Anti-Aging Effect of Nigella Sativa Fixed Oil on D-Galactose-Induced Aging in Mice

  • Objectives: Aging is an unconscious and gradual process that can lead to changes in biological systems. Induction of oxidative stress and apoptosis, hepatotoxicity and neurotoxicity are involved in the aging process. Regarding the antioxidant property of black seed oil, the aim of this study was to evaluate the anti-aging effect of Nigella sativa (N. sativa) oil on d-galactose-induced aging in mice. Methods: For induction of aging, D-galactose (500 mg/kg, subcoutaneously SC) was administrated to male mice for 42 days. Animals were treated with D-galactose alone or with b lack seed oil (0.1, 0.2, 0.5 mL/kg, intraperitoneally (ip)). Additionally, vitamin E (200 mg/kg) was used as a positive control. At the end of treatment, the malondialdehyde (MDA) and the glutathione (GSH) contents in brain and liver tissues were measured. Also, enzymes in serum, including aspartate aminotransferase (AST) and alanine amino transferase (ALT), were determined. The levels of the proteins Bax, Bcl2, caspase-3 (pro and cleaved) in brain and liver tissues were evaluated. Results: Administration of D-galactose (500 mg/kg, SC) for 42 days increased serum levels of ALT and AST, as well as the MDA content, in brain and liver tissues, but decreased the GSH content. Additionally, the levels of apoptotic proteins, including Bax, procaspase-3 and caspase-3 cleaved, were markedly increased. N. sativa oil (0.1 and 0.2 mL/kg) diminished the levels of the biochemical markers ALT and AST. Administration of black seed oil (0.1, 0.2 and 0.5 mL/kg) reduced lipid peroxidation and at doses 0.1 and 0.2 mL/kg significantly recovered the GSH content. The oil decreased Bax/Bcl2 levels and at 0.1 mL/kg down-regulated the expressions of caspase-3 (pro and cleaved) proteins in brain and liver tissues. Conclusion: Through its antioxidant and anti-apoptosis properties, black seed oil exhibited an anti-aging effect in a model of aging induced with D-galactose.

한인진 분획물의 항염증, 항균 효과 (Anti-inflammatory and Anti-bacterial Activities of Artemisia iwayomogi Kitamura Extract Fractions)

  • 한인진의 용매별 분획물에 관한 항염증, 항균효과를 조사하기 위하여 용매별로 추출하여 그 수율을 조사하여 본 결과, H2O 층에서는 64.20%로 가장 높은 수율을 나타내었으며, Hexane층(16.05%), Chloroform, Ethyl acetate 층(각각 11.11%)의 순으로 조사되었다. 항산화 활성을 알아보기 위한 DPPH와 ABTs 소거활성을 분석한 결과 Ethyl acetate층에서 각각 12.32 ppm, 7.058 ppm로 가장 높은 소거능을 나타내었고 페놀화합물 및 플라보노이드 함량 분석결과 역시 Ethyl acetate 674.13 mg GA/g, Ethyl acetate 232.5 mg GA/g으로 가장 높게 나타났다. 항균 테스트 결과 황색포도상구균인 Staphylococcus aureus 에서 한인진의 Hexane 층은 모든 농도에서 13-18 mm의 Inhibition zone을 보이며 메탄올 추출물에서 16-10 mm 나타냈고, Chloroform층에서도 1,000 ppm에서 15 mm로 나타났다. 염증세포인 RAW 264.7 세포에서 염증억제효과 및 독성을 확인 한 결과 물층을 제외하고 나머지 분획층에서는 염증유발 물질인 LPS를 처리한 군보다 2-9배 이상 효과를 보이는 것으로 확인되었다. 특히 Ethyl acetate층에서 대조군 보다 9배 이상의 높은 항염증효과를 보였으나 세포독성은 나타나지 않는 것으로 확인되었다.

