A Study on the Analgesic and Anti-convulsional Effect of Paeoniae Radix (작약류(芍藥類)의 진통(鎭痛) 진경(鎭痙)에 대(對)한 연구(硏究))

  • Both wild peony and cultivated peony demonstrated a slight effect and tranquilizer of anti-convulsion on the strychinine and picrotoxine but the wild plant was more effective. The methanol extract of cultivated peony root demonstrated a weak effect of muscle loosening but its mixture with water extractable fraction did not have this effect in contrast to the case wild type whose methanol extract mixture with water extractable fraction still retained a strong anti-convulsional muscle loosening effect. Therefore, it is concluded that wild peony root has more analgesic and muscle loosening effect than the cultivated counterpart has.

  • We have studied the possible mechanism of anti-asthmatic action of ethanolic extracts of dried seeds of Lepidium sativum (EXLS, 400 mg/kg) using various experimental models. EXLS produced an increase in the Pre-Convulsion Dyspnoea time induced by histamine and acetylcholine aerosol, a significant reduction in the elevated leucocyte counts in the Broncho-Alveolar Lavage fluid of sensitized guinea-pigs and reduction in the paw edema volume as compared to the control rats. Treatment with EXLS also produced decrease in the elevated histamine release from the sensitized guinea-pig lungs. The anti-asthmatic anti-inflammatory responses of EXLS was supported by improvement in microscopic changes like infiltration of inflammatory cells, submucosal edema, epithelial desquamation and reduced lumen size of the bronchi. The $pD_2$ values of histamine in tracheal chain and taenia-coli were significantly greater and that in lung strip was lower in the sensitized animals as compared to control. Treatment of sensitized guinea pigs with EXLS significantly decreased $pD_2$ values of histamine in all three preparations. Our data suggest the prevention of hyper-responsiveness in bronchial smooth muscles and inhibition of the immediate hypersensitive reaction, histamine release in the lungs and the infiltration of various inflammatory cells as the possible mechanisms of anti-asthmatic activity of EXLS.

Effects of Dansambohyultang on serum gastrin, secretin release and CNS (단삼보혈탕(丹蔘補血湯)이 혈청(血淸) gastrin, secretin 및 중추신경계(中樞神經系)에 미치는 영향(影響))

  • This study was conducted to investigate effects of Dansambohyultang on serum gastrin, secretin release and CNS in rats and mice. One group of seven rats was intactly in normal condition, the other group of seven rats was respectively heated, starved, and stressfully immobilized. The third group of seven rats was provided with Dansambohyultang before the stress, and the forth one after the stress. And, gastrin concentration and secretin concentration were measured. Mice were studied with regard to convulsion time, total sleep time and analegic effects on cental nerve system. The results were as follows: 1. Gastrin concentration was significantly decreased, and secretin concentration was significantly increased. 2. Analegic effect by acetic acid method was recognizably observed. 3. Effect of total sleep time done by pentobarbital-Na was recognizably observed. 4. Anti-convulsion effects induced by strychnine, picrotoxin and caffeine were recognizable. It is inferred from above results that Dansambohyultang inhibits gastin release, stimulates secretin release and palliate the pain.

  • An Experimental study were done to examine the clinical effects of Mokwhyangbinrang-Whan on gastrointestinal disease and undertakened by being carried out with rat. We work on the digestive system of Mokwhangbinrng whan. The following results have been obtained ; 1. Spontaneous mobilities in the isolated ilem wewe significantly suppressed and relaxative effects in the isolated ileum were recognized. 2. Significant preventure effects wewe recognized on the pylorus - lighted ulcer in rat(p<0.01) 3. Remarkable preventure effects were recognized on the indomethacin induced gastric ulcer(p<0.01) 4. Inhibitory effects on gastric juice was recognized(p<0.01). Inhibitory effects of free & total acidity were recognized(p<0.05). 5. Significantic analegegic effects were recognized by acetic acid in Mice(p<0.01). 6. Duration of Hypnosis induced by penthbarbital-Na was significantly prolonged (p<0.01). 7. Anti-convulsion action of fat induced by picrotoxin & caffeine was recognized (p<0.05). Notable preventure effects were revealed on the strychnine induced convulsion in rat(p<0.01). According to the above experimental results, it can be conclude that effects of references and clinical application of Mokwhyangbinrng-Whan are approximate.

  • Stephania hernandifolia belonging to the family Menispermaceae is the biggest storehouse of diphenylbisbenzylisoquinoline (DBBI) alkaloids. Exhaustive chemical processing of the bulb of S. hernandifolia by the application of modern separation techniques yielded a DBBI alkaloid which was identified as cycleanine using spectroscopic methods (UV, IR, $^1HNMR$. $^{13}CNMR$, Mass). Cycleanine showed significant anti-inflammatory activity against carrageenin induced paw oedema, comparable to that produced by diclofenac sodium, used as standard drug. It exhibited potent analgesic effects against chemical and thermal noxious stimuli. It was also found to possess anticonvulsive activity in the strychnine induced convulsion model.

