Uptake kinetics of Cd and Zn by leaves and rhizome of the seagrass Zostera marina were examined in controlled laboratory radiotracer experiments. Subsequently, acute toxicity of Cd, Cu and TBT on photosynthetic quantum yield (ΔF/Fm’ of Z. marina were determined, and the differential sensitivities of rapid light curve (RLC) to those harmful substances were also compared. All measurements on photosynthetic activity were determined by chlorophyll a fluorescence method using pulse amplitude modulation (PAM). Metal uptake by Z. marina was saturated with increasing exposure time in leaves and rhizomes. Uptake of Zn by Z. marina was faster than that of Cd. Metal uptake rates in Z. marina decreased with the increase of dissolved metal concentrations and also with the increase of biomass. The adverse effect of TBT on effective quantum yield was stronger than other pollutants. Average acute toxicity on the RLC of the seagrass exposed to TBT and two heavy metals (Cd and Cu) was going to decrease as follows: TBT > Cd > Cu. Our preliminary results in this study suggested that Z. marina potentially can be used as a biomonitor of harmful substances contamination in coastal waters.
This study was conducted to investigate the potential acute toxicity of Aralia elata by a single oral dose in ICR mice. Thirty mice of each sex were randomly assigned to three groups of 10 mice each. The test articles were administered once by the gavage to mice at dose levels of 0, 2,500 and 5,000 mg/kg body weight. The mortality and changes on body weight and clinical signs of gross observation were monitored for 14 days after dosing. At the end of 14-day observation period, all animals were sacrificed and complete gross postmortem examinations were performed. There were no dead animal and adverse effects on clinical signs, the body weight and the gross finding. As the results, we could not find any toxic effect at the dose levels of 2,500 or 5,000 mg/kg in mice and the minimal lethal dose was considered to be over 5,000 mg/kg body weight in mice.
The acute toxicity study of combined antibiotic (Cefatrizine / Clavulanic Acid), a formulation consisting of cafatrizine and clavulanic acid in a ratio of 2 : 1, was evaluated in rats. The antibiotic was orally administered with single dose in dose levels up to 5 g/kg (0, 1.25, 2.5, 5 g/kg). Treatment-related effects were limited to soft stool excretion and caecal dilatation, but histologically no morphological changes could be detected in caecum. In hematology, serum-chemistry parameters and histopathology, no drug-related changes were found. The results of the present study indicate that cefatrizine / clavulanic acid has a low toxic potential and the oral $LD_{50}$values exceed 5 g / kg in rats
The toxicity values heavy metals were evaluated by immobilization and chronic reproduction impairment tests, using Daphnia magna. Acute tests were evaluated by the inhibition of their mobilization after 24hrs without food addition. The tests of reproductive impairment were investigated for 21 days by food addition and exchange or water. The effect of each concentration was assessed by Probit analysis and t-test. The results obtained from this study were as follows : 1) The change of pH and DO was not significant in the acute tests, while, in the reproductive tests, pH was increased by 0.3~1.4 and DO also increased. 2) The $E_iC_{50}$ values of immobilization to Daphnia magna in artificial fresh water were $0.030mg/{\ell}(Cu),\;0.054mg/{\ell}(Cd),\;0.12mg/{\ell};(Cr),\;0.74mg/{\ell}(Pb),\;3.4mg/ {\ell}(As)$ and the $NOE_iC$ values were $0.010mg/{\ell}(Cu),\;0.018mg/{\ell}(Cd),\;0.010mg/{\ell}(Cr),\;0.10mg/{\ell}(Pb),\;and\;$1.8mg/{\ell}(As)$. 3) The $E_rC_{50}$ values of reproductive impairment to Daphnia magna were $13.8\mu\textrm{g}/{\ell}(Cu),\;2.9\mu\textrm{g}/{\ell}(Cd),\;15.5\mu\textrm{g}/{\ell}(Cr),\;61.7\mu\textrm{g}/{\ell}(Pb),\;759\mu\textrm{g}/{\ell}(As)$, and $NOE_rC$ values were $0.95\mu\textrm{g}/{\ell}(Cu),\;$0.54\mu\textrm{g}/{\ell}(Cd),\;1.2\mu\textrm{g}/{\ell}(Cd),\;$7.4\mu\textrm{g}/{\ell}(Pb),\;110mu\textrm{g}/{\ell}(As)$. The results of tests using OECD artificial culture water were more sensitive than natural water for culturing. The presented data show that an artificial culture water is suitable in the experiment of bioassay for assessing the toxicity of marterials.
