Characteristics of Byproducts during Anaerobic Hydrogen Fermentation Using Protein (단백질을 이용한 혐기성 수소 발효시 부산물 발생 특성 평가)

  • This study was performed to evaluate initial pH and substrate concentration on hydrogen fermentation of protein. The optimum initial pH and substrate concentration of hydrogen fermentation using protein was 8.0 and 1.0 g peptone/L, respectively. The maximum hydrogen yield at initial pH 8.0 and 1.0 g peptone/L was $19.2{\pm}0.8mL\;H_2/g$ peptone. As results of VFAs analysis, percentages of valerate was similar to hydrogen yield. Also, C. stickalandii, which was hydrogen and valerate producing bacteria, was dominated.

  • This paper deals with an economic evaluation of hydrogen production by fermentation. We evaluate the economic feasibility of domestic hydrogen production by fermentation utilizing glucose and waste water sludge in terms of hydrogen production prices. In addition, we make some sensitivity analysis of hydrogen prices by changing the values of input factors such as the price of glucose, the capital cost of the hydrogen production system, and the hydrogen production yields. The estimated hydrogen prices of the two-step dark-light hydrogen production by fermentation utilizing glucose was $5,347won/kgH_2$, and the single-step hydrogen production by anaerobic fermentation utilizing waste water sludge was $4,255won/kgH_2$, respectively. It is expected that the hydrogen production price by anaerobic fermentation can be reduced if we produce methane or hydrogen utilizing by-products such as alcohols and organic acids, or the government imposes some legal regulations on the treatment of waste water sludge.

Microalgae Removal and Energy Production by Combined Electro-flotation and Anaerobic Hydrogen Fermentation Processes (전기부상과 혐기성 수소 발효 공정의 결합을 통한 미세조류 제거 및 에너지 생산)

  • The algal bloom, resulting from eutrophication, has caused serious water quality problems in river and lake. Therefore, it has to be removed by any means including physicochemical or biological treatment for preserving water quality. This study was conducted to investigate the microalgae removal and energy production using combined electro-flotation and anaerobic hydrogen fermentation processes. The result showed that algae removal efficiency based on chlorophyll a removal increased with the current. At a current of 0.6A, the maximum microalgae removal efficiency of 95.9% was achieved. The treatability of anaerobic hydrogen fermentation was investigated to recover energy from microalgae removed by electro-flotation. The ultimate hydrogen yields of algae before and after ultrasonic pretreatment were 17.3 and 61.1 ml $H_2/g$ dcw(dry cell weight), respectively. The ultrasonic pretreatment of algae led to 3.4-fold higher $H_2$ production due to the increase of hydrolysis rate.

Effects of Short-Term Oxygen Exposure on Anaerobic Reductive Dechlorination and Formate Fermentation by Evanite Culture (혐기성탈염소화 혼합균주에서 산소 노출이 탈염소화 및 수소발생 발효에 미치는 영향)

  • Oxygen sensitivity and substrate requirement have been known as possible reasons for the intricate growth of Dehalococcoides spp. and limiting factors of for routinely applying bioaugmentation using anaerobic Dehalococcoides-containing microbes for remediating chlorinated organic compounds. To explore the effect of the short-term exposure of the short-term exposure of oxygen on Dehalococcoides capability, dechlorination performance, and hydrogen production fermentation from formate, an anaerobic reductive dechlorination mixed-culture (Evanite culture) including dehalococcoides spp. was in this study. In the results, once the mixed-culture were exposed to oxygen, trichloroethylene (TCE) degradation rate decreased and it was not fully recovered even addition of excess formate for 40 days. In contrast, hydrogen was continuously produced by hydrogen-fermentation process even under oxygen presence. The results indicate that although the oxygen-exposed cells cannot completely dechlorinate TCE to ethylene (ETH), hydrogen fermentation process was not affected by oxygen presence. These results suggest that dechlorinating microbes may more sensitive to oxygen than fermenting microbes, and monitoring dechlorinators activity may be critical to achieve an successful remediation of a TCE contaminated-aquifer through bioaugmentation using Dehalococcoides spp..

  • This study aimed to develop a novel anaerobic two-stage process converting food waste to $H_2$ and $CH_2$. The anaerobic two-stage process was devised by combining hydrogen fermentation with methane fermentation. At the high loading rate of 12.3 kg $Vs/m^3/d$, it could remove 72.5%of VS and convert $VS_{removed}$ to $H_2$ (28.2%) and $CH_4$ (69.9%) on COD basis in 8 days.

