Metformin ameliorates bile duct ligation-induced acute hepatic injury via regulation of ER stress

  • Cholestasis is a condition in which the bile duct becomes narrowed or clogged by a variety of factors and bile acid is not released smoothly. Bile acid-induced liver injury is facilitated by necrotic cell death, neutrophil infiltration, and inflammation. Metformin, the first-line treatment for type 2 diabetes, is known to reduce not only blood glucose but also inflammatory responses. In this study, we investigated the effects of metformin on liver injury caused by cholestasis with bile acid-induced hepatocyte injury. Static bile acid-induced liver injury is thought to be related to endoplasmic reticulum (ER) stress, inflammatory response, and chemokine expression. Metformin treatment reduced liver injury caused by bile acid, and it suppressed ER stress, inflammation, chemokine expression, and neutrophil infiltration. Similar results were obtained in mouse primary hepatocytes exposed to bile acid. Hepatocytes treated with tauroursodeoxycholic acid, an ER stress inhibitor, showed inhibition of ER stress, as well as reduced levels of inflammation and cell death. These results suggest that metformin may protect against liver injury by suppressing ER stress and inflammation and reducing chemokine expression.

  • The present paper reports the life time prediction of Acrylonitrile-Butadiene rubber (NBR) fuel cell gasket materials as a function of operational variables like acid concentration, ageing time and temperature. Both material and accelerated acid-heat aging tests were carried out to predict the useful life of the NBR rubber gasket for use as a fuel cell stack. The acid ageing of the gasket compounds has been investigated at 120, 140 and $160^{\circ}C$, with aging times from 3 to 600 h and increasing acid ($H_2SO_4$) concentrations of 5, 6, 7 and 10 vol%. Material characteristics the gas compound such as cross-link density, tensile strength and elongation at break were studied. The hardness of the NBR rubber was found to decrease with decreasing acid concentration at both 120 and $140^{\circ}C$, but at $160^{\circ}C$ interestingly the hardness of the NBR rubber increased abruptly in a very short time at different acid concentrations. The tensile strength and elongation at break were found to decrease with increase in both the acid concentrate ion & temperature. The life time of the compounds were evaluated using the Arrhenius equation.

Kinetic Studies on the Ageing of Aluminum Hydroxide Gel (수산화알루미늄.겔의 노화에 관한 연구)

  • The rats of thermal ageing at elevated temperatures and heats of activation of aluminum hydroxide gel have been determined. The ageing was expressed in terms of diminution of acid consuming capacity. Although the actual rates of ageing were different by the samples, the slopes of their Arrhenius plots were the same. This may indicate similarity of ageing mechanisms and heats of activation of samples. The heat of activation was the mangitude of 30 Kcal/mole. Thus, rates of ageing of aluminum hydroxide gel may be greatly accelerated at elevated temperatures.

Effect of Extraction Methods on the Types and Levels of Free Amino Acid of Beef Longissimus Muscle

  • The current study was carried out to investigate the impact of extraction conditions on the free amino acid level and type in beef longissimus muscle. The sample blocks were chiller aged for 1 d and 7 d at $4^{\circ}C$. There are three homogenization speeds (11,000, 19,000 and 24,000 rpm) for bigger and two speeds (11,000 and 13,000 rpm) for smaller homogenizer's dispersing tools were used for evaluation. Results showed that chiller ageing greatly (p<0.05) increased extractable free amino acids, except cystine. Homogenization with the bigger dispersing tool at 24,000 rpm resulted in the highest free amino acid levels for both 1 and 7 d samples. Significant differences (p<0.05) in the mean values of most amino acids due to the effect of speed and interactions between ageing times. However, the speed effect and interaction between ageing with homogenization speed were not significant (p>0.05) for most of the amino acids except valine and isoleucine when using the smaller dispensing tools. The current data indicated that a standardized method for free amino acid types and levels of aged beef samples. In addition, the results also suggested that utilization of a big dispensing tool at high homogenization speed is a better condition for releasing free amino acid contents in beef samples.

