Anti-microbial Effect on Streptococcus mutans and Anti-oxidant Effect of the Butanol Fractions of Sanguisorbae Radix (지유(地楡) 부탄올 분획물의 Streptococcus mutans 항균활성 및 항산화 효과)

  • Objective : Sanguisorbae Radix(SO). that belong to Rosaceae is widely distributed in Asia including Korea, Japan and China. It has been used as traditional medicine from old times as a treatment for anti-inflammatory drugs. This study was designed to investigate the effects of n-BuOH fractions of SO on anti-oxidant effect and anti-microbial effect on $Streptococcus$ $mutans$ ($S.$ $mutans$). Methods : The anti-microbial effect of $n$-BuOH fractions of SO. was assessed by the paper disk diffusion method and anti-oxidant effect was assessed by the DPPH radical scavenging effect, Superoxide anion radical scavenging effect and SOD like ability. Results : DPPH radical scavenging of the $n$-BuOH fraction from SO in 50 ug/mL was shown to be Fr-2 (89.9%), Fr-3 (88.7%), Fr-4 (76.3%), Fr-1 (59.4%), Fr-5 (56.2%). Superoxside anion radical scavenging activity of the n-BuOH fraction from SO. in 50 ug/mL for Fr-3 was shown to be 78%. SOD-like activity of n-BuOH fraction from the SO in 1000 ug/mL for Fr-3 were shown to be 76.1% respectively. The $n$-BuOH fraction from the SO had high anti-microbial effect on $S.$ $mutans$. Conclusion : As a result, the $n$-BuOH fraction from SO. has good anti-microbial and anti-oxidant effects in a concentration-dependent manner.

  • Moringa oleifera Lam. or munga is one of the most important plant widely cultivated in India. It belongs to family Moringaceae. This plant is widely used as nutritional herb and contains valuable pharmacological action like anti-asthmatic, anti-diabetic, hepatoprotective, anti-inflammatory, anti-fertility, anti-cancer, anti-microbial, anti-oxidant, cardiovascular, anti-ulcer, CNS activity, anti-allergic, wound healing, analgesic, and antipyretic activity, Moringa oleifera Lam. The plant is also known as Horse - radish tree, Drumstick tree. Every part of this plant contains a valuable medicinal feature. It contain rich source of the vitamin A, vitamin C and milk protein. Different types of active phytoconstituents like alkaloids, protein, quinine, saponins, flavonoids, tannin, steroids, glycosides, fixed oil and fats are present. This plant is also found in the tropical regions. Some other constituents are niazinin A, niazinin B and niazimicin A, niaziminin B. The present review discusses the phytochemical composition, medicinal uses & pharmacological activity of this plant.

The Functional Effects of Anti-microbial Activity and Anti-inflammatory of Seaweed polysaccharide Extracts (해조 다당류 추출물의 항균성 및 항염증 기능성평가)

  • This study aimed to evaluate seaweed polysaccharide extracts as a cosmetic material. To assess anti-microbial efficacy, Staphylococcus aureus (S. aureus) was treated with seaweed polysaccharide extracts and zones of inhibition were measured. In addition, the anti-inflammatory effect was confirmed in RAW 264.7cells, and seaweed polysaccharide extracts was applied to the dorsal skin of Sprague-Dawley (SD) rats to evaluate single-dose toxicity. As a results, seaweed polysaccharide extracts did not exhibit cytotoxicity at concentrations up to $1,000{\mu}g/mL$ in skin fibroblasts. Furthermore, when S. aureus was treated with 1% seaweed polysaccharide extracts, clear zones of $1.52{\pm}0.34cm$ formed, confirming sufficient anti-microbial activity. When RAW 264.7 cells were treated with seaweed polysaccharide extracts extract, nitric oxide (NO) production decreased in a concentration-dependent manner and the production of inflammation-related cytokines, such as interleukin 1 beta ($IL1{\beta}$), tumor necrosis factor alpha ($TNF{\alpha}$), and prostaglandin E2(PGE2), decreased. When seaweed polysaccharide extracts extract was applied at various concentrations to rats, symptoms did not change for more than 14 d, and there was no change in body or organ weights. In conclusion we found that seaweed polysaccharide extracts is not cytotoxic and has anti-microbial and anti-inflammatory effects. Therefore, it is suitable for use as a cosmetic material.

