Chemotype Discrimination and Rapid Identification of Angelica Roots by DART-TOF-MS

  • The Angelica root has been used as a medicinal herb in many Asian countries including Korea, China, and Japan. Angelica gigas, A. sinensis, and A. acutiloba have been considered as Angelicae radix in Korean, Chinese, and Japanese Pharmacopoeia, respectively. Since the origins of Angelicae radix differ from country to country, there is a need to develop an efficient analytical method to identify the origin of the Angelica root. In order to obtain chemical fingerprints, three different Angelicae Radices were analyzed by direct analysis in real time mass spectrometry (DART-MS). Significantly different DART-MS spectra were observed from three different species of Angelicae Radix. Strong peaks of decursin or decusinol angelate, and its dimer were exclusively found from A. gigas. Ligustilide and linoleic acid were detected as the major component from A. acutiloba. The strongest ligustilide peak was observed from A. sinensis. DART-MS fingerprinting is a promising method for the rapid identification and/or quality control of Angelicae Radix.

Anti-Photoaging Effects of Angelica acutiloba Root Ethanol Extract in Human Dermal Fibroblasts

  • The effects that ultraviolet rays elicit on collagen synthesis and degradation are the most common causes of wrinkle formation and photo-aging in skin. The objectives of this study were to evaluate the effects of Angelica acutiloba root ethanol extract (AAEE) to promote collagen synthesis and inhibit collagen degradation in human dermal fibroblasts. By examining total polyphenol and flavonoid contents, electron donating ability, radical scavenging activity, and superoxide dismutase-like activity, we found that AAEE exhibited fairly good antioxidant activity. Treatment with AAEE significantly increased type I procollagen production by cultured fibroblasts, as well as reduced ultraviolet-induced matrix metalloproteinase-1 (MMP-1) expression and MMP-2 activity in a dose-dependent manner (p < 0.05). In addition, AAEE significantly increased TIMP-1 mRNA expression (p < 0.05), although without an associated dose-dependent increase in TIMP-1 protein expression. In summary, we suggest that AAEE may be a potentially effective agent for the prevention or alleviation of skin-wrinkle formation induced by ultraviolet rays.

Comparison of essential oil composition between Angelica gigas and Angelica acutiloba

  • Two kinds of Angelica belong to Umbelliferae collected, the one is Angelica gigas that is inhabitant in Korea and the other is Angelica acutiloba that is indigenous in Japan at the field of Snyder Research and Extension Farm Rutgers University, New Jersey and was analyszed by GC and GC/MS. The composition of the essential oil of the different aerial parts of the Angelica has been studied. The oil yields obtained upon hydrodistillation were 0.18% (v/w) in Korean Angelica and 0.44% (v/w) in Japanese Angelica on dry root weight basis. By the growing stage in the Rutgers greenhouse condition, leaf and root of essential oil content a little decreased on 9 months later than 4 months later except for Angelica gigas leaf. Both of Angelica showed that amounts of essential oil content presented in order of leaf > petiole > root according to different plant part. The analysis of the essential oil from Angelica root led to the identification of 14 constituents totaling 64% in Korean Angelica and 13 constituents totaling 68% in Japanese Angelica. The major constituents of the Angelica root essential oil were ligustilide (47 %) and gamma terpi (14 %) in Korean Angelica, and alpha pinei (32 %) and nonane (25 %) in Japanese Angelica

한약재 추출물의 항산화작용 및 아질산염 소거작용 (Antioxidative and Nitrite Scavenging Abilities of Medicinal Plant Extracts)

