Influences of Protease on the Removal of Protein Soils from Cotton Fabrics -Emphasis on the Characteristics of Enzyme and Soil Substrates- (단백질 분해 효소가 세척에 미치는 영향 -효소와 오염 기질의 특성을 중심으로-)

  • The influences of protease on the removal of various protein soils from cotton fabrics were studied. The human epidermal stratum corneum, hemoglobin and casein were used as protein soils. The soiled fabrics were denatured by steaming for 30 min. before washing and laundered using Terg-O-Tometer under washing conditions. The removal efficiency was evaluated by analysis of protein on the fabrics before and after washing by means of copper-Folin method. The relations between the removal and the characteristics of protease were discussed. Also the degradation of protein were examined by microscopy. The seperation of human epidermal stratum corneum after hydrolysis was examined by SDS-PAGE. The results obtained were as follow : 1. The protein from the soiled cotton fabric was removed effectively by adding protease. The removal of protein was increased in proportion to increasing of the enzyme concentration up to a certain point, but it began to decrease above the point. The removal effect was high in the order of casein>human epidermal stratum corneum>hemoglobin. Especially the protein was more effectively removed in ADS solution(pH 9.5) containing enzyme. 2. When protease was used with ADS. the removal of protein was efficiently showed in relatively short time(5~15min.) compared to using ADS only. It is due to the properties of this enzyme that reacts with very short time. 3. Even at low temperature the removal efficiency of enzyme was relatively higher compared with the activity of enzyme. The removal of protein soil was increased up to a maximum near $50^{\circ}C$, and then decreased. 4. The removal of protein by protease was improved with the increase of alkalinity in the pH range from 9.5 to 11.0 but it began to decrease above pH 11.0. 5. According to the increase of mechanical agitation, the removal effect was increased. But the removal efficiency of protease was more effective compared with the agitation in detergency. 6. According to the SDS-PAGE separation and micrograph it was confirmed that the human epidermal corneum was effectively hydrolysed by the enzyme added. So the fragments of protein were removed more efficiently by means of the interfacial reaction of AOS.

Effect of Oxygen Transfer Rate and Dissolved Oxygen on the Production of PHBV by Azoto-bacter vinelandii UWD. (산소전달 속도와 용존산소가 Azotobacter vinelandii UWD의 생분해성 고분자(PHBV) 생산에 미치는 영향)

  • In a 20 L fermentor experiments the level of dissolved oxygen (D.O.) strongly affected growth and PHBV production of Azotobacter vinelandii UWD. A higher D.O. (5%) increased specific cell growth rate two folds but PHBV production was 17 folds higher (62.3 wt%) at a lower D.O.(1%) level. D.O. level was not a good criterion to evaluate the effect of aeration on fermentation characteristics of A. vinelandii UWD. This strain maintained an equal D.O. (5%) by decreasing its oxygen consumption rate when oxygen transfer rate (OTR) was decreased by changing agitation speed at a fixed aeration rate. OTR rather than D.O. was a criterion to explain the effect of aeration on the cell growth and PHBV production. At 5% D.O. with a lower 0TR cell growth rate decreased but PHBV production (57.3 wt%) approached to that (62.3 wt%) of the lower (1%) D.O.

  • An experimental study was carried out to investigate the effect of the inclination angle on the nucleate pool boiling of saturated water at atmospheric pressure. We considered an annulus with a gap of 5 mm and a bottom opening. The inner tube of the annulus was heated, and the outer diameter and the length of the tube were 25.4 mm and 500 mm, respectively. The inclination angle was varied from horizontal to vertical. The results were compared to those for an annulus with a larger gap and a single tube. In the small-gap annulus, the effect of the inclination angle on the heat transfer was not significant. However, an early onset of the critical heat flux was observed at 80 kW/$m^2$ when the annulus was horizontal. Liquid agitation and bubble coalescence were considered to be the major heat-transfer mechanisms.

  • The effects of stirring speed during filler modification by dual polymers on flocculation and reflocculation of PCC (precipitated calcium carbonate) particles and its effect on handsheet properties were elucidated. PCC surface was modified by adsorbing A-PAM (anionic polyacrylamide) and C-starch (cationic starch) in series at various stirring speeds. It was found that increasing stirring speed during filler modification decreased the initial floc size of PCC. Continuous stirring with the same speed for filler modification resulted in the decrease of a floc size, eventually reached a steady state. The variations in a floc size was influenced by the stirring speed during filler modification: the lower the stirring speed during filler modification, the larger the floc size variations. Conclusively, the stability of PCC floc could be improved by increasing the stirring speed. In addition, the stirring speed influenced the handsheet properties. The smaller the PCC floc, the lower the strength of handseet. However, too much larger floc size also deteriorated paper strength. There exists an optimum floc size in term of paper strength which shall be controlled by stirring speed during filler modification.

The Improvement of Cephalosporin C Production by Fed-batch Culture of Cephalosporium acremonium M25 Using Rice Oil

  • The objective of this study is to improve cephalosporin C (CPC) production byoptimization of medium and culture conditions. A statistical method was introduced to optimize the main culture medium. The main medium for CPC production was optimized using a statistical method. Glucose and corn steep liquor (CSL) were found to be the most effective factors for CPC production. Glucose and CSL were optimized to 2.84 and $6.68\%$, respectively. CPC produc­tion was improved $50\%$ by feeding of $5\%$ rice oil at day 3rd and 5th day during the shake flask culture of C acremonium M25. The effect of agitation speeds on CPC production in a 2.5-L bio­reactor was also investigated with fed-batch mode. The maximum cell mass (54.5 g/L) was obtained at 600 rpm. However, the maximum CPC production (0.98 g/L) was obtained at 500 rpm. At this condition, the maximum CPC production was improved about $132\%$ compared to the re­sult with batch flask culture.

