바이오짐의 에탄올 대사에 대한 영향 (Effects of Biozyme on the Ethanol Metabolism in vivo and in vitro)

  • Effects of $Biozyme_{R}$ and $\textrm{Business}_{R}$ on alcohol metabolism in rats, and on the activities of alcohol dehydrogenase(ADH) and acetaldehyde dehydrogenase(ALDH) were studied in vitro. Alcohol concentration in rat blood was decreased after the treatment of Business(3.3 mι/kg, Biozyme 1.67 mg/wι) and Biozyme(3.3 mι/kg, 1.67 mg/mι) prior to the administration of ethanol(25%, 0.83 g/kg). And the acetaldehyde concentration of rat blood was also decreased when compared with control values in the same condition. Effects of Biozyme on ADH and ALDH activity were also studied. While the ALDH activity was elevated in the presence of Biozyme(2 $\mu\textrm{g}$/assay), the ADH activity was not influenced by Biozyme at the concentration range from 2 $\mu\textrm{g}$/assay to 0.2 mg/assay. In summary, Biozyme accelerated the rate of ethanol metabolism and the acceleration might be due to the increase in ALDH activity.vity.

  • Spraque-Dawley계 응성흰쥐에 계통분획한 cornsilk의 각 분획물을 경구투여하고 혈청 Ethanol농도와 간의 Alcohol dehydrogenase활서에 미치는 효과를 검색 추적한 결과 알코올대사를 촉진시키는 성분은 주로 에탄올가용부에, 억제시키는 성분은 에탄올불용부에 주로 존재함을 추정할 수 있었다.

  • These studies were conducted to investigate the alcohol degradation effects of the extract of herbal composition (DTS20) containing Viscum album L., Lycium chinense L., Inonotus obliquus and Acanthopanax senticosus H., on the alcohol administered mice. To investigate anti-hangover effect, alcohol and alcohol dehydrogensae (ADH) concentration of blood were measured after oral administration of ethanol. The administration of DTS20 (200-500 mg/kg) had beneficial actions toward alcohol degradation in acute alcohol treated mice model. The oral administration of DTS20 showed decreased gastric mucous membrane damage produced in ethanol treated mice. In addition, intraperitoneal administration of DTS20 showed anti-inflammatory effects in inhibition tests of vascular permeability produced by acetic acid. DTS20 also reduced the concentration of nitric oxide (NO), tumor necrosis factor (TNF)-${\alpha}$ in macrophages that were activated by LPS. These results demonstrate that DTS20 possesses potential to stimulate the alcohol degradation and inhibit the inflammatory effects in mice.

Relationship between BrACs and BACs of Healthy Koreans for BAIIDs

  • This study aims to suggest the partition ratio (Q-factor) of healthy Koreans and the comparison results of breath alcohol concentration (BAC) measurements using two methods (photoacoustic and electrochemical methods) for developing breath alcohol ignition interlock devices (BAIIDs). Given the relationship between BACs and BrACs and the Q-factor, the alcohol metabolism of healthy Koreans (96 males and 91 females) is revealed for understanding the digestion of alcohol and surveying the fundamental data of alcohol-related problems, CO2 concentrations vs. alcohol concentrations, and the performance of alcohol sensors in the marketplace. The average Q-factor of healthy Korean males and females are 1,913 (95% confidence interval from 1,889-1,937) and 1,991 (95% confidence interval from 1,945-2,036). Photoacoustic measurements could be applied to predict the BACs of drinkers, which is confirmed by the Bland-Altman plots presented in this study. The biases based on the partition ratios (Q=1,913 and Q=1,991) in the Bland-Altman plots were -0.0004% (95% CI from -0.0011 to +0.0003% for males) and -0.0017% (95% CI from -0.020 to +0.017% for females).

생체 신호 기반 음주량 예측 및 음주량에 따른 운전 능력 평가 (Prediction of Alcohol Consumption Based on Biosignals and Assessment of Driving Ability According to Alcohol Consumption)

  • Drunk driving defines a driver as unable to drive a vehicle safely due to drinking. To crack down on drunk driving, alcohol concentration evaluates through breathing and crack down on drinking using S-shaped courses. A method for assessing drunk driving without using BAC or BrAC is measurement via biosignal. Depending on the individual specificity of drinking, alcohol evaluation studies through various biosignals need to be conducted. In this study, we measure biosignals that are related to alcohol concentration, predict BrAC through SVM, and verify the effectiveness of the S-shaped course. Participants were 8 men who have a driving license. Subjects conducted a d2 test and a scenario evaluation of driving an S-shaped course when they attained BrAC's certain criteria. We utilized SVR to predict BrAC via biosignals. Statistical analysis used a one-way Anova test. Depending on the amount of drinking, there was a tendency to increase pupil size, HR, normLF, skin conductivity, body temperature, SE, and speed, while normHF tended to decrease. There was no apparent change in the respiratory rate and TN-E. The result of the D2 test tended to increase from 0.03% and decrease from 0.08%. Measured biosignals have enabled BrAC predictions using SVR models to obtain high Figs in primary and secondary cross-validations. In this study, we were able to predict BrAC through changes in biosignals and SVMs depending on alcohol concentration and verified the effectiveness of the S-shaped course drinking control method.

