Accumulation of Dietary Conjugated Linoleic Acid (CLA) in Silkworm, Bombyx mori

  • Conjugated linoleic acid (CLA) exhibits potent anti-carcinogenic and other biological activities in several animal models. We report here that dietary CLA, chemically synthesized from corn oil, accumulates I silkworm, Bombyx mori, which is used as therapeutic agent for diabetes in Korea and Japan. Mulberry leaves treated with 0.1 or 10% CLA in ethanol were supplied to silkworms from the end of 3rd instar to the 3rd day of the 5th instar. Fresh mulberry leaves or leaves treated with 10% corn oil in ethanol were fed as a check treatment. the amount of total lipids in the larval body ranged from 17.4 to 19.1mg/g of body tissue, which was not significantly affected by the source of the diets. No CLA was found in the control silkworms. But the level of CLA significantly increased to 83.5 mg/g of fat, when fed with mulberry leaves treated with 10% CLA. Only trace amounts of CLA were detected in the larvae reared with check leaves and 0.1% CLA-treated leaves. Mulberry leaves treatd with corn oil or CLA were not palatable to the larvae, resulting in a reduction of larval weight. These results suggest that silkworms containing CLA in body lipids could be producted by dietary CLA.

Chemopreventive Potential of Coumarin in 7,12-dimethylbenz[a] anthracene Induced Hamster Buccal Pouch Carcinogenesis

  • The aim of the present study was to investigate the chemopreventive effect of coumarin against 7, 12-dimethylbenz[a]anthracene (DMBA)-induced hamster buccal pouch carcinogenesis by monitoring tumor incidence and histopathological changes as well as by analyzing the status of biochemical markers (lipid peroxidation, enzymatic and non-enzymatic antioxidants, phase I and phase II detoxification enzymes). Oral squamous cell carcinomas were induced in the buccal pouch of Syrian golden hamsters by painting with 0.5% DMBA in liquid paraffin three times a week for 14 weeks. We noted 100% tumor formation with marked abnormalities in the biomarkers status in hamsters treated with DMBA alone. Oral administration of coumarin at a dose of 100 mg/kg body weight (bw) to DMBA treated hamsters completely prevented the tumor formation as well as restored the staus of biochemical variables. The results of the present study thus suggest that the chemopreventive effect of coumarin is probably due to its anti-lipid peroxidative potential and modulating effect on carcinogen detoxification agents in favor of the excretion of ultimate carcinogenic metabolites of DMBA during DMBA-induced hamster buccal pouch carcinogenesis.

Effects of Dojeokseungki-tang on the apoptosis of Leukemia Cell (도적승기탕이 백혈병세포의 Apoptosis에 미치는 효과)

  • The purpose of this research was to investigate the anticancer effects of Dojeokseungki-tang(DJSKT) on the various leukemia cell lines. DJSKT treatment suppressed proliferation of cultured-HL60, Jurkat, L1210 cells and increased apoptosis of cultured-L1210, HL60, Molt4, Jurkat cells. DJSKT treatment induced apoptosis of Jurkat cells including the morphologic changes such as the 'ladder pattern' revealed by agarose gel electrophoresis of DNA in a dose-dependent manner. Administration of DJSKT induced apoptosis of transplanted-L1210 cells in vivo, and decreased of mitochondrial transmembrane potential of L 1210 and Jurkat cells in vitro. DJSKT treatment reduced the expression of bcl-2 proteins in Jurkat cells and increased ICE, c-myc, p53 mRNA expression in Molt4 cells. In conclusion, these results suggest that DJSKT might be usefully applied for anti-carcinogenic agent of leukemia.

Effect of Methyl Gallate on 1-Nitropyrene-Induced Keratinocyte Toxicity in a Human and Canine Skin Model

  • The skin, which is the largest organ of the human body, is in direct contact with pollutants in the surrounding atmosphere. Meanwhile, 1-nitropyrene (1-NP), the most abundant nitro-polycyclic aromatic hydrocarbon found in particulate matter, is known to have carcinogenic effects; however, studies on its toxicity in human and canine skin are still needed. In this study, we investigated 1-NP-induced apoptosis and inflammatory pathways in HaCaT cells. In addition, we also measured the cytoprotective effect of methyl gallate (MG), which is widely distributed in medicinal and edible plants and is well known for its anti-inflammatory and antioxidant properties. MG inhibited 1-NP-induced cell death and apoptosis pathways, including the cleavage of PARP and activation of caspase-3, -7, and -9. MG also suppressed 1-NP-induced COX-2 expression and phosphorylation of mitogen-activated protein kinases (MAPKs) and MAPK kinases (MAPKKs). Our findings suggest that 1-NP induces skin toxicity in human and canine through apoptosis and inflammatory responses, and moreover, that this can be prevented by treatment with MG.

The Possible Protective Role of Korean Ginseng on Ochratoxicosis: with Special References on Chromosomal Aberrations in Rats.

