반복적인 고온환경이 사전고온 적응한 육계의 생산성에 미치는 영향 (Effects of Repeated High Ambient Temperature on Performance in Broilers Heat-Conditioned at an Early Age)

  • 생후 초기에 고온을 경험하게 되면 이후 고온환경에 대하여 저항성을 획득하는 것으로 알려져 있다. 본 연구에서는 5일령의 병아리에게 단시간 고온을 경험하게 한 다음, 29일령에 1차 고온환경을 경험한 육계에서 반복된 고온환경이 생산성에 미치는 영향을 조사하였다. 아버에이커 초생추를 크기가 동일한 두 개의 사육실에 수용하였으며, 사료와 물은 자유채식토록 하였다. 조명 환경 조건은 점등 23시간, 소등 1시간이었다. 사전 고온 적응구는 5일령에 $37^{\circ}C$의 고온에 24시간 동안 노출시킨 후 정상 온도로 돌려졌고, 대조구의 감온 일정은 정상적으로 진행되었다. 21일령에 육계를 기존에 수용된 사육실에서 반을 다른 사육실로 옮겨, 한 사육실 내 대조군와 열 적응구가 각각 4개의 Pen에 배치되게 하였다. 1) 사전고온 적응 및 고온환경 양자 모두를 경험하지 않은 육계집단(CON+CON), 2) 사전고온 적응을 경험했지만 고온환경을 경험하지 않은 육계집단(HC+CON), 3) 사전고온 적응 없이 고온환경만 경험한 육계집단(CON+HS), 4) 사전고온 적응 및 고온환경 모두를 경험한 육계집단(HC+HS) 을 가지는 이원배치법(HC vs. HS) 으로 실험을 수행하였다. 29일령부터 3일 동안 고온환경에 노출된 육계들은 43일령부터 재차 고온환경을 경험하였다. 고온환경 처리구에서 사육실 온도를 $32^{\circ}C$($2^{\circ}C$/1 h)까지 올려 3일 동안 유지하였으며, 대조구는 $22^{\circ}C$로 유지되었다. 고온환경은 사료 섭취량, 음수량 및 체중을 현저하게 감소시켰으며, 직장 온도와 폐사율 및 corticosterone의 농도를 증가시켰고, 혈액 생화학 성분을 변화시켰다. 그러나 사전열적응한 육계에서는 비장의 상대무게가 유의적으로 높았다. 따라서 연구결과를 종합적으로 고려해볼 때, 사전고온 적응은 반복적으로 고온환경을 경험하는 육계에게 고온에 대한 저항성을 부여하지 않았다는 것을 의미한다.

Development of a Rapid PCR Test for Identification of Streptococcus agalactiae in Milk Samples Collected on Filter Paper Disks

  • Streptococcus (Strep.) agalactiae is one of the major pathogens of bovine mastitis and is the main cause of subclinical infection. This study attempted to develop a rapid PCR diagnosis procedure using milk samples collected on filter paper disks. Chromatographic filter paper was employed as the preservation media and kept at room temperature for one to four weeks. The revival rate of Strep. agalactiae kept on dried filter paper disks was affected by the pretreatment preservation time. The revival test suggested that not all the bacteria in artificially contaminated milk samples on the filter paper disks could be recovered. After that, a PCR based on the 16-23S intergenic spacer region of Strep agalactiae was performed. The results distinguished the strep. agalactiae from major pathogens of bovine mastitis at a $2{\times}10^2$ colony forming units (CFU)/ml level, which showed similar sensitivity to the results from liquid milk samples. The results also showed that milk samples collected on filter paper disks could be kept at room temperature for one to four weeks with little negative effect on sensitivity and specificity. The field test showed that the diagnostic sensitivity and specificity was 96.15% and 98.60%, respectively. In conclusion, the protocol will provide a rapid and economic procedure for the detection of bovine mastitis.

