Sliding wear mechanism of the high-nitrogen austenitic 18Cr-18Mn02Mo-0.9N steel (고질소 Fe-18Cr-18Mn-2Mo-0.9N강의 미끄럼 마멸 기구)

  • Sliding wear mechanism of a high nitrogen austenitic 18Cr-18Mn-2Mo-0.9N steel has been investigated. Dry sliding wear tests of the steel were carried out at various loads of 1N-10N under a constant sliding speed condition of 0.15m/s against AISI 52100 bearing steel balls. Solution $(1050^{\circ}C)$ and isothermal aging $(900^{\circ}C)$ heat treatments were performed on the steel and the effect of the heat treatments on the wear were investigated. Wear rates of the solution-treated steel specimen remained low until 5N, and then increased abruptly at loads above 5N. The rates of isothermally aged specimens were low and increased gradually with the applied load. Worn surfaces, their cross sections, and wear debris of the steel specimens were examined with a scanning electron microscopy. Phases of the heat-treated specimen and the wear debris were identified using XRD. Phases transformed underneath the sliding track during the wear were investigated and analyzed using TEM. Effects of the phase transformation during the wear and $Cr_2N$ precipitates formed during the isothermal aging on the wear of the austenitic steel were discussed.

Replicative Senescence in Cellular Aging and Oxidative Stress (세포 노화에 있어서 복제 세네센스 현상과 산화적 스트레스의 영향)

  • Explanted mammalian cells perform a limited number of cell division in vitro and than are arrested in a state known as replicative senescence. Such cells are irreversibly blocked, mostly in the G1 phase of cell cycle, and are no longer sensitive to growth factor stimulation. Thus replicative senescence is defined as a permanent and irreversible loss of replicative potential of cells. For this characteristic, replicative senescence seems to evolve to protect mammalian organism from cancer. However, senescence also contributes to aging. It seems to decrease with age of the cell donor and, as a form of cell senescence, is thought to underlie the aging process. Extensive evidence supports the idea that progressive telomere loss contributes to the phenomenon of cell senescence. Telomeres are repetitive structures of the sequence (TTAGGG)n at the ends of linear chromosomes. It has been shown that the average length of telomere repeats in human somatic cells decreases by 30∼200 bp with each cell division. It is generally believed that when telomeres reach a critical length, a signal is activated to initiate the senescent program. This has given rise to the hypothesis that telomeres act as mitotic clocks to regulate lifespan. One proposes that cumulative oxidative stress, mainly reactive oxygen species generated from mitochondria, may mainly cause telomere shortening, accelerating aging. Here, the biological importance and mechanism of replicative senescence were briefly reviewed. Also it was summarized that how oxidative stress affects replicative senescence and telomere shortening.

An approach of evaluation and mechanism study on the high and steep rock slope in water conservancy project

  • In this study, an aging deformation statistical model for a unique high and steep rock slope was proposed, and the aging characteristic of the slope deformation was better reflected. The slope displacement was affected by multiple-environmental factors in multiple scales and displayed the same tendency with a rising water level. The statistical model of the high and steep rock including non-aging factors was set up based on previous analyses and the study of the deformation and residual tendency. The rule and importance of the water level factor as a non-aging unit was analyzed. A partitioned statistical model and mutation model were established for the comprehensive cumulative displacement velocity with the monitoring study under multiple factors and multiple parameters. A spatial model was also developed to reflect and predict the whole and sectional deformation character by combining aging, deformation and space coordinates. A neural network model was built to fit and predict the deformation with a high degree of precision by mastering its feature of complexity and randomness. A three-dimensional finite element model of the slope was applied to approach the structure character using numerical simulations. Further, a three-dimensional finite element model of the slope and dam was developed, and the whole deformation state was analyzed. This study is expected to provide a powerful and systematic method to analyze very high, important and dangerous slopes.