A Study on the Anti-microbacterial Activity, Anti-inflammatory and Anti-allergic Effects of Samhwangseje gagambang(SHB) (삼황세제가감방(三黃洗劑加減方)의 항균, 항염 및 항알레르기 효과에 대한 실험적 연구)

  • Herbal mixture water extract of (Phellodendron amurense, Scuellaria baiklensis, Spphora flavescens, Lithospermum erythrorhizon, Mellaphis chinesis, Alumite, Zanthoxylum schinifolium, Glycyrrhiza uralensis), which exhibit several beneficial effects including acne and skin diseases, was tested for anti-microbial activity and anti-inflammation effects. The herbal mixture extract showed antimicrobial activity against Stapylococcus epidermis, Propionibacterium acne, and Malassezia furfur. The growth of Stapylococcus epidermis, Propionibacterium acne, and Malassezia furfur was inhibited allergy and LPS induced cyclooxygenase-2(COX-2)gene expression in RAW24.7macrophage. The results indicated th ear swelling and histamine release induced by compound 48/80 were dose-dependently reduced, ranging 11-38% and 11-56%, respectively. Furthermore the extract inhibited the expression of LPS-induced COX-2 proteins and mRNAs without an appreciable cytotoxic effects on RAW264.7 cells. The cytotoxicity of the extract using M7T assay showed the cytotoxicity of 7 and 18% against L929 cell line. Based on these results, it is concluded that the herbal mixture water extract can be applied to the acne and skin diseases therapy.

  • Herbal mixture water extract of (Rheum coreanum Scutellaria baikalensis, Phellodendron amurese), which exhibit several beneficial effects including acne and skin diseases, was tested for anti-microbial activity and anti-inflammation effects. The herbal mixture extract showed antimicrobial activity against Stapylococcus epidermis and Propionbacterium acne. The growth of Stapylococcus epidermis and Propionibacterium acne was inhibited completely by addition of 1.0% of the extract. Also in the present study we examined the mixture extract on compound 48/80 induced allergy and LPS induced cyclooxygenase-2(COX-2) gene expression in RAW264.7 macrophage. The results indicated the ear swelling and histamine release induced by compound 48/80 were dose-dependently reduced, ranging 18-36% and 10-61%, respectively. Furthermore the extract inhibited the expression of LPS-induced COX-2 proteins and mRNAs without an appreciable cytotoxic effects on RAW264.1 cells. The cytotoxicity of the extract using MTT assay showed the cytotoxicity of 6% and 13% against L929 cell line. Based on these results, it is concluded that the herbal mixture water extract can be applied to this acne and skin diseases therapy.

  • This study evaluated the possibility of Hoangtonogak Plus extracts as a bioactive ingredients for cosmetic products. Methanol(MN) and hot-water(WN) extracts were analysed by DPPH/ABTS radical scavenging activity, FRAP value for anti-oxidant activity, MTT assay for cell viability, inhibition of NO production and iNOS protein expression for anti-inflammatory effect, paper disc diffusion method for anti-microbial activity against Staphylococcus aureus, Staphylococcus epidermidis and Escherichia coli.. The contents of total polyphenol of MN and WN extracts were 2.92±0.01 mgGAE/g and 1.67±0.02 mgGAE/g, respectively. DPPH, ABTS and FRAP values of MN extracts were higher than WN at each concentration. No significant cytotoxicity was observed in RAW264.7 cells. Furthermore, NO production of MN and WN at 1 mg/mL concentration was measured as 11.69 μM, 20.4 μM, respectively. In addition, MN extracts showed anti-microbial effect only on S. epidermidis. Also MN extracts suppressed iNOS protein level in a concentration-dependent manner. According to our results, the MN extracts demonstrated its potential as a natural source of antioxidant with anti-microbial and anti-inflammatory properties.

Effects of Egg Shell Membrane Hydrolysates on Anti-Inflammatory, Anti-Wrinkle, Anti-Microbial Activity and Moisture-Protection

