  • In order to find out the effect of Anti-Histamine of the Sungmagalkuntang and Sungmagalkuntang-plus-samultang I've investigated the effect of the time to death, the peripheral vascular Pemeability, the amount of secretion of Gastric Juice, the total Acidity of Gastric Juice, the contraction of Intestine Tube by Histamine and the effect of the Skin-Reaction by the decrease of DNCB. These results are following ; 1. The effect of Mice's time to death by Histamine was that the injection group of sungmagalkungtang and the injection group of sungmagalkungtang-plus-samultang both have had the effect of the extention of the time to death, which indicated a statistical significance in comparison with comparative group. 2. The effect of the increased vascular permeability by Histamine was that the injection group of sungmagalkuntang and the injection group of sungmagalkuntang-plus-samultang both have had the effect of the restraint of the increased vascular permeability, which indicated a statistical significance in comparison with comparative group. 3. The effect of the the amount of the secretion of Gastric Juice by Histamine stimulus was that the injection group of sungmagalkuntang-plus-samultang only has had the effect of restraint of the increased of the secretion of Gastric Juice, which indicated a statistical significance in comparison with comparative group. 4. The effect of the change of total Acidity of Gastric Juice by Histamine's stimulus was that the injection group of sungmagalkuntang-plus-samultang only has had the effect of the restraint of the increase of total Acidity of Gastric Juice, which indicated a statistical significance in comparison with comparative group. 5. The effect of the contraction of Intestine Tube by Histamine was that sungmagalkuntang and sungmagalkuntang-plus-samultang both have had the effect of the contraction and the restraint. According to the increase of the density, the effect of the restraint was more clear. But the difference of the effects between two substances had little. 6. The effect of the reaction of skin by the decrease of DNCB was that the injection group of sungmagalkuntang and the injection group of sungmagalkuntang-plus-samultang have had the decreased effect of the Skin-Reaction in comparison with comparative group. sungmagalkuntang-plus-samultang aws more effective. In these results shown above, both sungmagalkuntang and sungmagalkuntang-plus-samultang was shown as more effective one.

  • IgE-dependent histamine-releasing factor (HRF) is found extracellularly to regulate the degranulation process of histamine in mast cells and basophils and known to play a predominant role in the pathogenesis of chronic allergic disease. HRF has been also identified in the intracellular region of the cell. Previously, we reported that HRF interacts with the 3rd cytoplasmic domain of the alpha subunit of Na,K ATPase and inhibits Na,K-ATPase activity. The predicated phosphorylation site in HRF by PKC was mapped to one serine residues (S98) by the computer analysis. In this study, we identified that S98 residue of HRF was phosphorylated using anti-HRFpS98 antibody which specifically recognizes the phosphorylated serine residue of HRF and HRFS98A mutant construct. We also performed $^{86}Rb^{+}-uptake$ assay to understand the role of HRF wild-type and HRFS98A mutants on the regulation of Na,K-ATPase activity. Dephosphorylation of HRF at serine 98 residue recovers slightly the inhibitory function of HRF, suggesting that phosphorylated HRF at serine 98 may not suppress the Na,K-hfpase activity.

  • Histamine-releasing factors (HRFs) are soluble mediators that can release histamine and other mediators from basophils and mast cells and their activity can vary, depending on the type of IgE. The activity of HRFs is affected by the presence of IgE, although HRF is thought to bind to a specific receptor other than IgE. Until now, HRF signaling pathway including its receptor remains unclear in spite of numerous studies. Since there had been many reports about reactive oxygen species (ROS) as a signaling molecule rather than as a by-product of metabolism, we investigated the possibility of ROS as an intracellular messenger involved in HRF-mediated histamine degranulation. In RBL-2H3 cells, ROS was generated by HRF using $H_2O_2$-sensitive fluorescence of fluorescent 2', 7'-dichlorofluorescein ($H_2DCFDA$). These effects were blocked by anti-oxidant N-acetylcysteine (NAC). These results suggest that ROS generation could play a role as an intracellular messenger in histamine release by HRF.