유암 처방에 사용되어온 한약재 복합 처방전의 항산화 및 항균활성에 관한 연구 (Anti-oxidant and Anti-microbial Activities of Herb-combined Remedies used in Traditional Korean Medicine for Treating Breast Cancer)

  • 본 연구에서 동의보감에 기록된 유암 치료에 사용되어온 5가지 한약재 처방전인 십육미류기음, 단자청피탕, 지패산 Ⅰ, 지패산 Ⅱ와 청간해울탕의 열수와 에탄올 추출물에 대한 총 페놀 함량의 비교 분석과 항산화능 및 항균활성능을 비교 조사하였다. 열수와 70% 에탄올 추출물의 수율은 각각 지패산 Ⅱ와 단자청피탕이 가장 높았으며, 총 페놀 함량은 다른 한약재에 비해 단자청피탕이 가장 높았다. 열수 추출물의 총 페놀 함량은 지패산 Ⅰ < 청간해울탕 < 지패산 Ⅱ < 십육미류기음 < 단자청피탕 순으로 높게 나타났다. 70% 에탄올 추출물의 총 페놀 함량은지패산 Ⅱ < 지패산 Ⅰ < 청간해울탕 < 십육미류기음 < 단자청피탕 순으로 풍부하였다. 그 중 단자청피탕과 십육미류기음의 환원력과 DPPH radical과 ABTS radical 소거능이 가장 뛰어났으며, 이들 시료의 총 페놀 함량과 그들의 항산화능과의 밀접한 상관도가 관찰되었다. 또한 Staphylococcus aureus 및 Escherichia coli에 대한 항균활성은 열수보다 70% 에탄올 추출물에서 효과가 높게 관찰되었다. 결론적으로 유암의 치료에 사용되었던 조사 대상 한약재 처방전은 항산화와 항균 활성은 그들의 총 페놀 함유량과 연관성이 높았으며, 본 연구의 결과는 각 처방전에 대한 구체적인 생리활성 비교를 위한 자료로서 활용될 것이다.

생리활성 펩타이드의 피부미용학적 특성 및 활용 (Characteristics and Applications of Bioactive Peptides in Skin Care)

  • Bioactive peptides (BAP) showed excellent cosmetic activity than bio-materials such as caffeic acid (CA), gallic acid (GA), and nicotinic acid (NA). Caffeoyl tripeptide-1 (CT-1) is a BAP that is stabilized with Gly-His-Lys (GHK) tripeptide and CA by using Fmoc solid phase peptide synthesis. Digalloyl tetrapeptide-19 (DT-19) is stabilized by combining Lys-Glu-Cys-Gly with GA and nicotinoyl tripeptide-1 (NT-1) is synthesized by GHK and NA. According to experiments, CT-1 has an excellent anti-oxidant function even with a very small amount of 10 ppm CT-1. DT-19's tyrosinase inhibition activity has the better effect of about 28.57% in 0.01% and 33.33% in 0.005% of concentration and about 7.89% in 0.001% concentration than vitamin-C. In addition, NT-1 is safer than the NA. Almost BAPs like pal-KTTKS, acetyl hexapeptide, and copper tripeptide-1 have the anti-wrinkle effect while DT-19 and NT-1 are applicable for potential BAPs focused on the whitening effect. The three kinds of BAPs like CT-1, DT-19, and NT-1 consisting of amino acids are safe to the skin, and have more excellent stability than bio-materials which are found to be unstable and cause skin irritation. Due to the high biological activity of BAP in the field of skin care, its utilization will increase constantly.

십이미관중탕(十二味寬中湯)의 노화(老化) 억제 효과에 대한 실험적(實驗的) 연구(硏究) (Anti-aging and Anti-Oxidative Effect of SYG (SipYiMiGwanJungTang) in aged Rats)

  • The purpose of this study is to investigate the anti-aging and anti-oxidative effects of oral administration with SYG (SipYiMiGwanJungTang) decoction in early aged rats. The results were as follows : 1. At the 52 week of the SYG group, the body weight gain was decreased. 2. At the 52 week of the SYG group, the activity of the SOD in the liver was significantly increased than the 10, 22 and 40 week. 3. At the 52 week of the SYG group, the level of the glutathione in the liver was significantly increased than the normal group. 4. At the 68 week of the SYG group, the activity of the catalase in the liver was significantly increased than the normal and control group. 5. At the 52 week of the normal group, the level of the NO in the liver was significantly increased than the 22 and 40 weeks. But, the level of the NO of the SYG group was not changed by the aging. 6. At the 52 and 68 weeks of the SYG group, the level of the MDA in the liver was significantly decreased than the normal and control groups. These results suggest that oral administration of SYG (SipYiMiGwanJungTang) decoction has anti-oxidative and anti-aging effects in early aged rats.

