Effects of Rehmanniae Radix and Radix Aconiti Water Extracts on Blood Pressure in Hypertensive Rats (숙지황, 부자 전탕액이 고혈압 백서의 혈압에 미치는 영향)

  • 한의학에 있어서 음양의 조절은 매우 중요하다. 숙지황은 음허를 치료하는 주약이며, 부자는 양허를 치료하는 주약이다. 한의학에서 신성고혈압의 발생은 음허와 관련이 있다고 하였다. 본 연구는 실험적 신성고혈압과 자연발증고혈압 백서에 숙지황과 부자 전탕액을 투여 후 혈압의 변동과 혈장 renin 활성도, aldosterone 및 심방이뇨호르몬 (atrial natriuretic hormone)의 변동을 관찰하였다. 숙지황 투여후 실험적 신성고혈압 백서의 혈압은 감소하였다. 부자 전탕액 투여 후 자연발증고혈압 백서의 혈압 상승이 억제되었다. 숙지황 및 부자 전탕액 투여 후 자연발증고혈압 백서의 혈장 renin 활성도는 감소하였다. 부자 전탕액 투여 후 신성고혈압 백서의 혈장 ANP 농도는 감소하였다. 부자 전탕액의 혈압강하 작용은 ANP 및 aldosterone 농도의 변동과 관련이 있다고 사료된다.

Upregulation of Renin-angiotensin, Endothelin and C-type Natriuretic Peptide in Rat Glomerulus with Bilateral Ureteral Obstruction

  • The present study was designed to investigate the effects renin-angiotensin-aldosterone system (RAAS), endothelin (ET) and local natriuretic peptide (NP) system for glomerulopathy induced in the experimental bilateral ureteral obstructive rats. Sprague-Dawley male rats ($200{\sim}220g$ body weight) were bilaterally obstructed by ligation of the proximal ureters for 24 hours. Control rats were treated in the same ways, except that no ligature was made. The glomeruli were isolated from cortex by graded sieve methods, and the mRNA expressions of local renin-angiotensin system (RAS), aldosterone synthase (CYP11B2), endothelin-1 (ET-1) and NP system were determined by real-time polymerase chain reaction. Following the bilateral ureteral obstruction, the mRNA expressions of renin, angiotensin converting enzyme 1 as well as ET-1 were increased, while that of angiotensin converting enzyme 2 was not changed. The expressions of CYP11B2 and angiotensin II receptors were not changed. C-type natriuretic peptide (CNP) expression was increased, while its receptors (natriuretic peptide receptor-B) were not changed. We suggest that the upregulation of local RAS and ET playa role in the progressive glomerular injury, and that the enhanced CNP activity also plays a compensatory role in obstructive uropathy in the glomerulus.

An Experimental Study on the Change of Stress-related hormone Contents by Prescription of Bunsimgieum (분심기음(分心氣飮) 투여(投與)에 따른 Stress 관련(關聯) Hormone 의 함량변화(含量變化)에 관(關)한 실험적(實驗的) 고찰(考察))

  • In order to study the anti-stress effect of Bunsimgieum, the changes of plasma catecholamine, aldosterone and cortisol levels were measured. In this study author gave immobilization stress to rats, and got the following results: 1. The contents of NE and EPI control group were $2.67{\pm}0.53$, $1.90{\pm}0.22\;ng/ml$, but in sample group the contents were $1.93{\pm}0.67$, $1.15{\pm}0.28\;ng/ml$. The real significance lies in thet only NE show the difference. 2. The contents of aldosterone in control group were $440.69{\pm}37.82\;Pg/ml$, but in sample group $174.64{\pm}42.41Pg/ml$ of aldosterone was measured. The difference was obvious. 3. The cortisol contents in control group were $0.17{\pm}0.03\;ug/dl$, but in sample group $0.28{\pm}0.03 \;ug/dl$ of cortisol was measured. Base on the results above, it may be concluded that Bunsimgieum is effective to prevent to prevent the changes by stress.

