata showed anti-oxidative effect.
To investigate the difference and change of ingredient and efficacy of Galgeun-tang (GGT) according to extraction solvent, water and 70% EtOH, the quantities of index components and the several in vitro activities of two kinds of GGT extract were compared. The contents of extracts were analyzed with HPLC. The biological activities such as anti-inflammatory and anti-obesity effects were measured through cell line-based in vitro assay. The cytotoxicity was measured by CCK-8 assay in RAW 264.7 cell, BEAS-2B cell, HaCaT cell and 3T3-L1 cell. We compared effects of two kinds of extract by measuring NO, PGE2, IL-6 and TNF-${\alpha}$ in RAW264.7 cell, RANTES in BEAS-2B cell, MDC and RANTES in HaCaT cell and GPDH activity and leptin level in differentiated 3T3-L1. 70% EtOH extract of GGT contained more compositions than water extract. The inhibitory effect of water extract of GGT on NO in RAW 264.7 cell and GPDH activity in 3T3-L1 was stronger than that of 70% EtOH. 70% EtOH has a stronger inhibitory effect on PGE2 in RAW264.7 cell, RANTES in BEAS-2B cell, MDC, RANTES in HaCaT cell, leptin in 3T3-L1 cell than water extraction. These results suggest that the ingredient and efficacy from 70% EtOH extract of GGT are more effective than water extract.
