Determination Times for Induction of Adventitious Shoots, Roots, Trichomes, and Calli from Segments of Arabidopsis thaliana by NAA and BA (애기장대(Arabidopsis thaliana)잎 절편에서 NAA와 BA에 의한 신초, 부정근, 모용 및 캘러스 형성 결정 시기)

  • The effect of NAA and Benzyladenine(BA) for determination times on the formation of adventitious shoots, roots trichmoes and calli in MS basal medium was investigated in leaf segments from ecotype 'Nosses' of Arabidopsis thalliana. Adventitious shoots, roots, trichomes and calli were formed fromed from leaf segments in a wede range of NAA and BA. The optimal combination of hormones for adventitious shoots formation, 20mg/L NAA for trichome formation, 100mg/L for callus formation. Inductive times for formation of adventitious shoots, roots, trichomes and calli were determined at 14, 4, 6 and 18 days respectively by periodical transfer of leaf segments from hormines containing media to hormone free medium.

  • Micropropagation and adventitious root production via the culture of Polygonatum odoratum were performed. Stem segments of seedlings of Polygonatum odoratum were the most efficient explants for adventitious shoot formation compared to leaf and root segments. Exogenous cytokinin treatment was required for adventitious shoot formation. Among the cytokinin (BA, Kinetin and Zeatin) tested, BA was most effective for shoot formation from stem segments. Auxin (NAA or IBA) in combination with cytokinin significantly enhanced adventitious shoot formation. Twenty five percent of explants produced adventitious shoots on medium with 2.0 mg/L BAP alone, while 83% of explants produced adventitious shoots on medium with the combination of 2.0 mg/L BAP and 0.1 mg/L IBA. Rooting of adventitious shoots was achieved after transferring to 112 MS medium supplemented with 0.1 mg/L IBA and 0.5 mg/L zeatin. When stem segments were cultured on MS medium with various kinds of auxin (IBA, NAA and 2,4-D), adventitious roots were formed from callus. frequency of adventitious root formation was highest in 2,4-D than NAA and IBA. When roots were in clusters together with parental stem segments, growth of roots actively occurred in hormone-free MS liquid medium. The above results represent that possible application for the mass production of roots and plantlets through in vitro culture system of Polygonatum odoratum.

Thidiazuron Induced High Frequency Adventitious Shoot Formation and Plant Regeneration in Capsicum annuum L.

  • An efficient procedure was developed for adventitious shoot bud induction and plantlet regeneration from various explants of the ten genotypes of Pepper (Capsicum annuum L.) using Thidiazuron (TDZ). Among various treatments at 1.0-3.0 mg/L TDZ Induced maximum number of adventitious shoots depending upon the explant type and genotype compared to other treatments. Among the explants tested, leaf induced maximum number of adventitious shoots than the cotyledons. TDZ-mediated organo-genesis was possible in 10 pepper cultivars, the extent of the response being genotype-dependent. Of the ten genotypes tested, C. annuum cvs CA960, $G_4$ and X-235 were produced maximum number of adventitious shoots and Sell was the least, and all other genotypes gave moderate response. Elongation of multiple shoots was observed on medium supplemented with SA (0.05 mg/L) in combination of IAA (0.05 mg/L). Differences in ability for in vitro shoot regeneration and elongation depend upon the variety and explant type. The elongated shoots were success. Fully rooted on MS medium containing at 1.0 mG/L IAA. Plantlets regenerated from different explants of ten genotypes were found to be diploid (2n=24) and were devoid of any chromosomal aberrations. Regenerated plants were successfully established in soil where 85-90% of them developed into morphologically normal and fertile plants.

