아미노 결합 정지상 HPLC 컬럼을 이용한 마 껍질의 allantoin 함량 분석 (Quantitative Analysis of Allantoin in Dioscorea japonica Peel Using an Amino Bonded-Phase HPLC Column)

  • 마에는 다양한 기능성 성분이 함유되어 있어서 식용 및 약용으로 다양하게 이용되고 있다. 특히 allantoin은 대표적인 마의 2차 대사산물로서 의약품 및 기능성 화장품 제조에도 활용되는 고부가가치 산물이다. 본 연구에서는 아미노 결합 정지상 HPLC 컬럼을 이용하여 마의 속과 껍질의 allantoin 함량을 분석하였다. Allantoin 표준용액의 검량선의 결정계수(R2)값은 0.9999로 높은 직진성을 보였으며, 검출한계(LOD)와 정량한계(LOQ)는 각각 0.0229 mg/mL 및 0.0694 mg/mL로 설정되었다. 마 속과 껍질의 건조중량 기준 allantoin 함량은 각각 3.09±0.02 mg/g, 3.91±0.11 mg/g으로 속보다 껍질에 더 많은 allantoin이 함유되어있음을 확인하였다. 따라서 이러한 결과를 토대로 향후 농업부산물인 마껍질이 새로운 고부가가치 기능성 소재로 활용될 수 있음을 확인하였다.

Quantification of Allantoin in Yams (Dioscorea sp.) Using a 1H NMR Spectroscopic Method

  • Allantoin is an abundant component of yams and has been known as a skin protectant due to its pharmacological activities. In previous methods for allantoin determination using high-performance liquid chromatography (HPLC), the separation was unsatisfactory. We herein developed a 1H quantitative nuclear magnetic resonance (qNMR) method for quantification of allantoin in the flesh and peel of yams. The method was carried out based on the relative ratio of signals integration of allantoin to a certain amount of the internal standard dimethyl sulfone (DMSO2) and validated in terms of specificity, linearity (range 62.5-2000 ㎍/ml), sensitivity (limit of detection (LOD) and quantification (LOQ) 4.63 and 14.03 ㎍/ml, respectively), precision (RSD% 0.02-0.26), and recovery (86.35-92.11%). The method was then applied for the evaluation of allantoin in flesh and peel extracts of four different yams cultivated in Korea.

Determination of Allantoin in Dioscorea Rhizoma by High Performance Liquid Chromatography Using Cyano Columns

  • An easy and reliable HPLC method was developed to determine allantoin in Dioscorea Rhizoma using cyano columns. Qualitative and quantitative analyses of allantoin were performed successfully by cyano columns (YMC-Pack CN column, Zorbax SB-CN column and Discovery$^{(R)}$ Cyano column). The intraday precision were 0.58 - 3.33% for YMC-Pack CN, 0.41 - 2.20 for Zorbax SB-CN and 0.45 - 1.93% Discovery$^{(R)}$ Cyano columns, while interday variations were 0.09 - 1.84%, 0.04 - 2.59% and 0.87 - 5.18% for YMC-Pack CN, Zorbax SB-CN and Discovery$^{(R)}$ Cyano columns. The recoveries of allantoin were in the range at 98.8 - 102.6% (RSD 1.1 - 1.6%) for YMC-Pack CN column, 99.7 - 110.5% (RSD 1.3 - 4.9%) for Zorbax SB-CN column, and 97.2 - 110.1% (RSD 1.8 - 5.7%) for Discovery$^{(R)}$ Cyano column. The contents of allantoin in four Dioscorea Rhizoma samples were determined by cyano columms and ranged at 4.1-7.1 mg/g dry weight. The present study indicated that HPLC method using cyano column for determining allantoin is a reliable method and this method can be applied to verify allantoin in Dioscorea Rhizoma.

PLASMA ALLANTOIN CONCENTRATION IN RESPONSE TO CHANGES IN NUTRITIONAL STATUS OF CALVES

  • Two experiments were conducted to search factor(s) affecting the plasma allantoin concentration in infant calves. In experiment 1, five male Holstein calves aged 1 week were given only milk replacer free from nucleic acids for 28 days Plasma allantoin concentration varied in a reverse proportion to daily amounts of milk replacer, and the concentration when calves received 750 g/d of milk replacer was significantly lower than that when they received 250 g/d. Contrary to plasma allantoin concentration, glomerular filtration rate(GFR) was directly proportional to daily amounts of milk replacer, leading to a constant filtration of allantoin across the glomeruli. Renal handling of allantion was also unaffected by the amount of milk replacer, resulting in the constant urinary excretion of allantoin. These results suggested that GFR, which was affected by the nutritional status, could affect plasma allantoin concentration. In experiment 2, the effect of age-related changes in nutritional status after weaning on GFR was examined in eight calves weaned at 5 weeks of age. The GFR expressed as body weight basis was lower immediately after weaning, but linearly increased up to the 19th week post-weaning. The present results suggested that the changes in GFR in response to nutritional status would be one of the possible causes of atypical plasma allantoin concentration immediately after weaning. We conclude that plasma allantoin concentration would not be a proper estimator of intestinal flow of microbial protein in cattle.