  • Ambers have been used mostly as beads, jewelry and ornaments from ancient times and excavated as a buried artifact. When excavated, they are severely weathered to be cracked, exfoliated and disintegrated. Monitoring of changes in composition of amber according to weathering is very important for diagnosing the condition of amber and applying conservation materials and techniques. In this study, we tried to find the components of amber by analyzing amber with pyrolysis/GC/MS. The changes in the composition of pyrolzates after artificial ageing for 60 days under heat and oxygen were also observed. Abietic acid was detected as a main component of fresh amber and monoterpene, alkene, aromatic hydrocarbon were detected as major pyrolyzates. Changes with artificial ageing was estimated by comparing the peak area ratio of 23 components, and it was found that abietic acid abruptly decreased in the presence of heat and oxygen together, revealing that oxygen is a key factor to the deterioration of amber. It was also tried to understand the weathered surface of original amber gemstone based on the result of this ageing experiment.

  • The annals of Joseon Dynasty has been known as one of the UNESCO's Memory of the World Heritage. The annals of Joseon Dynasty composed with two kind of books, wax coated book and non coated book. Especially the waxed book have been severely damaged by various reason. For the safety preservation of the annals of Joseon Dynasty waxed book, the thermal ageing mechanism of beeswax coated Hanji paper has been evaluated. The weight loss of waxed Hanji were rapidly increased until 30 days in three temperatures(105, 120 & $150^{\circ}C$) and stabilized after 30 days. The acid value and relative intensity of carbonyl groups in beeswax were increased with strong conditions. This means that the deterioration of beeswax should be progressed with oxidizing reaction. The physical strength and the crystallinity of dewaxed Hanji were decreased with ageing time.

  • The synthesis conditions of fibrous calcium sulfate hemihydrate were investigated by using phosphogypsum and calcium sulfate hemihydrate. The unstable organogel was deposited by adding methanol to the saturated solution with gypsum at ageing temperature, and it was crystallized to fibrous gypsum hemihydrate while methanol was removed by rapid filtrating. In case of using calcium sulfate hemihydrate, fibrous $\beta$-calcium sulfate hemihydrate was formed by adding methanol of 67% to saturated solution at 6$0^{\circ}C$ and ageing for 5 minutes and filtering with suction. Minor components in phosphogypsum did not affect the reaction.

Effects of a Dietary Chitosan-Alginate-Fe(II) Complex on Meat Quality of Pig Longissimus Muscle during Ageing

  • The current study was conducted to investigate the effects of dietary chitosan-alginate-Fe(II) complex (CAFC) supplementation on carcass and meat qualities of pig m. longissimus during chiller ageing. One hundred and twenty-two LYD (Landrace${\times}$Yorkshire${\times}$Duroc) pigs were sampled from an industrial population. Seventy-four pigs (32 gilts and 42 barrows) were administered 3 ml of dietary supplementation of CAFC per day from 25 to 70 days of age, while the remaining 48 pigs (20 gilts and 28 barrows) were fed the same commercial feeding regime without the supplementation. For assessing the dietary effects on pH, objective meat color, cooking loss, water-holding capacity (WHC), thiobarbituric acid reactive substances (TBARS), volatile basic nitrogen (VBN) and fatty acid composition during ageing, 20 barrows (10 of each treatment) were randomly sampled, and aged for 3, 7, 12, 16, 20 and 25 days in a $1^{\circ}C$ chiller. The results showed that CAFC-fed pigs required approximately 10 fewer feeding days than the control group. Furthermore, the treatment resulted in greatly higher carcass grade whereby the grade A was increased by approximately 35% and 7% for gilts and barrows, respectively. The treatment had no significant effect (p>0.05) on pH, meat color and WHC during ageing. On the other hand, the CAFC-fed pigs showed significantly (p<0.05) lower TBARS values from 20 days of storage. In addition, the sum of unsaturated fatty acids for the treated group was significantly (p<0.05) higher than that for the control group after the storage time. This implied that CAFC supplementation could reduce the formation of free radicals in fatty acids (i.e., lipid oxidation). The treatment also significantly (p<0.05) retarded VBN formation during ageing, indicating a significant reduction in protein degradation. However, as there was no difference in pH between the two groups, the result raised a possibility that antibacterial activity of the CAFC alone could cause reduction in the formation of TBARS and VBN. In this regard, although the treatment effectively slowed down the formation of TBARS and TBA during chiller ageing, it was not resolved whether that was associated with the direct effect of the antioxidant function of chitosan and/or alginate, or a consequence of their antibacterial functions.

