Effects of Minerals Added to Medicinal Plant Extracts on Alcohol-Induced Oxidative Stress and Alcohol Metabolism in Rats (약용식물 추출물이 첨가된 미네랄이 알코올에 의한 산화적 스트레스 및 숙취해소에 미치는 효과)

  • This study investigates the effects of a hangover beverage (MIX) that contains minerals (highly-salty mineral water, HSMW) and several medicinal plant extracts, on antioxidant and alcohol-metabolizing enzymes in alcohol administered Sprague-Dawley rats. HSMW is pumped from below the sedimentary rock layer of Dadaepo, Busan, South Korea, which is 1,050 m below the land surface; it tastes salty, like sea water. In terms of medicinal plant extracts, the total phenolic and flavonoid contents of Rubus coreanus and Cornus officinalis were measured as being significantly higher than those in Curcuma longa. The results suggest that treatment with MIX significantly increases superoxide dismutase (SOD) activity and DPPH radical scavenging activity. In the 10% HSMW-, for MIX- and company product (CP)-treated groups, the concentration of blood alcohol was significantly reduced 1~5 hr after alcohol loading, compared to that in the control group. In hepatic alcohol-metabolizing enzyme activities, alcohol dehydrogenase (ADH) activity was found to be higher in the MIX- and CP-treated groups than in controls, whereas acetaldehyde dehydrogenase (ALDH) activity was significantly higher in the CP-treated groups than other groups. This study concludes, therefore, that MIX (HSMW) minerals, like as Zn, Ca, Mg, Mn, and others stimulate alcohol-metabolizing enzymes, while the antioxidants of plant extracts prevent the damage otherwise incurred by alcohol toxicity. These results suggest that the hangover beverage (MIX) alleviates alcohol hangover symptoms by stimulating activities related to hepatic alcohol-metabolizing enzymes and antioxidant effects.

A Study on the Extraction and Efficacy of Bioactive Compound from Hovenia dulcis (헛개나무로부터 생리활성물질의 추출 및 효능에 관한 연구)

  • The biological activities of extracts from the fruit, stem, and leaf of Hovenia dulcis were examined. In the batch mode of operation, the fruit, stem, and leaf of Hovenia dulcis were extracted with hot water for 10 hr. The fruit extract of Hovenia dulcis gave the highest activity for decreasing alcohol concentration which was 138% of control. The equilibrium between bioactive compound in the fruit (size : 4 mm) and hot water solution was reached within 6 hr and the recovery was 95% by three-times extraction. The fruit extract of Hovenia dulcis showed significant alcohol decrease in blood and hepatoprotective activity against $CCl_4$-toxicity in rat. The fruit extract significantly inhibited the elevation of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), and lactate dehydrogenase (LDH) levels.

Protective effects of extracts from the aerial parts of hydroponically cultured ginseng on alcohol-induced liver damage in mice and quantitative analysis of major ginsenosides (알코올성 간손상을 유발한 마우스 모델에서의 새싹인삼 지상부 추출물의 간 기능 보호효과 및 지표성분 함량분석)

  • We studied the effects of the extract of aerial parts of hydroponically cultured ginseng (HGE) on alcohol-induced liver damage (AILD) in mice. AILD was induced by the oral administration of ethanol (EtOH) (25%; 5 g/kg body weight) for seven days in the study as well as EtOH-only groups. However, HGE (4 and 12 mg/kg) was orally administered (once daily for ten consecutive days) only to the study group, three days prior to the EtOH treatment. The HGE-treated group showed significantly lower levels of alanine aminotransferase and aspartate aminotransferase than the EtOH-only group. In addition, HGE administration decreased the level of serum lactate dehydrogenase, a known marker of liver damage. The effect of HGE on AILD was found to be dose dependent, and the consecutive administration of HGE showed no side effects in mice. Our study indicates that HGE treatment can potentially reduce oxidative stress and toxicity in the liver of alcohol-treated mice and that HGE can be a useful therapeutic agent for alcohol-induced hepatotoxicity. Additionally, a simple and efficient high-performance liquid chromatography-ultraviolet detection method was developed for determining the contents of four major ginsenosides in HGE. The aerial parts of hydroponically cultured ginseng were extracted using 70% fermented ethanol, and the contents of ginsenosides F5, F3, F1, and F2 in HGE were found to be 2.5, 4.4, 1.4, and 23.3 mg/g, respectively.

  • The toxicity of chlorothalonil on the growth of yeasts was investigated using several yeast strains. An alcohol tolerant yeast, Saccharomyces cerevisiae F38-1, was the most chlorothalonil-tolerant. The glutathione content and the glutathione S-transferase activity were related to the chlorothalonil-tolerant phenotype. Several thiol compounds affect the dissipation of chlorothalonil. However, there was no significant difference on the effects of chlorothalonil dissipation among the thiol compounds tested. The growth of yeast cells was arrested by chlorothalonil. It took about 13 h to dissipate 1 mg/l of chlorothalonil, and the growth was restored as the chlorothalonil content decreased. The glutathione content and glutathione S-transferase are suggested to be among the most important factors of yeast resistance to chlorothalonil.

