Production Conditions and Characterization of the Exo-biopolymer Produced by Submerged Cultivation of Ganoderma lucijum Mycelium (영지(Ganoderma lucidum) 균사체의 액체배양에 의한 세포외 생물고분자의 생산조건과 특성)

  • For the screening and the development of the new bio-material, cultural conditions for the exo-biopolymer (EBP) production throught the submerged cultivation of Ganoderma lucidum mycelium were investigated. Also, the fractionations and the purifications of the exo-biopolymer were carried out and the chemical compositions of the exo-biopolymer were examined. The optimal culture conditions for the exo-biopolymer production were pH 5.0, 30$^{\circ}C$ and 100 rpm of agitation speed in the medium containing of 5% (w/v) glucose, 0.5%(w/v) yeast extract, 0.1% (w/v) ($(NH_4)_2HPO_4$, and 0.05% (w/v) $KH_2PO_4$. In the flask cultivation for 7 days under these conditions, the concentration of the maximum exo-biopolymer and the cell mass were 15.4g/l and 18.8g/l, respectively. The specific growth rate was 0.039 $hr^{-1}$. In addition, the substrate consumption rate, and the exo-biopolymer production rate were 0.043$gg^{-1}$$hr^{-1}$ and 0.025$gg^{-1}$$hr^{-1}$, respectively. The exo-biopolymer was fractionated into BWS (water soluble exo-biopolymer) and BWI (water insoluble exo-biopolymer) by the water extraction, and the sugar contents of two fractions were higher than 97% (based on dry basis). The components sugar of BWS and BWI fractions were glucose, galactose, mannose, xylose, and fucose. Their molar ratios were 3.6:1.5:2.1:0.5: trace and 2.9:3.1:2.0:1.6:0.3, respectively.

Optimized Conditions for High Erythritol Production by Penicillium sp. KJ-UV29, Mutant of Penicillium sp. KJ81

  • To improve the erythritol productivity of Penicillium sp. KJ81, mutants were obtained using UV irradiation and NTG treatment Among these mutants, Penicillium sp. KJ-UV29 revealed no morphological changes, yet was superior to the wild strain in the following three points: (1) Penicillium sp. KJ-UV29 produced more erythritol than the wild strain under the same conditions, (2) no foam was produced during cultivation, unlike the wild strain, and (3) the mutant produced a Significantly lower amount of glycerol. Penirillium sp. KJ-UV29 produced as much as 15.1 g/L of erythritol, whereas the wild-type Penirillium sp. KJ81 only produced 11.7 g/L. Penicillium sp. KJ-UV29 only generated 6.1 g/L of glycerol, compared to 19.4 g/L produced by the wild strain. When investigating the optimal culture conditions for erythritol production by the mutant strain Penicillium sp. KJ-UV89, sucrose was identified as the most effective carbon source, and the mutant was even able to produce erythritol in a 70% sucrose-containing medium, although a 30% sucrose medium exhibited the highest productivity. The production of erythritol by Penirillium sp. KJ-UV29 was also significantly increased by the addition of ammonium carbonate, potassium nitrate, and sodium nitrate. Accordingly, under optimal conditions, Penicillium sp. KJ-UV29 produced 45.2 g/L of erythritol in a medium containing 30% sucrose, 0.5% yeast extract, 0.5% (NH$_4$)$_2$C$_2$O$_4$, 0.1% KNO$_3$, 0.1% NaNO$_3$, and 0.01% FeSO$_4$ with 1 vvm aeration and 200 rpm agitation at 37$^{\circ}C$ for 7 days in a 5-L jar fermentor.

  • This study is to explore the contents and interaction of residents voluntary network and propose the strategies to promote residents voluntary network. The grounded theory was utilized to attain our object. Total of seven social worker and 17 residents participated in the study. Data were collected through in-depth interviews and documents. The data were analyzed by using Strauss and Corbin's method. Results are the followings. In open coding 13 categories, 32 subcategories and 133 concepts were constructed. In axial coding causal conditions were qualitative ascent of needs, emergence of the right welfare consumer. Phenomenon was agitation of praxis ground and grope of exist. Contextual conditions were crisis resources, skepticism of welfare. Intervention conditions were maturation of welfare cognition and proliferation of the sense of community responsibility. Strategy were resocialization of voluntary organization and construction of field related service delivery system. Consequence were grass routing welfare strategic fitting service system. In selective coding we constructed the core category: The praxis revolution from bottom for break social welfare environment.