Anti-inflammatory and Anti-allergic Effects of Lentinula edodes Extract by UVIrradiation (UV-B 조사에 따른 버섯 추출물의 항염증 및 항알레르기 활성)

  • In this study, the effects of UV irradiation-enhanced ergocalciferol (vitamin D2) content containing Lentinula edodes extract on inflammation and allergic responses were investigated in vitro. The anti-inflammatory and anti-allergic effects of the mushroom extract were tested by estimating the cytokine secretions, such as TNF-α, IL-6, and IL-1β in LPS-activated macrophages (RAW 264.7), or histamine release in PMA and A23187-activated mast cells (RBL-2H3). Under the condition of macrophage activation with LPS, mushroom extract significantly reduced the secretions of pro-inflammatory cytokines, TNF-α and IL-6, and their mRNA expression also matched the observation. The current mushroom extract also significantly reduced the amount of mast cell degranulation-induced histamine secretion from PMA- and A23187-treated mast cells as well as the reduced secretion of IL-4. These results suggest that mushroom extract, which has increased ergocalciferol content by UV irradiation, inhibits the expression of cytokines in inflammation and allergic reactions; therefore, it can be used effectively for the prevention and treatment of inflammatory and allergic diseases.

Effect of Terminalia chebula fruit on anaphylaxis by anal therapy

  • The effect of aqueous extract of Terminalia chebula fruit (Combretaceae) (TCAE) by anal administration on mast cell-dependent immediate-type anaphylactic reactions was investigated. TCAE (0.005 to 1 g/kg) inhibited systemic anaphylaxis induced by compound 48/80 in mice. When TCAE was pretreated at the same concentrations with systemic anaphylaxis, the plasma histamine levels were reduced in a dose-dependent manner. TCAE (0.1 and 1 g/kg) also significantly inhibited local anaphylaxis activated by anti-DNP IgE. TCAE (0.001 to 1 mg/ml) dose-dependently inhibited the histamine release from rat peritoneal mast cells (RPMC) activated by compound 48/80 or anti-DNP IgE. Moreover, TCAE (0.01 and 0.1 mg/ml) had a significant inhibitory effect on anti-DNP IgE-mediated tumor necrosis $factor-{\alpha}$ $(TNF-{\alpha})$ production from RPMC. These results provide evidence that anal therapy of TCAE may be beneficial in the treatment of systemic and local mast cell-dependent anaphylaxis.

Effects of Sesami Semen on Anti-Allergic Inflammation Mechanism related with Atopic Dermatitis (흑지마(黑芝麻)가 알러지성 염증 반응에 미치는 영향)

  • This study aimed to investigate the effects of Sesami Semen on the itching and anti-allergic inflammation mechanism related with cytokine, chemokine, histamine, $\beta$-hexosaminidase, NF-$\kappa$B, and free radical, and it was concluded as follows : 1. Sesami Semen did not show any cytotoxicity at the range of con-centration (1-250 ${\mu}g/m\ell$) on the human fibroblast cell (hFCs). 2. Sesami Semen reduced the gene expressions of IL-8, TNF-$\alpha$, IL-6 mRNA comparing with control. 3. Sesami Semen reduced the levels of IL-6, IL-8, MCP-1 within THP-1 cell depending on the concentration, and especially significantly reduced the the levels of IL-6, MCP-1 at all the concentration. 4. Sesami Semen significantly decreased the histamine secretion on HMC-1 at all the concentration. 5. Sesami Semen decreased the $\beta$-Hexosaminidase secretion 6.2% at 10 ${\mu}g$/ml conc., 58.3% at 100 ${\mu}g$/ml conc. and 63.2% at 200 ${\mu}g$/ml conc., respectively. And IC50 (${\mu}g$/ml) was 158.25 ${\mu}g$/ml. 6. Sesami Semen significantly suppressed the activity of NF-$\kappa$B promoter depending on the concentration. 7. Sesami Semen decreased the production of reactive oxygen species (ROS) and DPPH generation depending on the concentration. As judged with above results, the effects of Sesami Semen on the anti-allergic inflamation would be recognized, and it could be applied on the medicinal sources for prevention or treatment of several allergy disease. And more studies are needed furthermore with the seperation of effective materials.

Inhibitory Effect of Saururus chinensis (Lour.) Baill Extracts on Allergy in Mouse Models (Mouse 모델 알레르기 반응에서 삼백초(三白草)가 미치는 영향)

  • We investigated the effect of Saururus chinensis (Lour.) Baill (SCB) on allergy in mice. We conformed compound 48/80-induced mesenteric mast cell degranulation, active systemic anaphylatic shock and histamine release. Also observed acetic acid-induced vascular permeability and anti-dinitrophenyl (DNP) IgE-mediated passive cutaneous anaphylaxis. SCB inhibited mesenteric mast cell degranulation and active systemic anaphylatic shock induced by compound 48/80 dose-dependently. When SCB was pretreated by intra-peritoneal injection, the plasma histamine levels were reduced. SCB also significantly inhibited acetic acid-induced vascular permeability and anti-DNP IgE-mediated passive cutaneous anaphylaxis. In addition, SCB reduced IL-10 mRNA expression of the lung on ovalbumin-induced allergy. These results indicate that SCB inhibits allergy.