Supplement of tauroursodeoxycholic acid in vitrification solution improves the development of mouse embryos

  • This study was performed to determine whether supplementation of tauroursodeoxycholic acid (TUDCA), an endoplasmic reticulum (ER) stress inhibitor, during vitrified cryopreservation enhances the development of frozen mouse embryos. Mouse 8-cell stage embryos were collected and exposed to a cryoprotectant solution containing TUDCA or TM (tunicamycin, an ER stress inhibitor) at room temperature and stored in liquid nitrogen following vitrification. The final concentration of TUDCA or TM was $50{\mu}M$. The survival and development rates of mouse 8-cell stage embryos exposed to TUDCA- or TM-containing solutions at room temperature or stored in liquid nitrogen following vitrification were measured. There were no significant differences in survival rate and blastocyst formation rate among control, TUDCA, and TM groups after embryos were exposed to vitrification solutions at RT. When mouse 8-cell stage embryos were treated with TUDCA or TM and then stored in liquid nitrogen, the survival rates of control and TUDCA groups were significantly higher than for the TM group. Blastocyst formation rate of the TUDCA group following in vitro culture was significantly higher than that in control or TM groups. The TM group showed a lower (p < 0.05) blastocyst formation rate than the other two groups. Our results indicate that TUDCA supplementation during cryopreservation of mouse embryos could enhance their development capacity.

겨울철 강제환기식 돈사 내 온실가스 (CH4, N2O) 배출 특성 연구 (Emission Characteristics of Greenhouse Gases (CH4, N2O) in Mechanically Ventilated Swine Farm during Winter Season)

  • The emission characteristics and emission factors were determined by measuring the concentration of methane (CH4) and nitrous oxide (N2O), the amount of ventilation, etc. in the two fattening rooms which have the same environment in winter. As a result of monitoring, the average concentration of CH4 and N2O was 20.7-26.7 ppm and 1.4-1.6 ppm. The average temperature inside the room was measured at 20.0-21.4℃, and the average ventilation was 1345.4-1567.3 m3/h. The daily emission of CH4 for the first 30 days showed a constant emission of 3.6-8.2 g/d/m2/pig, but thereafter, the emission increased rapidly. The daily emission of N2O was 0.7-1.3 g/d/m2/pig, showing stable emission during the test period, and relatively insignificant emission compared to the emission of CH4. After repeated test, it was confirmed that there was no significant difference between the two rooms. As a result, the CH4 6. 21 g/d/m2/pig and N2O 1.02 g/d/m2/pig average emission for each room was derived.

Physicochemical Properties and Freshness of Spent Hen's Meat under Frozen or Refrigeration Conditions after Thawing

  • This study was conducted in order to investigate the effect of storage temperature abuse on the freshness of refrigerated and frozen spent hen's meat. After a room temperature storage condition, two different storage temperature were followed: refrigeration and frozen storage. All parts of the spent hen's meats were thawed at 4 d intervals up to 3 times (2, 6, and 10 d) for 24 h. The level of bacteria on the different parts of the refrigerated and frozen meats was higher than 6 Log CFU/g under the following storage conditions: refrigerated - breast, 12 h; leg, 6 h; wing, 12 h at the $1^{st}$ analysis, frozen - breast, 12 h at the $2^{nd}$ analysis; leg, 24 h at the $1^{st}$ analysis; wing, 12 h at the $1^{st}$ analysis. The pH value for the leg meat was higher than breast and wing meats. In the color measurements, under the room temperature storage condition, lightness and redness values decreased but the yellowness increased in refrigerated meats (p<0.05). The K-value regarding refrigerated leg meats exceeded 60%, which is the threshold value to evaluate the degree of freshness, during the $1^{st}$ analysis (p<0.05). The VBN value of refrigerated leg meat was the highest and reached up to 96.93 mg%. Thus, studies regarding the possible decline in quality according to the change of storage temperature could be used in order to establish a basic database for the quality control of spent hen meat being traded with other countries.

