Effect of Zinc-enriched Yeast FF-10 Strain on the Alcoholic Hepatotoxicity in Alcohol Feeding Rats

  • The possible protective effects of highly zinc-containing yeast Saccharomyces cerevisiae, FF-10 strain, isolated from tropical fruit rambutan on acute alcoholic liver injury in rats were evaluated. Zinc concentration in this strain was 30.6mg%. The activities of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), and $\gamma$-glutamyl transpeptidase ($\gamma$-GTP) were highly increased when alcohol was treated, relative to the normal rats. Also, a highly significant increase in the blood alcohol and acetaldehyde levels by alcohol treatment was observed. Administration of FF-10 strain markedly prevented alcohol-induced elevation of the activities of serum ALT, AST, and $\gamma$-GTP, and the levels of blood alcohol and acetaldehyde, and these reduced levels reached to that of normal rats. As compared with alcohol treated control rats, the FF-10 strain supplementation showed highly decreased the triglyceride concentration in serum. Alcohol treatment induced the marked accumulation of small lipid droplets, hepatocytes necrosis, and inflammation, but FF-10 strain administration attenuated to alcohol-induced accumulation of small lipid droplets and hepatocyte necrosis in the liver. Therefore, the current finding suggests that zinc-enriched yeast FF-10 strain isolated from tropical fruit rambutan may have protective effect against alcohol-induced hepatotoxicity.

오미자 열매의 물추출물이 알콜대사에 미치는 효과 (Effects of Water Extracts in fruits of Omija (Schizandra chinensis Baillon) on Alcohol Metabolism)

  • To assess the effects of water extracts in fruits of Omija (Schizandra chinensis Baillon) on alcohol metabolism, rats were orally administrated with alcohol (25% alcohol, 0.75g/200g B.W., 40% alcohol, 0.8g/200g B.W.). The level of metabolites and enzyme activities of the serum and liver were unchanged by the 25% ethanol or 40% ethanol treatment with acute orally administration. Blood alcohol level was markdely decreased by the treatment with water extracts in fruits of Omija. The serum level of Urea nitrogen, Free fatty acid, GPT and LDH were tended to decreased, level of GOT was unchanged. Contents of hepatic microsomal protein, glycogen, pyruvate in the liver were increased by water extracts in fruits of Omija. In conclusion, the present study clearly demonstrates that water extract in fruits of Omija promotes the overall metabolism and detoxication of alcohol.

유기성 폐수의 오존전해처리에 관한 연구 (A Study on the Treatment of Organic Wastewater by Ozone Electrolysis)

  • To treat certain wastewater that has alcohol and phenol, we performed the ozone electrolysis by using the titanium electrode. In this experiment, we examined decomposition voltage of organics, time for electrolysis, and removal efficiency of organics. In addition we compared the ozone oxidation electrolysis. The followings are results; 1. When it comes to the alcohol treatment in wastewater, ozone electrolysis showed higher removal efficiency than ozone oxidation or electrolysis. 2. After comparing the decomposition rate of methylalcohol, ethylalcohol, and prophylalcohol in ozone electrolysis, we knew the fact that increasing carbon number made the decomposition rate slow. 3. According to the treatment of alcohol by ozone electrolysis, decomposition voltage was 50V, time for electrolysis was three hours, and treatment acidity was neutral (pH 6.5 - 8.1). 4. Ozone electrolysis was effective to the phenol treatment. When we treated phenol by using ozone electrolysis for three hours, TOC treatment efficiency was 95%. However, ozone oxidation just showed 45% treatment efficiency.

알코올 장기투여시 비타민 또는 Piracetam의 병합투여가 백서 해마와 소뇌 신경세포의 미세형태학적 구조에 미치는 영향 (Effects of Long-Term Alcohol Treatment Combined with Vitamins or Piracetam on the Ultrastructure of Rat Hippocampal and Cerebellar Neurons)

