Screening of Biological Activities of the Extracts from Basil (Ocimum basilicum L.) (Basil(Ocimum basilicum L.) 추출물의 생리활성 탐색)

  • Physiological functionalities of water and ethanol extracts from Basil were determined. The concentration of total phenolic compounds of the water and ethanol extracts were $286.0\;{\mu}g/ml$, $250.0\;{\mu}g/ml$. Antioxidant activities of Basil extracts were determined using 2,2'-azinobis (3-ethyl benzothiazoline-6-sulfonic acid) radical cations (ABTS), 2,2-diphenyl-1-picryl hydrazyl radicals (DPPH), antioxidant protection factor and thiobarbituric acid reactive substances. The total antioxidant activities of Basil extracts using ABTS were 96.8% in the water extracts and 94.7% in the ethanol extract, DPPH were 87.0%, 93.9%, PF were 0.69, 1.16 and TBARS were $0.2{\times}10^{-3}\;{\mu}M,\;0.6{\times}10^{-3}\;{\mu}M$. Angiotensin converting enzyme inhibitory activity and xanthine oxidase inhibitory activity of Basil were higher in ethanol extracts (99.7%, 100.0%) than those of water extracts (39.9%, 54.7%). Phenolic profiles in Basil extracts were analyzed using HPLC. The result was that among the 6 phenolics, rosemarinic acid was the highest in ethanol extracts.

Screening of Biological Activity of Caragana sinica Extracts (골담초(Caragana sinica) 추출물의 생리활성 탐색)

  • In this study, extracts from Caragana sinica flowers and leaves were tested for antioxidant and angiotensin converting enzyme inhibitory activities, along with xanthine oxidase, tyrosinase, elastase, and astringent effects. Total phenolic compounds of acetone extracts from Caragana sinica flowers and leaves were the highest at 3.42 and 2.98 mg/g, respectively, when various extraction solvents were used. Optimal conditions for extraction of phenolic compounds from Caragana sinica leaves and flowers were 70% ethanol for 18 hr. DPPH scavenging activities were the highest in 70% ethanol extracts of Caragana sinica. ABTS radical cation decolorization values of 70% ethanol extracts were higher than those 60% ethanol extracts at 74%. Antioxidant protection factor was 1.2 PF in 70% ethanol extracts from Caragana sinica flowers and leaves. TBARS was lower than that of control (0.54 ${\mu}M$) in all sections. Angiotensin converting enzyme inhibitory activity of Caragana sinica flower extract was 80~90% at a phenolic concentration of 0.2~1.0 mg/mL, whereas xanthin oxidase inhibitory activity of Caragana sinica leaf extract was higher than that of flower extract. Tyrosinase inhibitory activity, which is related to skin-whitening, was above 20%, whereas elastase inhibitory activity related to anti-wrinkle effect was above 50% at a phenolic concentration of 0.8 mg/mL. Astringent effects of Caragana sinica flower and leaf extracts were higher than tannic acid as a control at an equivalent concentration. This result suggests that extracts from Caragana sinica flowers and leaves are suitable as functional foods having anti-hypertension, anti-gout, and medicinal cosmetic activities, including whitening and anti-wrinkle effects.

Biological Activities of Ethanol Extracts from Monascus-Fermented Chinese Yam (발효 홍국마 에탄올 추출물의 생리활성 효과)

  • This study was conducted to investigate antioxidative and physiological activities of ethanol extracts concentration from Monascus-Fermented Chinese Yam (MFCY). The ethanol extracts from MFCY were measured to examine pigment, monacolin K content, total polyphenol content, DPPH radical scavenging activity and angiotensin converting enzyme inhibitory effects. In this study, the results show that Monascus sp. MK2, with Dioscorea batatas Dence. as the substrate, can produce pigments (red, orange, and yellow), monacolin K content, total polyphenol content and DPPH radical scavenging activity at 13.48 (red), 11.66 (orange), 12.77 (yellow), 462.78 (mg/kg), 658.8 (mg/kg) and 92.8% in EtOH extract, respectively. In addition, ACE inhibitory activity was shown to be 74.55% in EtOH extract. Therefore, it can be concluded that Dioscorea batatas Dence is the best fermentative substrate for Monascus species to produce secondary metabolites as biomedicinal substances.

