The Function of Multiple Pribnow Box on the Aerobic-Anaerobic Switch Control of aeg-46.5 Gene Expression

  • The gene aeg-46.5, which is expressed under anaerobic condition, has putative triple -10 regions and four transcription start sites. The mRNA transcription level and its start point change depending on the aerobic/anaerobic growth condition. RNA polymerase and its regulatory proteins must choose which of three -10 region to use. The putative triple 10 region was mutated to make only one of them function with consensus -10 region sequence (TATAAT) and the other two as non-functional region. The results show that the second and third -10 regions are used for the aerobic/anaerobic expression. The third -10 region is responsible for the high aerobic to anaerobic switch ratio. This suggests that only the last two of the putative triple -10 region have functions on aeg-46.5 gene expression switch control. The phenotype of the mutated promoter was tested in the wild type cell and narL - cell. The results indicate that the control by NarL is independent from the selection of -10 region. The expression patterns on multi-copy plasmids and on single-copy chromosome were compared. These results show that the aerobic/anaerobic switch control of aeg-46.5 is through the choice of -10 region. The mechanism of choosing different -10 region remains to be seen.

포장방법에 따른 양념갈비의 저장 중 총균수, 전자공여능 및 지방산화의 감마선 조사효과 (Effect of Gamma Irradiation on Microbial Growth, Electron Donating Ability, and Lipid Oxidation of Marinated Beef Rib (Galbi) with Different Packaging Methods)

  • 양념갈비의 품질 안정성 및 저장성 확보를 위한 일환으로 시판되는 양념갈비에 감마선 조사(0, 2.5, 5, 7.5 kGy)하여 포장 방법(함기 vs 진공) 및 저장 기간에 따른 양념갈비의 총균수, pH, 수분활성도, 전자공여능 및 지방산패도를 측정하였다. 감마선 조사한 양념갈비의 미생물 생육은 조사선량이 증가할수록 저하되었고 포장방법별로는 함기포장구보다 진공포장구가 미생물 생장억제에 더욱 효과적임을 확인하였다. pH는 함기포장구에서 저장 7일째 다소 감소하는 경향을 보였으나 포장방법과 조사선량에 따른 유의 차가 나타나지 않았다. 감마선 조사한 양념갈비의 전자공여능은 저장기간에 따라 감소하는 경향을 보였으나 그 감소폭은 함기포장구가 진공포장구보다 크게 나타났다. TBARS값은 진공포장구의 경우 감마선 조사선량과 저장기 간에 따라 유의적인 차이는 없었다. 그러나 함기포장구는 조사선량이 증가할수록 그리고 저장기간에 따라 유의적으로 증가하여 저장 7일째 진공포장구보다 2배의 높은 값을 나타내었다. 따라서 시판되는 양념갈비의 저장 중 유통안전성을 확보하기 위한 감마선 조사의 사용에서 품질을 유지하기 위해서는 진공포장으로 유통하는 것이 바람직하다고 판단된다.

Model for Estimating CO2 Concentration in Package Headspace of Microbiologically Perishable Food

  • Levels of carbon dioxide gas, a metabolite of microbial growth, have been reported to parallel the onset of microbial spoilage and may be used as a convenient index for a packaged food's shelf life. This study aimed to establish a kinetic model of $CO_2$ production from perishable food for the potential use for shelf life control in the food supply chain. Aerobic bacterial count and package $CO_2$ concentration were measured during the storage of seasoned pork meat at four temperatures (0, 5, 10 and $15^{\circ}C$), and their interrelationship was investigated to establish a mathematical model. The microbial growth at constant temperature was described by using model of Baranyi and Roberts. $CO_2$ production from the stored food could be explained by taking care of its yield and maintenance factors linked to the microbial growth. By establishing the temperature dependence of the microbial growth and $CO_2$ yield factor, $CO_2$ partial pressure or concentration in package headspace could be estimated to a limited extent, which is helpful for controlling the shelf life under constant and dynamic temperature conditions. Application and efficacy of the model needs to be improved with further refinement in the model.

효모에 의한 남은 음식물의 호기성 액상발효 (Aerobic Liquid Fermentation of Food Wastes by Using Yeast)

  • 남은 음식물을 기질로 생균효모사료를 만들기 위해 Kluyveromyces marxianus 효모의 호기적 액상발효가 $35^{\circ}C$에서 시도되었다. 먼저 액상으로 시료를 충분히 마쇄시킨 시료의 효모 생균수를 증가시키기 위한 적정발효조건이 진탕배양을 통해 연구되었다. 시료로 사용된 남은 음식물의 고형분 함량을 수분첨가로 5%, 10%와 15%로 다르게 조정하여 발효시킨 결과 고형분 함량 10%에서 생균수가 가장 높은 것으로 나타났다. 효모의 생균농도를 증가시키기 위하여 Aspersillus oryzae를 2리터용 jar fermenter를 이용하여 혼합배양한 결과 증식속도를 증가시켜 주었으나 최고 생균수는 $3.5{\times}10^9/ml$로서 단독 배양시보다 근소하게 높은 결과를 보여주었다.