  • In order to search for new antibiotics from anaerobic bacteria, a large number of samples from domestic soil were collected and processed by apropriate methods. A potential strain, Streptococcus sp. An-21-1, was found to produce antimicrobial compounds. The Results were as follows; 1. During fermentation, the bacteria grew rapidly up to 20hr, thereafter entered the death phase. The optimal temperature and pH for the bacterial growth were $37^{\circ}C$ and pH 7.0, respectively. 2. Antibiotics were purified from culture broth by solvent extraction, silica gel column chromatography and Sepadex L.H 20 column. 3. Physicochemical properties of Ap-1 and Ap-2 were similar ; Their melting points were between $234-237^{\circ}C$. Color reactions of ninhydrin, 2,7-dichlorofluorescein, 4-dimethylaminobenzaldehyde, Dragendroffs reagent and 20% $H_2SO_4$, were positive. Therefore, we assumed that these antibiotics have amine group, immine group, alkaloid, and lipid components. These were stable to heat. UV spectrophotometry showed two peaks at 210 nm and 260 nm. From above results, we assumed these antibiotics are belong to the peptide antibiotic family.

  • Chemical production by anaerobic bacterial fermentations was an important microbiological topic in the past due to both fundamental and applied aspects related to acetone-butanol production prior to its replacement by chemical synthetic routes from petroleum. Presently, the depletion and price-escalation of petroleum has regenerated a great interest in the potential of anaerobic bacteria to transform the renewable resouces such as biomass and wastes into chemical feed-stocks and fuels.

  • Studies on the carbohydrate metabolism of yeast as influenced by gamma-irradiation from cobalt-60 have been carried, then the mechanisms of radiation effect on respiration and fermentation were discussed under considerations of permeable changes of irradiated cell membrane. The cells of baker's yeast (Saccharomyces cerevisiae) which had been gamma-irradiated of 240 k.r. doses for an hour, then were put into aerobic oxidation and anaerobic fermentation without substrate. Total and fractionated carbohydrates of irradiated yeast cells were determined by calorimetric method with anthrone and orcinol reagents, the amounts of total carbohydrate, trehalose, RNA-ribose, PCA-soluble glycogen, alkali-soluble glycogen, acetic acid-soluble glycogen, mannan and glucan were determined according to the course of aerobic oxidation and anaerobic fermentation. It is found that the carbohydrates of irradiated cells leak out and amount of the losses teaches eleven times more than that of control, the volume of losses are seems to be replaced by water, it can be suggested the damage of gamma-irradiation occurs in the site of passive transport of cell membrane. The endogeneous aerobic respiration of irradiated cells are increased much more than control, the synthesis of reserve glycogen, glucan and RNA-ribose promoted much more than control. The anaerobic fermentation of irradiated cells are also increased than that of control, but the breakdown of carbohydrate is less than endogeneous respiration of irradiated cells. The synthetic rate is also less than that of aerobic oxidation. In irradiated yeast cells, trehalose is revealed to be primary substrate for endogeneous carbohydrate metabolism, so it is proved that the enzymic patterns are not changed but the activities of enzymes relating endogeneous respiration and autofermentation is activated. It is to be considerable to distiguish endogeneous respiration and autofermentation from exogeneous respiration and fermentation on irradiation, for membrane permeability changes and loses out carbohydrate by ionizing radiation.

  • This study was aimed at evaluating the effects of carbohydrate, protein and lipid content of substrate on hydrogen yields and microbial communities. The hydrogen yields were linearly correlated to carbohydrate content of substrates while others (content of proteins and lipids) did not make a significant contribution. The chemical composition of substrates produced effects on the final products of anaerobic hydrogen fermentation. Acetate and butyrate were the main fermentation products, with their concentration proving to correlate with carbohydrate and protein content of substrates. The result of microbial community analysis revealed that the relative abundances of Clostridium butyricum increased and Clostridium perfringens decreased as the carbohydrate content increased.

Effects of Pretreatment Time and pH low set value on Continuous Mesophilic Hydrogen Fermentation of Food Waste (열처리 시간과 pH 하한값이 음식물쓰레기 연속 중온 수소 발효에 미치는 영향)

  • Since 2005, food waste has been separately collected and recycled to animal feed or aerobic compost in South Korea. However, the conventional recycling methods discharge process wastewater, which contain pollutant equivalent to more than 50% of food waste. Therefore, anaerobic digestion is considered as an alternative recycling method of food waste to reduce pollutant and recover renewable energy. Recent studies showed that hydrogen can be produced at acidogenic stage in two-stage anaerobic digestion. In this study, the authors investigated the effects of pretreatment time and pH low set value on continuous mesophilic hydrogen fermentation of food waste. Food waste was successfully converted to $H_2$ when heat-treated at $70^{\circ}C$ for 60 min, which was milder than previous studies using pH 12 for 1 day or $90^{\circ}C$. Organic acid production dropped operational pH below 5.0 and caused a metabolic shift from $H_2/butyrate$ fermentation to lactate fermentation. Therefore, alkaline addition for operational pH at or over 5.0 was necessary. At pH 5.3, the result showed that the maximum hydrogen productivity and yield of 1.32 $m^3/m^3$.d and 0.71 mol/mol $carbohydrate_{added}$. Hydrogen production from food waste would be an effective technology for resource recovery as well as waste treatment.
