Anti-Wrinkling Effect of Noni (Morinda citrifolia) by Antioxidant and Anti-Inflammatory Properties

  • Noni has been used for medicinal purposes for more than 2,000 years in South Pacific Polynesia, China and India, and has been heavily ingested as an extract for its excellent antioxidant and anti-inflammatory effects. However, a recent study found that the noni extract causes digestive disorders, kidney problems, and liver diseases, which made it necessary to use it for other purposes than as an extract. In this study, we want to evaluate the potential of noni as an anti-oxidant, anti-inflammatory and anti-wrinkling agent. Methods: Noni was freeze-dried, extracted in water, and concentrated. Skin cells were treated with the noni extract for 24 hrs and then were exposed to UVB (55 mJ/cm2). After 48 hrs of incubation, pro-inflammatory cytokine, elastase, MMP-1 and type-1 procollagen levels were measured by ELISA. Results: To find out the antioxidant effect of the noni extract, the DPPH and ABTS radical scavenging activity experiments were conducted and the noni extract showed 97.0 % and 92.0 % antioxidant efficacy at 200 ㎍/mL respectively. The noni extract (50 and 100 ㎍/mL) decreased IL-6 and TNF-α in RAW 264.7 cells induced by LPS in a concentration-dependent manner. In the RT-PCR experiment involving NO production, the noni extract (50 and 100 ㎍/mL) inhibited NO production by strongly inhibiting iNOS mRNA expression, and also inhibited the elevation of MMP-1 and elastases caused by UVB irradiation by 25.0 % and 7.0 % respectively. In addition, type-1 procollagen was elevated by 20.0 % by the noni extract treatment in HaCaT cells. Conclusion: The noni extract has photoprotective ability by reducing proinflammatory mediators, elastase and MMP-1 production, and elevation of collagen synthesis. Our findings suggest that the noni extract might be a good natural substance to protect against UVB-induced premature skin aging.

Anti-arthritic Effects of Oplopanax elatus in a Rat Model of Rheumatoid Arthritis (Adjuvant-induced Arthritis)

  • The stems of Oplopanax elatus (OE) have long been used to treat inflammatory disorders in herbal medicine, and in the previous investigation, OE was found to possess anti-inflammatory activity in lipopolysaccharide-treated macrophages, RAW 264.7 cell. OE reduces inducible nitric oxide (NO) synthase-induced NO production, and interferes with mitogen-activated protein kinase activation pathways. In the present study, the pharmacological action of the water extract of OE was examined to establish anti-arthritic action, using a rat model of adjuvant-induced arthritis (AIA). The water extract of OE administered orally inhibited AIA-induced arthritis at (100 - 300) mg/kg/day. The paw edema was significantly decreased, in combination with reduced production of pro-inflammatory cytokines. The action mechanism includes an inhibition of MAPKs/nuclear transcription factor-κB activation. These new findings strongly suggest that OE possesses anti-arthritic action, and may be used as a therapeutic agent in inflammation-related disorders, particularly in arthritic condition.

Anti-inflammatory effects of porcine placenta in forced swimming tested fatigue mice and RAW264.7 cells

  • Inflammation has been linked to various diseases. Especially, fatigue is a frequent symptom in several inflammatory disorders. Therefore, blocking inflammatory process is effective in fatigue. We investigated whether Denmark porcine placenta (DPP) alleviates fatigue by inhibiting inflammatory reaction using forced swimming test (FST) animal model and RAW264.7 cells. In FST-induced fatigue animal model, the mice which received the DPP for 21 days showed decreases of interleukin $(IL)-1{\beta}$ and IL-6 serum levels. Furthermore, our data revealed that lipopolysaccharide (LPS)-induced $IL-1{\beta}$, IL-6, and tumor necrosis $factor-{\alpha}$ secretion were markedly inhibited by DPP in RAW264.7 cells without inducing cytotoxicity. LPS-enhanced nitric oxide secretion and inducible nitric oxide synthase expression were inhibited by DPP. The present study also figured out that these effects of DPP were mediated by blockade of caspase-1 and nuclear $factor-{\kappa}B$ activation. Taken together, our results indicated that DPP could be alleviating fatigue as candidate of anti-inflammatory agent.