  • 본 연구는 3종류의 약용식물(당귀, 목통, 골담초)을 물과 $70\%$ 에탄올로 추출하여 전자공여능과 아질산염 소거능을 조사하여 건강 식품의 제조에 필요한 기초자료를 얻고자 하였다. 3종류의 한약재중 물추출물의 수율은 목통($35.63\%$)>골담초($31.36\%$)>당귀($8.98\%$)의 순이었으며 에탄을 추출물의 수율은 목통($31.82\%$)>당귀($14.15\%$), 골담초($5.20\%$)의 순으로서 목통이 가장 높은 수율을 나타내었다. 한약재 추출물의 전자공여능은 물추출물 300-1,000 ppm에서 당귀가 $62-88\%$로 가장 높았으며, 에탄을 추출물 300-1,000 ppm에서는 목통이 $72-87\%$로 가장 높은 전자공여능을 나타내었다. 아질산염 소거능은 pH 1.2에서 가장 높았고 pH가 증가할수록 감소하였다. pH 1.2에서 아질산염 소거능은 물추출물 1,000 ppm에서 $35-40\%$로 골담초가 가장 높았고, 에탄을 추출물 1,000 ppm에서 $33-42\%$로서 목통이 가장 높았다. 3종류의 한약재 중에서 목통은 추출물의 수율이 가장 높고 전자공여능과 아질산염 소거능이 가장 우수하여 식품에 항산화 및 아질산염 소거의 목적으로 첨가할 수 있는 적합한 약재로 생각된다.

한국.중국.일본 당귀가 cyclophosphamide로 유발된 흰쥐의 빈혈에 미치는 영향의 차이 (Differences of Hematopoietic Effects of Angelica gigas, A. sinensis and A. acutiloba Extract on Cyclophosphamide-induced Anemic Rats)

  • 항암제인 cyclophosphamide를 흰쥐에 투여하여 유발된 빈혈증상에 대하여 한국당귀, 중국당귀, 일당귀의 투여가 빈혈회복에 미치는 효능을 비교한 연구로서, Sprague-Dawley계 수컷 흰쥐에게 cyclophosphamide를 30mg/kg BW/day로 3회 복강 투여 후, 참당귀, 중국당귀와 일당귀 추출물을 1 g/kg BW/day로 14일 동안 경구 투여하였다. Cyclophosphamide 투여로 체중감소는 대표적인 전신증상으로 나타났으나 당귀추출물의 투여가 cyclophosphamide에 의한 체중감소 부작용을 저해함으로써 체중증가를 유도한 것으로 볼 수 있다. 참당귀 투여군은 적혈구 용적(hematocrit, Hct) 변화, 적,백혈구수 변화, 혈색소 농도 변화, 혈청 iron, transferrin 포화도 등의 항목에 대하여 유의성 있는 회복 효과를 나타내었고, 중국 및 일당귀 투여군은 백혈구수 감소에 대하여 유의성 있는 회복효과를 나타내었다. Cyclophosphamide 투여 후 비타민 $B_{12}$ 농도는 정상군에 비해 유의적으로 낮았고, 중국당귀, 참당귀, 일당귀 순으로 비타민 $B_{12}$ 농도가 유의적으로 증가하는 것을 관찰하였다. 이는 당귀가 비타민 $B_{12}$의 농도를 증가시키면서 혈액생산작용에 도움을 줄 것으로 여겨진다. 결론적으로 참당귀추출물 투여로 인해 혈액수치가 향상된 결과는 암 환자가 화학요법으로 인하여 생긴 빈혈치료에 도움이 될 것이라 생각된다.

토당귀와 일당귀의 화학성분 비교 (Comparision of Chemical Components of Angelica gigas Nakai and Angelica acutiloba Kitagawa)

  • 국내에서 재배 수확되는 토당귀와 일당귀를 한방이나 가공시에 기초 자료로 활용될 수 있도록 기본적인 화학 성분을 분석하였다. 일반성분은 건물기준으로 토당귀와 일당귀의 조단백질은 각각 18,1, 13.4%, 조지방은 8.9, 4.3%, 조섬유는 8.6, 9.4%,조회분은 7.4, 8.2%, 당질은 57.0, 64.7%이었다. 두 종류의 당귀 속에 함유된 무기질 함량은 칼륨이 각각 2,740.0, 2,582.4 mg%로 가장 높았으며, 유리당 조성은 sucrose가 각각 0.4, 0.3%, fructose와 glucose는 검출되지 않았다. 토당귀와 일당귀의 지방산 조성은 linoleic acid가 각각 60.8, 59.9%, palmitic acid는 17.4, 15.3%. oleic acid는 8.8, 7.7%이었는데 두 시료간 뚜렷한 차이점은 발견할 수 없었다. 토당귀와 일당귀의 주요 구성아미산으로 아르기닌이 각각 2599.8, 1543.4 mg% 로 가장 많았고, 티로신이 각각 113.7, 84.9 mg%로 가장 적게 함유되어 있었다. 비타민 $B_1$,는 토당귀와 일당귀에 각각 10.5, 12.2 mg%, 비타민 $B_2$,는 각각 0.1, 0.04 mg%, 지표성분인 decursin은 각각 4.3, 0.8%이었다 탄닌은 토당귀, 일당귀에서 각각 988.0, 900.0 mg%로 분석되었다.