  • This study was carried out to investigate the direct neurotoxicity of alcohol on CNS and the effects of piracetam or vitamins on ultrastructural changes of the rat cerebellar and hippocampal neurons during long-term alcohol treatment. To evaluate the results, quantitative analysis were done for light and electronic microscopic findings. On the light microscopy, red degeneration of pyramidal cells and Purkinje cells was found more apparently in the alcohol only treated group than in the control group. On the electron microscopy, increased lipofuscin pigments were found in cerebellum and hippocampus. In quantitative analysis, vitamins significantly reduced red degeneration in both hippocampus and cerebellum. However, piracetam significantly reduced red degeneration in cerebellum but not in hippocampus. Lipofuscin pigments in Purkinje cells and pyramidal cells were significantly reduced in the alcohol with piracetam treated group than the alcohol only treated group. However, vitamins had no significant reducing effect of lipofuscin pigments in Purkinje cells and pyramidal cells. According to the results, it is concluded that vitamins deficiency might cause red degeneration of pyramidal cell after long-term alcohol treatment, but increment of lipofuscin pigments in pyramidal and Purkinje cell may be caused by alcohol itself or its metabolite rather than vitamins deficiency. Piracetam seems to improve cognitive function impairment caused by alcohol consumption.

알코올 의존 환자에서 한국어판 알코올 충동 척도(Korean Alcohol Urge Questionnaire, AUQ-K)의 신뢰도와 타당도 연구 (The Study on Reliability and Validity of Korean Alcohol Urge Questionnaire(AUQ-K) for Alcohol Dependence)

  • Objectives : The Alcohol Urge Questionnaire(AUQ) has been used in alcohol dependence treatment and research. The goal of this study is to develop of the Korean Alcohol Urge Questionnaire(AUQ-K). Methods : To examine the AUQ-K's psychometric properties, responses from 104 patients admitted in alcohol dependence treatment facility were investigated. Results : The internal consistency of the 8-item AUQ-K, measured by coefficient ${\alpha}$, was high(Cronbach's ${\alpha}$=0.78). AUQ-K scores showed significant correlation when the retest interval was 1 day(p<0.01). The AUQ-K's validity was investigated using correlational analyses with two other craving scales[the Obsessive Compulsive Drinking Scale(OCDS) and the Visual Analogue Scale(VAS)]. The high correlations were obtained between total AUQ-K scores and total OCDS scores, and between total AUQ-K scores and the VAS scores(p<0.01, respectively). Conclusion : The AUQ-K is a reliable and valid short scale for measurement of self-reported alcohol craving. This scale may offer significant advantages over existing single-item measures of alcohol craving in the fields of alcohol dependence treatment and research.

Pine Needle Oil and Korean Medicinal Herb Complex Protect Hyperlipidemia and Liver Cell Damage Induced by Alcohol

  • The effect of treatment with pine needle oil complex (complex of pine needle oil and Korean medicinal herbs) upon rat hepatocytes exposed to alcohol was investigated. We compared body weight gain and ratios of liver and kidney to body weight and the serum biochemistry of rats administered both alcohol and Pine needle oil complex to control rats treated with alcohol alone. Pine needle oil complex treatment resulted in a significant reduction in the levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT) and triglycerides (TG) compared to the control rats. These data suggest that Pine needle oil complex represents an excellent candidate for protection of rat hepatocytes from alcohol-mediated damage.

  • Objectives: This study identified the effects of Korean medicine treatment on a patient with alcohol-associated cirrhosis. Methods: A 50-year-old male with alcohol-associated cirrhosis was treated with Cheongganhaeju-tang from 10 November 2023 to 18 January 2024 to reduce fatigue, dyspepsia, anorexia, and weight loss and to improve laboratory findings, such as liver enzymes. We observed changes in the patient's symptoms and laboratory findings during a treatment period of approximately 2 months. Results: With Cheongganhaeju-tang treatment, serum levels of liver enzymes remained stable, clinical symptoms improved, and alcohol cravings decreased. Conclusion: This case points to Cheongganhaeju-tang as a therapeutic option to manage alcohol-associated cirrhosis.

