Objective : The purpose of this study was to investigate the anti-cancer effects of Sophorae Radix and the effects of 5-Fluorouracil (5-FU) in human gastric adenocarcinoma cells (AGS). Method : We used human gastric adenocarcinoma cell line, AGS cells. We examined cell death by MTT assay and caspase 3 assay with Sophorae Radix. To examine the inhibitory effects of Sophorae Radix, cell cycle (sub G1) analysis was done the AGS cells after three days with Sophorae Radix. The reversibility of Sophorae Radix was examined on one day to five days treatment with 100 ${\mu}g/ml$ Sophorae Radix. Result : Sophorae Radix inhibited the growth of AGS cells in a dose-dependent fashion. Also we showed that Sophorae Radix induced apoptosis in AGS cells by MTT assay, caspase 3 assay and sub-G1 analysis. Sophorae Radix combined with 5-FU markedly inhibited the growth of AGS cells compared to Sophorae Radix or 5-FU alone. After 3 days treatment of AGS cells with Sophorae Radix, the fraction of cells in sub-G1 phase was much higher than that of the control group. Conclusion : Our findings provide insight into unraveling the effects of Sophorae Radix in human gastric adenocarcinoma cells and developing therapeutic agents against gastric cancer.

A wide variety of cancer chemotherapeutic agents have been shown to induce programmed cell death (PCD, APOPTOSIS) in various tumor cell lines in vitro. cis-Malonato [(4R,5R)-4,5-bis(aminomethyl)-2-isoprpopyl-1,3-dioxolane] platinum(II) (heptaplatin), which is a new drug approved by KFDA in 1999, in a novel platinum-based antitumor agent with clinical potential against stomach cancer and the 3rd generation of the cisplatin. This study was performed to know how heptaplatin and cisplatin and sunpla (mixture of heptaplatin and mannitol) affect on SK-MEL-28 cell line, and how they induce the apoptosis. At EM analysis, the morphology of the cell was changed by treatment of the cisplatin, heptaplatin and sunpla. Apoptotic body formed around plasma membrane, and chromatin condensation represented in nucleus. This phenomenon is one of the characteristic of the apoptosis. The DNA of SK-MEL-28 cell line truncated by cisplatin and sunpla treatment was identified on 2% agarose gel electrophoresis. TUNEL assay was performed to know whether SK-MEL-28 cell die as apoptosis or necrosis by cisplatin, heptaplatin and sunpla. At this result, fluorescence intensity increased according to increase of time and concentration. Therefore, it was identified that cislatin, heptaplatin and sunpla induced apoptosis. Fas expressed on SK-MEL-28 cell membrane by cisplatin, heptaplatin and sunpla was identified by using flow cytometer and the expression of bcl-2(anti-apoptotic gene) decreased according to increase of concentration of the cisplatin, heptaplatin and sunpla. Cisplatin, heptaplatin and sunpla induced apoptosis against SK-MEL-28 cell line, and the apoptotic mechanism was identified as Fas-mediated apoptosis and decreased bcl-2 expression.
Background: In recent years, there has been considerable research on recycling of agro-industrial waste for production of bioactive compounds. The food processing industry produces large amounts of citrus peels that may be an inexpensive source of useful agents. Objective: The present work aimed to explore the phytochemical content, antioxidant, anticancer, antiproliferation, and antigenotxic activities of lemon, grapefruit, and mandarin peels. Materials and Methods: Peels were extracted using 98% ethanol and the three crude extracts were assessed for their total polyphenol content (TPC), total flavonoid content (TFC), and antioxidant activity using DPPH (1, 1-diphenyl-2-picrylhydrazyl). Their cytotoxic and mitogenic proliferation activities were also studied in human leukemia HL-60 cells and mouse splenocytes by CCK-8 assay. In addition, genotoxic/antigenotoxic activity was explored in mouse splenocytes using chromosomal aberrations (CAs) assay. Results: Lemon peels had the highest of TPC followed by grapefruit and mandarin. In contrast, mandarin peels contained the highest of TFC followed by lemon and grapefruit peels. Among the extracts, lemon peel possessed the strongest antioxidant activity as indicated by the highest DPPH radical scavenging, the lowest effective concentration 50% ($EC_{50}=42.97{\mu}g\;extract/mL$), and the highest Trolox equivalent antioxidant capacity (TEAC=0.157). Mandarin peel exhibited moderate cytotoxic activity ($IC_{50}=77.8{\mu}g/mL$) against HL-60 cells, whereas grapefruit and lemon peels were ineffective anti-leukemia. Further, citrus peels possessed immunostimulation activity via augmentation of proliferation of mouse splenocytes (T-lymphocytes). Citrus extracts exerted non-cytotoxic, and antigenotoxic activities through remarkable reduction of CAs induced by cisplatin in mouse splenocytes for 24 h. Conclusions: The phytochemical constituents of the citrus peels may exert biological activities including anticancer, immunostimulation and antigenotoxic potential.