Optimal Conditions for the Production of (+)-Jasmonic acid by Diplodia gossypina ATCC10936 (Diplodia gossypina ATCC10936 균주를 이용한 (+)-Jasmonic acid의 생산조건 최적화)

  • Diplodia gossypina ATCC10936 produced chiral specific (+)-jasmonic acid (JA) that is the most biologically active form. (+)-JA is a plant growth hormone and also one of the most important aroma compounds responsible for jasmin-like aroma note. In order to develop a commercial bioprocess for the production of (+)-JA, optimal culture conditions for D. gossypina ATCC10936 were investigated. D. gossypina produced (+)-JA using either fructose and glucose as a sole carbon source. As a nitrogen source, $NaNO_3$ gave relatively high (+)-JA production. The optimal temperature for the production of (+)-JA by D. gossypina was $28^{\circ}C$, and optimal agitation was found to be 200 rpm. D. gossypina produced (+)-JA upto 600 mg/L in SM medium, although the highest level of biomass was obtained in PDMYS medium.

The Improvement of Cephalosporin C Production by Fed-batch Culture of Cephalosporium acremonium M25 Using Rice Oil

  • The objective of this study is to improve cephalosporin C (CPC) production byoptimization of medium and culture conditions. A statistical method was introduced to optimize the main culture medium. The main medium for CPC production was optimized using a statistical method. Glucose and corn steep liquor (CSL) were found to be the most effective factors for CPC production. Glucose and CSL were optimized to 2.84 and $6.68\%$, respectively. CPC produc­tion was improved $50\%$ by feeding of $5\%$ rice oil at day 3rd and 5th day during the shake flask culture of C acremonium M25. The effect of agitation speeds on CPC production in a 2.5-L bio­reactor was also investigated with fed-batch mode. The maximum cell mass (54.5 g/L) was obtained at 600 rpm. However, the maximum CPC production (0.98 g/L) was obtained at 500 rpm. At this condition, the maximum CPC production was improved about $132\%$ compared to the re­sult with batch flask culture.

Physiochemical Changes and Optimization of Phosphate-Treated Shrimp (Litopenaeus vannamei) Using Response Surface Methodology

  • The objective of this study was to determine the factors responsible for the changed physiochemical properties of unpeeled shrimp treated in cold phosphate solution ($2{\sim}4^{\circ}C$) with the intervention of 4 factors: phosphate concentration, dipping time, rotation speed, and volume of brine solution. Response surface analysis was used to characterize the effect of the phosphate treatment on shrimps by running 33 treatments for optimizing the experiment. For each treatment, phosphate amount, moisture content, and weight gain were measured. The results showed that phosphate concentration is the most important factor than other factors for facilitating phosphate penetration in the meat of the shrimp and for getting the best result. The optimum condition of phosphate-treated shrimp in this study was 110 to 120 min dipping time, 500 to 550 mL brine solution for 100 g shrimp sample, and 190 to 210 rpm agitation speed. The studied conditions can be applied in fisheries and other food industries for good phosphate treatments.