Effect of Beverage Containing Fermented Akebia quinata Extracts on Alcoholic Hangover

  • The present study was conducted to investigate the effects of beverages containing fermented Akebia quinata extracts on alcoholic hangover. For this study, 25 healthy young men were recruited. All participants consumed 100 mL of water (placebo), commercial hangover beverage A or B, fermented A. quinata leaf (AQL) or fruit (AQF) extract before alcohol consumption. After 1 h, all participants consumed a bottle of Soju, Korean distilled liquor (360 mL), containing 20% alcohol. Blood was collected at 0 h, 1 h, 3 h, and 5 h after alcohol consumption. The plasma alanine transaminase (ALT) activity was highest in the placebo group. Compared with the control group, the AQL and AQF groups showed decreased ALT activity at 5 h after alcohol consumption. Plasma ethanol concentration was increased after alcohol intake and peaked at 3 h after alcohol consumption. Compared with the control group, the A group showed a higher plasma ethanol concentration at 1 h (P<0.05). At 3 h after alcohol consumption, the AQF group showed the lowest mean plasma ethanol concentration compared to the other groups; however, there were no statistical differences. After 5 h of alcohol consumption, the AQL and AQF groups showed lower plasma ethanol concentrations compared with the B group. The sensory evaluation score for the fermented A. quinata fruit extract was lower than for the commercial hangover beverages. In conclusion, the present intervention study results suggest that fermented A. quinata extracts alleviate alcoholic hangover and reduce plasma ethanol concentrations.

효모 K. marxianus에 의한 돼지감자 착즙에 대한 에탄올 발효 특성 (Ethanol Fermentation Characteristics of K. marxianus on Jerusalem Artichoke tuber Extract)

  • 돼지감자 착즙의 당농도를 50,80,110,190,250g/l로 하고 K.marxianus의 비증식속도,균체수율,알콜수율등을 발효액주으이 생성알콜농도및 당 소모량의 함수로 구명한 결과 다음의 결론을 얻었다. 생성알콜농도에 대한 비증식속도 및 알콜의 비생성속도의 함수관계는 알콜생성량이 낮을때 선형 함수관계를, 알콜생성량이 높을때는 지수 함수관계를 나타내었다. 균체의 증식은 생성알콜농도가 50g/l이상에서, 그리고 당의 소모량이 100g/l이상에서 잔여 당농도에 무관하게 정지되는 경향을 나타내었다. 비증식속도와 당 소모량사이에는 선형 함수관계를 나타내었으며,알콜 비생성속도와 당소모량사이의 함수관계는 초기당농도가 낮을때 선형 함수관계를 나타내었으나 초기당농도가 높아질수록 지수함수관계로 천이되었다. 균체수율및 알콜수율은 초기당농도에 무관하게 생성알콜농도 25g/l 부근에서 최대치를 나타내었으며, 알콜생성량이 증가함에 따라 초기 당농도가 증가할수록 완만하게 감소하였다. 이상의 함수관계로부터 K.marxianus의 알콜발효에 있어 한계 당농도는 110g/l근방으로, 한계 알콜농도는 50g/l근방인 것으로 추정되었다.

알코올의 섭취가 식이내 Selenium과 Vitamin E 수준을 달리한 흰쥐의 체내 Selenium 수준과 심장 및 간조직에 미치는 영향 (The Effect of Alcohol Administration on Selenium Concentration and Cell Morphology of Heat and Liver of Rats Fed with the Different Levels of Selenium and Vitamin E)