  • Ochratoxin A (OA) is a potent mycotoxin causing considerable health hazard and economic loss- e,i. OA is of concern as it is hepato-nephrotoxic, mutagenic, and carcinogenic to a great variety of animals. LDso of crude OA was 8.5 mgf kg.b.w., i.p. The clinical symptoms, mortalities and necropsy were recorded in rats injected with OA (LD5o, i.p.) during 10 days of daily treatment. Ginseng treatments (20 mg 1 kg. b.w., i.p.) : before, mixed with, or after OA dose, completely prevented the mortality in rats. OA-treated animals showed microcytic normochromic anaemia, lucocytosis, hypoproteinaemia and elevation of serum ALT, AST, AP, urea, and creatinine values. These findings were declined near the normal levels when ginseng injected with OA. OA (115 LDso) induced chromosomal aberrations (65.66%) compared to the control. When ginseng given 10 min before OA injection, chromosomal aberrations were reduced to be 31.66% compared to OA-treated animals. In conclusion: ginseng has a protective effect against ochratoxicosis, it has anti-genotoxic activity and it can repair the chromosomal damage induced by ochratoxin A. Physiological Characteristics and Anti-diabetic Effect of Lactobacillus plantarum KI69 (Lactobacillus plantarum KI69의 생리적 특성 및 항당뇨 효과)

  • This study aimed to investigate the physiological characteristics and anti-diabetic effects of Lactobacillus plantarum KI69. The α-amylase and α-glucosidase inhibitory activity of L. plantarum KI69 was 91.17±2.23% and 98.71±4.23%, respectively. The propionic, acetic, and butyric acid contents of the MRS broth inoculated with L. plantarum KI69 were 8.78±1.12 ppm, 1.34±0.07% (w/v), and 0.876±0.003 g/kg, respectively. L. plantarum KI69 showed higher sensitivity to penicillin-G, oxacillin, and chloramphenicol among 16 different antibiotics and showed the highest resistance to ampicillin and vancomycin. The strain showed higher β-galactosidase, β-glucosidase, and N-acetyl-β-glucosaminidase activities than other enzymes. Additionally, it did not produce carcinogenic enzymes, such as β-glucuronidase. The survival rate of L. plantarum KI69 in 0.3% bile was 96.42%. Moreover, the strain showed a 91.45% survival rate at pH 2.0. It was resistant to Escherichia coli, Salmonella Typhimurium, Listeria monocytogenes, and Staphylococcus aureus with the rates of 15.44%, 50.79%, 58.62%, and 37.85%, respectively. L. plantarum (25.85%) showed higher adhesion ability than the positive control L. rhamnosus GG (20.87%). These results demonstrate that L. plantarum KI69 has a probiotic potential with anti-diabetic effects.

Antioxidant and Whitening Effects of the Fermentation of Barley Seeds (Hordeum vulgare L.) Using Lactic Acid Bacteria (유산균을 이용한 보리의 발효를 통한 항산화 및 미백 효과)

  • Barley (Hordeum vulgare L.), of the Poaceae/Gramineae family, is a common grain in the surrounding area. It has been used in Ancient Egyptian medicine and it has been used worldwide for many years as food and as an ingredient in beer. Barley has been reported to have anti-inflammatory, anti -carcinogenic and anti-diabetic effects. So far, a lot of research has been done on barley but the effects of fermented barley seeds with lactic acid bacteria have not been studied largely. In this study, we investigated the effects of ethanol-extracted barley seeds after their fermentation with lactic acid bacteria. The biological activities of fermented barley seeds with lactic acid bacteria and non-fermented barley seeds were analyzed for total polyphenol content, total flavonoid content, DPPH radical scavenging, superoxide dismutase-like activity and tyrosinase inhibition. These results showed that fermented barley seeds with lactic acid bacteria have more advanced anti-oxidant and whitening properties than non-fermented barley seeds. Hence, we suggest that fermenting barley seeds with lactic acid bacteria can be an impressive material in the food and cosmetic industries.

Induction of Cdk inhibitor p21 and inhibition of cyclooxygenase-2 by resveratrol in human lung carcinoma A549 cells. (Resveratrol에 의한 A549 인체 폐암세포의 증식억제 및 apoptosis 유발에 관한 연구)

