The effects of Pongamia pinnata on osteogenic differentiation and mineralization of human stem cells derived from the gingiva

  • Objectives: The aim of the present study is to evaluate the effects of the extract of Pongamia pinnata on the morphology, viability, and differentiation potential of human stem cells derived from the gingiva. Methods: Stem cells obtained from gingivae were cultured in an osteogenic medium in the presence of methanol extract of Pongamia pinnata (PPT) at concentrations ranging from 0.001 to 1%. Evaluations of cell morphology and cellular viability were done at Day 1. Alkaline phosphatase activity assays and Alizarin red S staining were performed to evaluate the osteogenic differentiation of stem cells. Results: The morphology of stem cells in the presence of PPT at final concentrations of 0%, 0.001%, 0.01%, 0.1%, and 1% did not produce any noticeable changes when compared with the untreated control group. Application of PPT produced a significant increase in alkaline phosphatase activity when compared to the control group. The results of the Alizarin Red S staining showed a significant increase of absorbance with the 0.001% group. Conclusions: Based on these findings, it was concluded that PPT could produce beneficial effects on mesenchymal stem cells with enhanced osteogenic differentiation.

20(S)-Protopanaxadiol 및 20(S)-Protopanaxatriol이 활성화된 비만세포로부터의 염증 매개체 유리에 미치는 영향 (Effects of 20(S)-Protopanaxadiol and 20(S)-Protopanaxatriol on the Inflammatory Mediators Release from the Activated Mast Cells)

  • 인삼 사포닌은 면역계에 다양한 약리 효과를 발휘한다. 20(S)-프로토파낙사다이올 (PPD) 및 20(S)- 프로토파낙사트리올 (PPT)은 장내 세균에 의하여 생성되는 인삼 대사체의 일종이며 생체 내 투여 시 순환계에서 탐지된다. 활성화된 비만 세포로부터의 염증 매개체 유리에 미치는 20(S)-프로토파낙사다이올 (PPD) 및 20(S)-프로토파낙사트리올 (PPT)의 영향을 평가하였다. 인삼 사포닌 대사체를 처치 후, 히스타민 유리는 활성화된 해명 폐 비만세포에서 평가하였으며, 인터루킨-4, 인터루킨-8, 및 종양괴사인자-알파 유리는 HMC-1 비만세포에서 평가하였다. 결과는 다음과 같다. PPT는 최고 $100\;{\mu}M$ 농도에서 PMA에 의하여 자극된 HMC-1 세포로부터의 인터루킨-4 유리를 완전히 차단하였다. 또한, 이는 HMC-1 세포로부터의 인터루킨-8의 유리를, PMA와 DMSO동시 처치 시얻어진 수치를 기준으로 대략 40-50% 정도 억제하였다. PPD는 최고 $100\;{\mu}M$ 농도에서 해명 폐 비만세포로부터의 히스타민 유리를 초래하였으나 통계적 유의성은 없었다. PPD는 HMC-1 세포에 PMA와 DMSO 동시 처치 시 얻어진 수치를 기준으로 할 때, 인터루킨-4의 유리를 대략 89% 정도 억제하였으나, 인터루킨-8의 유리에는 유의적인 효과를 초래하지 않았다. 그러나 PPD 및 PPT 모두, PMA에 의하여 자극된 HMC-1 세포로부터의 종양괴사 인자-알파의 유리에는 전혀 효과를 나타내지 않았다. 그러므로 본 연구 결과는 PPD와 PPT가 경구로 투여된 인삼 추출물의 면역조절 작용을 담당하는 장내 인삼 대사체 중의 한 종류임을 제시한다.

