Study on the Development of Measuring System for Fermentation Degree of Liquid Swine Manure Using Visible Ray (가시광선을 이용한 돈분뇨 액비 부숙도 측정장치 개발에 관한 연구)

  • This study was conducted to develop an measuring system and method for fermentation degree of liquid swine manure by visible ray. The constituent changes of liquid swine manure were examined. pH gradually increased with time, but EC gradually decreased. Malodor strength decreased gradually with aeration treatment with time. Control needed more time to decrease malodor strength than aeration treatment. In aeration treatment, there was no germination of seeds (radish, chinese cabbage) up to 6 weeks and germination rate at 15th week was over 50%. However, in control, there was no germination up to end of experiment. Circular chromatography method showed that there was change after 10th week in aeration treatment but there was no change up to end of experiment in control. As a result, the fermentation degree of liquid swine manure would have relations among pH, EC, germination rate, malodor concentration, and reaction of circular chromatography. The simple analytical instrument for liquid swine manure consisted of a tungsten halogen and deuterium lamp for light source, a sample holder, a quartz cell, spectrometer for spectrum analyzer, a malodour measuring device, a software, etc. Results showed that the simple analytical instrument that was developed can approximately predict the fermentation degree of liquid swine manure by visible ray. Generally, the experiment proved that the simple analytical instrument was reliable, feasible and practical for analyzing the fermentation degree of liquid swine manure.

  • Nitrifier consortium immobilized in polyvinyl alcohol was used for the removal of ammonia nitrogen from synthetic aquaculture water in the airlift bioreactor. At the aeration rate fo 0.15 vvm and bead packing volume fraction of 20%, airlift bioreactor was operated effectively for a removal of ammonia nitrogen and for a stability of operation. Ammonia nitrogen removal rate by airlift bioreactor was continuously increased with decreasing hydraulic residence time. At the HRT(hydraulic residence time) of 0.3 hour, ammonia nitrogen removal rate was 84.3 g/$m^3$.d and the highest ammonia nitrogen removal rate was 130.8 g/$m^3$.d when HRT was 0.1 hour.

  • Studies were made to investigate effects of the scale-up of stirred tank bioreactors on cell growth and alkaloids production for suspension cultures of Eschscholtzia californica. In the 1.5 L STR, cell lysis was observed at 110 rpm or higher agitation speed. The agitation speed of 30 L STR was 43.7 rpm to maintain the same shear stress developed in 1.5 L STR of 100 rpm. As a result of scale-up from 1.5 L to 30 L STR, the specific growth rate was decreased from 0.12 to 0.07 day-1. The alkaloids productivity was also decreased from 0.24 to 0.14 mg/L-day. Changes of mixing performance and oxygen transfer were studied to explain the decrease of cell growth and alkaloids production. Decreased oxygen transfer rate coefficient(KLa) and increased mixing time by the scale-up was observed at various aeration rates.

  • Process intensification without any increase in bed requires the exploitation of fluid mechanical phenomena as the basis for elegant solutions to the process engineering problems which result from the need to retain and control the immobilized biomass, and for biomass recovery. The fluidized bed biological reactor provides a solution to these needs. The wastewater treatment characteristics of the fluidized bed was filled with sand media. Indirect aeration were studied experimentally. The researcher was filled with sand particle size(0.60~0.42mm) in three reactors with different section area(A)/height(H), in the state BOD loading 4.5kg-$BOD_5/m^3$ㆍd, and under the fixed state of hydraulic retention time for around 32 minutes.

  • Optimal agitation speed and aeration rate for the production of gellan by Sphingomnas paucibilis NK2000 in a 7 l bioreactor were found to be 400 rpm and 1.0 vvm. The best time for glucose feeding into the medium for enhanced production of gellan by S. paucibilis NK2000 was 36 hr after cultivation. The concentrations of gellan produced by S. paucibilis NK2000 from 1) 20.0 g/l glucose without additional feeding, 2) 20.0 g/l glucose with feeding of 200.0 g/l glucose at 36 hr, in which the final concentration in the medium was 10.0 g/l, 3) 20 g/l glucose with feeding of 200.0 g/l glucose and a shift in an agitation speed from 400 to 600 rpm, 4) 20.0 g/l glucose with feeding of 200.0 g/l glucose at 36 hr and shifts in an agitation speed from 400 to 600 rpm and an aeration rate from 1.0 to 1.5 vvm, 5) and 20.0 g/l glucose with feeding of 200.0 g/l glucose at 36 hr and shifts in an agitation speed from 400 to 600 rpm and an aeration rate from 1.0 to 2.0 vvm, were 5.19, 5.74, 6.73, 7.93, and 9.40 g/l, respectively, and their conversion rates from glucose were 26.0, 19.1, 22.4, 26.4, and 31.3%, respectively. Compared to those developed using a normal process, production of gellan by S. paucibilis NK2000 from 20.0 g/l glucose was 1.81 times higher, and and its conversion rate was 1.20 times higher when the optimized process developed in this study was used.