  • This study was conducted to examine the effects of eggshell membrane hydrolysates (ESMH) on the anti-inflammatory, anti-wrinkle, anti-microbial activity, and moisture-protection for cosmetic use. Whole ESMH (before fractionation), and fraction I (>10 kDa), fraction II (3-10 kDa), and fraction III (<3 kDa) of the hydrolysates were assessed in this experiment. As lipopolysaccharide (LPS) and IFN-${\gamma}$ caused the inflammation on Raw264.7 cell, whole ESMH and fraction I showed to be effective in inhibiting the induction of cell inflammation depending on the concentration, and also showed outstanding effect to suppress the skin inflammation. Fraction I inhibited collagenase and elastase activities to a greater extent than the other fractions, while all fractions had antibiotic effects at concentrations of 10 mg/disc and 20 mg/disc. In addition, it showed the moisture protection effects of skin on the holding amount and losing amount of moisture in upper-inner arm of the human body with a relatively low loss rate in skin, which confirmed that the hydrolyzed fractions of ESM helps to form the superior protective layer of moisture. It was concluded that ESMH fractions with different molecular weights, especially the 10 kDa fraction, have anti-lipopolysaccharide, anti-IFN-${\gamma}$-induced inflammation, anti-collagenase and elastase activities, and thus can be used as a cosmetic agent to protect skin.

Beauvericin, a cyclic peptide, inhibits inflammatory responses in macrophages by inhibiting the NF-κB pathway

  • Beauvericin (BEA), a cyclic hexadepsipeptide produced by the fungus Beauveria bassiana, is known to have anti-cancer, anti-inflammatory, and anti-microbial actions. However, how BEA suppresses macrophage-induced inflammatory responses has not been fully elucidated. In this study, we explored the anti-inflammatory properties of BEA and the underlying molecular mechanisms using lipopolysaccharide (LPS)-treated macrophage-like RAW264.7 cells. Levels of nitric oxide (NO), mRNA levels of transcription factors and the inflammatory genes inducible NO synthase (iNOS) and interleukin (IL)-1, and protein levels of activated intracellular signaling molecules were determined by Griess assay, semi-quantitative reverse transcriptase-polymerase chain reaction (RT-PCR), luciferase reporter gene assay, and immunoblotting analysis. BEA dose-dependently blocked the production of NO in LPS-treated RAW264.7 cells without inducing cell cytotoxicity. BEA also prevented LPS-triggered morphological changes. This compound significantly inhibited nuclear translocation of the $NF-{\kappa}B$ subunits p65 and p50. Luciferase reporter gene assays demonstrated that BEA suppresses MyD88-dependent NF-${\kappa}B$ activation. By analyzing upstream signaling events for $NF-{\kappa}B$ activation and overexpressing Src and Syk, these two enzymes were revealed to be targets of BEA. Together, these results suggest that BEA suppresses $NF-{\kappa}B$-dependent inflammatory responses by suppressing both Src and Syk.

Effects of bamboo salt on dental caries prevention (임상가를 위한 특집 2 - 죽염을 이용한 우식예방)

  • Bamboo salt is a special processed salt by Korean traditional recipe. Recent study results showed that bamboo salt or bamboo salt with some other materials like herbal extracts have the anti-microbial activity, inhibition effects of dental plaque and gingival inflammation. Bamboo salt also showed anti-cariogenic effects; remineralization and acid resistance. Compare to fluoride toothpaste, bomboo salt toothpaste with fluoride showed the more effective remineralization on inner part of the early dental caries lesion. It increased the surface hardness and decreased lesion depth of early dental caries lesion. Thus, it is suggested thai bamboo salt could be used as a anti-microbial, anti-plaque, anti-inflammatory and anti-cariogenic material for oral disease prevention. Especially, bamboo salt dentifrice with fluoride can be recommanded as a useful remineralizing agent.

Synthesis and Pharmacological Evaluation of Some Novel 2-Mercapto Benzimidazole Derivatives

  • The present study is synthesis of derivatives of N'-(4-amino-5-sulfanyl-4H-1,2,4-triazole-3-yl)-2-(1H-benzimidazole-2-ylsulfanyl) acetohydrazide (IV). Antibacterial activity tested against the E. coli and A. Substilis. Biological activities conducted by disc diffusion method. Compound $2MB_1$, $2MB_3$, $2MB_5$ inhibit the appreciable microbial growth while rest of the compound possess the moderate activities. Anti-inflammatory activity tested by reduces local edema induced in the rat paw by injection of phlogestic agent. Compound $2MB_1$, $2MB_8$, $2MB_5$, $2MB_3$ and $2MB_6$ exhibit satisfying anti-inflammatory activity while analgesic activity conducted by acetic acid induced writhing effect in mice while compound $2MB_1$, $2MB_4$ and $2MB_7$ having the good analgesic activity. The chemical structures of all newly synthesized compounds were confirmed by their IR, $^1H$ NMR and mass spectral data.