Antihypertensive, Antimicrobial and Antifungal Activities of Buckwheat Hydrolysate (메밀 가수분해물의 항고혈압, 항균 및 항곰팡이 활성)

  • Antibacterial, antifungal, and Angiotensin-I-converting enzyme (ACE) inhibitory activities of buckwheat (Fagopyrum esculentum and F. tataricum) hydrolyzed by Viscozyme L and Alcalase 2.4 L were investigated. The Alcalase 2.4L-hydrolyzed buckwheat showed highest yield of 22.10-24.65%. F. esculentum hydrolysate treated with Viscozyme L from Salmonella typhimurium (clear zone: 3-4.7 mm) and Listeria monocytogenes (clear zone: 4-7.2 mm) showed highest antimicrobial activity among enzymes used. F. esculentum hydrolysate treated with Trichoderma reesei showed strongest antifungal activity among enzymes used (clear zone: 3.7-12 mm). Alcalase 2.4L-hydrolyzed F. esculentum and F. tataricum showed strong ACE inhibitory activities (61.19% and 94.48%, respectively).

Quality characteristics and physiological activities of strawberry vinegars using Acetobacter pasteurianus SRCM60009 (Acetobacter pasteurianus SRCM60009로 발효한 딸기 식초의 품질특성 및 생리활성)

  • Strawberries fermented with Acetobacter pasteurianus SRCM60009 were prepared, and the quality characteristics and physiological activity were measured. As the fermentation period increased, viable cell counts increased, pH decreased, and total acidity increased from 1.09% to 4.20%. The organic acid content of strawberry through acetic acid fermentation was confirmed in the following order: acetic acid, succinic acid, citric acid, and lactic acid. Measurement of α-glucosidase and pancreatic lipase inhibitory activities and angiotensin converting enzyme inhibitory activity showed significantly increased physiological activity owing to fermentation. The use of strawberry vinegar as a functional material was confirmed by measuring the anti-diabetic, anti-obesity, and anti-hypertensive physiological activities through acetic acid fermentation of strawberry. Thus, fermented strawberry vinegar can be used as a functional material in vinegar and other foods.

Partial Purification of Mussel Adhesive Protein for Mytilus Edulis and Preparation of Mussel Protein Hydrolysates

  • Mussel adhesive protein (MAP) was extracted from Korean Mytilus edulis and then partially purified using Sephacryl S-300 gel permeation chromatography and reversed-phase high performance liquid chromatography. As an indicator of adhesiveness, is 3,4-dihydroxyphenylalanine (DOPA) content was determined. Its DOPA/protein ratio of 0.19 was higher than those of other reports, indicating a good adhesive. The partially purified MAP was confirmed by acid-urea polyacrylamide gel electrophoresis using cetylpiridinium bromide as a cationic detergent. Sea mussel hydrolysates were prepared using three commercial proteases to provide value-added functional materials and their angiotensin converting enzyme (ACE) inhibitory activities were determined. Among hydrolysates of sea mussel, Protamex was the best and further purification would improved ACE inhibitory activity.

  • Bovine hide and pig skin gelatins were prepared and their molecular weight profiles were examined by SDS-PAGE. The major molecular weights of bovine hide gelatin were 220 kDa, 140kDa, and 130kDa and the weights of pigskin gelatin were 210 kDa, 135kDa and 120kDa. Also , as a typical parameter of rheological property of the gelatin , viscosities of gelatin were measured under various conditions. Gelatin hydrolysates were prepared using typical commerical proteases and their angiotensin converting enzyme inhibitory activities were examined.