National Genetic Evaluation (System) of Hanwoo (Korean Native Cattle)

  • Hanwoo (Also known as Korean native cattle; Bos taurus coreanae) have been used for transportation and farming for a long time in South Korea. It has been about 30 yrs since Hanwoo improvement began in earnest as beef cattle for meat yield. The purpose of this study was to determine the trend of improvement as well as to estimate genetic parameters of the traits being used for seedstock selection based on the data collected from the past. Hanwoo proven bulls in South Korea are currently selected through performance and progeny tests. National Hanwoo genetic evaluations are implemented with yearling weight (YW), carcass weight (CW), eye muscle area (EMA), backfat thickness (BF) and marbling score (MS). Yearling weights and MS are used for selecting young bulls, and EMA, BF, and MS are used for selecting proven bulls. One individual per testing room was used for performance tests, and five individuals per room for progeny tests. Individuals tested were not allowed to graze pasture, but there was enough space for them to move around in the testing room. Feeds including roughages and minerals were fed ad libitum, and concentrates were provided at the rate of about 1.8% of individual weight. Overall means of the traits were $352.8{\pm}38.56$ kg, $335.09{\pm}44.61$ kg, $77.85{\pm}8.838\;cm^2$, $8.6{\pm}3.7$ mm and $3.293{\pm}1.648$ for YW, CW, EMA, BF and MS. Heritabilities estimated in this study were 0.30, 0.30, 0.42, 0.50 and 0.63 in YW, CW, EMA, BF and MS, respectively, which are similar to results from previous research. Yearling weight was 315.54 kg in 1998, and had increased to 355.06 kg in 2011, resulting in about 40 kg of improvement over 13 yrs. YW and CW have improved remarkably over the past 15 yrs. Breeding values between 1996 and 2000 decreased or did not change much, but have moved in a desirable direction since 2001. These improvements correspond with the substantial increase in use of animal models since the late 1990s in Korea. Hanwoo testing programs have practically contributed to the improvement in aspects of quality and quantity. In sum, the current selection system is good enough to accommodate circumstances where fewer sires are used on many more cows. Although progeny tests take longer and cost more, they seem to be appropriate under the circumstances of the domestic market with its higher requirement for better meat quality. Consequently, accumulative data collection, genetic evaluation model development, revision of selection indices, as well as cooperation among farms, associations, National Agricultural Cooperative Federation, universities, research institutes, and government agencies must be applied to the Hanwoo selection program. All these efforts will assist the domestic market to secure a competitive position against imported beef under Free Trade Agreement trade system and will provide farmers with higher profits as well as the public with a higher quality of beef.

완충공간의 설치가 이유자돈사 온·습도변화 및 자돈 폐사율에 미치는 영향 (Effects of Buffering Zone Installation on the Temperature and Humidity of Pig House and Mortality of Weaned Pig)

  • This survey was conducted to collect basic data about the effect of buffering zone installation in weaned pig house. Buffering zone was installed either inside or outside of pig house to compare changes of temperature, humidity and air velocity of pig room. The body weight and mortality of weaned pigs in house with buffering zone was also measured. There was no difference in temperature, humidity and air velocity between inside and outside buffering zone. Mortality of weaned pig in house with buffering zone was drastically decreased compare to pigs in house without buffering zone that could be useful to maintain constant temperature and decrease mortality.

  • The aim of this study is to evaluate the microbial safety and physical qualities of Korean jerky, and the effect of nisin during storage. Jerky processed packages with or without nisin (100 IU or 500 IU) were stored at room temperature (25 $^{\circ}C$) for 60 days, and samples measured for quality at regular intervals throughout this storage period. In the case of 25$^{\circ}C$ storage, the number of mesophilic microorganisms in seasonedbeef packages without nisin increased markedly, but with nisin there was no observed increase. B. cereus cells showed similar trends, although coliform was not detected in all samples. At 25$^{\circ}C$ storage, changes in the cutting force of packages containing nisin showed no significant change, packages without nisin decreased markedly.