  • Biosurfactant have a low toxicity, good biodegradable, various structure. It can be used for a broad range of purposes in a large variety of different applications. Sorbitol mono alkyl ester synthesized by the Porcine Pancreatic lipase catalyzes transesterification reaction between a sugar alcohol and plant oils. We are examined the surface tension, emusion stability and critical micelle concentration were evaluated. Consequently It has a good properties of biosurfactant.

  • The water extracts of Hovenia dulcis Thunb. showed significant hepatoptotective activity against D-galactosamine/LPS and $CCl_4-toxicity$. The extracts significantly inhibited the elevation of serum ALT, AST and LDH levels. The concentration of blood alcohol were decreased from 2hr after injected in rats with Hovenia dulcis extracts.

Suppressive Effect of Lactic Acid Bacteria on the Toxicity of Bisphenol A in Rats

  • We have examined if lactic acid bacteria could suppress the toxic effect of bisphenol A. Lactobacillus casei YA-70 was chosen as representative. Thirty rats were divided into two groups (immature and mature) according to the weight. Each group was divided again into the control group (only alcohol treatment), bisphenol A treated group, and bisphenol A plus Lactobacillus casei YA-70 treated group. When 500 ppm of bisphenol A was fed everyday, 83% of immature group and 50% of mature group died within 3 weeks. Their internal organs, mainly livers and lungs, were changed in color and severely damaged. In the intestine of 5 ppm-fed group tumor-like nodules were observed. However, their number and size were markedly decreased when Lactobacillus casei YA-70 was supplemented in diet. This study strongly indicates that Lactobacillus casei YA-70 might play an important role to suppress the toxic effect of endocrine disruptor.

  • Objective : This research was carried out to compare the effect of Gorynju and a Soju extract Psoraleae fructus on melanin synthesis of B16 melanoma cells. Methods : To investigate melanin synthesis of B16 melanoma cells, this research was measured cell survival, tyrosinase activity, melanin synthesis, western blot. Results : Both Gorynju and Soju extract Psoraleae fructus, cell toxicity depended on the density. Tyrosinase activity depended on the density of Gorynju extract Psoraleae fructus and statistic was showed significant(0.5, 1, 2, 3 ${\mu}g/ml$), in a Soju extract Psoraleae fructus, 1 ${\mu}g/ml$ were showed significant. Melanin synthesis was showed significant in a Soju extract Psoraleae fructus(3, 4 ${\mu}g/ml$). Western blot was showed to depend on the density of Gorynju and a Soju extract Psoraleae fructus. Conclusions : In a tyrosinase activity and a melanin synthesis, the intermediate alcohol of Gorynju and a Soju may be suitable to use.

Anti-inflammatory Modulating Effect of Rengyolone in Rat

  • Hepatitis refers to inflammation of hepatocytes and liver tissue, and is mainly caused by viruses, alcohol, and drugs. Forsythiae Fructus has traditionally been used as a diuretic, anti-inflammatory and antipyretic. Research on rengyolone, a bioactive substance extracted from Forsythiae Fructus, is rarely found in Korea and abroad. First, an acute animal toxicity test for rengyolone was conducted for the animal experiment. 4 week-old SD rats were injected intraperitoneally with acetaminophen for 2 weeks to induce chronic liver inflammation. Rengyolone was orally administered into two groups during 4 weeks: pre-inflammatory group and post-inflammatory group. Oral doses were also divided into 1 mg/kg and 5 mg/kg. Liver function tests (ALT, AST, ALP), western blot analysis of liver tissue, and level of inflammatory cytokine were performed to evaluate the improvement of hepatitis. Experimental results showed that rengyolone inhibited the development of acute inflammation and thus could reduce hepatitis symptoms.

Effects of Monascus-fermented Angelica gigas Nakai on the Contents of Serum Lipid and Tissue Lipid Peroxidation in Alcohol Feeding Rats (알코올성 간독성 흰쥐의 혈청 지질 및 조직 과산화지질 농도에 미치는 발효당귀의 영향)

  • The effect of Monascus-fermented Angelica gigas Nakai (AFAG) on the contents of serum lipids and tissue lipid peroxidation was investigated in alcohol feeding rats (Alc group). The serum contents of total lipid and free fatty acid in the alcohol feeding rats were significantly increased, but these increases tended to decrease in the AFAG group. The content of serum triglyceride was also significantly decreased in the AFAG group compared to the other groups. The serum content of total-cholesterol was not significantly different between the normal group and the AFAG group. The content of HDL-cholesterol in serum was slightly increased in the AFAG group compared to the Alc group. The content of thiobarbituric acid reactive substances (TBARS) in the liver, heart, spleen, and testis were significantly increased in the Alc group compared to the normal group, but these increases were significantly decreased in the AFAG group. The content of liver zinc was decreased in the Alc group and it was significantly increased in the AFAG group, which suggested that the lipid peroxidation contents are inversely correlated with the liver zinc content. The hepatic glutathione concentration was significantly decreased in the Alc group, but this content was significantly increased in the AFAG group, and it showed the antioxidant ability of glutathione. These activities were also compared to the standard silymarin drug treatment. Thus, the findings of the present study indicated the significant antioxidant and antihyperlipidemic activity of Monascus-fermented Angelica gigas Nakai against ethanol-induced toxicity.