Aging and Recovery of HTV Silicone Rubber Used for Outdoor Insulator (옥외용 HTV 실리콘고무 절연재료의 열화 및 회복특성)

  • This paper presents a study on the aging and recovery of HTV (high temperature vulcanized) silicone rubber used for outdoor insulators. UV irradiation, corona discharge and water immersion were employed as factors of the artificial aging. The effects of changes derived from these stresses on the tracking and arc resistance of silicone rubber were examined. We have investigated the aging phenomena of HTV silicone rubber by the above stresses using the surface energy calculation with contact angle measurement, solvent-extraction, and surface/volume resistivity and so on. These results showed that UV irradiation and corona discharge lead to nearly the same surface oxidation, but the percentage change of mobile low molecular weight by these stresses was different. Furthermore, the oxidized layer induced under UV irradiation restricted the recovery of hydrophobic surface. Water immersion little lowered hydrophobicity level and leaded to a loss of tracking and arc resistance. The degradation mechanism based on our results was discussed.

Molecular Mechanism of Dietary Restriction in Neuroprevention and Neurogenesis: Involvement of Neurotrophic Factors

  • Dietary restriction (DR) is the most efficacious intervention for retarding the deleterious effects of aging. DR increases longevity, decreases the occurrence and severity of age-related diseases, and retards the physiological decline associated with aging. The beneficial effects of DR have been mostly studied in non-neuronal tissues. However, several studies have showed that DR attenuate neuronal loss after several different insults including exposure to kainate, ischemia, and MPTP. Moreover, administration of the non-metabolizable glucose analog 2-deoxy-D-glucose (2DG) could mimic the neuroprotective effect of DR in rodent, presumably by limiting glucose availability at the cellular level. Based on the studies of chemically induced DR, it has been proposed that the mechanism whereby DR and 2DG protect neurons is largely mediated by stress response proteins such as HSP70 and GRP78 which are increased in neurons of rats and mice fed a DR regimen. In addition, DR, as mild metabolic stress, could lead to the increased activity in neuronal circuits and thus induce expression of neurotrophic factors. Interestingly, such increased neuronal activities also enhance neurogenesis in the brains of adult rodents. In this review, we focus on what is known regarding molecular mechanisms of the protective role of DR in neurodegenerative diseases and aging process. Also, we propose that DR is a mild cellular stress that stimulates production of neurotrophic factors, which are major regulators of neuronal survival, as well as neurogenesis in adult brain.

Comparison of Anti-cancer Potentials of Water Extracts of Bigihwan, Daechilgithang and Mokwhyangbinranghwan in Human Hepatocellular Carcinoma Cells (인체 간암세포에서 비기환(肥氣丸), 대칠기탕(大七氣湯) 및 목향빈랑환(木香檳榔丸) 열수 추출물의 항암 활성 비교)

  • Objectives : We selected three herb-combined remedies, Bigihwan (BGH), Daechilgitang (DCGT) and Mokwhyangbinranghwan (MHBRH), through Donguibogam text-mining analysis, and evaluated their anti-cancer effects on human hepatocellular carcinoma Hep3B cells. Methods : Cytotoxicity was assessed by an MTT assay. Apoptosis rate, autophagy and ROS level were detected by flow cytometry. The autophagy was also observed by Cyto-ID staining fluorescence microscopy. The expression of autophagy, mitophagy and pexophagy regulatory proteins was detected by Western blot analysis. Results : BGH showed the strongest effect among the three prescriptions in inhibiting Hep3B cell viability, which was associated with the induction of apoptosis and autophagy. Autophagy blockers improved cell viability and reduced apoptosis after BGH and DCGT treatments, indicating that autophagy by these prescriptions enhanced Hep3B cells against their cytotoxicity. However, MHBRH enhanced the reduction of cell viability and apoptosis by autophagy blockers. Induction of autophagy by BGH treatment was associated with mitophagy due to mitochondrial dysfunction than DCGT and MHBRH-treated cells. In addition, induction of apoptosis by BGH treatment was ROS-dependent and showed the possibility of pexophagy involvement. Conclusion : Although further studies need to be conducted to study the efficacy and mechanism of accurate anticancer activity, the present results will serve as important sources of understanding the mechanism of action of herbal remedies prescribed for liver disease as documented in Donguibogam.