The purpose of this study was to investigate the anti-cancer effects of Sophorae Radix (SR) and doxorubicin (DOX) in human gastric and colorectal adenocarcinoma cells. We used the human gastric and colorectal adenocarcinoma cell lines (MKN-45 and WIDR cells, respectively). We examined cell death by using the MTT(3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide) assay and the caspase 3 assay with SR. To examine the inhibitory effects of SR, we performed a cell cycle (sub G1) analysis for the MKN-45 and WIDR cells after three days with SR. The reversibility of SR was examined for one-day to five-day treatments with SR. SR inhibited the growth of MKN-45 and WIDR cells in a dosedependent manner. Also, we showed that SR induced apoptosis in MKN-45 and WIDR cells by using the MTT assay, the caspase 3 assay and the sub-G1 analysis. SR combined with DOX markedly inhibited the growth of MKN-45 and WIDR cells compared to SR or DOX alone. After 3 days of treating MKN-45 and WIDR cells with SR, the fraction of cells in the sub-G1 phase was much higher than that of the control group. Our findings provide insights into unraveling the effects of SR on human gastric and colorectal adenocarcinoma cells and into developing therapeutic agents for use against gastric and colorectal adenocarcinomas.
Transcriptional silencing is regulated by promoter methylation and histone modifications such as methylation and acetylation. We constructed a Schizosaccaromyces pombe reporter strain, KCT120a, to identify modifiers of transcriptional silencing, by inserting the $ura4^+$ gene into a heterochromatic telomere region. Two compounds inhibited the activity of histone deacetylases, induced acetylation of histone H3 and caused apoptotic cell death in HeLa cells. Expression of gelsolin and $p21^{waf1/cip1}$ also increased, as it does in response to HDAC inhibitors such as TSA. Therefore, these compounds appear to be potent inhibitors of HDACs, and hence potential anti-cancer drugs. Our observations suggest that a yeast cell-based assay system for transcriptional silencing may be useful for identifying histone deacetylase inhibitors and other agents affecting chromatin remodeling.
소아의 종양성질환은 출생 첫해에 가장 많고, 2-3세경에 두 번째로 많이 나타난다. 소아에서는 급성 백혈병, 임파선암, 뇌종양, 연조직 종양 그리고 신장 종양이 일반적이다. 조기진단과 의학의 발달로 치료결과가 향상되고 치료후 생존율 또한 높아지는 추세이나, 항암화학요법, 방사선 및 조혈모세포 이식 등의 치료과정에 동반되는 전신 및 국소적합병증 또한 심각한 문제로 나타난다. 구강조직은 항암치료의 독성에 특히 예민하고 구강내 병소가 나타나는 경우가 많다. 소아치과의사는 종양성질환으로 다양한 항암치료를 받는 어린이의 치료 전후에 삶의 질에 영향을 줄 수 있는 치과적 문제를 진단하고 예방 및 관리에 매우 중요한 역할을 담당한다. 따라서 소아치과의사를 포함한 치과종사자는 환자의 병력, 치료과정, 건강상태에 따른 합병증을 예방 또는 처치하기 위한 개별적 구강관리지침을 제공하는 것이 필요하다. 이 논문에서는 소아에서 종양성질환을 치료중인환자, 특히 항암화학요법, 방사선 요법 및 조혈모세포이식환자에 대한 치료전후의 구강관리에 대하여 알아본다.