Barium Nitrate Single Crystals Growth by Aqueous Solution Method

  • The growing conditions of barium nitrate $Ba(NO_3)_2$ single crystals using the aqueous solution method have been studied. Supersaturation can be calculated by measuring the temperature of the solution and its equilibrium temperature. Supersaturation of $Ba(NO_3)_2$ was 0.7% at $32.0^{\circ}C$ and about 3% at $34.0^{\circ}C$. The obtained single crystals have three kind of morphology: tetrahedral, cubic, and, rarely, dodecahedral. The normal growth rate is proportional to the supersaturation; it is necessary to make the solution below 5% supersaturation in order to obtain transparent $Ba(NO_3)_2$ single crystals. The normal growth rate for {1$\bar{1}$1} faces was $2.51{\times}10^{-6}$ mm/s for the 0.7% supersaturation condition ($32.0^{\circ}C$), $6.43{\times}10^{-6}$ mm/s for the the condition of 3.0% supersaturation, and $7.01{\times}10^{-6}$ mm/s for the condition of 5.0% supersaturation. The quality of the grown crystals depends on the nature of the seed, the cooling rate employed, and the agitation of the solution. The faces of the obtained crystals have been identified uising an X-ray diffractometer. The surface diffusion is responsible for the low growth rates of the {1$\bar{1}$1} faces.

Attempted Suicide by Nitrogen Gas Asphyxiation: A Case Report (질소 가스를 사용한 자살 시도 1례: 증례보고)

  • Nitrogen is an inert gas that is harmless to humans under normal conditions. While it is not inherently toxic, nitrogen gas becomes dangerous when it displaces oxygen, resulting in suffocation. Herein, we report a case of a 34-year-old man who attempted suicide by nitrogen asphyxiation who presenting with decreased mental function and agitation. Lactic acidosis and hyperammonemia were observed on presentation at the emergency department, but these improved after a few hours. After 2 days, the patient regained full consciousness, and was discharged without any complications. Survival after asphyxiation due to nitrogen gas is very rare, and these patients are more likely to have poorer outcomes. There is a potential for the increasing use of nitrogen gas as a method of committing suicide because of the ease of access to this gas.

The Effects of Water-and Oil-Repellent Finishes on the Deposition and Removal of Oily Soils from Polyester Fabrics (폴리에스테르 직물의 발수발유 가공처리가 유성오구의 부착 및 제거에 미치는 영향)

  • The effects of water-and oil-repellent finishes on the deposition and removal of oily soils from polyseter fabrics were investigated in this study. The polyester fabrics treated with three kinds of fluoropolymers(TG-410H, TG-990) were soiled with mixed oily soils and washed by using Terg-O-Tometer at various conditions. It was found that TG-410H and TG-527 treated polyester fabrics reduced significantly the deposition of oily soils than untreated and TG-990 treated polyester fabrics. The removal of oily soils from polyester fabrics was increased in porportion to increasing of surfactant concentration up to a certain point(0.2% owb) but it levelled off above the point. The removal effect was high in the order of TG-990 treated polyester fabrics > untreated polyester fabrics > TG-527 treated polyester fabrics > TBG-410H treasted polyester fabrics. Especially the removal of oily soils from polyester fabrics was more effectively removed in triton X-100 solution. In general the removal of oily soils from polyester fabrics treated with fluoropolymers was increased up to a miximum near 6$0^{\circ}C$ and than decreased above 6$0^{\circ}C$. And efficient removal could be achieved within relatively short time (30min) The removal of oily soils from polyester fabrics was increased in proportion to increasing of pH range up to a certain point(9.5 or 11.0) but it began to decrease above the point. Furthermore the removal of oily soils was increased with th increase of mechanical agitation, but it began to decrease above 160 rpm.

  • The purified biosurfactant $3.16g/{\ell}$ was obtained after cultivation for 104hr at $25^{\circ}C$ with an optimal agitation speed of 200rpm, an aeration rate of 2vvm in a $14{\ell}$ fermenter containing $5.5{\ell}$ of LB medium and 1%(w/v) olive oil as a carbon source. For the kinetic studies, the optimal substrate concentration was analyzed on different olive oil concentrations(0.1, 0.5, 1.0, 1.5, 2.0%(w/v)) and optimal culture conditions(MLBM, 200rpm, 2vvm at $25^{\circ}C$) in a $14{\ell}$ jar fermenter. The results obtained indicate that $K_s$=0.0086 $g/{\ell}$, $q_s$= 0.664 $g/g{\cdot}h$, $q_p$= $4.2{\times}10^{-3}$ $g/g{\cdot}h$, and ${\mu}_{max}$ was determined as $0.1449h^{-1}$.