Quantitative Analysis of Allantoin in Various Rice Varieties

  • Identification and quantification of allantoin in rice grain of selected varieties were investigated. Allantoin was isolated from Jeokjinjubyeo, and its structure has been elucidated on the basis of spectral data. Allantoin was extracted with a 70% acetone and analyzed by high performance liquid chromatographic methods without a previous chemical derivatization. The concentration of allantoin in selected rice varieties was in a narrow range from 2 to 18 mg per 100 g of brown rice. The highest content was detected in Heugkwang with 18.6 mg per 100 g brown rice, while the lowest was 2.59 mg in Yeomyung.

A major component isolated from this herb consist of dioscin, diosgenin, allantoin, starch, alanin, mucilage, choline, amylase, and glycoprotein. Studies on Milk Allantoin and Uric Acid in Relation to Feeding Regimens and Production Performance in Buffaloes

  • Allantoin and uric acid were estimated in milk to study the association between the levels of these purine derivatives and milk production per day under given feeding regimens. Keeping the stage of lactation, parity and initial milk yield in view thirty lactating buffaloes were randomly selected from early lactating group. All the animals were fed 30 kg green, 2 kg straw and 5 kg concentrate mixture on per animal/day basis at basal level up to 8 1 produce. 1 kg concentrate mixture, soaked cotton seed and boiled cotton seed was fed for every 2 I milk, respectively in Group I (control), Group II and Group III animals. Average milk Allantoin and Uric acid levels were $120{\pm}11.7g/ml$ and $4.03{\pm}0.63g/ml$, respectively in milk. Cotton seed feeding enhanced the milk production significantly (p<0.01) in comparison to concentrate mixture fed control group animals. A significant difference (p<0.01) in milk allantoin levels was found over the different feeding management at higher level of production group animals. Study also revealed a significant negative correlation between the milk allantoin and production per day r=-0.43 (p<0.05).

  • 토양 및 양액의 산도와 질소시용이 대두의 근류착생과 식물체중의 allantoin태질소 및 전질소함량과 주요무기성분함량에 미치는 영향을 구명하여 산도에 따른 질소반응을 밝히기 위하여 장백콩과 두엽콩을 시험하여 산도를 산성(pH 5)과 중성(pH 7) 조건으로 하고 질소시용량을 4수준으로 하여 양액재배 및 포장시험을 실시하였는데 그 결과를 요약하면 다음과 같다. 1. 근류의 착생은 산성조건에서 적었고 질소증시에 의하여 감소하는 경향이었는데 무질소구는 표준질소구보다 떨어졌으며 특히 근류의 건물중은 중성조건에서 질소증시에 의하여 현저히 감소하였다. 2. 식물체중의 allantoin 태 질소함량은 엽보다 줄기중에 현저히 많았는데 산성조건과 질소증시에 의하여 감소하였고 특히 중성조건에서 질소증시에 의한 allantoin 태 질소함량의 감소가 현저하였으며 이와같은 경향은 단엽콩에서 더욱 현저하게 나타났다. 3. 근류의 착생정도는 식물체중의 allantoin 태 질소함량과 유의적인 상관관계를 나타내었다. 4. 식물체중의 전질소함량은 pH가 낮은 산성조건에서 감소하였고 질소증시에 의하여 증가하였는데 그 정도는 산성조건에서 현저하였고 특히 장백콩에서 잘 나타났다. 5. 식물체중의 K, Ca, MG, Mn 등의 함량도 산성조건에서 감소하였고 칼리함량은 질소시용량의 증가에 의하여 감소하는 경향이었다. 6. 상성조건에 의한 근류의 착생, 식물체중의 allantoin 태질소 및 전질소함량 등의 감소정도는 단엽콩에 비하여 장백콩에서 적게 나타나 내산성 품종의 특성을 나타내었다.

Renal and Salivary Excretions of Plasma Purine Derivatives in Swamp Buffaloes and Zebu Cattle

  • This study compared the recovery rate of intrajugular-administered allantoin in the urine and saliva between swamp buffaloes and zebu cattle to examine whether it could explain the lower excretion rate of urinary purine derivatives (PD) in the buffaloes. Three male swamp buffalo yearlings, with an average body weight of $349{\pm}40.35$ kg, and three Thai native cattle ($154{\pm}3.26$ kg) of similar age and sex were used in the study. Animals were kept in individual pens and fed at a maintenance energy level with a diet containing 65% monk bean husk (Vigna radiata) as roughage and 35% concentrates. Allantoin solution was infused into the jugular vein in four incremental rates equivalent to 0, 5, 10 and 15 mmol/d and urine was collected daily in acidified form. Daily PD excretion was linearly correlated with intrajugular allantoin infusion in both species. The relationship between daily urinary PD excretion (Y, mmol/d) and intrajugular allantoin infused (X, mmol/d) was $Y=0.75{\pm}0.318X+22.45{\pm}2.98$ ($r^2$ = 0.36, n = 12, MSE = 38.02, CV = 21.9, p<0.01) for swamp buffaloes and $Y=0.96{\pm}0.10X+15.93{\pm}0.92$ ($r^2$ = 0.91, n = 12, MSE = 3.60, CV = 8.27, p<0.01) for zebu cattle. The salivary PD concentration was not correlated with intrajugular allantoin infusion in both species, with values for buffaloes numerically lower than those for cattle. The present study reconfirmed previous studies that buffaloes have a lower plasma PD excretion rate via the renal route and a significant proportion (22%) of the plasma PD loss is via the saliva. However, results of our present and previous studies suggest that differences in purine base (PB) metabolism between buffaloes and zebu cattle occur before the purine compounds reach the plasma pool.
