Molecular Networking-based De-replication Strategy Leads to the Isolation of a New Chromone from Pleosporales sp.

  • A new chromone analogue (1) was isolated from an EtOAc-extract of Pleosporales sp. culture medium, together with five known chromones (2 - 6). The isolation workflow was guided by a Molecular Networking-based dereplication strategy. The chemical structure of the new compound was elucidated using NMR and MS spectroscopy, and the absolute configuration was established by the Mosher's method. All isolated compounds were evaluated for their inhibitory effects on lipopolysaccharide-induced nitirc oxide production in RAW 264.7 macrophages. Compound 1 showed marginal inhibitory activity with an IC50 value of 118.7 μM.

Comparison and optimization of deep learning-based radiosensitivity prediction models using gene expression profiling in National Cancer Institute-60 cancer cell line

  • Background: In this study, various types of deep-learning models for predicting in vitro radiosensitivity from gene-expression profiling were compared. Methods: The clonogenic surviving fractions at 2 Gy from previous publications and microarray gene-expression data from the National Cancer Institute-60 cell lines were used to measure the radiosensitivity. Seven different prediction models including three distinct multi-layered perceptrons (MLP), four different convolutional neural networks (CNN) were compared. Folded cross-validation was applied to train and evaluate model performance. The criteria for correct prediction were absolute error < 0.02 or relative error < 10%. The models were compared in terms of prediction accuracy, training time per epoch, training fluctuations, and required calculation resources. Results: The strength of MLP-based models was their fast initial convergence and short training time per epoch. They represented significantly different prediction accuracy depending on the model configuration. The CNN-based models showed relatively high prediction accuracy, low training fluctuations, and a relatively small increase in the memory requirement as the model deepens. Conclusion: Our findings suggest that a CNN-based model with moderate depth would be appropriate when the prediction accuracy is important, and a shallow MLP-based model can be recommended when either the training resources or time are limited.

Hygrolansamycins A-D, O-Heterocyclic Macrolides from Streptomyces sp. KCB17JA11

  • Six ansamycin derivatives were isolated from the culture broth of Streptomyces sp. KCB17JA11, including four new hygrolansamycins A-D (1-4) and known congeners divergolide O (5) and hygrocin C (6). Compounds 1-5 featured an unusual six-membered O-heterocyclic moiety. The isolation workflow was guided by a Molecular Networking-based dereplication strategy. The structures of 1-4 were elucidated using NMR and HRESIMS experiments, and the absolute configuration was established by the Mosher's method. Compound 2 exhibited mild cytotoxicity against five cancer cell lines with IC50 values ranging from 24.60 ± 3.37 µM to 49.93 ± 4.52 µM.

  • Two new phenanthrenes, namely, (1R,2R)-1,7-hydroxy-2,8-methoxy-2,3-dihydrophenanthrene-4(1H)-one (1) and 2,7-dihydroxyphenanthrene-1,4-dione (2), were isolated from the ethyl acetate-soluble fraction of Dendrobii Herba, together with the eleven known compounds, densiflorol B (3), 6,7-dimethoxyphenanthrene2,5-diol (4), dehydroorchinol (5), 1,5,7-trimethoxy-2-phenanthrenol (6), denthyrsinin (7), ephemerantol A (8), lusianthridin (9), moscatilin (10), gigantol (11), 3-[(1E)-2-(3-hydroxyphenyl)ethenyl]-5-methoxyphenol (12) and (-)-syringaresinol (13). Structures of 1 and 2 were elucidated by analyzing 1D and 2D NMR as well as HR-ESI-MS data. The absolute configuration of compound 1 was confirmed by ECD spectroscopic method. In cytotoxicity evaluation using FaDu human hypopharyngeal squamous carcinoma cell line, compounds 3-6, 8, 10 and 12 showed activities, with IC50 values ranging from 2.55 to 17.70 μM. Among them, compound 10 exhibited remarkable cytotoxic activity with IC50 value of 2.55 μM. This is the first report on the anticancer mechanistic study of compound 10 in HNSCC including FaDu cells.

영화색채 하양의 활용 양상과 문화적 의미 (The Style and Cultural Significance of Film Color White)

