Anti-aging Effects of Sweroside Isolated from Nymphoides indica (어리연꽃으로부터 분리한 Sweroside의 항노화 효과)

  • To develop a new functional agent for cosmetics, we investigated the anti-aging activities in fibroblasts using sweroside isolated from Nymphoides indica. The anti-aging effect of sweroside was validated in CCD-986sk cells. Results showed that sweroside inhibited expressions of UVB-induced ROS and increased pro-collagen. Furthermore, sweroside had inhibited MMP-1 expression. Taken together, the results suggested that sweroside has considerable potential as a new cosmetics ingredient with an anti-aging effect.

  • One of the most effective ways of preventing skin aging is to protect the skin from UV radiation, which was identified as the primary cause of photoaging. Therefore, it is necessary to develop natural and environment-friendly materials to the human skin. This study examined the effects of MAAs extracted from Chlamydononas hedleyi on UV protection and anti-inflammation in human skin cells. The function of porphyra-334 in the skin, which was isolated and purified from MMAs mixture, was tested in terms of its UV protective ability and anti-inflammation. As a result, porphyra-334 played a role in protecting the skin from UV radiation and anti-inflammation through the suppression of COX-2 expression. These results suggest that porphyra-334 can be a useful material in cosmetic products because it can protect the skin from UV radiation and anti-inflammation.

Anti-oxidant and Anti-microbial Activities of Herb-combined Remedies used in Traditional Korean Medicine for Treating Breast Cancer (유암 처방에 사용되어온 한약재 복합 처방전의 항산화 및 항균활성에 관한 연구)

  • Sipyukmiryuki-eum (SYMYKE), Danjacheongpi-tang (DJCPT), Jipae-san Ⅰ (JPS Ⅰ), Jipae-san Ⅱ (JPS Ⅱ), and Chungganhaeul-tang (CGHUT) are representative herb-combined remedies used in traditional Korean medicine for treating breast cancer patients, as mentioned in "Dongeuibogam." In this study, we investigated the total phenolic contents (TPCs) and the anti-oxidant and anti-microbial activities of hot water and 70% ethanol extracts of these herbal prescriptions. Among the five herb-combined remedies, the extraction yields of the hot water extracts and 70% ethanol extracts were the highest in JPS Ⅱ (34.30%) and DJCPT (30.50%), respectively. The TPCs of the hot water extracts from the herb medicines were rich in the order of JPS Ⅰ < CGHUT < JPS Ⅱ < SYMYKE < DJCPT. In addition, the 70% ethanol extracts from the herb medicines were rich in the order of JPS Ⅱ < JPS Ⅰ < CGHUT < SYMYKE < DJCPT. Among them, DJCPT and SYMYKE displayed a strong anti-oxidant capability, which was determined using ferric-reducing anti-oxidant power and scavenging of 2,2'-diphenyl-1-picrylhydrazyl and 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonate) cationic radical activity assays. In addition, anti-microbial activities against Staphylococcus aureus and Escherichia coli were stronger in the 70% ethanol extracts than in the hot water extracts. Together, these findings reveal a positive relationship between TPCs and their anti-oxidant activities.

Development of Anti-Wrinkle Materials using Galloyl-Peptide Derivatives (갈릭산 펩타이드 유도체를 이용한 주름개선 소재 개발)

  • Conjugating a phytochemical, a strong antioxidant, with a functional peptide not only compensates for its stability, but also improves its solubility and anti-wrinkle effects, thereby contributing to the possibility of becoming an excellent cosmetic ingredient. Thus, in this study we examined the potential cosmetic use of a phytochemical-peptide derivative using gallic acid, a phytochemical with antioxidant, anti-inflammatory, and anti-cancer effects. To evaluate the antioxidant and wrinkle-improving efficacy of 5 synthesized gallic acids conjugated with LVH, IVH, KTTKS, YGGFM, and YGGFLRKYP respectively, we observed the expression of genes related to wrinkle improvement using DPPH radical scavenging activity and real-time PCR. As a result, all 5 derivatives had excellent free radical scavenging effects. The expression level of genes involved collagen synthesis also increased, and the secreted peptides during collagen production contributed to their antioxidant and wrinkle improving effects. These results mark the potential use of gallic acid peptide derivatives as a cosmetic ingredient for anti-oxidation and wrinkle improvement.