해양구조물 수중부의 해양생물 부착을 방지하는 효과적인 방법으로 방오도료를 사용하고 있다. 트리부틸틴(Tributyltin, TBT) 화합물은 우수한 방오성능을 가져 지금까지 광범위하게 사용해 되어 왔으나, 유해물질 사용금지에 따라 새로운 기술을 적용한 방오도료 개발이 진행되고 있다. 신규 방오도료는 낮은 독성을 가지면서도 우수한 방오성능을 가져야 한다. 본 논문에서는 상용 TBT-free 방오도료 3종(아산화동 함유 자기마모형 도료(Cu SPC AF), 아산화동을 함유하지 않는 자기마모형 도료(Cu-Free SPC AF), Foul-release 실리콘 도료(Foul release AF)의 용출수가 가지는 환경영향성을 조피볼락과 알테미아를 사용하여 평가하였다. 용출수에 대한 급성독성을 조사한 결과 방오도료 용출수의 농도와 생물종의 생존율은 반비례하는 경향을 나타내었으며, 자가마모형 도료가 Foul-release 실리콘 도료보다 상대적으로 높은 급성독성을 가지는 것을 확인할 수 있었다.
To provide information on the safety of crude antifungal compounds produced by Bacillus subtilus SN7 isolated from Meju, we carried out an acute (single) oral dose toxicity test and 4 week repeated oral dose determination test on crude antifungal compounds in male and female Sprague Dawley rats. In the acute toxicity test, rats were treated with crude antifungal compounds produced by Bacillus subtilus SN7 orally at increasing dose levels (500, 1,000, and 2,000 mg/kg) and observed for 2 weeks. In the repeated-dose 28-day oral dose determination study, rats were orally administered doses of 500, 1,000, and 2,000 mg/kg daily for 4 weeks. There were no test article-related deaths or abnormal clinical signs in the two studies. In the 4 week repeated oral dose determination test, there were also no significant differences in clinical signs, body and organ weight changes, or any other hematological and biochemical parameters between the control and treated groups. The results suggest that the crude antifungal compounds produced by Bacillus subtilus SN7 up to a dosage level of 2,000 mg/kg are not toxic in male and female rats.

A drought-tolerant transgenic rice (Agb0103) was developed using a pepper methionine sulfoxide reductase (CaMsrB2) under the control of rice Rab21 promoter with a selection marker, the phosphinothricin acetyltransferase (PAT) gene. Commercialization of genetically modified (GM) crops will require the evaluation of risks associated with the release of GM crops. With the potential problems associated to GM crops safety testing, the investigation of their effects on non-target organisms is necessary for environmental risk research. This study was carried out to assess acute toxicity of a GM crop using the water flea (Daphnia magna) for non-target organism risk evaluation. The effect of acute toxicity on Daphnia magna of Agb0103 rice and a non-GM rice, Ilmibyeo, were investigated at different concentrations (0, 625, 1,250, 2,500, 5,000, and 10,000 mg/L). The Agb0103 rice used for the test was confirmed to express the CaMsrB2/PAT gene by the PCR and ELISA. Daphnia magna feeding tests showed no significant differences in cumulative immobility or abnormal response with either Agb0103 rice or non-GM rice. The 48hr-EC50 values showed no difference between Agb0103 rice (2243 mg/L) and non-GM rice (2694 mg/L). These results suggest that there is no significant difference in toxicity to Daphnia magna between Agb0103 rice and its non-GM counterpart.