  • 이 글은 한국영화 가운데 색채를 의도적이거나 관습적으로 활용한 사례들을 분석하였다. 장르영화 대부분이 관례적 활용 빈도가 높으며, 색채를 의도적으로 부각시킨 미학적 선택 또한 예술영화에 한정되지 않고 다양한 양식으로 나타난다. 영화색채의 시각적 인상만큼이나 그것의 분석과 해석은 주관적일 수 있다. 보기의 객관성을 위해 색채에 관한 오랜 문화적 관점을 채택하고, 유사성과 차이에 따라 발생하는 의미를 제시하고자 하였다. 영화색채 하양의 활용 양상의 분류는 보편적이고 일반적인 절대선, 특수한 사례로서의 강박과 치유, 성의 구분 없이 여성성의 특성을 보여주는 전형과 시각적 쾌락, 미학적 용어인 프레임의 경계를 넘어서는 유령적 사유라는 범주에서 접근하였다. 특정한 장르, 작가, 영화를 세밀하게 분석하는 미시적 방법보다는 하양을 시각장치로 활용하는 영화들의 유형별 특성을 분류해보고, 색채 활용의 미학적이고 문화적인 의미를 고찰하였다. 첫째, <악인전>(2019), <범죄와의 전쟁: 나쁜 놈들 전성시대>(2011), <불한당: 나쁜 놈들의 세상>(2016), <아수라>(2016), <나쁜 녀석들>(2019) 같은 영화들에서 하양이 절대선을 표현하는 시각의 전형으로 기능한다. 둘째, <기생충>(2019)의 공포와 불안, <곡성>(2016)의 악몽, <슈퍼맨이었던 사나이>(2008)의 과대망상, <기억의 밤>(2017)의 신경쇠약, <마더>(2009)의 광기, <윤희에게>(2019)의 강박, <밀양>(2007)의 히스테리 등이 가학과 피학을 시각적으로 강조하는 사례들이다. 셋째, <태극기 휘날리며>(2004), <포화 속으로>(2010), <마이웨이>(2011>, <고지전>(2011), <명량>(2014), <연평해전>(2015), <봉오동전투>(2019), <장사리: 잊혀진 영웅들>(2019) 등에서 여성 인물에 부여되는 하양이 전통적인 이미지를 고착시킨다. 넷째, 사회정치적 변화를 반영하는 <박하사탕>(2000), <변호인>(2013), <택시운전사>(2017), <1987>(2017) 등에서 하양이 역사의 순간을 기억하고 기록한다.

Deep Neural Network 기반 프로야구 일일 관중 수 예측 : 광주-기아 챔피언스 필드를 중심으로 (Deep Neural Network Based Prediction of Daily Spectators for Korean Baseball League : Focused on Gwangju-KIA Champions Field)

  • 본 연구는 Deep Neural Network(DNN)을 이용하여 광주-기아 챔피언스 필드의 일일 관중 수를 예측함으로써 이를 통해 구단과 관련기업의 마케팅 자료제공 및 구장 내 부대시설의 재고관리에 자료로 쓰임을 목적으로 수행 되었다. 본 연구에서는 Artificial Neural Network(ANN)의 종류인 DNN 모델을 이용하였으며 DNN 모델의 과적합을 막기 위해 Dropout과 Batch normalization 적용한 모델을 바탕으로 총 4종류를 설계하였다. 각각 10개의 DNN을 만들어 예측값의 Root Mean Square Error(RMSE)와 Mean Absolute Percentage Error(MAPE)의 평균값을 낸 모델과 예측값의 평균으로 RMSE와 MAPE를 평가한 Ensemble 모델을 만들었다. 모델의 학습 데이터는 2008년부터 2017년까지의 관중 수 데이터를 수집하여 수집된 데이터의 80%를 무작위로 선정하였으며, 나머지 20%는 테스트 데이터로 사용하였다. 총 100회의 데이터 선정, 모델구성 그리고 학습 및 예측을 한 결과 Ensemble 모델은 DNN 모델의 예측력이 가장 우수하게 나왔으며, 다중선형회귀 모델 대비 RMSE는 15.17%, MAPE는 14.34% 높은 예측력을 보이고 있다.

요양병원 외래 및 중앙진료부의 의료영역 간 연결관계에 관한 연구 (Study on the Network System between of the Outpatient and Central Treatment Department of Long Term Care Hospitals)

  • Purpose: As our population ages and becomes an elderly society the number of elderly care hospitals is rapidly increasing. Because physical functions and spatial perception in the elderly decrease with age, these hospitals require more systematic and intelligent space designs. The design of these spaces are even more complex because they must accommodate medical programs to treat various different diseases and ailments and also because there are many first time patients and irregular short term patients that seek out outpatient treatment services. Also by analyzing the spatial configuration systems and systematic relationships between each of the functional spaces of the outpatient treatment service departments for hospitals specialized in care for the elderly by focusing on the hallway and corridor systems of these hospitals, the according characteristics and trends were examined. Methods: Based on preceding research, the types of hallway and corridor systems of these hospitals were categorized into five types, including gallery corridors, middle corridors, hall-type, mixed type and cyclic type corridors, and into six types according to function including by medical diagnosis, patient registration, examination, administration and convenience and shared common space to derive any interconnecting relationships between the corridor systems. Also by comprehensively examining the types and combined utilization of the corridor types and the integration and the intelligibility of the space syntax, any trends within the corridor system were derived. The elderly care hospitals examined in this research study were twelve hospitals that opened after the year 2000 in Korea with more that 150 sick beds with areas larger than $1000m^2$ and with all outpatient medical service related rooms located entirely on a single floor of the hospital. Results: The following results could be confirmed based on this research study. 1) The spaces where medical diagnosis and examination occurred were adjacent, and the movement lines for first time patients and re-visiting patients were taken into consideration by separating the treatment space. 2) This research study confirmed that the larger the size of the hospital was, there were more detailed categorizations of treatment services and that there was a tendency for treatment areas to be separated and independent from examination areas. 3) There was a tendency for integration and intelligibility to decrease the more complex and diverse the combination of hall types designed into the corridor systems of these hospitals was. cyclic type corridors dramatically decreased the intelligibility of the corridor systems of these hospitals. 4) The priority rank of these spaces were confirmed to be highest in the order of registration, diagnosis, examination, treatment, administration and shared common spaces. However it was confirmed for the local integration that the diagnosis scope had the highest priority rank. Implications: There were exceptional cases confirmed where the number of unit spaces did not have an absolute effect on integration and intelligibility. These results can be interpreted to mean that this can be overcome through efficient architectural planning.

