Amperometric Detection of DNA by Electroreducation of O2 in an Enzyme-Amplified Two-Component Assay

  • The two-component type enzyme amplified amperometric DNA assay is described to use an ambient $O_2$ of the substrate of the DNA labeling enzyme. Although the assay detects DNA only at > 0.5M concentration, a concentration $\~10^6$ fold higher than the sandwich-type enzyme amplified amperometric DNA assay, it can be run with an always available substrate. The assay utilizes screen-printed carbon electrodes (SPEs) which were pre-coated by a co-electrodeposited film of an electron conducting redox hydrogel and a 37-base long single-stranded DNA sequence. The DNA in the electron conducting film hybridizes and captures, when present, the 37-base long detection-DNA, which is labeled with bilirubin oxidase (BOD), an enzyme catalyzing the four-electron reduction of $O_2$ to water. Because the redox hydrogel electrically connects the BOD reaction centers to the electrode, completion of the sandwich converts the film from non-electrocatalytic to electrocatalytic for the reduction of $O_2$ to water when the electrode is poised at 200 mV vs. Ag/hgCl. The advantage or the assay over the earlier reported sandwich type enzyme amplified amperometric DNA assay, in which the amplifying enzyme was horseradish peroxidase, is that it utilizes ambient $O_2$ instead of the less stable and naturally unavailable $H_2O_2$.

  • The nitrogen oxides, NO and NO2, abbreviated usually as NOx, emitted from combustion facilities such as power plants and automobiles are the typical air-pollutants causing acid rain and photochemical smog. In order to solve the NOx-related pollution problems effectively, we need efficient techniques to monitor NOx in the combustion exhausts and in environments. Development of solid-state electrochemical devices for detecting NOx is demonstrated based on various combination of solid electrolytes and auxiliary sensing materials. The object of this research is to develop various sensor performance for solid state amperometric sensor, and to test gas sensor performance manufactured. So we try to present a guidance for developing amperometric gas sensor. We concentrated on development of manufacturing process and performance test.

Amperometric Nitrogen dioxide sensor was fabricated using NASICON and an $NaNO_2$ layer deposited on the counter electrode. The current response was almost linear with Nitrogen dioxide concentration in the range 1-350 ppb at $150^{\circ}C$.

  • A dipstick-type immunochemical biosensor for the detection of the organophosphorus insecticide fenthion was developed using a screen-printed electrode system as an amperometric transducer with polyclonal antibodies against fenthion as a bioreceptor. The assay of the biosensor involved competition between the pesticide in the sample and pesticide-glucose oxidase conjugate for binding to the antibody immobilized on the membrane. This was followed by measurement of the activity of the bound enzyme by the supply of the enzyme substrate (glucose) and amperometric determination of the enzyme reaction product ($H_2O_2$). The activity of the bound enzyme was inversely proportional to the concentration of pesticide. The optimized sensor system showed a linear response against the logarithm of the pesticide concentration ranging from $10^{-2}$ to $10^3\;{\mu}g/L$.

  • A ISFET-based glucose sensor has inherent problems such as low sensitivity, drift effect and long response time. For that reason, a amperometric actuation technique was introduce to make a highly sensitivity of the ISFET glucose sensor with a Pt actuator, which electrolyzes $H_2O_2$, one of the by a by-products of the oxidation reaction of glucose. Moreover, a potential-step measurement method detecting response by only the electrolysis of $H_2O_2$ was developed for eliminating a drift problem. The operation characteristics of ISFET-based glucose sensor was improved by using the amperometric actuation and a measurement techniques. The fabricated ISFET glucose sensor is shown good operation such as characteristics(30mM PBS, about 26mV/decade) and linearity. A portable glucose meter with a highly resolution by using the fabricated ISFET-based glucose sensor with Pt actuation was developed and its characteristics investigated.

  • Recently, FET type dissolved oxygen sensor was proposed to overcome the disadvantages of the amperometric Clark-type sensor. The inherent problem of the proposed sensor, however, is the flow dependence of the sensor performances since the proposed sensor detects the pH change in close proximity to the working electrode. In this study, we decided the direction which minimize the flow effect in FIA(flow injection analysis) system. And a hydrodynamic buffer layer which can reduce the flow dependence were proposed. The suggested buffer-layers were formed onto sensing area and working electrode with mixed polymer matrix of TEOS(tetraethylorthosilicate) and DEDMS(diethoxydimethylsilane).

Disposable Strip-Type Biosensors for Amperometric Determination of Galactose

  • A development of disposable strip-type galactose sensor for point-of-care testing (POCT) was studied, which was constructed using screen-printed carbon electrodes. Galactose levels were determined by the redox reaction of galactose oxidase in the presence of potassium ferricyanide as an electron transfer mediator in a small sample volume (i.e., less than 1 µL). The optimal performance of biosensor was systematically designated by varying applied potential, operating pH, mediator concentration, and amount of enzyme on the electrode. The sensor system was identified as a highly active for the galactose measurement in terms of the sensitivity (slope = 4.76 ± 0.05 nA/µM) with high sensor-to-sensor reproducibility, the linearity (R2 = 0.9915 in galactose concentration range from 0 to 400 µM), and response time (t95% = <17 s). A lower applied potential (i.e., 0.25 V vs. Ag/AgCl) allowed to minimize interference from readily oxidizable metabolites such as ascorbic acid, acetaminophen, uric acid, and acetoacetic acid. The proposed galactose sensor represents a promising system with advantage for use in POCT.

  • The planar-type amperometric creatinine biosensor employing an additional enzyme layer containing creatine kinase and adenosine triphosphate was developed to eliminate severe interference from creatine. In the additional enzyme layer, an interfering substance, creatine is converted to noninterfering product, phosphocreatine. Furthermore, the carbon electrode electroplated with Pt black(Pt-B) was employed to fabricate creatinine biosensors with improved sensor performance(e.g., sensitivity, reliability, and reproducibility). The creatinine levels in an unknown sample were determined within less than 5% errors using creatinine microsensors equipped in a flow-cell cartridge.

Disposable Glucose Sensor Based on Platinised Carbon Paste Electrode (백금 도금된 탄소반죽 전극을 이용한 일회용 글루코오스 센서)

  • Disposable, amperometric glucose sensor was constructed using platinised carbon paste electrode. The sensor response was studied by amperometry and cyclic voltammetry applying sample solutions on the strip-type electrode. Platinization of screen-printed carbon paste electrode effectively improved the electrochemical reversibility of a mediator and the analytical characteristics of the sensor. The heterogeneous rate constant for $[Fe(CN)_6]^{4-/3-}$ was $1.45\times10^{-2}cm{\cdot}s^{-1}$. An applied potential of 0.3V vs. Ag/AgCl resulted in the best selectivity for glucose. The apparent Michaelis-Menten constant for glucose on the strip sensor, $K_m^{app}$, was 24.5 mM. To evaluate the analytical performance of the glucose sensor strip, a correlation study was performed with the NOVA S.P, Ultra M analyzer for 30 serum samples containing $80\~297mg/dL$ of glucose: the correlation coefficient value was 0.983. It can be seen that the strip sensor has satisfactory precision and accuracy.