한우사 내 송풍팬의 최적 설치에 관한 연구 (Study on Optimum Installation of Fan in Standard Hanwoo Loose Barn)

  • The fans installed in standard Hanwoo loose barns (room size : 10 m (width) $\times$ 5 m (length)) are frequently used to reduce Hanwoo's heat stress during hot weather and to dry the wet floor. However, the most effective method of installing fans has not been suggested yet. Therefore, this study was carried out to evaluate two methods of installing fans under the ceiling of Hanwoo loose barn by using CFD (Computational Fluid Dynamics) code, FLUENT and to recommend the optimum fan installing method. The fan installation options were fan tilting angles of $45^{\circ}$ and $0^{\circ}$ (horizontal). The fans of 1 m diameter were installed at 3 m above floor. A velocity scale on 10 cm and 110 cm above floor and air flow pattern were used as the parameters to evaluate the fan installing methods. The fans tilted at $45^{\circ}$ angle produced higher wind at 10 cm and 110 cm above floor and more uniform air flow pattern, compared with the fans installed horizontally. Based on these results, fans tilted at $45^{\circ}$ angle may help to reduce Hanwoo's heat stress and will dry the floor better than fans installed horizontally. Therefore, it is suggested that the fans of 1 m diameter in a standard Hanwoo loose barn should be installed at a $45^{\circ}$ tilt angle and 3 m above floor with spacing of 5 m at the center of a room column.

Degradation kinetics of vitamins in premixes for pig: effects of choline, high concentrations of copper and zinc, and storage time

  • Objective: The present work was undertaken to evaluate the effects of storage time, choline chloride, and high concentrations of Cu and Zn on the kinetic behavior of vitamin degradation during storage in two vitamin premixes and four vitamin-trace mineral (VTM) premixes. Methods: Two vitamin premixes (with or without 160,000 mg/kg of choline) were stored at 25℃ and 60% humidity. Besides, four VTM premixes were used to evaluate the effects of choline (0 vs 40,000 mg/kg) and trace minerals (low CuSO4+ZnO vs high CuSO4+ZnO) on vitamin stability in VTM premixes stored in room, and the VTM premixes were stored in room temperature at 22℃. Subsamples from each vitamin and VTM premix were collected at 0, 1, 2, 3, 6, and 12 months. The retention of vitamin A (VA), vitamin D3 (VD3), vitamin E (VE), vitamin K3 (VK3), vitamin B1 (VB1), vitamin B2 (VB2), vitamin B3 (VB3), vitamin B5 (VB5), and vitamin B6 (VB6) in vitamin premixes and VTM premixes during storage was determined. The stability of vitamins in vitamin premixes and VTM premixes was determined and reported as the residual vitamin activity (% of initial) at each sampling point. Results: The effect of choline on VK3 retention was significant in vitamin premixes (p<0.05). The negative effect of storage time was significant for the retentions of VD3, VK3, VB1, VB2, VB5, and VB6 in vitamin premix (p<0.05). For VTM premixes, negative effect of storage time was significant (p<0.05) for the losses of vitamin in VTM premixes. Choline and high concentrations of Cu and Zn significantly increased VA, VK3, VB1, and VB2 loss during storage (p<0.05). The supplementation of high concentrations of Cu and Zn significantly decreased the concentrations of VD3 and VB6 (p<0.05) in VTM premixes at extended storage time. Conclusion: The maximum vitamin stability was detected in vitamin and VTM premixes containing no choline or excess Cu and Zn. The results indicated that extended storage time increased degradation of vitamin in vitamin or VTM premixes. These results may provide useful information for vitamin and VTM premixes to improve the knowledge of vitamin in terms of its stability.