기준중위소득 방식을 반영한 보장시설생계급여 지급기준 논리 구성과 시설균등화지수 합리화에 따른 급여수준 (Logical Configuration of Livelihood Benefit Standard for the Institutionalized Recipients under the Standard Median Income Scheme and the Level of Benefit by the Adjusted Equivalence Scale of the Institution)

  • 본 연구는 국민기초생활보장제도의 맞춤형 개별급여체계로의 개편에 따른 보장시설 생계급여 지급기준 논리구성을 논의하고, 시설생계급여 수준을 제도의 정합성 차원에서 제시하는 것을 목적으로 한다. 2015년 7월, 국민기초생활보장제도의 개편에 따라, 일반수급자에게는 기준중위소득의 일정 비율에 따른 맞춤형 개별급여 지급기준이 설정되었다. 하지만, 보장시설에서 생활하는 시설수급자들에 대해서는 기존의 필수비목 비용을 반영한 절대적 최저생계비 개념이 적용되고 있었다. 이에 본 연구는 국민기초생활보장제도 개편 취지에 맞도록 시설생계급여 지급기준을 기준중위소득과 연동되도록 하였다. 이를 위해, 첫째, 시설수급자의 생계비목을 문헌연구와 FGI를 통해 도출하고, 둘째, 가계동향조사 4분위 이하 소비자료를 통해 일반생계급여의 반영비를 구하였다. 셋째, 대규모 시설에 불합리하게 적용되었던 시설규모별 균등화지수를 재조정한 뒤, 2019년 시설생계급여를 산출하였다. 향후 이러한 반영비 방식이 지속되기 위해서는 생계비 비목의 소비 성향 변화와 반영비의 안정성을 정기적으로 검토해나가야 하는 과제가 있다.

Zynq SoC를 이용한 초음파 신호처리 시스템 HW/SW co-design (HW/SW Co-design For an Ultrasonic Signal Processing System Using Zynq SoC)

  • 본 연구에서는 Xilinx의 Zynq SoC(system on chip)를 이용하여 초음파신호의 포락선을 검출하기 위한 신호처리 시스템을 설계하였다. 설계 툴로 Vivado IDE(integrated design environment)를 이용하여, 초음파 신호처리를 위한 전체 과정을 계층적 블록의 형태로 설계하였다. 제안된 시스템은 Zynq-7010의 내장 ADC, FIR(finite impulse response) 밴드패스 필터, 절대값 계산모듈, FIR 로우패스 필터 및 Kalman 필터 등으로 구성되며, 최종 단으로서 FIR 로우패스 필터를 사용하는 HW design 방식과 Kalman 필터를 사용하는 HW/SW co-design 방식에 대해 성능과 유효성을 비교하였다. 비교결과, 포락선 검출 성능에 있어서는 두 방식이 서로 유사한 특성을 갖지만, 시스템 개발에 소요되는 시간 측면에서는 HW/SW co-design 방식이 HW design 방식에 비해 훨씬 더 효율적임이 확인되었다.

Anti-adipogenic Pregnane Steroid from a Hydractinia-associated Fungus, Cladosporium sphaerospermum SW67

  • A pregnane steroid, 3α-hydroxy-pregn-7-ene-6,20-dione (1), was isolated from a Hydractinia-associated Cladosporium sphaerospermum SW67 by repetitive column chromatographic separation and high-performance liquid chromatography (HPLC) purification. The planar structure of 1 was elucidated from the analysis of the spectroscopic data (1D and 2D NMR spectra) and LC-MS data. The absolute configuration of 1 was determined by interpretation of ROESY spectrum of 1, together with the comparison of reported spectroscopic values in previous studies. To the best of our knowledge, this is the first report of the identification of the pregnane scaffold from C. sphaerospermum, a natural source. Compound 1 was evaluated for its effects on lipid metabolism and adipogenesis during adipocyte maturation and showed that compound 1 substantially inhibited lipid accumulation compared to the control. Consistently, the expression of the adipocyte marker gene (Adipsin) was reduced upon incubation with 1. Further, we evaluated the effects of 1 on lipid metabolism by measuring the transcription of lipolytic and lipogenic genes. The expression of the lipolytic gene ATGL was significantly elevated upon exposure to 1 during adipogenesis, whereas the expression of lipogenic genes FASN and SREBP1 was significantly reduced upon treatment with 1. Thus, our findings provide experimental evidence that the steroid derived from Hydractinia-associated C. sphaerospermum SW67 is a potential therapeutic agent for obesity.