갈릭산 펩타이드 유도체를 이용한 주름개선 소재 개발 (Development of Anti-Wrinkle Materials using Galloyl-Peptide Derivatives)

  • 항산화 효능이 우수한 phytochemical에 콜라겐 합성능이 우수한 기능성 펩타이드의 결합을 통해 소재 자체의 안정성뿐만 아니라 수용액 상태에서의 분산성을 증대시켜 주름개선 효능과 더불어 우수한 화장품 소재로서의 가능성을 가진다. 이에 본 연구는 항산화, 항염 및 항암 등의 효능을 가진다고 알려져 있는 phytochemical인 갈릭산을 이용해 phytochemical-펩타이드 유도체를 개발하여 화장품 소재로서의 가능성을 시험하였다. 갈릭산에 LVH, IVH, KTTKS, YGGFM, YGGFLRKYP 총 5종의 펩타이드를 각각 합성하여 만든 갈릭산 펩타이드 유도체의 항산화 및 주름개선의 효능을 평가하고자 DPPH radical scavenging activity와 real-time PCR로 주름개선과 관련된 유전자의 발현을 확인하였다. 그 결과 5종 유도체 모두 우수한 자유 라디컬 소거능을 보였다. 또한 collagen 합성에 관여하는 유전자의 발현이 증가하였고, collagen 생성시 분비되는 펩타이드의 증가를 통해 항산화뿐만 아니라 주름개선에도 효과가 있었다. 이를 통해 갈릭산 펩타이드 유도체가 항산화 및 주름개선을 위한 화장품 소재로서의 가능성을 확인하였다.

Skin Anti-Aging Activities of Bacteriochlorophyll a from Photosynthetic Bacteria, Rhodobacter sphaeroides

  • In this work, the anti-aging skin effects of bacteriochlorophyll a isolated from Rhodobacter sphaeroides are first reported, with notably low cytotoxicity in the range of 1% to 14% in adding 0.00078 (% (w/w)) of the extracts, compared with the normal growth of both human dermal fibroblast and keratinocyte cells without any treatment as a control. The highest production of procollagen from human fibroblast cells (CCD-986sk) was observed as 221.7 ng/ml with 0.001 (% (w/w)) of bacteriochlorophyll a, whereas 150 and 200 ng/ml of procollagen production resulted from addition of 0.001 (% (w/w)) of the photosynthetic bacteria. The bacteriochlorophyll-a-induced TNF-α production increased to 63.8%, which was lower secretion from HaCaT cells than that from addition of 0.00005 (% (w/w)) of bacteriochlorophyll a. Additionally, bacteriochlorophyll a upregulated the expression of genes related to skin anti-aging (i.e., keratin 10, involucrin, transglutaminase-1, and MMPs), by up to 4-15 times those of the control. However, crude extracts from R. sphaeroides did not enhance the expression level of these genes. Bacteriochlorophyll a showed higher antioxidant activity of 63.8% in DPPH free radical scavenging than those of water, ethanol, and 70% ethanol extracts (14.0%, 57.2%, and 12.6%, respectively). It was also shown that the high antioxidant activity could be attributed to the skin anti-aging effect of bacteriochlorophyll a, although R. sphaeroides itself would not exhibit significant anti-aging activities.

Oligonol promotes anti-aging pathways via modulation of SIRT1-AMPK-Autophagy Pathway

  • BACKGROUND/OBJECTIVES: Oligonol, mainly found in lychee fruit, is an antioxidant polyphenolic compound which has been shown to have anti-inflammatory and anti-cancer properties. The detailed mechanisms by which oligonol may act as an anti-aging molecule have not been determined. MATERIALS/METHODS: In this study, we evaluated the ability of oligonol to modulate sirtuin (SIRT) expression in human lung epithelial (A549) cells. Oligonol was added to A549 cells and reactive oxygen species production, mitochondrial superoxide formation, and p21 protein levels were measured. Signaling pathways activated upon oligonol treatment were also determined by western blotting. Furthermore, the anti-aging effect of oligonol was evaluated ex vivo in mouse splenocytes and in vivo in Caenorhabditis elegans. RESULTS: Oligonol specifically induced the expression of SIRT1, whose activity is linked to gene expression, metabolic control, and healthy aging. In response to influenza virus infection of A549 cells, oligonol treatment significantly up-regulated SIRT1 expression and down-regulated viral hemagglutinin expression. Oligonol treatment also resulted in the activation of autophagy pathways and the phosphorylation of AMP-activated protein kinase (AMPK). Furthermore, oligonol-treated spleen lymphocytes from old mice showed increased cell proliferation, and mRNA levels of SIRT1 in the lungs of old mice were significantly lower than those in the lungs of young mice. Additionally, in vivo lethality assay revealed that oligonol extended the lifespan of C. elegans infected with lethal Vibrio cholerae. CONCLUSIONS: These data demonstrated that oligonol may act as an anti-aging molecule by modulating SIRT1/autophagy/AMPK pathways.