Effect of Ageing Temperature and Time on the Meat Quality of Longissimus Muscle from Hanwoo Steer (숙성온도와 기간이 한우 거세우 배최장근의 육질 특성에 미치는 영향)

  • This study was conducted to investigate the effect of ageing temperature(0 and $4^{\circ}C$) and time(2, 7, 14, 21, 28, and 49 days) on the meat quality of longissimus muscle from Hanwoo steer. The pH, Warner-Bratzler shear force(WBSF), myofibrillar fragmentation index(MFI), cooking and purge loss, sensory, morphological changes, and amino acid composition were assessed. The pH of longissimus muscle increased significantly(p<0.05) for 21 days at $0^{\circ}C$ and 14 days at $4^{\circ}C$, but decreased thereafter. Longissimus muscle aged from 14 days to 28 days at $4^{\circ}C$ had a lower WBSF than longissimus muscle aged at $0^{\circ}C$(p<0.05). The WBSF of longissimus muscle decreased significantly(p<0.05) with ageing time. The MFI, cooking and purge loss, tenderness, and juiciness scores were higher for longissimus muscle aged at $4^{\circ}C$ than at $0^{\circ}C$. As ageing time increased, the MFI, cooking and purge loss, tenderness, juiciness, and amino acid composition of longissimus muscle increased significantly(p<0.05) with ageing at 0 or $4^{\circ}C$. Transmission electron microscopy showed Z-disk degradation and morphological changes of the myofibrils correlating with increased tenderness during the ageing time. Based on these results, the proper ageing conditions of longissimus muscle of Hanwoo steer were 14 days of ageing at $0^{\circ}C$ or 7 days of ageing at $4^{\circ}C$. However, further study on the application of ageing technology to this industry is necessary for the optimization of ageing conditions for the sub-primals or muscles from Hanwoo beef.

Age-Related Changes of Adult Neural Stem Cells in the MouseHippocampal Dentate Gyrus

  • This study was designed to investigate the changes in the properties of the neuronal setm cells or progenitor cells associated with age-related decline in neurogenesis of the hippocampal dentate gyrus (DG). Active whole cells cycle marker Ki67 (a marker of whole cell cycle)-positive and S phase marker bromodeoxyuridine (BrdU)-positive. Neural stem cells gradually were reduced in the hippocampal subgranular zone (SGZ) in an age-dependant manner after birth (from P1 month to P1 year). The ratio of BrdUpositivecells/Ki67-positive cells was gradually enhanced in an age-dependent manner. The ratio of Ki67-positive cells/accu-mulating BrdU-positive cells at 3 hrs after BrdU injection was injected once a day for consecutive 5 days gradually decreased during ageing. TUNEL- and caspase 3 (apoptotic terminal caspase)-positive cells gradually decreased in the dentate SGZ during ageing and immunohistochemical findings of glial fibrillary acid protein (GFAP) were not changed during ageing. NeuN, a marker of mature neural cells, and BrdU-double positive cells gradually decreased in an age-dependent manner but differentiating ratio and survival rate of cells were not changed at 4 wks after BrdU injection once a day for consecutive 5 days. The number of BrdU-positive cells migrated from the hippocampal SGZ into granular layer and its migration speed was gradually declined during ageing. These results suggest that the adult neurogenesis in the mouse hippocampal DG gradually decrease through reducing proliferation of neural stem cells accompanying with cells cycle change and reduced cells migration rather than changes of differentiation.
