SR proteins regulate V6 exon splicing of CD44 pre-mRNA

  • CD44 pre-mRNA includes 20 exons, of which exons 1-5 ($C_1-C_5$) and exons 16-20 ($C_6-C_{10}$) are constant exons, whereas exons 6-15 ($V_1-V_{10}$) are variant exons. $V_6$-exon-containing isoforms have been known to be implicated in tumor cell invasion and metastasis. In the present study, we performed a SR protein screen for CD44 $V_6$ splicing using overexpression and lentivirus-mediated shRNA treatment. Using a CD44 $V_6$ minigene, we demonstrate that increased SRSF3 and SRSF4 expression do not affect $V_6$ splicing, but increased expression of SRSF1, SRSF6 and SRSF9 significantly inhibit $V_6$ splicing. In addition, using a constitutive exon-specific primer set, we could not detect alterations of CD44 splicing after SR protein-targeting shRNA treatment. However, using a $V_6$ specific primer, we identified that reduced SRSF2 expression significantly reduced the $V_6$ isoform, but increased $V_{6-10}$ and $V_{6,8-10}$ isoforms. Our results indicate that SR proteins are important regulatory proteins for CD44 $V_6$ splicing.

Generation of Hydrogen Peroxide by Single-Atom Clusters Pd Anchored on t-BaTiO3 for Piezoelectric Degradation of Tetracycline

  • Single-atom Pd clusters anchored on t-BaTiO3 material was synthesized using hydrothermal and ultrasonic methods for the effective piezoelectric catalytic degradation of pollutants using vibration energy. XRD patterns of BaTiO3 loaded with monoatomic Pd were obtained before and after calcining, and showed typical cubic-phase BTO. TEM and HAADF-STEM images indicated single-atom Pd clusters were successfully introduced into the BaTiO3. The piezoelectric current density of the prepared Pd-BaTiO3 binary composite was significantly higher than that of the pristine BaTiO3. Under mechanical vibration, the nanomaterial exhibited a tetracycline decomposition rate of ~95 % within 7 h, which is much higher than the degradation rate of 56.7 % observed with pure BaTiO3. Many of the piezo-induced electrons escaped to the Pd-doped BaTiO3 interface because of Pd's excellent conductivity. Single-atom Pd clusters help promote the separation of the piezo-induced electrons, thereby achieving synergistic catalysis. This work demonstrates the feasibility of combining ultrasonic technology with the piezoelectric effect and provides a promising strategy for the development of ultrasonic and piezoelectric materials.

Survey of Korean Elementary School Children's Happiness (한국 초등학교 아동의 행복감 실태조사)

  • This study investigated Korean elementary school children's happiness by gender and grade and by attendance in after-school programs during weekdays and weekends. Participants were 3,608 children in Korean provincial areas except Jeju-do; they responded to the Happiness Scale developed by the researchers. Data were analyzed by frequency, percent, and t-test. Results were (1) children's happiness level was high; girls' happiness level was higher than boys'. As they moved up in school grade, happiness level decreased. (2) Their attendance in after-school programs during weekdays and weekends decreased their happiness level in leisure and extra academic activity, but it increased happiness in school/community life and sibling relations. Conclusions were that reduction in the amount of children's after-school programming would increase children's happiness.

Development and Evaluation of a Finding Meaning in Life CD Program for Life-esteemed Education of Older School-age Children (초등학생의 생명존중교육을 위한 삶의 의미발견 CD 프로그램 개발 및 효과)

  • Purpose: The purpose of this study was to develop a finding meaning in life CD program about life-esteemed education and to identify the effect of the program. Methods: The life-esteemed education philosophy and the concepts of logotherapy were applied as a theoretical framework for this program. This program was developed through the process of planning, designing, developing, and evaluating with a content validity test. To identify the effect of the program, one experimental group design was applied to 54 students. Data were collected before the program started and one week and five weeks after the program finished. Results: The program was developed based on the students' needs and evaluation of the CD's content and consists of five periods: Dinosaur Park of Promise, Hill of Fragrance, Garden of Love, Forest of Acceptance, and My Lake. Each post-test score of knowledge, attitude, and practice on meaning of life was significantly higher than pretest scores. Conclusion: This program can be effective for life-esteemed education in elementary school students. Moreover, it is encouraged that the program will be utilized in more life-esteemed education for elementary school students.

  • This study was a quasi-experimental study conducted in ${\ulcorner}$Design of Home Life${\lrcorner}$ unit of high school Home Economics. The purposes of this study was to analyze the effects of gender equity instruction on gender equity consciousness of high school students, and to raise gender equity consciousness of students. The summarized results of this study were as follows; 1. After experiment. gender equality consciousness of experimental group was higher than that of comparison group in all four areas, that is, home life, school life, vocational life and socio-cultural life. So there was statistically significant differences in gender equity consciousness between two groups. 2. The gender equity consciousness of the experimental group was higher In post-test than pre-test, so there was statistically significant differences between pre-test and post-test. On the other hand, there was no statistically significant difference between pre-test and post-test in the comparison group. 3. After experiment, the change-rate of the gender equality consciousness In the experimental group was higher than that of in the comparison group, so there was statistically significant differences between two groups in the change-rate. As results of this study, in the ${\ulcorner}$Design of Home Life${\lrcorner}$ unit of high school Home Economics, it was proved that the gender equity class was effective on raising the gender equality consciousness of high school students in all four areas of home life, school life, vocational life and socio-cultural life.