Depigmenting Effects of Mistletoe (Viscum album var. coloratum) Extracts (겨우살이 추출물의 미백 효과)

  • Melanin pigments are the main cause of skin color. They are produced in melanocytes and then transferred to keratinocytes, which eventually gives the skin surface a variety of colors. Although many skin-lightening or depigmenting agents have been developed, the demand for materials to reduce pig- mentation is still increasing. Here, we tried to find materials for skin-lightening or depigmentation using natural compounds and found that mistletoe (Viscum album var. coloratum) extracts (ME) had an inhibitory effect on tyrosinase activity. As a result, ME significantly reduced pigmentation in human primary melanocytes. In addition, a promoter reporter assay revealed that ME inhibited the transcription of microphthalmia-associated transcription factor (MITF), melanophilin (MLPH), tyrosinase-related protein-2 (TRP-2), and tyrosinase (TYR) genes in HM3KO melanoma cells. In addition, ME decreased the protein level for pigmentation-related molecules, such as TYR and TRP-1. Furthermore, it markedly inhibited the melanogenesis of zebrafish embryos, an in vivo evaluation model for pigmentation. To elucidate the action mechanism of ME, we investigated its effects on intracellular signaling. Eventually, the ME dramatically decreased the phosphorylation of the cAMP responsive element binding protein (CREB), AKT, and ERK. The data suggest that ME may inhibit the melanogenesis pathway by regulating the signaling pathway related to pigmentation. Taken together, these data propose that ME can be developed as a depigmenting or skin-lightening agent.

Effect of Artemisia annua Linne callus induced by plant cell culture technology on wound healing (식물세포배양기술을 이용한 약용식물 개똥쑥 세포주 유도 및 세포주 추출물의 wound healing effect)

  • Currently, many countries have an interest in developing cosmetics materials using native plants. In this aspect, there is increasing need to develop cosmetics materials using native plants in our county. In the present study, calluses were induced from Artemisia annua Linne, which was highlighted because of its useful effects, such as anti-cancer, anti-fungal and anti-inflammation. Water and ethanol extractions were performed from the calluses of Artemisia annua Linne. After the mass production of Artemisia annua Linne's calluses, water and ethanol extraction was performed to examine its functional roles in healing wounds and inflammation. The differences in the effective elements were observed in the ethanol extract. The callus showed anti-inflammation activity through the suppression of the inflammation-related gene, COX-2, and ethanol extracts showed their ability to heal wounds. Overall, these results suggest that the extract of Artemisia annua Linne's calluses is a natural and environment-friendly material, and can be used as medical supplies associated with anti-inflammation and healing wounds.

  • Recently, with the remarkable increase in elderly persons and improvement of skin care technique, the requirements of customer for anti-aging care have become very important. Also, after cosmeceutical-product law was done in 2001, the major interesting field in cosmetic industry has been focused to developing anti-aging materials having safety and efficacy by basic science. In this review, we ill introduce the trend of anti-aging research and materials to prevent wrinkle or improve elasticity of skin.