Caspase-11 Promoter-GFP Construct as a Dual Reporter of Cytotoxicity and Inflammation

  • Caspase-11 has been known as a dual regulator of apoptosis and inflammatory response. An unusual feature of caspase-11 is that its expression is induced by apoptotic or proinflammatory stimuli. Utilizing these unusual features of caspase-11, we have developed a simple and sensitive assay method to screen pro- or anti-apoptotic/inflammatory molecules. To develop this assay method, we generated a reporter construct where GFP expression is regulated by caspase-11 promoter. When several types of cultured cells were transfected with this reporter construct and subsequently treated with various apoptotic or proinflammatory molecules, expression of GFP by the activation of caspase-11 promoter was easily detected by fluorescence microscopy or spectrofluorometry. In addition, a reduction of the GFP fluorescence was detected when an agent reported to suppress caspase-11 induction was applied. These results suggest that our reporter system can be used to screen pro- or anti-apoptotic/inflammatory molecules.

해수산 클로렐라(Chlorella ellipsoidea) 유기용매 추출물의 항염증 효과 (Anti-Inflammatory Effect of Chlorella ellipsoidea Extracted from Seawater by Organic Solvents)

  • Chlorella has been reported to have certain beneficial physiological effects, including hypocholesterolemic, antihypertensive, antioxidative, and anti-tumor activities in animal and human studies. The aim of this study was to determine the anti-inflammatory activities of an 80% methanol extract(CE-Met), hexane fraction (CE-Hex), and ethyl acetate fraction (CE-EA) of Chlorella ellipsoidea in lipopolysaccharide (LPS)-stimulated RAW 264.7 murine macrophage. Treatment with various concentrations of the C. ellipsoidea extract resulted m a significant, dose-dependent reduction in nitric oxide (NO) production by LPS-induced macrophages. The C. ellipsoidea extract significantly inhibited LPS-induced NO production accompanied by an attenuation of IL-6 and TNF-$\alpha$ formation in macrophages. These results suggest potent inhibit이y effects on the production of inflammatory mediators by a C. ellipsoidea extract. Thus, C. ellipsoidea extract may be a potent anti-inflammatory agent for troubled skin.

탱자 추출물에서 항염증 활성을 갖는 쿠마린 유도체의 효능에 관한 연구 (A Study on the Efficacy of the Coumarine Derivatives with Anti-Inflammatory Activity in the Trifoliate Orange Extract)

  • 탱자에는 여러 종류의 monoterpenes, limonoids, flavonoids과 coumarins 등의 구성성분으로 이루어져 있다. 탱자의 구성 성분중에 7-제라닐옥시쿠마린는 7개의 탄소로 구성된 제라닐옥시기의 곁사슬을 가진 물질이다. 그리고 7-제라닐옥시쿠마린은 여러 약리적인 효과들을 나타낸다고 알려져 있다. 본 연구에서는 탱자의 구성성분인 7-제라닐옥시쿠마린과 이들의 다양한 유도체를 합성하였다. 그리고 항염증에 대한 항염증 효능을 알아보기 위하여 염증을 유발하는 일산화질소 억제 cytokine을 측정한 결과는 6-제라닐옥시쿠마린의 성분이 사이토카인인 인터루킨-6가 $1{\mu}M$농도에서 68.9% 를, $10{\mu}M$에서 72.6% 의 저해효과를 나타냈다.

  • 본 연구에서는 왕호장 열매 추출물(FSR)의 항염증 효과를 측정하였다. 왕호장 열매 추출물은 RAW 264.7 macrophage 세포에서 염증 사이토카인($1L-1{\beta}$, IL-6, TNF-${\alpha}$)의 발현을 억제하였으며, 농도 의존적으로 NO 생성을 억제하고, 그 효과는 $100{\mu}g/mL$ 농도에서 51%로 나타났다. 뿐만 아니라, iNOS와 COX-2의 mRNA와 단백질 발현을 모두 억제함을 확인하였다. 따라서 왕호장 열매 추출물은 항염증 효능을 갖는 화장품 소재로서의 개발 가능성이 클 것으로 기대된다.