미각센서와 DNA 염기서열을 이용한 당귀류 비교 (Comparison of Angelica Species Roots Using Taste Sensor and DNA Sequencing Analysis)

  • Objectives : Angelica Gigantis Radix is prescribed as the root of different Angelica species on the pharmacopoeia in Korea, Japan and China. Chemical components and their biological activities were also different according to their species. A study for the development of simple method to compare Angelica roots was needed. In order to classify them, the methods such as DNA sequencing analysis and taste sensor were applied to three Angelica species like Angelica gigas, Angelica acutiloba and Angelica sinensis. Methods : PCR amplification of intergenic transcribed spacer (ITS) region was performed using ITS1 and ITS4 primer from nine Angelica roots, and then nucleotide sequence was determined. Taste pattern of samples were measured using the taste-sensing system SA402B equipped with a sensing unit, which consists of artificial lipid membrane sensor probes of anionic bitterness, astringency, saltiness, umami, and cationic bitterness (C00, AE1, CT0, AAE, and AN0, respectively). Results : As a result of comparing the similarity of the ITS region sequences, A. sinensis was discriminated from the others (A. gigas and A. acutiloba). Equally this genetic result, A. gigas and A. acutiloba showed similar taste pattern as compared to A. sinensis. Sourness, bitterness, aftertaste of bitterness, astringency, and aftertaste of astringency of A. sinensis were significantly high as compared with A. gigas and A. acutiloba. In contrast, richness was significantly low. Conclusions : These taste pattern can be used as a way of comparison of Angelica species and this technic could be applied to establish a taste pattern marker for standardization of herbs in various purposes.

Pharmaceutical Studies on "Dang-Gui" in Korean Journals

  • A crude drug "Dang-Gui", belonging to the genus Angelica, has been used as a traditional herbal medicine in Asia. Various studies have investigated the chemical components and pharmacological activities of Dang-Gui worldwide. However, domestic research results published in Korean are undervalued in international academia due to language barriers. Therefore, it is necessary to summarize the domestic research findings systematically for greater accessibility. This review focuses on the results published in four Korean pharmaceutical journals between 1970 and 2018, which detail the botanical, phytochemical, and pharmacological properties of three Angelica species (A. gigas, A. sinensis, and A. acutiloba) used as "Dang-Gui" in Korea, China, and Japan.

일본에서의 본초자원 재배 관리현황 -사물탕(당귀, 천궁, 작약, 지황)구성약재를 중심으로- (Current status of medicinal plant cultivation in Japan)

  • Objectives : In this report, we were studied the current status and future about cultivation of medicinal plants in Japan. Methods : To analysis the management and production of medicinal crops in Japan, variation of importation and cultivation of four medicinal crops such as Angelica acutiloba Kitagawa, Cnidium officinale Makino, Paeonia lactiflora Pallas and Rehmannia glutinosa Liboschitz var. purpurea Makino within 22 years ($1981{\sim}2002$) were investigated. Also we analyzed the herbal drug market in Japan using literature. Results : In the results, cultivated area and amount of Angelica acutiloba Kitagawa and Paeonia lactiflora Pallas reduced more than 50% within 22 years ($1981{\sim}2002$). Production decrease of medicinal crops is alloting through contract production in China. The cultivation of Rehmannia glutinosa Liboschitz var. purpurea Makino is enforcing minimum procluction for seed security. Conclusion : We hope that this report stimulates research to investigate for the establishment of cultural practices in Korea.