  • Xanthoceras sorbifolium Bunge (yellowhorn) is a woody tree in the soapberry family, Sapindaceae, native to northern China. This species has been identified as a major woody bioenergy plant for bio-diesel production because of high oil content in seed. But the flowers do not bear fruit well while the many flowers blooming. This study was performed to regenerate in vitro plantlet using adventitious shoot formation. To establish the protocol of plant regeneration, adventitious shoots formation rate in the culture of cotyledon of immature zygotic embryos was 68.6% in 1/2 MS medium with 0.5 mg l-1 BA and 3% sucrose (w/v). In the culture of cotyledons of mature zygotic embryos, induction of adventitious shoots was needed to contain high sucrose in pre-culture medium and the frequency of shoot induction was 64.4%. Multiple shoots were induced in 0.5 mg l-1 TDZ, and rooting of shoot was induced 4.0 mg l-1 IBA. Flow cytometry analysis revealed that all the regenerated plantlets were diploid.

  • When the leaves, roots and stem segments of seedling of Polygonatum odoratum were cultured on Murashige and Skoog medium with 2.0mg/l BAP, stem segments were the most efficient explants for adventitious shoot inductino. To observe the efficient combination of growth regulators on the adventitious shoot formation , stem segments were cultured on MS medium with various kinds of cytokinins (BAP, kinetin, zeatin). From this experiment, cytokinin treatement was prerequisite for theadventitious shoot formatino,especially BAP was the most effective. Auxin (NAA or IBA) in combination with cyotokinin highly enhanced the adventitious shoot formation. Twenty five percents of explants produced the adventitious shoots on medium with 2.0mg/l BAP solely, while 83% of explants produced the adventitious shoots on medium with 2.0mg/l BAP and 0.1mg/l IBA. Root formationform adventitious shoot was promoted after transfer to 1/2 MS medium supplemented with 0.1mg/l IBA and 0.5mg/l zeatin, thereafter the plantlets with shoots and roots were cultured on 1/2MS medium lacking growth regulators. When the stem segments were cultured to MS medium with 1.0mg/l 2,4 NAA and IBA , yellow and nodulous cali were formed from the stem segments which were developed into adventitious roots. These roots were actively grew after transferred to MS liquid medium lacking growth regulators.

  • Adventitious organs (shoots and roots) from mature leaves of Populus deltoides (Eastern cottonwood) clones were induced on WPM (Woody Plant Medium) containing fructose as a carborn source and different combinations of either BA (Benzyl aminopurin) or TDZ (Thidiazuron) with NAA (Naphthaleneacetic acid). The morphologies of adventitious shoots were different depending on the kinds of growth regulators and the length of shoots induced on the medium containing BA were generally longer than that of TDZ. BA 0.5 mg/L and TDZ 0.01 mg/L combined with either 0.05 or 0.1 mg/L NAA showed the highest values in the frequency of regeneration (63% to 100%) the number of shoots per explant and in the organogenic sites per explant among tested three clones (A, B and C). Although there was a minor clonal difference in the development of adventitious shoots from leaf segments, their responses to growth regulators were very similar.

  • The effect of auxins and cytokinins on the formation of adventitious shoots, adventitious roots, trichomes, and calli in MS basal medium was investigated in leaf segments from ecotype Columbia of Arabidopsis thaliana. Adventitious shoots, adventitious roots, trichome, and calli were formed from leaf segments by a wide range of hormone concentrations and combinations. Adventitious shoots were formed respectively in treatment with 0.1mg/L IAA and 10 mg/L BA. Adventitious roots were formed in treatments with low concentration of IAA and NAA. Trichomes and calli were formed by increasing the concentration of IAA and NAA. The optimal combination was 0.5mg/L NAA and 0.1mg/L BA for trichome formation, 10mg/L NAA and 10mg/L BA for calli formation. When NAA was treated alone in culture media, adventitious roots were formed in 0.1mg/L, trichomes were formed in 2.0mg/L, and calli were formed in 10mg/L. Inductive time for formation of adventitious roots, trichomes and calli were determined at 6,7 and 18 days respectively by periodical transfer of leaf segments from NAA containing medium to NAA free medium.