  • Hypokalemia usually reflects total body potassium deficiency, but less commonly results from transcellular potassium redistribution with normal body potassium stores. The differential diagnosis of hypokalemia includes pseudohypokalemia, cellular potassium redistribution, inadequate potassium intake, excessive cutaneous or gastrointestinal potassium loss, and renal potassium wasting. To discriminate excessive renal from extrarenal potassium losses as a cause for hypokalemia, urine potassium concentration or TTKG should be measured. Decreased values are indicative of extrarenal losses or inadequate intake. In contrast, excessive renal potassium losses are expected with increased values. Renal potassium wasting with normal or low blood pressure suggests hypokalemia associated with acidosis, vomiting, tubular disorders or increased renal potassium secretion. In hypokalemia associated with hypertension, plasam renin and aldosterone should be measured to differentiated among hyperreninemic hyperaldosteronism, primary hyperaldosteronism, and mineralocorticoid excess other than aldosterone or target organ activation. Hypokalemia may manifest as weakness, seizure, myalgia, rhabdomyolysis, constipation, ileus, arrhythmia, paresthesias, etc. Therapy for hypokalemia consists of treatment of underlying disease and potassium supplementation. The evaluation of hyperkalemia is also a multistep process. The differential diagnosis of hyperkalemia includes pseudohypokalemia, redistribution, and true hyperkalemia. True hyperkalemia associated with decreased glomerular filtration rate is associated with renal failure or increased body potassium contents. When glomerular filtration rate is above 15 mL/min/$1.73m^2$, plasma renin and aldosterone must be measured to differentiate hyporeninemic hypoaldosteronism, primary aldosteronism, disturbance of aldosterone action or target organ dysfunction. Hyperkalemia can cause arrhythmia, paresthesias, fatigue, etc. Therapy for hyperkalemia consists of administration of calcium gluconate, insulin, beta2 agonist, bicarbonate, furosemide, resin and dialysis. Potassium intake must be restricted and associated drugs should be withdrawn.

Effect of Korean Gu-Gi-Ja Tea on Plasma Hormone in Cd-administered Rats (구기자가 카드뮴 급여에 의한 흰쥐의 혈 중 호르몬에 미치는 영향)

  • This study was designed to investigate the effects of Korean Gu-Gi-Ja tea on plasma hormone such as renin and aldosterone water in cadmium administered rats. The cadmium administered rats were given 50 ppm and 100 ppm of CdCl$_2$.2$H_2O$ disolved in the distilled water. Sixty male Sprague-Dawley rats weighing 100$\pm$10 g were divided into 6 groups according to body weight. The control group was fed standard diet without cadmium. The experimental groups, which were fed standard diet containing 50 ppm and 100 ppm cadmium and Gu-Gi-Ja tea group. The results of this study were as follows; food intake, body weight gain and kidney weight content in cadmium administered groups were lower than those in Gu-Gi-Ja tea group. The contents of cadmium in kidney of the rats were determined by using ICP (Inductively Coupled Plasma Spectrcphotometer). In kidney accumulation of Gu-Gi-Ja tea groups were lower than those in cadmium administered group. Plasma levels of renin activity was increased by Cadmium administration group, compared with Gu-Gi-Ja tea and cadmium administred group. Plasma levels of aldosterone activity was increased by cadmium administration group, compared with Gu-Gi-Ja tea and cadmium administred group. This results suggested that Gu-Gi-Ja tea has a lowering effects on the accumulation of cadmium in kidney and it is believed that the Gu-Gi-Ja tea has some protective effects to cadmium administered lenin and aldosterone hormone in rats, but the mechanism of these effects was obscure.

  • Hypertension is characterized by multiple genetic and environmental factors. To establish the genetic basis of hypertension in Koreans, we investigated the genetic variations of two candidate genes (aldosterone synthase (CYP11B2), ${\gamma}$ subunit of the amiloride-sensitive epithelial sodium channel (ENaC) in the Korean patients with hypertension and normotensive controls. There were no significant differences in the genotype and allele frequencies between two groups, respectively. However, there was the significant difference between Korean and Caucasian populations in allele frequency of RFLPs in the two candidate genes. Therefore, these studies also need to be confirmed in other ethnic groups, although our results do not support a possible role of these genes on hypertension in Korean population

  • Objectives : This study was done to investigate the effect of Gamigehyuldeung-tang on hypertension. Methods : After administering Gamigehyuldeung-tang extract to SHR(Spontaneous Hypertensive Rats) for 5 weeks, changes in blood pressure, pulse rate, aldosterone and catecholamine levels in plasma were examined, and immunohistochemical changes and scanning electron microscopic changes were observed. Results : The following results were obtained; blood presure decreased significantly as well as levels of aldosterone, dopamine and epinephrine in SHR. But levels of norepinephrine were unaffected. No capillary vessel dilation was observed. A decrease in cell damage was seen in microscope investigation. Conclusions : These results support a role for Gamigehyuldeung-tang might be usefully applied in treatment of hypertension.

Effect of Ganyangsanghang-bang on Hypertension (간양상항방(肝陽上亢方)이 고혈압(高血壓)에 미치는 영향)

  • This study was done to investigate the effect of Ganyangsanghang-bang(GYSHB) on hypertension. After administering GYSHB extract to SHR for 5 weeks, changes in blood pressure, pulse rate, aldosterone and catecholamine levels in plasma were examined, and immunohistochemical changes were observed, and liver function test was done. The following results were obtained; blood presure decreased significantly as well as levels of aldosterone and norepinephrine in SHR. But levels of dopamine were unaffected. No capillary vessel dilation in adrenal cortex was observed. Safety against hepatic toxicity was showed. These results support a role for GYSHB might be usefully applied in treatment of hypertension.