Cloning of aprE86-1 Gene Encoding a 27-kDa Mature Fibrinolytic Enzyme from Bacillus amyloliquefaciens CH86-1

  • A gene encoding the major secreted fibrinolytic protein of Bacillus amyloliquefaciens CH86-1 was cloned from genomic DNAs. DNA sequencing showed that the gene, aprE86-1, could direct the synthesis of a mature protein 275 amino acids in length after processing. When aprE86-1 was introduced into B. subtilis, a mature 27-kDa protein was produced as expected. The fibrinolytic activity of the B. subtilis transformant (TF) was higher than that of B. amyloliquefaciens CH86-1, showing the possibility of increasing the fibrinolytic activity of Bacillus strains through genetic engineering.

Knockdown of SMYD3 by RNA interference inhibits cervical carcinoma cell growth and invasion in vitro

  • Elevated expression of SMYD3 is a frequent genetic abnormality in several malignancies. Few studies knocking down SMYD3 expression in cervical carcinoma cells have been performed to date. In this paper, we established an inducible short hairpin RNA expression system to examine its role in maintaining the malignant phenotype of HeLa cells. After being induced by doxycycline, SMYD3 mRNA and protein expression were both reduced, and significant reductions in cell proliferation, colony formation and migration/invasion activity were observed in the SMYD3-silenced HeLa cells. The percentage of cells in sub-G1 was elevated and DNA ladder formation could be detected, indicating potent induction of apoptosis by SMYD3 knockdown. These findings imply that SMYD3 plays crucial roles in HeLa cell proliferation and migration/invasion, and that it may be a useful therapeutic target in human cervical carcinomas.

The Spatio-Temporal Characteristics of Primary School Children's Daily Life Activities (초등학생 일상생활활동의 시.공간적 특성)

  • The daily life activities of primary school children have been increasingly diversified. This research examined the spatio-temporal characteristics of primary school children's daily life activities. The time allocation of primary school children's daily activities was monotonous. They tended to spend most of their time in school activities, private lessons after school, and studying at home. The daily trip to school has been diversified as the result the of increase in the number of double-income families, car owners, facilities for pastime, and educational facilities for private lessons after school. The selection of educational facilities for private lessons was strongly influenced by accessibility to the residence because of the limited time budget of primary school children and the controlled trip behavior by parents. The leisure activities were considered as a minor function and took place in the interval of performances of study-related activities. The high proportion of leisure activities was found in the indoor play. The amount of the time for leisure activity was very insufficient. The spatial range and time budget were strongly influenced by the institutional conditions of parents.

Factors Influencing Health-related Quality of Life among Knee or Hip Arthroplasty Patients (하지 인공관절 치환술 환자의 건강 관련 삶의 질 영향요인)

  • Purpose: The purpose of this research was to identify factors that influence health-related quality of life of patients who have had knee or hip arthroplasty. Factors included sleep disorders, pain, activities of daily living, and depression. Methods: The participants in this research were 97 patients within one year after receiving knee or hip arthroplasty surgery and who regularly visited a hospital outpatient department. Data were analyzed using descriptive statistics, t-test, ANOVA, and stepwise regression analysis with the SPSS program. Results: Sleep disorders, pain, activities of daily living, and depression had significant correlations with health related quality of life. Factors influencing health-related quality of life among arthroplasty patients were the length of time since surgery, sleep disorders, activities of daily living, and depression. These factors explained 58% of the participants' health-related quality of life. Conclusion: The results show that various factors can affect patients' health-related quality of life following joint replacement surgery. The findings indicate that sleep disorders and depression should receive continued attention after the arthroplasty. It is also necessary to develop appropriate nursing intervention programs to lower depression and increase patient participation in daily living activities.

The Effects of After-school Service Use on Self-care of Children: Focusing on the Types of Service, the Number of Types, and the Patterns of Service Use (방과후 서비스 이용이 아동의 자기보호에 미치는 영향: 이용하는 방과후 서비스의 유형, 유형의 개수, 혼합이용 패턴을 중심으로)

  • This study explored the effects of after-school services on whether they reduced the possibility of self-care of children. In terms of characteristics of the use of after-school services, it focused on the 1) types, 2) diversity, and 3) use patterns of the after-school services. For the analysis, it used data of the cohort of elementary-school children from the Korean Children and Youth Panel Survey. The main findings are as follows. First, private mentoring and after-school services provided by welfare centers reduced the possibility of self-care. Second, when the total amount of educational time was controlled, the diversity of participating after-school services did not explain the possibility of self-care. Third, when the total amount of educational time was controlled, the use patterns of after-school services did not explain the possibility of self-care. Based on these findings, it suggested policy implications and future research.