TNF ligand 군에 속하는 Tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL/Apo2L)은 death receptor를 통한 세포사멸을 유도하는 것으로 알려졌다. TRAIL은 정상세포에서는 세포사를 일으키지 않고 암세포에서만 특이적으로 세포사멸을 유도함으로써 잠재력 있는 항암제로 주목을 받고 있다. 그러나, 최근 연구에 의하면 악성 신장암과 간암과 같은 일부 암에서는 TRAIL에 의한 세포사에 저항성을 가지는 것으로 알려져 있다. 그러므로, TRAIL 만으로는 다양한 악성종양을 위한 치료법으로 적절하지 않다. TRAIL에 대한 저항성을 가지는 분자적 기전을 이해하고, TRAIL 저항성을 극복할 수 있는 증감제를 밝혀내는 것이 보다 효율적인 TRAIL을 이용한 암세포 치료 전략에 필요하다. 화학치료제들이 TRAIL 수용체인 death receptor의 발현을 증가시키고, 세포 내의 TRAIL에 의한 신호전달 체계를 활성화 시키는 것으로 알려져 있고, 이러한 기전을 통하여 다양한 화학치료제들이 TRAIL에 의한 세포사멸을 증가시키는 것을 확인하였다. 이 논문에서, 우리는 TRAIL에 의한 세포 사멸을 증가시키기 위한 생물학적 약물을 정리하고, 그 분자적 기전을 고찰한다.
연구배경 : Arsenic trioxide($As_2O_3$)은 재발성 또는 불응성 급성전골수성백혈병의 치료제로 쓰이는 항암제로서 비소세포폐암을 포함한 다른 암세포주에도 효과가 있는 것으로 되어있다. NSAIDs는 항암 예방약제로 사용되고 있고, 세포고사를 통해 다른 항암제나 방사선치료의 반응성을 강화시키는 것으로 알려져 있다. 저자들은 NCI-H157 세포주에서 $As_2O_3$와 sulindac의 병합치료가 그것들의 세포고사를 배가시키는지 여부를 알아보고자 하였다. 방 법 : 세포 독성은 MTT 방법으로 측정하였고, 세포고사를 알아보기 위해 핵산 염색과 유식세포 분석을 시행하였다. 세포고사의 기전을 보기 위해 caspasefamily의 활성을 보았고, PARP와 ICAD의 분절을 western blotting으로 확인하였다. 또한 Fas와 Fas-L의 발현유무를 western blotting을 통해 관찰하였다. 결 과 : NCI-H157 폐암세포에 $As_2O_3$와 sulindac을 병합치료시 단독치료군에 비해 생존율이 의미 있게 감소하였고, 이러한 세포사는 핵산염색을 통한 염색사의 응축과 핵 분절 유도와 유식세포 분석에 의한 $sub-G_0/G_1$ DNA분획의 증가현상을 통해 세포고사에 의해 매개됨을 알 수 있었다. 세포고사의 유도에는 caspase 3, 8, 9를 통한 활성화와 이에 의한 PARP와 ICAD의 절단을 확인하였다. 또한 caspase-8 protease의 활성화에는 Fas와 Fas/L 단백질의 발현증가가 유도되었음을 알 수 있었다. 결 론 :NCI-H157 폐암세포주에 $As_2O_3$와 sulindac의 병합요법은 Fas/FasL 신호전달계의 활성화와 caspase 단백질 활성화 의해 세포고사가 유도되었다.
이상의 연구 결과로 미루어 볼 때, 참식나무 기주 참나무겨우살이(TY-NS-B)는 $GSK3{\beta}$에 의한 ${\beta}$-catenin 인산화에 의존하지 않는 ${\beta}$-catenin의 분해 유도를 통해 암세포의 생육과 관련된 c-Myc의 발현을 억제하며, 이것은 비소세포성 폐암의 생육억제와 관련이 있는 것으로 판단된다. 또한, 본 결과는 항암을 위한 대체보완소재로 참식나무 기주 참나무겨우살이의 활용이 가능할 것으로 판단된다. 그러나 추가적 연구를 통해 참식나무 기주 참나무겨우살이가 어떻게 ${\beta}$-catenin의 분해 유도에 관여하는지에 대한 기전연구와 관련 활성물질의 분석연구가 필요할 것으로 사료된다.