  • 알코올의 섭취가 Se가 vitamin E 수준을 달리한 흰주의 체내 Se 수준 및 심장, 간세포 형태에 미치는 영향을 살펴보기 위하여 평균 체중이 58~62g인 Sprague-Dawley계의 젖을 갖뗀 숫쥐 72마리를 12군으로 나누어 Se의 수준을 식이 kg당 0mg, 0.4mg,10mg으로 달리하고 vitamin E는 식이 Kg당 0mg, 150mg으로 하여 7주간 사육시켰다. 알코올 섭취는 실험시작 3주째부터 ethanol을 급수물에 10% 맞추어 섭취군과 비섭취군으로 나누어서 섭취군에서는 제한없이 먹게 하였다. 얻어진 결과를 요약하면 다음과 같다. 혈액과 소변의 Se의 수준은 식이내 Se 수준의 차이에 따라 유의적인 차이를보여 식이내 Se수준이 높은 H-군, 정상적으로 섭취한 C-군, 결핍된 L-군의 순으로 높았으며 알코올 섭취군들은 낮은 경향을 보였다. 간장, 신장의 Se의 농도는 식이 Se 수준에 비례하여 H-, C-, L- 순으로 높았으며 알코올 섭취로 인한 Se농도의 변화는, 알코올을 섭취한 군둘의 Se 농도가 낮은 경향이나 신장에서는 반대로 높은 경향이었다. 전자현미경을 통한 관찰에서 심근세포 조직형태의 변화는 식이내 Se수준이 높은 H-군들, Se수준이 낮은 L-군에서 lysosome의 증가 및 지방적 출현, mitochondria의 종창, 특히 모세혈관의 내피세포에 심한 종창이 일어났고, 알코올을 섭취한 군들에도 심근세포의 종창, lysoscme의 증가, mitochondria 변화를 보였다. Se을 과잉으로 섭취하고 vitamin E를 정상으로 섭취한군(HC-)에서는 특이한 변화가 없었으나 알코올을 투여한 HCA군에서 지방적이 나타났다. 특히 Se과 vitamin E이 결핍되고 알코올을 섭취한 LLA군에서는 lysosome의 증가 및 지방적이 많이 증가 하였다.

비열 및 열처리에 따른 보리의 알코올발효 특성 (Properties of Alcohol Fermentation from Barley Treated with Non-steam and Steam)

  • 본 연구에서는 비열 및 열처리에 따른 5종류의 보리 품종별 알코올발효 특성을 조사하였다. 비열처리구에서 알코올 함량은 쌀보리, 찰쌀보리 및 흰찰쌀보리가 각각 1.7, 2.9 및 4.3%로 나타났으며 보리와 찰보리는 발효가 되지 않았다. 열처리구에서 비열처리구에 비해 전반적으로 골은 알코올생성능을 보였다. 당도는 비열처리구의 보리 및 찰보리가 $9.0\;^{\circ}Brix$ 전후의 높은 당도를 나타내었고 쌀보리, 찰쌀보리, 흰찰쌀보리가 $5.0\;^{\circ}Brix$ 전후의 당도로 나타났다. 열처리구의 당도는 알코올생성능과 반비례 경향이었으며 알코올발효 종료 후 쌀보리를 제외하고는 당함량이 50%이상 감소되었다. 비열처리구에서 산도는 흰찰쌀보리가 0.51%로 나타났으며 나머지 시료들은0.80%이상의 수치를 보여 산패의 가능성을 보였고, 열처리구에서는 모든 시료들이 0.3%이하의 수치로 나타났다. 유리당은 비열처리구의 보리와 찰보리가 알코올을 생성하지 않았으므로 유리당 총함량은 4,000.0 mg%전후로 높게 나타났고 쌀보리, 찰쌀보리 및 흰찰쌀보리는 300.0mg%전후의 함량으로 나타났다. 열처리구에서는 당도가 높을수록 유리당 총 함량이 높게 나타났으며 이상의 결과 보리는 열처리에 비해 비열처리 알코올 발효에는 적합하지 않았다.

Effect of Lactobacillus fermentum MG590 on Alcohol Metabolism and Liver Function in Rats

  • Alcohol consumption has numerous health consequences for the human body. For example, heavy drinking on a daily basis causes liver diseases, and certain products such as acetaldehyde produced from alcohol metabolism are more toxic than alcohol itself. Accordingly, the current study evaluated the role of Lactobacillus fermentum MG590 to enhance the removal of the toxic effect of alcohol in alcohol metabolism. The maximum activities of the alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) by L. fermentum MG590 were observed after 6 h of culture. The production of ADH and ALDH by L. fermentum MG590 was also confirmed by SDS-PAGE. Six hours after the addition of alcohol to a culture broth of L. fermentum MG590, the alcohol concentration decreased from 7.5 to 2.7%. From an in vitro evaluation based on hepatocytes, the viability of hepatocytes in a medium containing alcohol and the cytosol of L. fermentum MG590 was higher than that in a medium containing only alcohol. From an in vivo test using SD rats fed a 22% alcoholic drink, the blood alcohol concentration (BAC), glutamic-oxaloacetic transaminase (GOT), and glutamic-pyruvic transaminase (GPT) in the rats fed a medium containing L. fermentum MG590 were lower than those in the rats fed a medium containing only the alcohol drink. These results demonstrate that the ADH and ALDH produced by L. fermentum MG590 play an important role in detoxicating alcohol in vivo. Therefore, a fermentation broth of L. fermentum MG590 could be used as an effective alcohol detoxification drink.