학령기용 음소지각검사 표준화를 위한 기초연구: 청각장애아동을 대상으로 (A preliminary study on standardization of phoneme perception test for school-aged children : Focused on hearing impaired children)

  • 본 연구는 학령기용 음소지각검사(Phoneme Perception Test for School-Aged Children, PPT-S)를 활용하여 보청기와 인공와우를 착용하고 있는 청각장애아동의 자음지각능력과 오류를 분석하고, 청각장애아동에 대한 검사 목록의 적합성을 확인하고자 하였다. 연구결과, 청각장애아동들은 초성에 비해 종성을 지각하는 데 어려움을 더 느끼는 것으로 나타났다. 목표어와 보기어의 조음방법과 조음위치가 유사한 PPT-S의 어려운형을 쉬운형보다 더 어렵게 느꼈다. 초성은 기식음에 대한 오답률이 더 높았고, 종성의 경우 'ㄷ'와 'ㅁ'에 대한 오답률이 상대적으로 더 높았다. 검사자의 성별에 따른 정답률에 유의미한 차이는 나타나지 않았다. 이상의 결과는 청각장애아동 대상 음소지각검사의 표준화와 청능재활 전·의 중재효과를 평가하기 위한 기초자료로 유용하게 사용될 수 있을 것이다.

기능성 소화불량 환자의 복진진단 객관화를 위한 임상연구 - 알고미터를 이용한 심하비경 진단 - (Clinical Study for Objectification of Abdominal Examination with Functional Dyspepsia - Epigastric Diagnosis using Algometer)

  • Objectives: Using algometer, measure the pressure pain threshold (PPT) of the epigastric pain(心下痞硬) and calculate the cut-off value, and this can serve as the basis for prognostic diagnosis of functional dyspepsia so we would like to evaluate its diagnostic value. Methods: We investigated 353 patients with functional dyspepsia symptoms who admitted Gangnam Weedahm Oriental Hospital from February 1, 2021 to February 27, 2021. At the time of the patient's visit, an oriental medical doctor measured the pressure at the first pain point on the Algometer of (CV14), twice each, at 1minute intervals. The ROC (receiver operating characteristic) curve and the optimal cut-off value derived through the diagnosis of the (CV14) PPT value for epigastric pain(心下痞硬) and the gold standard of oriental medical doctor, it was evaluated through. Results: In 353 patients, the area under the ROC curve (AUC) was 0.909 (p=0). In addition, the optimal cutting value was 10.05 (kg/cm2), which was statistically significant. Additionally, the sensitivity of the Algometer's PPT measurement was 0.704 and the specificity was 0.884. As a result, if the PPT value of the Algometer exceeds 10.05 (kg/cm2) in terms of the optimal cutting value, it can be seen that epigastric pain(心下痞硬) is lost. Conclusion: Algometer's PPT value measurement can be a reliable test method for quantification of epigastric pain(心下痞硬) diagnosis and can be useful as an objective indicator.

Determination of Ginsenosides Content in Korean Ginseng Seeds and Roots by High Performance Liquid Chromatography

  • A high performance liquid chromatography (HPLC) method has been successfully developed to identify and quantify major ginsenosides in Korean ginseng seeds and roots. Using gradient elution of acetonitrile and water without buffer, the 6 major ginsenosides ($Rb_1,\;Rb_2$, Rc, Rd, Re, and $Rg_1$) were identified. Compared with ginseng roots, the amount of ginsenoside Re and Rd in ginseng seeds were significantly higher than those in ginseng roots (p<0.05). In ginseng seeds, the content of protopanaxtriol (PPT) was higher than that of protopanaxdiol (PPD) and the ratio of PPT and PPD was approximately 2.2 : 1. However, the content of PPT was lower than that of PPD in ginseng roots. It should be mentioned that both content of PPT and PPD in ginseng seeds were much higher than those in ginseng roots.

  • Recently, With explosive national land developments construction fields ate increasing, and slope failures are disastrous when they occur in mountainous area adjoining highways. but public institutions and supervisors, to manage slope need GIS for managing it more quickly and correctly. For manage slope, it is nesessary to use new IT, especially wireless internet. The current domestic population using cell phones exceeded 30 million and the domestic wireless Internet environment has matured, but implementation of mobile technology for government services isnot matured enough. In this paper the author proposed a new system of mobile PPT monitoring for managing slope inspection for Ubiquitous Environment. This new system manages data in real time and reduces human power when applied to current working environments. This research was supported by a grant(NEMA-06-NH-05) from the Natural Hazard Mitigation Research Group, National Emergency Management Agency.