L-histidine and L-carnosine exert anti-brain aging effects in D-galactose-induced aged neuronal cells

  • BACKGROUND/OBJECTIVES: Brain aging is a major risk factor for severe neurodegenerative diseases. Conversely, L-histidine and L-carnosine are known to exhibit neuroprotective effects. The aim of this study was to examine the potential for L-histidine, L-carnosine, and their combination to mediate anti-brain aging effects in neuronal cells subjected to D-galactose-induced aging. MATERIALS/METHODS: The neuroprotective potential of L-histidine, L-carnosine, and their combination was examined in a retinoic acid-induced neuronal differentiated SH-SY5Y cell line exposed to D-galactose (200 mM) for 48 h. Neuronal cell proliferation, differentiation, and expression of anti-oxidant enzymes and apoptosis markers were subsequently evaluated. RESULTS: Treatment with L-histidine (1 mM), L-carnosine (10 mM), or both for 48 h efficiently improved the proliferation, neurogenesis, and senescence of D-galactose-treated SH-SY5Y cells. In addition, protein expression levels of both neuronal markers (β tubulin-III and neurofilament heavy protein) and anti-oxidant enzymes, glutathione peroxidase-1 and superoxide dismutase-1 were up-regulated. Conversely, protein expression levels of amyloid β (1-42) and cleaved caspase-3 were down-regulated. Levels of mRNA for the pro-inflammatory cytokines, interleukin (IL)-8, IL-1β, and tumor necrosis factor-α were also down-regulated. CONCLUSIONS: To the best of our knowledge, we provide the first evidence that L-histidine, L-carnosine, and their combination mediate anti-aging effects in a neuronal cell line subjected to D-galactose-induced aging. These results suggest the potential benefits of L-histidine and L-carnosine as anti-brain aging agents and they support further research of these amino acid molecules.

Efficacy of callus induced from Ullengdo niche plants for skin protection (식물세포배양기술을 이용한 울릉도 자생식물의 세포주 개발 및 피부세포 효능)

  • Many countries in the world have protected their native plants and utilized them as industrial materials in each country. In this aspect, it is increasingly important to develop cosmetics materials using native plants in Korea. Cosmetic materials have been developed with niche plants, such as Campanula takesimana Nakai, Dianthus superbus, Aster spathulifolius in Ullengdo, in which a specific plant distribution by distinct climate and environment was present. Water and ethanol extractions were performed from the calluses of Campanula takesimana Nakai, Dianthus superbus, Aster spathulifolius. HPLC analysis revealed different compositions and functions of effective elements in each ethanol extract. For example, all types of ethanol extracts showed an ability to heal wounds. In particular, the expression of the inflammation-related gene, COX-2, was decreased in response to the ethanol extracts of Dianthus superbus. These results indicate that the ethanol extracts from niche plants' calluses in Ullengdo are natural and environmentally-friendly compounds, and can be used as medical supplies associated with anti-inflammation and wound healing.

  • Inflammatory and oxidative stimuli play a critical role not only in the process of transforming normal cells into cancer cells, but also in the proliferation process of cancer cells. Sipyukmiryukieum (SYMRKU), a traditional Korean herb-combined remedy, is composed of 16 kinds of herbal medicines, which were recorded for "Ongjeo" treatment in "Dongeuibogam". In this study, we investigated the inhibitory effect of SYMRKU against inflammatory and oxidative responses in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. Our results showed that SYMRKU significantly inhibited LPS-induced secretion of pro-inflammatory mediators including nitric oxide (NO) and prostaglandin $E_2$ without showing any significant cytotoxicity. Consistent with these results, SYMRKU down-regulated LPS-induced expression of their regulatory enzymes such as inducible NO synthase and cyclooxygenase-2. SYMRKU also inhibited LPS-induced production and expression of pro-inflammatory cytokines such as tumor necrosis factor-${\alpha}$, interleukin (IL)-$1{\beta}$ and IL-6. In addition, SYMRKU significantly reduced the production of reactive oxygen species by LPS and showed a strong, which was associated with induction of nuclear factor erythroid 2-related factor 2 and heme oxygenase-1 expression. Although further studies are needed to fully understand the anti-inflammatory effects associated with the antioxidant capacity of SYMRKU, the findings of the current study suggest that SYMRKU may have potential benefits by inhibiting the onset and/or treatment of inflammatory and/or oxidative diseases.