  • Resveratrol, a phytoalexin found at high levels in grapes and in grape products such as red wine, has been reported to possess a wide range of biological and pharmacological activities including antioxident, anti-inflammatory, anti-mutagenic, and anti-carcinogenic effects. According to recent studies, this compound is an effective inhibitor of cell growth in general, triggers partial arrest of the cell cycle and induce apoptosis. In this study, the anti-proliferative effects of resveratrol in A549 human lung carcinoma cells were investigated. It is shown that resveratrol induced the growth inhibition in a time-dependent manner and morphological changes of A549 cells, which were associated with induction of S phase arrest of the cell cycle and apoptotic cell death. The Bcl-$X_L$levels were markedly down-regulated in resveratrol treated cells, however, Bax and Bcl-2 were remained unchanged. Resveratrol treatment induced the proteolytic degradation of Sp-l and proliferating cell nuclear antigen protein, and inhibited the expression of $\beta$-catenin protein. Resveratrol treatment also induced a marked up-regulation of cyclin-dependent kinase (Cdk) inhibitor p21 and inhibited the kinase activities of Cdk2 and Cdk4. In addition, resveratrol treatment inhibited the levels of cyclooxygenase (COX)-2 mRNA and protein, and the release of prostagladin E2 without alteration of COX-1 expression. Taken together, these findings suggest that resveratrol may be a potential chemotherapeutic agent for the control of human lung carcinorma cells.

Effect of Herbal Acupuncture with Sang-hwang(Phellinus linteus) on High Fat Diet-induced Obesity in Rats (지방식이로 유도된 비만동물모델에서의 약침의 효과)

  • Acupuncture has fairly good weight-reducing effect in treating simple obesity due to the neuroendocrine regulation. In this study, the antiobesity effects of herbal acupuncture(HA) with Sang-hwang(phellinus linteus) at Fuai(SP16) were investigated in the rat fed on high-fat(HF) diet. Sang-hwang mushroom has been proven to have anti-carcinogenic effects and Sang-hwang extracts are highly effective in treatment and preventive treatment of AIDS, diabetes and high blood-pressure. To determine whether the Sang-hwang herbal acupuncture may have the anti-obesity effect, male Sprague-Dawley(4-wk-old) rats were fed a HF diet for 5 wk, which produced significant weight gain compared to rats were fed a normal diet, and then herbal acupuncture were treated for 3 wk in HF diet group. The body weight, food consumption, food effeciency ratio(FER), body fat mass, plasma nitric oxide(NO) were investigated in rats fed on normal diet, HF diet, and HF diet with HA(HF-diet-HA) groups. NO has been proposed to be involved in the regulation of food intake. In addition, the expression of appetite peptides such as orexigenic peptide neuropeptide Y(NPY) and the anorectic peptide cholecystokinin(CCK) were observed in the hypothalamus. HF-HA group reduced body weight gain, FER, body fat contents and NO concentration compared to HF diet group. The expression of NPY was reduced in arcuate nucleus(ARC), and CCK was increased in the paraventricular nucleus(PVN) after treatment of HA. In conclusion, Sang-hwang HA reduced adipocity, plasma NO and hypothalamic NPY, but increased CCK expression in the HF dietinduced obesity rat, therefore HA may have anti-obesity action through regulating body weight and appetite peptide of the central nervous system.

Effects of Dietary Proteins and Inositol Hexaphosphate on the Preneoplastic Lesions and Antioxidant Enzymes of Hepatocellular Carcinogenesis in Rats (식이 단백질의 종류 및 Inositol Hexaphosphate가 간세포 암화과정에서 전암성 병변의 지표 및 항산화 효소계에 미치는 영향)

  • Six-week-old Sprague Dawley rats were fed the diets of 20% casein or soy protein. Two weeks after the feeding, hepatocellular chemical carcinogenesis was initiated by diethylnitrosamine(DEN), and promoted by the diet containing 0.01% 2-acetylamino-fluorene(AAF) and two-thirds partial hepatectomy(PH). The animals were sacrificed at 8 weeks after the DEN injection. The area of placetal glutathione S-trnasferase(GST-P) positive foci, the activities of several enzymes in cellualr antioxidant enzyme systems and glucose 6-phosphatase were determined to investigate the mechanism of the anticarcinogenic effect by the dietary proteins. In another set of experiments, the drinking water of rats fed casein was supplemented with 1.5% inositol hexaphosphate(InsP6) to elucidate whether it has the comparable anticancer action of soy protein. The area and number of GST-P positive foci in the soy protein group were significantly(p<0.05) lower than those inthe casein group. The livers of rats fed casein showed moderate fattydegeneration and larger hyperplastic nodules than those of rats fed soy protein. In another set of experiments, the area and number of GST-P positive foci in the rats fed casein supplemented with InsP6 were not significantly different from those in the rats fed casein or soy protein. The lipid peroxidation of rats fed different protein sources showed no significant difference. Glutathione S-transferase(GST) activities were increased significantly(p<0.05) by carcinogen treatment in all dietary groups. Glucose 6-phosphatase(G6Pase) activities were decreased by carcinogen treatment, and hence showed a reverse relationship(r=-0.695, p<0.01) to the GST-P positive foci. Therefore, the activities in the rats fed casein were lower than those in the rats fed soy protein. These results suggest that the soy protein seems to be more anti-carcinogenic than casein by decreasing the preneoplastic lesion and by increasing the membrane stability but inositol hexaphosphate, a component of soy protein, may not be protective against hepatocarcinogenesis.
