Effects of Sinomenium acutum Extract on Body Weight Gains and the DNA Chip Expression of Obese Rats. (방기(防己)의 투여가 비만 유발 쥐의 생리기능과 DNA Chip을 통한 유전자 발현에 미치는 영향에 대한 연구)

  • Purpose: This study is to examine anti-obesity effect and cytotoxicity of the long-term oral administration of Sinomenium acutum (Bang-gi, SA) Methods: Using diet-induced obesity C57BL/6 mouse model, anti-obesity effect and DNA chip expression and cytotoxicity of the long-term oral administration of this herbal extract were investigated. Results: The herbal extract treated groups were arrested in weight increment only when they were lodged together. Such effects were abolished when they kept individually. SA fed mice behaved very rudely and violently. On the basis of histological studies of liver tissues and also in vitro cytotoxicity tests of the liver and kidney cell lines, no significant toxicity was found by 14 weeks of SA treatments. However, we found significant changes in gene expression profile in SA treated group by micro-array analysis. In case of SA group, up-regulated genes were 1,213 and down-regulated ones were 2,558. Some of lipid metabolism related genes also significantly changed in both treatment groups. Conclusion: SA had effects of increasing the basal metabolic rate by stimulating the sympathetic nervous systems.

Analysis of studies on Bangpungtongseong-san (Fangfengtongsheng-san) to establish the fundament for Evidence Based Medicine (EBM) (EBM 기반구축을 위한 방풍통성산 실험 연구 분석)

  • Objective : To establish the fundament for EBM of Traditional Korean Medicine, the papers on Bangpungtongseong-san (Fangfengtongsheng-san) frequently used in medical institutions of Traditional Korean Medicine were analyzed through researching domestic and international papers. Methods : The papers were classified by the registration of domestic or international journals, the year of publishment, experimental fields and the kinds of studies on biological activities. Results : Among 324 volumes of papers on Bangpungtongseong-san (Fangfengtongsheng-san), 38 volumes were selected according to selective criteria. 18 volumes were published in domestic journals, 20 volumes were in Japanese journal. The papers on instrumental analyses reported the quantification of standard compounds of herbal medicines in Bangpungtongseong-san (Fangfengtongsheng-san) using HPLC method. The papers on biological activities of Bangpungtongseong-san (Fangfengtongsheng-san) showed inhibitory effect to obesity, arteriosclerosis, allergic response, inflammation, pain, hyperlipidemia, hypertension, atopy, diabete mallatus and improvement of gastrointestinal activity and cerebral blood circulation. Among biological activities, papers on anti-obesity effect were reported mostly. Conclusions : Further studies including anti-obesity activity need to be preceeded to establish the fundament for EBM of Bangpungtongseong-san (Fangfengtongsheng-san).

Effect of Crude Saponin of Korea Red Ginseng on High Fat Diet-induced Obese Rats (고지방식이(高脂肪食餌)로 유발(誘發)된 비만(肥滿) 흰쥐의 체지방 및 Leptin과 Neuropeptide-Y(NPY)에 대한 홍삼(紅蔘) 조사포닌의 효과(效果))

  • The purpose of this study was to investigate the anti-obesity effects of crude saponin (CS) in Korean red ginseng (KRG) on rats fed a high fat (HF) diet. Methods: Male Sprague-Dawley (SD) rats were divided into a normal diet group and a high-fat (HF) diet group. The HF diet group became obese from being fed a HF diet over five weeks. The normal diet group were fed a normal diet. Each CS group of the normal diet group and HF diet group was administered CS (200 mg/kg, i.p.) for three weeks, the control group of both types was administered normal saline (1ml/kg, i.p.) instead of CS. Each group had 5 rats. Results: After administration of CS, the body weight, food consumption, adipose tissues, and expression of appetite peptides such as leptin and neuropeptide-Y (NPY) were investigated in the HF diet group as well as the normal diet group. Administration of CS reduced body weight, food intake, and fat content in the HF and normal diet groups. After CS administration, NPY expression and leptin were lower in the HF diet group. Conclusions: Our results suggest that CS may be useful in the treatment of obesity, especially of type qixu (氣虛).

