Pollutant Load Characteristics of a Rural Watershed of Juam Lake (주암호 농촌 소유역 오염부하특성)

  • A monitoring study has been conducted to identify hydrologic conditions, water quality and nutrient loading characteristics of small watershed in Juam Lake. Climate data of the watershed were collected; flow rate was measured and water quality sampling was conducted at the watershed outlet for this study. Water quality data revealed that T-P concentrations meet I grade of lake water quality standard during non-storm period, but degraded up to II-III grade of lake water quality standard during storm period. The observed T-N concentrations always exceeded lake water quality standard. Therefore, T-P was identified as limiting chemical constituent for eutrophication of Juam Lake. T-P concentration of non-storm period also revealed that point source pollution is not serious in the watershed. Three year monitoring results showed that the observed T-N losses were $10.85\~18.88$ kg/ha and T-P losses were $0.028\~0.323$ kg/ha during six month (Mar. - Oct.), respectively. Major portion of runoff amount discharged by a few storm events a year and nutrient load showed apparent seasonal variation. Huge runoff amounts were generated by intense storms, which make application of water treatment or detention facilities ineffective. Monitoring results confirmed that water quality improvement by abating nonpoint source pollution in rural watershed of monsoon climate should be focused on source control. T-P losses from paddy field seemed to consist of significant amount of total load from study watershed. Therefore, management of drainage from paddy field is considered to be important for preventing algal blooming problem in Juam Lake.

Salts Requirement of Moderately Halophilic Bacterium, Kordia algicida gen. nov., sp. nov. (호염성세균, Kordia algicida gen. nov., sp. nov.의 염류요구특성)

  • Moderately halophilic bacterium, Kordia algicida gen. nov., sp. novo was isolated from seawater of Masan Bay, Korea, during algal blooming caused by Skeletonema costatum. This bacterium was grown on the ZoBell 2216e medium supplied aged seawater, but not grown the medium supplied $3\%$ NaCl. This bacterium showed absolute requirements for mono and divalent cations such as $Na^+,\;Mg^{2+}\;and\;Ca^{2+}$, since no growth was observed in the medium, which is not supplemented with one of $Na^+,\;Mg^{2+}\;and\;Ca^{2+}$ ions. In kinetic studies for three kinds of cation, Km values of $Na^+,\;Ca^{2+}\;and\;Mg^{2+}$ were determined to 0.202 M, 0.089 mM, and 0.189 mM, respectively. Also, $V_{max}({\mu}max)$ was 0.442 h, 0.411 hand 0.316 h. The bacterial cells were quickly lysed in the condition limited by the cations. This result should be suggested that Kordia algicida originated from marine.

  • In this study GIS based pre- and post-processing tool for the Visual MODFLOW that is specific software to model groundwater flow is developed. This tool not only makes input data scientifically but also manages input and output data in terms of the groundwater flow analysis. In addition it can storage project products systematically into Oracle database. The most characteristic figure of this processing tool is to provide the module, which automatically or semi automatically develops various grid cell sizes using GIS ArcView and also produces DXF files reflecting various boundary conditions in the modeling zone. In particular, eminences of this research are to create 3 dimensional hydrogeological structures with 2 dimensional GIS ArcView and to conduct pre- and post- processing along with same topology and data format of the MODFLOW.

CONTROL OF DIATOM BY PREOXIDATION AND COAGULATION IN WATER TREATMENT

  • Conventional coagulation is still the main treatment process for algae removal in water treatment. The coagulation efficiency can be significantly improved by the preoxidation of algae-containing water. Jar test was conducted to determine the optimal condition for the removal of diatoms, especially Cyclotella sp. by preoxidation and the subsequent coagulation. The effects of various concentration of PAC (Polyaluminum chloride) on coagulation with and without preoxidation using chlorine or potassium permanganate at different pHs (7.7 and 9.0) were evaluated. At pH 7.7, preoxidation with 2ppm $Cl_2$ followed by coagulation with 7.5 ppm PAC coagulant could reduce Cyclotella sp. concentration by 86%. At pH 9.0, preoxidation with 1 mg $KMnO_4/L$ followed by coagulation with 12.5 ppm PAC coagulant reduced Cyclotella sp. concentration by 85%. Non-linear regression was applied to determine the optimal condition. At pH 7.7 and 9.0, R was over 0.9, respectively. The pH of algal blooming water is over 9.0. Algae (diatom; Cyelotella sp.) can be controlled in the following ways: preoxidation with 1 mg $KMnO_4/L$ followed by coagulation with 12.5 ppm PAC coagulant can remove 80% algae from water. If water pH is adjusted to 7.7, it was expected that less amount of coagulant (7.5 or 10 mg PAC /L) after preoxidation ($Cl_2$ 2 ppm or $KMnO_4$ 0.33, 1 ppm) would be needed to achieve similar level of algae removal. The oxidation with 0.33ppm $KMnO_4$ followed by coagulation with 7.5 ppm PAC coagulant was preferable due to cost-effectiveness of treatment condition and color problem after treatment.