Substrate Removal Condition in Activated Sludge Process of Wastewater from Acetaldehyde Manufacturing Plant (Acetaldehyde폐수의 활성오이법에 의한 기질제거조건)

  • This study is conducted to investigate treatability by activated sludge process for wastewater from acetaldehyde manufacturing plant. The optimum hydraulic retention time in aeration tank for removal of high strength substrate were measured. The removal efficiency were checked out by hydraulic retention time : 35hr., 40hr. and 45hr., respectively. $COD_{Cr}$, like substances were removed in all hydraulic retention time zone directed for efficiency, but non-biodegradable substances were remained. $COD_{Cr}$ biomass loading was 0.81kg $COD_{Cr}/kgMLVSS$ . day at 35hr. of retention time, 0.34 kg$COD_{Cr}$/kg MLVSS . day at 40hr., and O.l9kg$COD_Cr$/kgMLVSS . day at 45hr. And the mean $COD_{Cr}$, removal efficiency was 65.5%, 81.6% and 83.0%, respectively. And also $COD_{Cr}$, volume loading was 1.01kg$COD_{Cr}/m^3$ day, 0.87kg$COD_{Cr}/m^3$ - day, and 0.79kg$COD_{Cr}/m^3{\cdot }$day, respectively. The basic design parameter obtained is as fallows. The value of Specific substrate removal rate coefficient (k), Yield coefficient(Y) and Decay coefficient($k_d$) was $0.0013day^{-1}$, $0.505kgMLVSS/kgCOD_{Cr}$ and $0.040day^{-1}$, respectively.

Treatment of Dairy Wastewater by the Combination of RBC and tapered Aeration (RBC와 점감포기조의 복합구성에 따른 유가공폐수 처리에 관한 연구)

  • The treatment efficiency of rotating biological contactors (RBCs) for the high strength of dairy wastewater was investigated. Two different systems were conducted composing of a single RBC with tapered aeration reactors for the system A and a sequential RBCs following tapered aeration reactors for the system B. Experiments using dairy wastewater were conducted for 50 days period of time, in which hydraulic rates were maintained at the constant ratios of 346L per day and variable BOD concentrations were at the range from 1,358mg/L to 829mg/L, the $COD_{cr}$, concentration of the range were from 2,384mg/L to 1,329mg/L, the range of T-N concentrations was from 66mg/L to 38mg/L, and 50% of internal recycle and 50% of sludge return were performed. Results indicated that the removal efficiencies of the system B were higher than those of the system A. The removal efficiencies of system A for the BOD, $COD_{cr}$, T-N and T-P were 97.8%, 96.7%, 87.2% and 82%, respectively. The removal efficiencies of system B for the BOD, $COD_{cr}$, T-N and T-P were as of 98.5%, 98.5%, 91.3% and 89%.

Drying Characteristics of Oak Mushroom Using Conveyer Far Infrared Dryer - Down Draft Air Flow Type - (컨베이어 원적외선 건조기를 이용한 표고버섯의 건조특성 - 하향 송풍방식 -)

  • This study was conducted to develop a new drying method far reducing the drying cost and time and to investigate the drying characteristics of oak mushroom. A far infrared dryer of down draft air flow type used for this experiment can control the drying parameters, such as far infrared heater temperature and aeration velocity. The far infrared drying tests were performed at aeration velocities of 0.3 and 0.6m/s under the temperature of 90 and 100$^{\circ}C$ in for infrared heater, respectively. The results were compared and analyzed with those of an heated air drying method used as a control in terms of properties representing the drying characteristics. such as shrinkage rate, color, energy consumption amino acid components, drying rate and moisture ratio. The results obtained from this research can be summarized as follows. 1. The drying rate of far infrared drying was faster than that of heated air drying. With high temperature of far infrared heater and slow aeration velocity, the far infrared drying of down draft air flow type was superior to the heated air drying. 2. Most of far infrared drying conditions required less energy consumption than heated air drying. 3. The shrinkage rates of heated air drying and far infrared drying were decreased by 17.0% and 18.2∼19.8%, respectively. 4. The difference of color on oak mushroom surface before and after drying can be represented as $\Delta$E. $\Delta$E values of far infrared drying and heated air drying were 2.39∼4.55 and 6.77, respectively. 5. The amounts of free amino acids were higher in the far infrared than in the heated air drying. In addition the amounts of Gln and Glu generally were increased and those of Ala, Leu, and Val were decreased in order.