Ramie Leaf Extracts Suppresses Adipogenic Differentiation in 3T3-L1 Cells and Pig Preadipocytes

  • The present study was carried out to evaluate the anti-obesity effect of different concentrations of extracts of hot air-dried ramie leaf (HR) and freeze-dried ramie leaf (FR) in 3T3-L1 cells and pig preadipocytes. To analyze the effect on cell proliferation, cells were treated with $25{\mu}g/mL$ or $100{\mu}g/mL$ HR or FR extract for 2 days. Cell differentiation was evaluated by measuring glycerol-3-phosphate dehydrogenase and lipoprotein lipase (LPL) activities and intracellular triglyceride content. Treatment with either HR or FR extracts inhibited the proliferation of 3T3-L1 cells and pig preadipocytes in a dose-dependent manner. HR extract treatment inhibited the differentiation of both cell types more effectively than FR treatment. The extent of triglyceride accumulation decreased significantly in both cells following either HR or FR treatment. Furthermore, LPL activity significantly decreased after treatment with HR or FR extract. These results indicated that HR and FR extracts may inhibit proliferation and differentiation of 3T3-L1 cells and pig preadipocytes. Further studies are needed to explore the anti-obesity effect of HR and FR extracts.

Inhibitory Effect of Crude Extracts from a Brown Alga Sargassum siliquanstrum on 3T3-L1 Adipocyte Differentiation (꽈배기모자반 추출물이 3T3-L1 지방세포 분화에 미치는 영향)

  • In this study, the potential capacity of the crude extract and its solvent fractions from S. siliquanstrum against adipocyte differentiation were evaluated in 3T3-L1 adipocytes. The anti-adipogenic effect of S. siliquanstrum was evidenced by the fact that its crude extract decreases the lipid accumulation of differentiating cells and the expression levels of crucial adipogenesis markers, peroxisome proliferator-activated receptor $(PPAR){\gamma}$ and CCAAT/enhancer-binding protein $(C/EBP){\alpha}$. All solvent fractions except the water fraction showed an observable decrease in lipid accumulation and $PPAR{\gamma}$ and $C/EBP{\alpha}$ expressions. In conclusion, these results suggest that S. siliquanstrum possesses obesity inhibiting components, which may possibly be used as a valuable anti-obesity agent for reducing the risk of obesity.

Anti-adipogenic Effect of Hydrolysate Silk Fibroin in 3T3-L1 Cells

  • Hydrolysate silk fibroin (HSF) is a fibrous protein composed of parallel $\beta$-structures and is made from pure silk elements including 18 amino acids, with glycine, alanine, and serine comprising of over 80% of the amino acids. Numerous studies have documented a range of effects of HSF, including moisturizing, antioxidant activity, nervous system disorders, and many more. We investigated whether HSF has anti-obesity effects in vitro. The effects of HSF inhibition on lipid accumulation and acceleration of lipid degradation in 3T3-L1 cells were studied. Treatment of 3T3-L1 cells with HSF caused significant inhibition of cell viability, an increase in glycerol release, and a decreased in adipocyte differentiation. Moreover HSF stimulated downregulated of adipogenic enzyme expressions (PPAR${\gamma}$ and C/EBP${\alpha}$) and up-regulated of fatty oxidation enzyme expressions (CPT-1 and UCP-2). Based on these results, hydrolysate silk fibroin can be suggested as a potential therapeutic substance as part of a prevention or treatment strategy for obesity.