Effect of Kamihyungbangjihwang-tang on Immediate-type Allergic Reaction (가미형방지황탕이 즉시형 알러지반응에 미치는 영향)

  • The purpose of this research was to investigate the effects of Kamihyungbangjihwang-tang (KHT) on immediate-type allergic reaction. KHT was composed of hyungbangjihwangtang (HJT) and Rubus coreanus Miquel (RC) and Aspalathus linear is (AL). KHT and HJT (500 mg/kg) inhibited the systemic anaphylaxis induced by compound 48/80 and inhibited the passive cutaneous anaphylaxis (PCA) induced by anti-dinitrophenyl (DNP)-IgE and DNP-human serum albumin (HSA) in vivo. In addition, KHT, HJT, RC and AL inhibited the release of histamine and increased the release of cAMP from rat peritoneal mast cells. The anti-allergic action of KHT was more potent than those of HJT. These results indicate that KHT may be useful for the prevention and treatment of type I allergy related disease via inhibition of histamine release from mast cells.

Inhibitory Effect of Immediate-Type Hypersensitivity of Syzygium aromaticum extract by Anal Therapy (肛腸療法에 의한 丁香의 卽刻型 過敏反應 抑制效果)

  • Cloves are the dried flower buds of Syzygium aromaticum (L.) Mere et Perry (Myrtaceae). They have been successfully used for the management of various allergic disorders by oral administration in Korea. In this study, the author investigated the effect of an aqueous extract of Syzygium aromaticum on immediate-type hypersensitivity by anal administration. Anal administration of Syzygium aromaticum showed a marked inhibition rate in systemic hypersensitivity with a dose of 1 mg/kg 1 hr before intraperitoneal injection of compound 48/80. Anal administration of Syzygium aromaticum significantly reduced plasma histamine contents induced by compound 48/80. Anal administration of Syzygium aromaticum (1 mg/kg) also inhibited to $61.4\%$ (P<0.01) local a1lergic reaction activated by anti-dinitrophenyl (DNP) IgE. In addition, Syzygium aromaticum dose-dependently inhibited the histamine release from the peritoneal mast cells by compound 48/80 or anti-DNP IgE. When Syzygium aromaticum was added, the level of cAMP in peritoneal mast cells transiently and significantly increased about 47-fold at 10 second compared with that of basal cells. These results provide evidence that anal therapy of Syzygium aromaticum may be beneficial in the treatment of systemic and local immediate-type hypersensitivity by inhibition of histamine release from mast cells in vivo and in vitro.

  • Anal therapy is another way of taking medicine It is a traditional pathway but not available in common situation. Nevertheless. It has many benefit and usefulness, it has not treated so much. The dried immature fruit of Poncirus trifoliata L(Ji-Sil) is widely used to treat urticaria, itch and indigestion in traditional Korean Medicine. So this study was carried out to examine the effect of an aqueous extract from immature fruit of Poncirus trifoliate L. on immediate-type allergic reaction by anal administration. Anal administration of Poncirus trifoliata L. showed a marked inhibition rate in systemic immediate-type allergic reaction with a dose of 0.1 - 1 g/kg 1 hr before intraperitoneal injection of compound 48/80. Anal administration of Poncirus tnjoliata L. (0.1 -1 g/kg) significant1y reduced plasma histamine contents induced by compound 48/80. Anal administration of Poncirus trifoliata L. (10 g/kg) also inhibited to $44.6\%$ (P<0.01) local allergic reaction activated by anti-dinitrophenyl (DNP) IgE. In addition, Syzygium aromaticum dose-dependently inhibited the histamine release from the peritoneal mast cells by compound 48/80 or anti-DNP IgE. These results provide evidence that anal therapy of Poncirus trifoliata L. may be beneficial in the treatment of systemic and local immediate-type hypersensitivity by inhibition of histamine release from mast cells in vivo and in vitro.