  • Snoezelen, multi-sensory stimulation, provides stimulation through the visual, aural, tactile, olfactory, gustatory and proprioceptive senses. Its aim is to be a relaxing, feeling of safety, novelty and stimulation with no expectations for performance. Agitated behavior of people with dementia causes significant problems and distress for cares. The objective of this research study was to test the effects of Snoezelen room on physical and verbal agitated behaviors of people with dementia. Four patients participated five sessions in Snoezelen room of 'F' dementia care facility in Sweden. Assessment instrument was basically used Cohen-Mansfield Agitation Inventory-Short Form to be rated by caregivers on a 5-point frequency scale. Pre-trial and post-trial assessments were carried out during $27^{th}$ Aug.-$27^{th}$ Sept. 2007. Result shows that Snoezelen room has some short-term positive effect on agitated behavior even though there was a considerable variation individuals responded. This result adds to the increasing evidence that Snoezelen could be a therapeutic environment for the elderly with dementia. Further research study, with larger number of participation, and a control group is required to establish more scientific evidence of Snoezelen effects on health outcome.

Physiochemical Changes and Optimization of Phosphate-Treated Shrimp (Litopenaeus vannamei) Using Response Surface Methodology

  • The objective of this study was to determine the factors responsible for the changed physiochemical properties of unpeeled shrimp treated in cold phosphate solution ($2{\sim}4^{\circ}C$) with the intervention of 4 factors: phosphate concentration, dipping time, rotation speed, and volume of brine solution. Response surface analysis was used to characterize the effect of the phosphate treatment on shrimps by running 33 treatments for optimizing the experiment. For each treatment, phosphate amount, moisture content, and weight gain were measured. The results showed that phosphate concentration is the most important factor than other factors for facilitating phosphate penetration in the meat of the shrimp and for getting the best result. The optimum condition of phosphate-treated shrimp in this study was 110 to 120 min dipping time, 500 to 550 mL brine solution for 100 g shrimp sample, and 190 to 210 rpm agitation speed. The studied conditions can be applied in fisheries and other food industries for good phosphate treatments.

The Effects of Water-and Oil-Repellent Finishes on the Deposition and Removal of Oily Soils from Polyester Fabrics (폴리에스테르 직물의 발수발유 가공처리가 유성오구의 부착 및 제거에 미치는 영향)

  • The effects of water-and oil-repellent finishes on the deposition and removal of oily soils from polyseter fabrics were investigated in this study. The polyester fabrics treated with three kinds of fluoropolymers(TG-410H, TG-990) were soiled with mixed oily soils and washed by using Terg-O-Tometer at various conditions. It was found that TG-410H and TG-527 treated polyester fabrics reduced significantly the deposition of oily soils than untreated and TG-990 treated polyester fabrics. The removal of oily soils from polyester fabrics was increased in porportion to increasing of surfactant concentration up to a certain point(0.2% owb) but it levelled off above the point. The removal effect was high in the order of TG-990 treated polyester fabrics > untreated polyester fabrics > TG-527 treated polyester fabrics > TBG-410H treasted polyester fabrics. Especially the removal of oily soils from polyester fabrics was more effectively removed in triton X-100 solution. In general the removal of oily soils from polyester fabrics treated with fluoropolymers was increased up to a miximum near 6$0^{\circ}C$ and than decreased above 6$0^{\circ}C$. And efficient removal could be achieved within relatively short time (30min) The removal of oily soils from polyester fabrics was increased in proportion to increasing of pH range up to a certain point(9.5 or 11.0) but it began to decrease above the point. Furthermore the removal of oily soils was increased with th increase of mechanical agitation, but it began to decrease above 160 rpm.

Effect of the rheological properties of aqueous xanthan gum solution on chemical absorption of carbon dioxide with diisopropanolamine

  • Absorption rate of carbon dioxide was measured in the aqueous xanthan gum (XG) solution in the range of 0~0.15 wt% containing diisopropanolamine (DIPA) of $0{\sim}2\;kmol/m^3$ in a flat-stirred vessel with an impeller of 0.05 m and agitation speed of 50 rpm at $25^{\circ}C$ and 101.3 kPa. The volumetric liquid-side mass transfer coefficient ($k_La$) of $CO_2$, which was obtained by the measured physical absorption rate, was correlated with the viscosity and the elastic behavior of XG solution such as Deborah number as an empirical formula. The chemical absorption rate of $CO_2$ ($R_A$), which was estimated by the film theory using the measured $k_La$ and the known kinetics of reaction between $CO_2$ and DIPA, was compared with the measured rate. The aqueous XG solution with elastic property of non-Newtonian liquid made $k_La$ and $R_A$ increased compared with Newtonian liquid based on the same viscosity of the solution.

  • Effect of immobilization on the production of human granulocyte-macrophage colony-stimulating factor (hGM-CSF) by Nicotiana tabacum cells was investigated using polyurethane foam as immobilization matrices. The cell activity and the hGM-CSF production were maintained for 16 days in spite of 3 times of media exchange. Under the same conditions of temperature and agitation rate, maximum concentrations of hGM-CSF in a 500-mL spinner flask and 100-mL Erleuneyer flasks were 17.3 ${\mu}g/L$ and 9.8 ${\mu}g/L$, respectively. Consequently high hGM-CSF production could be possible in spinner flask when the rate and amount of media exchange were optimized.