BACKGROUND: The disease resistant (OsCK1) rice was generated by inserting choline kinase (CK1) and phosphinothricin acetyltransferase (PAT) genes isolated from Oryza sativa and Streptomyces hygroscopicus into the genome of the rice, Nakdongbyeo. With the potential problems of safeties, the evaluations on non-target organisms are essentially required for the environmental risk assessment of genetically modified (GM) crops. In the present study, we conducted the evaluation of acute toxicity on Daphnia magna that commonly used as a model organism in ecotoxicological studies for non-target organism evaluation. METHODS AND RESULTS: Effect of acute toxicity to Daphnia magna by each concentration were investigated in the disease resistant (OsCK1) rice and non-genetically modified (non-GM) rice, Nakdongbyeo, as concentration (0, 1,000, 1,800, 3,240, 5,830, 10,500 and 20,000 mg/L). The OsCK1 rice used for the test was confirmed to express the OsCK1/PAT gene by the PCR(Polymerase chain reaction) and western blot analysis. Feeding test showed that no significant differences in cumulative immobility and abnormal response of Daphnia magna fed on OsCK1 rice or non-GM rice. The 48hr-$EC_{50}$ values showed no difference between OsCK1 rice (3,147.18 mg/L) and non-GM rice (3,596.27 mg/L). CONCLUSION: This result suggested that there was no significant difference in toxicity to Daphnia magna between OsCK1 rice and non-GM counterpart.
생활수준이 향상됨에 따라 세제의 사용량이 큰 폭으로 증가하고 있으며, 하수처리공정에서 완전히 처리되지 못한 합성세제가 하천이나 강으로 흘러들어 지표수의 오염을 가중시키고 있다. 이와같은 세제는 수중생물에 해로운 여러 독성물질을 함유하고 있으며, 현재까지는 주로 화학적 분석에 의하여 그 독성을 측정하고 있다. 그러나 오염물질 상호간의 작용 등으로 화학분석만으로 적정기간동안에 독성을 평가하는 것은 상당히 어려운 실정인 바, 본 논문에서는 물벼룩인 Daphnia magna와 형광성 박테리아인 Photobacterium phosphoreum을 이용하여 생물학적으로 주방용 세제와 의류용 세제의 급성 독성을 평가하였다. 본 평가에 사용된 세제는 세탁용 세제와 주방용 세제로 구분하여 국내 사용량이 가장 많은 7종류를 선택하였으며, 조사결과 세탁용 세제의 평균 24hr, 48hr-LC50은 각각 4.25%, 2.50%이며, 주방용 세제의 평균 24hr, 48hr-LC50은 각각 2.01%, 1.36%이었다. 또한 세탁용 세제의 평균 5min, 15min-EC50은 각각 1.83%, 1.02%이며, 주방용 세제의 평균 5min, 15min-EC50은 각각 1.57%, 1.21% 이었다. 이를 통하여 국내에서 사용되는 합성세제중 주방용 세제가 세탁용 세제보다 지속적 독성이 큰 것을 알 수 있었으며 세제마다 독성의 정도가 차이가 있었다.
Perfluorinated Compounds (PFCs) are anthropogenic compounds found in trace amounts in many environmental compartments far from areas of production. Along with the highly persistent nature of PFCs, there are increasing concerns over the potential adverse effects of them on the ecosystems. Most of highly fluorinated compounds degrade into PFOS and PFOA that are very stable compounds hard to break down. So, in this study, we tried to determine the toxicity of PFOS and PFOA in the terrestrial invertebrate. Acute toxicity test using earthworm, Eisenia fetida, was performed according to the OECD test guideline 207 (Earthworm, Acute Toxicity Tests). In the 14 day acute toxicity tests, the highest concentration causing no mortality and the lowest concentration causing 100% mortality of PFOS were 160 and 655 mg/kg (dry weight), respectively. And the highest concentration causing no mortality and the lowest concentration causing 100% mortality were 500 and 1,690 mg/kg (dry weight), respectively in the PFOA-exposure group. 14 day-LC50 values were estimated at the level of 365 and 1,000 mg/kg (dry weight) in the PFOS and PFOA-exposed group. These results indicate that under laboratory conditions PFOS is about 3 times more toxic to earthworms than PFOA. Based on known environmental concentrations of PFOS in the soil of Korea, which occur in the 0.42~0.73 ng/L range, there is no apparent risk to terrestrial invertebrate, earthworms. However, further work is required to investigate long-term effects on these and other terrestrial organisms.