알콜과 식이지방량이 흰쥐의 간 지질조성과 간조직형태에 미치는 영향 (Effects of Alcohol Consumption and Fat Content in Diet on Chemical Composition and Morphology of Liver in Rat)

  • Effects of alchohol and fat content in a balanced diet on chemical composition and morphology of liver were investigated in growing rats. Fourth eight male rats of Sprague-Dawley strain weighing about 160g were divided into 4 groups ; high fat diet group, alcohol-administered high fat diet group, low fat diet group and alcohol-administered high fat diet group, low fat diet group and alcohol-administered low fat diet group. High and low fat diets supplied 30% and 12%, respectively, of total calorie intake from fat, and alcohol was given by adding ethanol in drinking waster at 10%. Diets contained adequate amounts of all nutrients required for rats, including lipotrpoic agents(choline and methionine) to minimize effects of factors other than alcohol on liver damage. Ratios of liver weight to body weight were statistically different among groups. Liver/dody weight ratios alcohol-administered rats were significantly higher than those of non-alcohol groups after 6 weeks treatment. Although total lipid and triglyceride per gram liver were increased in alcohol-administered rats, especially low fat diet fed rats, the values were not significantly different. Opticmicroscopical observation revealed increase in cell size and no change in morphology of liver. Examination of hepatocytes by electron microscopy showed that fat droplets were observed in all groups but enlarged in the alcohol-administered low fat diet fed rat. Contents of protein, cholesterol and phospholipid were not affected by alcohol consumption. The level of lipid peroxide was significantly lower in the livers of alcohol-administered rats than in the livers of non-alcohol groups. The results of this study indicate that even moderate alcohol drinking and dietary fat content did not affect any significant change in composition and morphology of liver until 6 week treatment but that even moderate alcohol drinking caused some signs of steatosis of liver.

A Prospective, Randomized, Controlled Trial to Assess the Efficacy of a Multi-Disciplinary Screening, Brief Intervention and Referral to Treatment Program for Patients with Fractures of the Oral and Maxillofacial Region Because of Alcohol-Related Injuries in the Emergency Department

  • Purpose: We have implemented a multi-disciplinary Screening, Brief Intervention and Referral to Treatment (SBIRT) protocol to prevent individuals who sustained alcohol-related traumatic injuries. We therefore conducted this single-center, prospective, randomized, controlled trial (RCT) to assess its efficacy. Methods: All the enrolled patients (n=30) were randomized to either the SBIRT group or the control group. In the current RCT, the proportion of the patients who reduced the amount of alcohol consumption and those who received a specialized treatment served as primary outcome measures. Moreover, changes in a 3-item version of the Alcohol Use Disorders Identification Test Consumption (AUDIT-C), Severity of Dependence Scale (SDS) and Kessler Psychological Distress Scale (K-6) scores at 3 months from baseline served as secondary outcome measures. Results: At 3 months, the proportion of the patients who reduced the amount of alcohol consumption was significantly higher in the SBIRT group as compared with the control group (86.7% vs. 57.1%, p=0.02). Moreover, the proportion of the patients who received a specialized treatment was also significantly higher as compared with the control group (26.7% vs. 1.4%, p=0.01). Furthermore, there were significant differences in changes in the AUDIT, SDS and K-6 scores at 3 months from baseline between the two groups (p<0.05). Conclusions: In conclusion, our results indicate that the SBIRT is effective in reducing hazardous and harmful levels of drinking, the degree of alcohol dependence and that of psychological distress in at-risk drinkers.

Alcohol Impairs learning of T-maze Task but Not Active Avoidance Task in Zebrafish

  • The aim of this study is to investigate whether alcohol alters learning and memory processes pertaining to emotional and spatial factors using the active avoidance and T-maze task in zebrafish. In the active avoidance task, zebrafish were trained to escape from one compartment to another to avoid electric shocks (unconditioned stimulus) following a conditioned light signal. Acquisition of active avoidance task appeared to be normal in zebrafish that were treated with 1% alcohol for 30 min for 17 days until the end of the behavioral test, and retention ability of learned behavior, tested 2 days later, was the same as control group. In the T-maze task, the time to find a reservoir was compared. While the latency was similar during the 1 st training session between control and alcohol-treated zebrafish, it was significantly longer in alcohol-treated zebrafish during retention test 24 h later. Furthermore, when alcohol was treated 30 min after 2nd session without prior treatment, zebrafish demonstrated similar retention ability compared to control. These results suggest that chronic alcohol treatment alters spatial learning of zebrafish, but not emotional learning.