The Study on the Korean and Western Medical Literatures for Skin Aging wrinkle, hyperpigmentation, dry skin, facial flush (피부 노화 현상에 대한 동서의학적 고찰 주름, 과색소침착, 피부건조, 안면홍조를 중심으로)

  • Objective : The purpose of this study is to understand conspicuous features of geroderma with visceral manifestation theory(臟象論). Methods : We categorized skin aging into wrinkles, hyperpigmentation, dry skin and face flush. After investigating the reason, histological changes and mechanism of each classification in western medicine, we interpreted them according to the malfunction of five viscera(五臟) in Korean medicine. Result : The results are as follows. 1. Pathologic change of dermis and subcutaneous fat makes wrinkles. We consider wrinkles as the malfunction of the spleen(脾). 2. Irregular synthesis and disproportion of melanin makes hyperpigmentation. We consider hyperpigmentation as the malfunction of the liver(肝). 3. Dry skin is attributed to a subtle disorder of epidermal maturation. We consider dry skin as the malfunction of the lung(肺). 4. Facial flush is detected in rosacea and menopausal hot flush, which are both related with blood vessel abnormality. We consider facial flush as the malfunction of the heart(心) Conclusion : We interpreted the pathologic changes and mechanism of skin aging in western medicine as the decrease of five viscera(五臟) in visceral manifestation theory(臟象論) of Korean medicine. Further studies are needed to apply these hypothesis to clinical diagnosis and treatment.

Epigenetic Characterization of Aging Related Genes (노화 관련 유전자의 후성유전학적 특성 분석)

  • Gene expression is regulated by a wide range of mechanisms at the DNA sequence level. In addition, gene expression is also regulated by epigenetic mechanisms through DNA methylation, histone modification, and ncRNA. To understand the regulation of gene expression at the epigenetic level, we constructed aging related gene database and analyzed epigenetic properties that are focused on DNA methylation. The DNA methylation of promoter or upstream region of the genes induces to repress the gene expression. We compared and analyzed distribution between whole human genes and aging related genes in the epigenetic properties such as CGI distribution, methylation motif pattern, and TFBS (transcription factor binding site) distribution. In contrast to methylation motif pattern, CGI and TFBS distributions are positively correlated with epigenetic regulation of aging related gene expression. In this study, the epigenetic data about DNA methylation of the aging genes will provide us to understand phenomena of the aging and epigenetic mechanism for regulation of aging related genes.

Aging of Immune System (면역 반응체계의 노화)

  • Immune system provides defense integrity of body against external invaders. In order to accomplish the important defending role immune system is composed of many different components which are regenerated continuously during lifespan. The key components are professional killing cells such as macrophage, neutrophil, natural killer cell, and cytotoxic T cell and professional blocking molecule, antibody, which is produced by plasma cell, the terminal differentiated B cell. Immune response is orchestrated harmoniously by all these components mediated through antigen presenting cells such as dendritic cells. Immune responses can be divided into two ways: innate immune response and adaptive immune response depending on induction mechanism. Aging is a broad spectrum of physiological changes. Likewise other physiological changes, the immune components and responses are wane as aging is progressing. Immune responses become decline and dysregulating, which is called immunosenescense. Immune components of both innate and adaptive immune response are affected as aging progresses leading to increased vulnerability to infectious diseases. Numbers of immune cells and amounts of soluble immune factors were decreased in aged animal models and human and also functional and structural alterations in immune system were reduced and declined. Cellular intrinsic changes were discovered as well. Recent researches focusing on aging have been enormously growing. Many advanced tools were developed to bisect aging process in multi-directions including immune system area. This review will provide a broad overview of aging-associated changes of key components of immunity.

A Consideration of Degradation Mechanism of a Effect of the Nitric Solution on the Aging Characteristics of the Outdoor Silicone Rubber (열화 메커니즘 규명을 위한 옥외용 실리콘고무의 산성비 성분 열화특성 연구)

  • In this paper, aging characteristics of outdoor silicone rubber insulator resulting from the acid rain have been investigated with regards to nitrate ion $({NO_{3}}^{-1})$. For this purpose, silicone rubber was aged by immersing into the nitric solutions with different level pH during 1500 hours and various tests (tracking, $tan{\delta}$, breakdown voltage, SEM, EDS, contact angle) were performed. It was observed that the degradation was increased to pH 2.5 slightly and at pH 1.5 highly. And thus, it could be expected that there is certain critical point between pH 1.5 and 2.5, which leads to throught degradation.