간헐포기소화의 비용 평가 (Cost Estimation of Intermittent Aerobic Digestion)

  • 간헐포기방식의 슬러지 소화 기술에 대한 비용을 분석하였다. 포기비율은 간헐포기소화의 가장 중요한 설계인자이며, 포기 비율에 따라 시설비와 동력비가 달라진다. 목표 SS 제거율을 35%로 설정하고, 철근콘크리트형식의 SBR 간헐포기소화조를 설계하였으며, 이로부터 시설비와 동력비를 산출하였다. 비용 평가는 할인율과 경제성장율을 고려하는 현재가치로 환산하는 방법을 사용하였다. 포기비율이 낮은 경우는 시설비가 높지만 동력비는 적게 소요되며, 포기비율이 높으면 반대로 시설비는 낮지만 동력비는 증가한다. 초기시설비는 호기성 소화가 간헐포기소화보다 더 저렴하다. 그러나, 총비용면에서 내용연수 약 10년 이상의 운전에서는 간헐포기소화가 호기성 소화보다 더 유리하다. 특히, 내용연수가 길수록 최적 포기비율은 낮아지며, 내용연수가 45년 정도일 때 최적포기비율은 0.3 정도이고, 이 경우 총비용은 호기성소화의 64% 수준이다.

Efficacy of Sodium Hypochlorite and Acidified Sodium Chlorite in Preventing Browning and Microbial Growth on Fresh-Cut Produce

  • The use of suitable sanitizers can increase the quality of fresh-cut produce and reduce the risk of foodborne illnesses. The objective of this study was to compare the washing effects of 100 mg/L sodium hypochlorite (SH) and 500 mg/L acidified sodium chlorite (ASC) on the prevention of enzymatic browning and the growth of microbial populations, including aerobic plate counts, E. coli, and coliforms, throughout storage at $4^{\circ}C$ and $10^{\circ}C$. Fresh-cut zucchini, cucumbers, green bell peppers, and root vegetables such as potatoes, sweet potatoes, carrots, and radishes were used. Compared to SH washing, ASC washing significantly (p<0.05) reduced microbial contamination on the fresh-cut produce and prevented browning of fresh-cut potatoes and sweet potatoes during storage. More effective inhibition of aerobic plate counts and coliforms growth was observed on fresh-cut produce treated with ASC during storage at $10^{\circ}C$. Polyphenol oxidase (PPO) activity of fresh-cut potatoes and sweet potatoes was more effectively inhibited after washing with ASC. The use of 500 mg/L ASC can provide effective antimicrobial and anti-browning treatments of fresh-cut produce, including processed root vegetables.

겨울철 소매점 및 냉장 닭고기의 미생물학적 평가 (Microbiological Evaluations of Retail and Refrigerated Chickens in Winter)

  • Aerobic plate counts(APC) gram-negative bacterial counts (GNC) and sensory evaluations on chic-ken carcasses during retail and refrigerated storages (3$\pm$1$^{\circ}C$ and 1$0^{\circ}C$) were evaluated. APC and GNC on whole chicken in retail store after storage of 7 days at 3$\pm$1$^{\circ}C$ increased to 3.11 and 3.89 log units com-pared to the initial controls. APC and GNC on whole chicken after storage of 7 days at 1$0^{\circ}C$ increased to 5.43 and 5.03 log units. Sensory scores of chicken carcasses obtained from retail store were in the "liked less" category after storage of 7 days compared to fresh controls. These results indicated that chicken carcasses during refrigerated (1$0^{\circ}C$) storages rapidly allowed the growth of aerobic spoilage bacteria dur-ing storage period which cluld not be microbiologically acceptable after of 7 days of 7 days.

Mitochondria in Cancer Energy Metabolism: Culprits or Bystanders?

  • Cancer is a disease characterized by uncontrolled growth. Metabolic demands to sustain rapid proliferation must be compelling since aerobic glycolysis is the first as well as the most commonly shared characteristic of cancer. During the last decade, the significance of metabolic reprogramming of cancer has been at the center of attention. Nonetheless, despite all the knowledge gained on cancer biology, the field is not able to reach agreement on the issue of mitochondria: Are damaged mitochondria the cause for aerobic glycolysis in cancer? Warburg proposed the damaged mitochondria theory over 80 years ago; the field has been testing the theory equally long. In this review, we will discuss alterations in metabolic fluxes of cancer cells, and provide an opinion on the damaged mitochondria theory.

Physiological Response of Escherichia coli W3110 and BL21 to the Aerobic Expression of Vitreoscilla Hemoglobin

  • The aerobic growth and metabolic performance of Escherichia coli strains BL21 and W3110 were studied when the Vitreoscilla hemoglobin (VHb) was constitutively expressed in the chromosome. When VHb was expressed, acetate production decreased in both strains and was nearly eliminated in BL21. Transcriptional levels of the glyoxylate shunt genes decreased in both strains when VHb was expressed. However, higher transcription of the α-ketoglutarate dehydrogenase genes were observed for W3110, while for BL21 transcription levels decreased. VHb expression reduced the transcription of the cytochrome bo3 genes only in BL21. These results are useful for better selecting a production host.

Quorum Sensing of Rhodobacter sphaeroides Negatively Regulates Cellular Poly-$\beta$-Hydroxybutyrate Content Under Aerobic Growth Conditions

  • The community escape response of Rhodobacter sphaeroides is exerted through the action of CerR and CerI, which code for a LuxR-type regulatory protein and acylhomoserine lactone synthase, respectively. Deletion of chromosomal DNA including cerR and cerI (mutant RI) or insertional interruption of cert (mutant AP3) resulted in two-fold increase in the cellular poly-${\beta}$-hydroxybutyrate (PHB) content In comparison with the wild-type under aerobic growth conditions. The PHB synthase (PhbC) activities of the cer mutants were doubled, and the enzyme expression was regulated at the level of phbC transcription. Thus, CerR, possibly in response to autoinducer (AI), appears to modulate the PHB content of aerobically grown cells by downregulating phbC transcription.