Effect of Coicis Semen on Starvation Stress in Mice (억이인이 생쥐의 기아 Stress에 미치는 영향)

  • Objective : In 2001, the rate of obesity in Korea reached 30.6%. There are many therapeutic ways to reduce body weight, such as low and very low calorie diet, exercise therapy, behavior modification therapy, etc. However, in many cases the patients feel stress under obesity treatment because of starvation. This study was aimed to evaluate the anti-starvation stress effect of Coicis Semen on mice. Methods : First, the mice were divided into 6 groups : Normal (group with no starvation), Control (administrated normal saline 6 times before starting 36 hours starvation), and Samples A, B, C, and D (administrated 0.25, 0.5, 1.0, 3.0 g/kg Coicis Semen respectively 6 times before starting 36 hours starvation). Then the plasma corticosterone level and rectal temperature were measured. The norepinephrine, dopamine, DOPAC (dihydroxy-phenylacetic acid), 5-HT (5-hydroxytryptamine) and 5- HIAA (5-hydroxy-indole-acetic acid) in the hypothalamus were measured by the HPLC method. Result : I. The rectal temperature in Sample group D showed a significant difference (P<0.05) compared with the Control group. 2. The DOPAC in Sample groups A, C and D showed the significant difference (P<0.05) compared with the Control group. Conclusion : It might be recognized that Coicis Semen has an anti-starvation stress effect.

The Effect of Selaginella tamariscina on Inhibition of Pancreatic Lipase and Lipid Accumulation (부처손(Selaginella tamariscina) 추출물의 리파아제 저해 활성 및 지질 축적 억제 효과)

  • The objective of this study was to evaluate novel usability as natural anti-obesity supplement of Selaginella tamariscina extract. The total phenol contents and total flavonoid contents were $60.29{\pm}3.11GAE\;mg/g$ and $14.90{\pm}0.34QE\;mg/g$, respectively. To evaluate anti-obesity activity of Selaginella tamariscina extract, pancreatic lipase inhibition activity as well as its inhibition effect of lipid accumulation in adipocytes were conducted by Oil Red O staining and lipolysis assay. The result of pancreatic lipase inhibition activity of S. tamariscina extract showed a wide range between 40 and 73% dose dependently. While the incubation of 3T3-L1 cells with S. tamariscina extract inhibited differentiation of preadipocytes and reduced lipid accumulation, the level of released free glycerol into culturing medium was increased in multiple concentrations. These results showed that S. tamariscina extract inhibit adipogenesis and pancreatic lipase activity. Thus, S. tamariscina extract can be a candidate for regulating lipid accumulation in obesity.

The Retrospective Analysis of Obesity and Overweight Female Patients with Clinical Treatment including Herbal Medicine (비만 및 과체중 여성 환자에서 한약 처방의 치료 효과에 대한 후향적 분석)

  • Objectives: The aim of this study was to investigate the effect of herbal medicine on obesity and overweight female patients and to gather the information of a proper treatment goal for anti-obesity program in Korean medicine clinic.Methods: 230 subjects were recruited and those data were analyzed for body-mass index (BMI), treatment duration, weight, body fat, muscle volume and body fat/weight loss ratio. It was compared among age groups and severity of obesity.Results: It was achieved to loss 12-13 kg for 105.8 days thru obesity program of Korean medicine clinic. There were no significant differences for BMI loss, weight loss, body fat loss, muscle volume gain and treatment duration among age groups. It was found that there were significant differences among overweight, obesity and severe obesity group for BMI loss, weight loss, body fat loss and muscle volume gain except treatment duration. It was assumed the weight loss for 1 month was 3.2 kg, 4.2 kg, 4.5 kg for each severity group respectively.Conclusions: It was found herbal medicine treatment would be effective for female obesity and expected the patients could achieve 12-13 kg weight loss for 15 weeks treatment including 8-12 weeks herbal medicine administration and BMI loss per month could be 1.22 kg/m2, 1.60 kg/m2 and 1.70 kg/m2 for severity of obesity respectively. Hereafter, further controlled study with more numbers of patients should be needed to determine the goal of medical treatment for obesity.