Corni Fructus attenuates testosterone-induced benign prostatic hyperplasia by suppressing 5α-reductase and androgen receptor expression in rats

  • BACKGROUND/OBJECTIVES: Benign prostatic hypertrophy (BPH) is a major cause of abnormal overgrowth of the prostate mainly in the elderly. Corni Fructus has been reported to be effective in the prevention and treatment of various diseases because of its strong antioxidant effect, but its efficacy against BPH is not yet known. This study was designed to evaluate the therapeutic efficacy of Corni Fructus water extract (CF) in testosterone-induced BPH rats. MATERIALS/METHODS: To induce BPH, rats were intraperitoneal injected with testosterone propionate (TP). Rats in the treatment group were orally administered with CF with TP injection, and finasteride, which is a selective inhibitor of $5{\alpha}$-reductase type 2, was used as a positive control. RESULTS: Our results showed that the increased prostate weight and histopathological changes in BPH model rats were suppressed by CF treatment. CF, similar to the finasteride-treated group, decreased the levels of testosterone and dihydrotestosterone by TP treatment in the serum, and it also reduced $5{\alpha}$-reductase expression and concentration in prostate tissue and serum, respectively. In addition, CF significantly blocked the expression of the androgen receptor (AR), AR co-activators, and proliferating cell nuclear antigen in BPH rats, and this blocking was associated with a decrease in prostate-specific antigen levels in serum and prostate tissue. CONCLUSIONS: These results suggest that CF may weaken the BPH status through the inactivation of at least $5{\alpha}$-reductase and AR activity and may be useful for the clinical treatment of BPH.

항노화 상품의 신뢰형성요인 척도 개발과 실증연구 (Development of Scale and Empirical Study of Trust-Forming Factors for Anti-aging Products)

  • 본 연구는 항노화 상품의 신뢰형성요인을 발굴하고 측정변수를 개발하여 결과변수인 신뢰와 구매의도 간의 영향관계를 실증적으로 검증하였다. 기존의 신뢰형성요인인 지각된 품질, 지각된 평판, 브랜드 인지도, 맞춤화에 더하여 새롭게 발굴된 요인은 '공인된 인증'이다. 우리나라의 항노화산업은 소비자 건강에 직접 영향을 미치는 산업이기 때문에 기존의 신뢰형성요인이 소비자 신뢰 구축에 한계가 있는 것으로 판단하여 새로운 척도를 발굴하였다. 본 연구는 항노화 상품 소비자를 대상으로 설문조사를 실시하여 문항의 견실성과 관계성을 분석하였다. 분석결과, 항노화 상품 신뢰형성요인 가운데 지각된 품질, 브랜드 인지도, 맞춤화, 공인된 인증이 신뢰에 정(+)의 영향을 미치는 것으로 나타났고, 지각된 평판은 신뢰에 부(-)의 영향을 미치는 것으로 나타났다. 후행변수인 신뢰와 구매의도 간에는 정(+)의 영향을 미치는 것으로 나타났다. 본 연구는 전략적으로 실효성 높은 유의미한 신뢰형성요인 발굴과 측정문항을 개발하여 실증적, 실무적으로 검증함으로써 신뢰형성요인과 신뢰와의 유의미한 관계를 입증하였다. 동시에 항노화 산업의 마케팅에 중요한 신뢰형성 핵심요인 발굴을 통해 향후의 항노화 상품관련 마케팅전략 수립에 본 연구가 상당부분 기여할 것으로 판단된다.