  • Red tide is a natural phenomenon temporary blooming harmful algal with changing sea color from normal to red, which fish and shellfish die en masse. It also give a bad influence to coastal environment and sea ecosystem. The damage of sea farming by a red tide has been occurred each year which it cost much to prevent disasters of red tide blooms. Red tide damage and prevention cost of red tide disasters can be minimized by means of prediction of red tide blooms. In this paper, we proposed the red tide blooms prediction method using ensemble train. The proposed method use the bagging and boosting ensemble train methods for enhancing red tide prediction and forecast. The experimental results demonstrate that the proposed method achieves a better red tide prediction performance than other single classifiers.

Axenic Culture Production and Growth of a Dinoflagellate, Cochlodinium polykrikoides (적조 와편모조류, Cochlodinium polykrikoides의 순수분리 및 성장)

  • To know the antibiotic specificity of a Dinoflagellate, Cochlodinium polykrikoides, we investigated the survival time of C. polykrikoides against several concentrations of antibiotics and judged the selective specificity of antibiotics based on the $LT_50$ ($50\%$ of lethal time). The result showed that C. polykrikoides was sensitive to tetracycline and chloramphenicol, and resistant to polymixin-B, ampicillin, penicillin-G, dihydrostreptomycin, and neomycin. In the case of sensitive antibiotics to C. polykrikoides, tetracycline and chloramphenicol, the safety concentrations of both antibiotics were determined and the antibiotic specificity based or the plotted survival curve was analyzed. Before antibiotic treatment, we tested the antibiotic susceptibility of the contaminated bacterial population in tile culture of C. polykrikoides, and decided the proper kinds of antibiotics and concentrations before percoll-centrifugation. By percoll-centrifugation, we reduced bacteria, removed fungi, collected the algal pellet, and made axonic culture by antibiotic cascade procedure based on the result of antibiotic susceptibility test. We observed that axonic C. polykrikoides culture entered the logarthmic phase of growth when cell density was over 740 cells/ml and propagated to 5,800 cells/ml maximally. Divisions per day, k value of C. polykrikoides represented a good index for growth at the low density of cells. There was a highest k value shift before reaching to the logarithmic phase. We suggested that the preceeding highest k value shift stage is a good indicator for accurate broadcasting for red. tide blooming in the field, and the stage is also a good time for controlling red tide blooming in the filed, either.

Nitrogen and Phosphorus Uptake and Growth Kinetics of Microcystis aeruginosa Cultured under Chemostats (연속배양에서 Microcystis aeruginosa의 질소 인 흡수와 생장 특성)

  • As unialgal cultures to examine the growth kinetics of an algal species, Microcystis aeruginosa was grown in chemostats with nitrogen and phosphorus limitation. The nutrient concentrations of $NH_4\;^+\;and\;PO_4\;^{3-}$ to limit the growth of M, aeruginosa were approximately 200 ${\mu}M$ and 7 ${\mu}M$, respectively. Cell size of the algae decreased towards the $NH_4$-nitrogen limitation under a constant dilution rate, while it increased in the $PO_4$-limitaion. The cell quota of nitrogen under nitrogen-limited conditions was 6.1 ${\mu}mol$ mg $C^{-1}$ and, under nitrogen sufficient conditions, ranged from 9.5 ${\mu}mol$ mg $C^{-1}$ to 12.4 ${\mu}mol$ mg $C^{-1}$. In addition to the cell quota, the half-saturation constants for nitrogen uptake ($K_s$) and the growth rate (${\mu}_m$) was 36 ${\sim}$ 61 ${\mu}M$ and 0.28 ${\sim}$ 0.35 ${\mu}mol$ cell ${\cdot}$ $hr^{-1}$ to show high values in comparison with other algal species. As the limiting concentration, cell quota and uptake rate of M. aeruginosa were higher than those of any other species, the its nitrogen requirement would be great. In the other side, as the half saturation constant ($K_s$) for nitrogen uptake was higher, and the ratios ofmaximum uptake rate ($V_m$) and $K_s$ was relatively low, the species would have the low competitive ability in the low nitrogen concentration in the ambient water. However, the low concentration of nitrogen in the Nakdong River during the Microcystis outbreak would be the inevitable results of the algal blooms. In the lower Parts of the Nakdong River, the nutrient status was coupled with the growth kinetics of the blooming algae to have clear seasonal variations through a year.