Anti-inflammatory Activity of Peel fractionation in Potato cv. Seohong

  • Potatoes were first introduced outside the Andes region four centuries ago, and have become an integral part of much of the world's food. Potatoes were first introduced into Europe in the 16th century and Korea in the early 19th century. It is the world's fourth-largest crop, following rice, wheat, and maize. In the nutritional aspects, potatoes contain abundant vitamins and minerals, as well as an assortment of phytochemicals such as carotenoids and natural phenols. Chlorogenic acid constitutes up to 90% of potato natural phenols. Due to the high content of potato functional compounds, it has known that potatoes are effective in the prevention of various human diseases. Recently, potato 'Seohong' was developed by RDA, and it has reported that they have a high-yield and dry matter content. Processing industry of potato generates high amounts of peel as a byproduct. It was reported as a good source of several beneficial functional ingredients including antioxidant effect. This study was conducted to enhance the utilization of the peel of Potato 'Seohong'. The anti-inflammatory effects on solvent fraction was evaluated. The anti-inflammatory activities of Ethylacetate fraction was evaluated for inhibitory activities against lipopolysacchride (LPS) induced nitric oxide (NO) in RAW264.7 cell lines. The fraction inhibitory activity for tests with $IC_{50}$ values showed in the ranges of $50{\mu}g/ml$. This result revealed that n-butanol fraction of 'Seohong's peel is expected to be good candidate for development into source of anti-inflammatory agent.

딜레니아 추출물의 항염증 효능과 세포독성안전성 (Anti-Inflammatory and Cytotoxic Effects of Dillenia Extracts)

  • Dillenia turbinata Finet & Gagnep. has been known to be used for couch cold, Clerodendrum japonicum (Thunb.) Sweet for pimple, Garcinia tinctoria (DC.) Dunn for oedema. and Garcinia cowa Roxb. for detoxification for a long time. To determine whether these plants have anti-inflammatory effects, we performed nitric oxide (NO) assay. Raw264.7 cells were stimulated with LPS and treated with methanol extracts of these plants. Garcinia cowa Roxb. did not relatively show anti-inflammatory effect, compared to Redcharm extract control as previously used. However, Dillenia, Clerodendrum and Garcinia tinctoria demonstrated significantly higher anti-inflammatory effects in NO assay. We then tested whether these extracts have cytotoxic effect with MTT assay in Raw264.7 cells. Dillenia, Clerodendrum, Garcinia tinctoria and Garcinia cowa Roxb. showed almost similar cell survival rate. This implies that all these plants have almost no significant cytotoxic effects. Taken together, these results suggest that Dillenia, Clerodendrum, and Garcinia tinctoria, but not Garcinia cowa Roxb. are a good anti-inflammatory agent to be developed for medical applications.

찔레나무뿌리(Rosa multiflora root)의 항산화 및 항염증효과 (Anti-Oxidant and Antiinflammatory Effects of Rosa multiflora Root)

  • 찔레나무뿌리를 열수, 에탄올, 메탄올, 아세톤에서 추출하여 항산화 및 항염증 효능효과를 확인하였다. 그 결과 아세톤추출물에서 가장 많은 67.05${\pm}$0.56 mg/g의 폴리페놀 함량을 확인하였으며, SOD 유사활성능을 제외한 DPPH, ABTS, superoxide anion 라디컬소거능 확인 결과 우수한 항산화 효과를 확인하였다. 찔레나무뿌리 추출물의 HAase 저해능을 측정한 결과 에탄올, 메탄올, 아세톤 추출물 500 ${\mu}g/ml$ 에서 60% 이상의 높은 HAase 저해 효과를 확인할 수 있었다. NO 생성량을 확인한 결과, RAW 264.7 cell에서 LPS에 의해 유도된 NO 생성을 농도의존적으로 뚜렷하게 감소시키는 것을 확인하였으며, 에탄올 추출물에서 NO 생성억제 효과가 가장 우수한 것을 확인할 수 있었다. 위의 사실에 기초하여 NO 생성저해의 기전을 알아보기 위하여 iNOS의 발현을 분석한 결과, 에탄올추출물 100 ${\mu}g/ml$에서 40%의 iNOS protein 발현 감소를 확인하였으며, iNOS의 발현억제가 NO생성억제와 유사한 경향을 나타냄으로 NO생성억제는 iNOS의 발현저해를 경유한 것임을 확인할 수 있었다. 이러한 결과들은 찔레나무뿌리 추출물이 항산화 및 항염증 연구의 기초 자료로 활용될 것으로 예상된다. 또한 추후 산업적 응용도 가능함으로 기능성화장품의 가능성을 제시하고 있다.