Plant Regeneration via Adventitious Shoot Formation from Hypocotyl Explants of Groundcherry (Physalis angulata L.) (땅꽈리(Physalis angulata L.) 하배축 절편으로부터 신초 형성을 통한 식물체 재분화)

  • In the present study, plant regeneration through adventitious shoot formation from hypocotyl segments of in vitro seedlings of groundcherry (Physalis angulata L.) was investigated to determine the optimum culture conditions for highly efficient regeneration of the species. Adventitious shoots in hypocotyl segments were efficiently induced on MS media with low concentrations of BAP, specifically, with 0.5-1.0 mg/L BAP singly or in combination with 0.1-0.5 mg/L NAA. The 1.0 mg/L BAP single treatment was most effective for forming multiple adventitious shoots. When the induced shoots were transferred to the root induction media, low concentrations of NAA, IBA, and IAA enhanced the development of adventitious roots from adventitious shoots, suggesting that low concentrations of auxins were optimal for producing regenerated plantlets. The number of roots per shoot was large (> 2.0), and the root length exceeded 8.0 cm. In particular, the development and the overall shape of the roots were ideal. Furthermore, the number and length of shoots exceeded 2 and 6.0 cm, respectively. When the regenerated plantlets were transferred to compost soil, the root and shoot systems had developed well to the point that all of the regenerated plantlets acclimated successfully, resulting in normal morphology and growth characteristics, similar to those of the mother plant. Therefore, plant regeneration via adventitious shoot formation is expected to be one of the main methods for producing groundcherry on a large scale for a stable supply of the raw materials.

Plant Regeneration and Mutagenesis from Organogenic Callus of Dianthus Distributed in Gangwon Province (강원지역 패랭이꽃속의 캘러스로부터 식물체 재분화와 돌연변이체 유발)

  • Useful Dianthus species were collected and selected from two native and seven foreign species distributed in Gangwon province. For in vitro breeding,. callus was induced from the explants of apical meristem, leaf, stem and the in vitro adventitious shoots on MS basal medium with 2.0 mg/L 2,4-D and 0.5 mg/L BA at 27$^{\circ}C$ under continuous light. After 3 weeks of culture, calli initiated the most highly from the leaf explants of D. chinensis Organogenic calli were able to be selected from the adventitious shoot-derived calli. For shoot regeneration, these organogenic calli were cultured on MS medium with the combination of 0.1 mg/L NAA+1.0 mg/L BA under continuous light. Multiple shoots were proliferated with low frequency (about 30%) from those adventitious shootderived calli. Also, shoots initiated directly from the adventitious shoot explants without callus formation at high frequency of 52% when cultured on N6 medium containing 0.1 mg/L NAA and 1.0 mg/L BA in D. gratianopol. Multiple shoots and plantlets grew well and rooted on MS medium supplemented with 0.1 mg/L NAA. Regenerants with well-developed roots were transferred to 8-cm pots containing vermiculite at 85% relative humidity and 27$^{\circ}C$ These plantlets were acclimatized in artificial soil mixture and transferred to the greenhouse for flowering with normal phenotypes. M28 Mutant line was selected with white flowers from 0.03M EMS-treated organogenic calli derived from in vitro adventitious shoot explants of D. chinensis and set seeds.

In vitro Anti-inflammation Effect of Adventitious Shoots of Toona sinesis in Propionibacterium acnes-induced Skin Dermatitis

  • Toona sinensis (TS) leaf is known to antinociceptive, antioxidative stress and skin moisturizing effects. Acnes vulgaris is a chronic skin disease with various symptoms including itchiness, pain and interruption of normal skin function. Propionibacterium acnes (P. acnes) is a major factor in the occurrence of inflammatory acnes. This study evaluated the antioxidant and anti-inflammation effects by TS extract from adventitious shoots. TS extract showed anti-inflammatory activities by suppression of pro-inflammation mediators (iNOS and COX-2) in LPS-stimulated RAW264.7 cells. TS extract also has anti-inflammatory activities by inhibiting the secretion of pro-inflammatory cytokines on P. acnes-stimulated HaCaT cells. These effects were regulated by MAPK signaling pathway. Therefore, we suggest that TS extract from adventitious shoots might have applications as a medicine for treating P. acnes-induced skin diseases.