Removal of Geosmin and 2-methylisoborneol in Drinking Water by Powdered Activated Carbon (분말 활성탄에 의한 먹는 물 내의 이취미 물질 제거)

  • Geosmin and 2-methylisoborneol (2-MIB) produced by cyanobacteria during algal blooming in surface water are the major taste-and-odor-causing compounds in drinking water and need to be removed. Activated carbon is often used in treatment plants for the mitigation of odor problem. However, there is a lack of information on the effect of pore size distribution and particle size of activated carbon for adsorption of both odor compounds. Therefore, we studied the effect of pore size distribution and particle size of activated carbon on the adsorption of geosmin and 2-MIB. When comparing the adsorption of geosmin and 2-MIB between activated carbon fiber (ACF), powdered activated carbon (PAC) and granular activated carbon (GAC), the order of removal efficiency was PAC > ACF > GAC. As a result of comparing PACs with various pore distribution characteristics, well-developed micropores on activated carbon were found to be favorable for adsorption of geosmin and 2-MIB. For particle size, smaller was more effective for adsorption of geosmin and 2-MIB.

Changes of the Nutrients and Water Trophic States in Upo Wetland (우포늪의 영양염과 수질 영양 상태 변화)

  • In the four swamps of Jjokjibeol, Mokpo, Upo and Sajipo in the Upo wetland, the nitrogen nutrients, phosphorus nutrients and chl-$\alpha$ had been observed during the period from April 2005 to December 2009 on monthly basis. Based on the results, the fluctuations of trophic state in the Upo wetland were estimated. Measurements of the nitrogen nutrients such as $NO_3$-N, $NH_3$-N and T-N showed to be generally decreased in comparison with those in the precent studies. Yet the T-N was still considerably higher than the general concentration level of eutrophication and algal blooming. $PO_4$-P and T-P showed to have reduced considerably in comparison to precedent studies. However, T-P also turned out to be dissolved over the nutrient standard. Nitrogen nutrients and phosphorus nutrients were the lowest in Jjokjibeol in the Upo wetland. The chl-$\alpha$ concentrations were the highest at summer periods in Jjokjibeol and Mokpo. However, the highest at non-summer periods in Upo and Sajipo. Among the four swamps, Upo had the highest density on average of chl-$\alpha$, and Mokpo the lowest. Through TRIX (Trophic Index) analysis evaluating trophic state of the Upo wetland, all four swamps were estimated of poor water quality (eutrophication).

Effect of pH Adjustment by CO2 on Coagulation and Aluminum Elution in Water Treatment (CO2 주입에 의한 pH 조정이 정수장 응집효율 및 알루미늄 용출에 미치는 영향)

  • In this study, a pH control method by carbon dioxide ($CO_2$) was applied to coagulation process in water treatment plant (WTP) to investigate the coagulation efficiency and residual dissolved aluminum when high pH raw water is flowing into the plant during algal blooming. Existing coagulant dose (1 mg/L in raw water) resulted in the pH reduction of 0.0384 by LAS, 0.0254 by PAC, 0.0201 by A-PAC, and 0.0135 by PACS2, respectively. And then the concentration of dissolved aluminum was 0.02 mg/L at pH 7.44, 0.07 mg/L at pH 7.96, 0.12 mg/L at pH 8.16, 0.39 mg/L at pH 8.38 showing the concentration increase with pH in the coagulation process. It was noteworthy that rapid increase was observed at pH above 8.0 next the rapid mixing. Therefore it is necessarily required to control pH below 7.8 in the coagulation process in order to meet drinking water quality standard of aluminum for high pH raw water into WTP, $CO_2$ injection could control pH successfully at about 7.3 even for the raw water of high pH above 8.0. In addition it was found that the pH control by $CO_2$ injection was significantly effective for coagulation in terms of turbidity removal, coagulant dosage, and residual dissolved aluminum concentration.