Anti-inflammatory activity of jakyakgamcho-tang on Lipopolysaccharide-Stimulated BV-2 Microglia Cells (LPS로 유도된 미세아교세포에서 작약감초탕의 항염증 효과)

  • Objectives : Jakyakgamcho-tang (JGT) has been traditionally used to treat muscular convulsion and pain in South Korea. According to recent studies, JGT has been reported to have anti-depression, anti-inflammation, anti-oxidative, anti-diabetics, anti-spasm and analgesic effects, but studies on its anti-neuroinflammatory and neuroprotective effect have not been deeply conducted. Thus, we investigated the anti-neuroinflammatory activity of JGT on lipopolysaccharide (LPS)-stimulated mouse microglia cells. Methods : To investigate the anti-neuroinflammatory effects of JGT on BV2 microglial cells, we examined the production of nitric oxide (NO) using griess assay, and mRNA expressions of pro-inflammatory cytokines such as interleukin (IL)-1𝛽, IL-6, and tumor necrosis factor (TNF)-𝛼 using real time RT-PCR. Furthermore, to determine the regulating mechanisms of JGT, we investigated the heme oxygenase (HO)-1 by real time RT-PCR. Results : Pre-treatment of JGT effectively decreased NO production in LPS-stimulated BV2 cells at concentrations without cytotoxicity. Additionally, JGT significantly suppressed the production of IL-1𝛽, IL-6, and TNF-𝛼 in LPS-stimulated BV2 cells. Furthermore, JGT activated the HO-1 expression, which is one of the immunomodulatory signaling molecules. And the abolishment of HO-1 by tin protoporphyrin IX (SnPP, the HO-1 inhibitor) reversed the anti- inflammatory activity of JGT in LPS-stimulated BV2 cells. Conclusions : Our results suggest that the JGT has anti-neuroinflammatory effect through the activation of HO-1 in LPS-stimulated BV2 cells. Thereby, JGT could expected to be used for the prevention and treatment of neurodegenerative disease related to neuroinflammation.

Anti-epilepsy Effect of Methanol Extract of Morinda officinalis Augments Pentylenetetrazol-Induced Convulsion Behaviors (파극천 추출물이 펜틸렌테트라졸로 유도된 실험동물에서 항 뇌전증 작용)

  • Morinda officinalis (MO) is a oriental medicinal herb which has been used traditionally for the treatment of impotence, anti-inflammatory, menstrual irregularity action and various brain diseases including antidepressant and anti-stress. In order to examine the mechanism of anticonvulsive effect, we treated the methanol extract of MO (100, 200 mg/kg, P.0) to the sleeping time and pentylenetetrazol (PTZ)-induced convulsive mice. The methanol extract of MO prolonged sleep time by pentobarbital. Dose-dependent of methanol extracts of MO were effected the concentration of GABA and GABA-T activity in the brain of PTZ-induced mice. Methanol extracts of MO significantly inhibited the convulsion state as well as the level of lipid peroxidation in the brain. The butanol and dichloromethane fraction of methanol extracts among the others effectively inhibited in vitro lipid peroxidation dose dependently ($5.0{\times}10^{-2}{\sim}20.0{\times}10^{-2}g/ml$).

  • Chichul-Tang, a combined preparation of crude drug, which was composed of Atractylodis Rhizoma alba and Ponciri Fructus, has been used widely for digestive disorder. Each herb extract of Atractylodis Rhizoma and Ponciri Fructus, the mixture of both herb extracts were examined for the effects on isolated smooth muscle, intestinal propulsion, anti-cathartic action and gastric secretion. The results were summarized as follows: Chichul-Tang showed the inhibitory effects of the convulsion of smooth muscle. Anti-cathartic effect was shown and gastric secretion was inhibited.

An Experimental Studies on the Effects of Chungsimondamtang (청심온담탕(淸心溫膽湯)의 효능(效能)에 관(關)한 문헌적(文獻的) 고찰(考察))

  • In order to study oriental medical literature and clinical effects of Chungsimondamtang, the experiments of analgesic action, sedative action, anticonvulsive action, action on the isolated ileum, action on the circulatory system and hyperlipidaemia were observed. The results were as follows: 1. The analgesic effects were significantly noted. 2. The prolongaton of hypnotic time was noted. 3. The inhibitory effects on the convulsion induced by strychnine, picrotoxin and caffeine were significantly recognized. 4. The spontaneous momentum of isolated ileum was evidently inhibited, and recognized anti Ach. and anti Ba. effects. 5. The effects of descending blood pressure in normal rats and anesthetized rabbits were strongly recognized. 6. The effects of inhibition on the increase of TG. and TC. levels in hyperlipidaemia rats induced by Triton WR-1339 was significantly recognized. 7. The effects of inhibition on the increase of TG. and TC. levels in hyperlipidaemia rats induced by 75% fructose was significantly recognized. With the genelalization of the above-mentioned experimental results, literatual and clinical effects of Chungsimondamtang were approximate to actual experimental results.