Exopolysaccharide Production and Mycelial Growth in an Air-Lift Bioreactor Using Fomitopsis pinicola

  • For effective exopolysaccharide production and mycelial growth by a liquid culture of Fomitopsis pinicola in an air-lift bioreactor, the culture temperature, pH, carbon source, nitrogen source, and mineral source were initially investigated in a flask. The optimal temperature and pH for mycelial growth and exopolysaccharide production were $25^{\circ}C$ and 6.0, respectively. Among the various carbon sources tested, glucose was found to be the most suitable carbon source. In particular, the maximum mycelial growth and exopolysaccharide production were achieved in 4% glucose. The best nitrogen sources were yeast extract and malt extract. The optimal concentrations of yeast extract and malt extract were 0.5 and 0.1%, respectively. $K_2HPO_4\;and\;MgSO_4{\cdot}7H_2O$ were found to be the best mineral sources for mycelial growth and exopolysaccharide production. In order to investigate the effect of aeration on mycelial growth and exopolysaccharide production in an air-lift bioreactor, various aerations were tested for 8 days. The maximum mycelial growth and exopolysaccharide production were 7.9 g/l and 2.6 g/l, respectively, at 1.5 vvm of aeration. In addition, a batch culture in an air-lift bioreactor was carried out for 11 days under the optimal conditions. The maximum mycelial growth was 10.4 g/l, which was approximately 1.7-fold higher than that of basal medium. The exopolysaccharide production was increased with increased culture time. The maximum concentration of exopolysaccharide was 4.4 g/l, which was about 3.3-fold higher than that of basal medium. These results indicate that exopolysaccharide production increased in parallel with the growth of mycelium, and also show that product formation is associated with mycelial growth. The developed model in an air-lift bioreactor showed good agreement with experimental data and simulated results on mycelial growth and exopolysaccharide production in the culture of F. pinicola.

High-Rate Nitrogen Removal using a Submerged Module of Sulfur-Utilizing Denitrification (침지형 황 탈질 모듈을 이용한 고속의 질소제거)

  • This study aims to develop a sulfur-using denitrification process which is possible a renovation to advanced treatment plant submerging a simple module in activated sludge aeration tank. At first, the impact factor of sulfur-using denitrification was appreciated by the batch test. Secondly, reflecting a dissolved oxygen effect of sulfur-using denitrification that was confirmed by the batch test, in a continuous nitrification/sulfur-using denitrification, high-rate nitrogen removal reaction was induced at optimum condition controlling DO concentration according to phases. Also, inside and outside of sulfur-using denitrification module was covered with microfilter and the module was considered as an alternative of clarifier. Result of batch test for sulfur-using denitrification, $NO_2{^-}N$ was lower for consumption of alkalinity and sulfur than that of $NO_3{^-}-N$. These results revealed the accordance of theoretical prediction. In continuous nitrification/sulfur-using denitrification experiment, actual wastewater was used as a influent, and influent nitrogen loading rates were increased 0.04, 0.07, 0.11, $0.14kg\;N/m^3-day$ by changing hydraulic retention times. At this time, nitrogen loading rates of packed sulfur were increased 0.23, 0.46, 0.69, $0.93kg\;N/m^3-day$. As a result, nitrification efficiency was about 100% and denitrification efficiency was 93, 81, 79, 72%. Accordingly, nitrogen removal was a high-rate. Also the module of sulfur-using denitrification covered with microfilter did not make a fouling phenomena according to increased flux. And the module was achieved effluent suspended solids of below 10 mg/L without a clarifier. In conclusion, it is possible a renovation to advanced treatment plant submerging a simple module packed sulfur in activated sludge aeration tank of traditional facilities. And the plant used the module packed sulfur is expected as a effective facilities of high-rate and the smallest.