Screening of Biological Activities of Extracts from Rhododendron mucronulatum Turcz. Flowers (진달래꽃(Rhododendron mucronulatum Turcz. Flower) 추출물의 생리활성 탐색)

  • Extracts from Rododendron mucronulatum Turcz. flowers were tested for antioxidant and their inhibitory activities of ${\alpha}$-amylase, ${\alpha}$-glucosidase and angiotensin converting enzyme (ACE). Total contents of phenolics were found as $30.6{\pm}0.14mg/g$ (60% EtOH extract) and $23.2{\pm}0.21mg/g$ (water extract). Electron donation ability (EDA), ABTS [2,2azinobis(3-ethylbenzothiazoline-6-sulfonic acid)] radical decolorization, Antioxidant protection factor (PF) and thiobarbituric acid reactive substance (TBARs) were measured for the antioxidative activity of the extracts from Rododendron mucronulatum Turcz. flowers. The water extract were determined as 97.5% at ethanol extract showed 83.2% and 60% EtOH extract were 89.7% in EDA. The water extract showed higher antioxidant activity than 60% EtOH extract when evaluated by ABTS radical decolorization and antioxidant PF. The TBARS of water extracts and 60% EtOH extracts were shown as $0.29{\times}10^2{\mu}M\;and\;0.28{\times}10^2{\mu}M$, respectively, and were lower than control. ACE inhibitory activity in water extract (67.6% inhibition) was higher than that of 60% EtOH extract (46.7% inhibition) at $200{\mu}g/mL$. Water extracts had higher inhibitory activities on ${\alpha}$-amylase and ${\alpha}$-glucosidase than 60% EtOH extracts. The result suggests that the water extract from Rododendron mucronulatum Turcz. flowers will be useful as natural antioxidants and functional foods.

ACE Inhibitory and Antioxidative Activities of Silkworm Larvae (Bombyx mori) Hydrolysate (번데기 가수분해물의 ACE 저해활성과 항산화활성)

  • In order to utilize the silkworm larvae (Bombyx mori) protein, defatted silkworm protein was hydrolysed by four enzymes (pepsin, trypsin, neutrase and alcalase) at various hydrolysis times (6, 12, 18, 24 and 30 hr) and suspension concentrations (2, 5, 10, 15 and 20%). Protein solubility index, ACE (angiotensin converting enzyme) inhibitory activity and antioxidative activity of silkworm protein hydrolysates were investigated. The optimum condition of hydrolysis was 10% suspension concentration and 18 hr. Protein solubility index of trypsin treatment was higher than other enzyme treatments. ACE inhibitory activity and $IC_{50}$ value of antioxidative activity of neutrase treatment were 86.16% at $100\;{\mu}g/mL$ and $352.75\;{\mu}g/mL$, respectively; also, these values were higher than other enzyme treatments.

In Vitro Screening of the Physiological Activities of lshige foliacea Extracts (넓패(Ishige foliacea) 추출물의 생리활성 탐색)

  • Ishige foliacea belongs to class Phaeophyceae and family Ishigeaceae. This study investigated the physiological activities of the Korean marine algae I. foliacea. Its solvent extracts were prepared with 70% ethanol, 80% methanol, and distilled water. The ethanol and methanol extracts had higher α-glucosidase (half-maximal inhibitory concentration, IC50: 0.67-0.73 mg/mL), xanthine oxidase (IC50: 0.25-0.28 mg/mL), and angiotensin-converting enzyme (IC50: 25.29-38.28 ㎍/mL) inhibitory activities than those of the water extract. The ethanol and methanol extracts possessed high acetylcholinesterase inhibitory activity (IC50: 0.78c0.97 mg/mL). Conversely, the water extract exhibited the highest β-secretase inhibitory activity (IC50: 0.48 mg/mL). These results indicate that I. foliacea may be useful as a functional substance in food and pharmaceuticals with anti-diabetic, anti-gout, anti-hypertension, and anti-dementia properties.