염분함유수에서의 잉어사육시험 (REARING EXPERIMENT OF COMMON CARP IN BRACKISH WATER)

  • 체장 약 10cm 되는 잉어 치어를 이용하여 염도 $20\%_{\circ}$까지의 범위내에서 실험실내 수조에서 그들의 저항성과 성장에 관한 일련의 실험을 한 결과는 다음과 같았다. 염분도 $12\%_{\circ}$까지는 담수에서 바로 옮겨도 죽지 않았으며, 낮은 염분에서 적응시켜 높은 데로 옮기니 $15\%_{\circ}$까지 대부분 잘 살아 남았다. 실험기간의 수온은 실온하에서 $14\~27^{\circ}C$의 범위내였다. 성장실험에서는 $2\~8\%_{\circ}$ 범위서는 생사료인 실지렁이를 사용했을 때나, 건조 pellet 사료를 사용했을 때나 다 같이 성장률과 사료효율이 대단히 높았다. 그러나, $12\%_{\circ}$에서는 실지렁이를 주었을 때는 아주 좋지 못했지만, pellet에서는 성장율은 조금 낮았지만 사료효율은 대단히 높았다.

Effects of Protopanaxatriol-Ginsenoside Metabolites on Rat $N$-Methyl-D-Aspartic Acid Receptor-Mediated Ion Currents

  • Ginsenosides are low molecular weight glycosides found in ginseng that exhibit neuroprotective effects through inhibition of $N$-methyl-D-aspartic acid (NMDA) receptor channel activity. Ginsenosides, like other natural compounds, are metabolized by gastric juices and intestinal microorganisms to produce ginsenoside metabolites. However, little is known about how ginsenoside metabolites regulate NMDA receptor channel activity. In the present study, we investigated the effects of ginsenoside metabolites, such as compound K (CK), protopanaxadiol (PPD), and protopanaxatriol (PPT), on oocytes that heterologously express the rat NMDA receptor. NMDA receptor-mediated ion current ($I_{NMDA}$) was measured using the 2-electrode voltage clamp technique. In oocytes injected with cRNAs encoding NMDA receptor subunits, PPT, but not CK or PPD, reversibly inhibited $I_{NMDA}$ in a concentration-dependent manner. The $IC_{50}$ for PPT on $I_{NMDA}$ was $48.1{\pm}4.6\;{\mu}M$, was non-competitive with NMDA, and was independent of the membrane holding potential. These results demonstrate the possibility that PPT interacts with the NMDA receptor, although not at the NMDA binding site, and that the inhibitory effects of PPT on $I_{NMDA}$ could be related to ginseng-mediated neuroprotection.

정보기술 환경하에서 대학생들의 학습방법 선호도에 대한 실태분석 (A Investigation on University Students Preferred Lesson-Methods Under IT Environment)

  • 본 연구는 대학생들이 학습에서 선호하는 정보활용매체를 평가하였다. 즉, 강의를 위한 효율적인 학습방법이 무엇인가를 분석한 것이다. 연구목적을 달성하기 위하여 정보매체와 관련된 이론적 검토와 관련 선행연구를 하였다. 다양한 정보매체를 이용하는 강의를 기준으로 학생들에게 가장 이해도가 높은 학습방법을 분석하였다. 자료수집은 부산시내의 대학생 82명을 대상으로 하였고, 수집된 자료분석을 위하여 명목척도와 등간척도를 이용하였다. 본 연구결과, 첫째, "PPT"를 학습방법을 선호하는 학생들이 IT 및 컴퓨터 활용능력수준에 대한 이해도가 높았다. 둘째, "PPT"학습방법을 "WRT"보다 더 선호하는 것으로 나타났다. 셋째, "PPT"집단이 "WRT" 집단에 비해 보다 학습효과에서 효과적인 것으로 나타났다. 마지막으로 두집단 모두 전체평균값은 3.08과 2.95로 수업집중도에는 큰 차이는 보이지 않았다. 그러나 본 연구는 면담에 의한 자료수집이 갖는 한계를 안고 있다. 향후 각 매체에 의한 학습효율성 연구와 정보매체에 의한 다양한 집단간 비교연구가 필요할 것이다.

Thrust Characteristics of a Laser-Assisted Pulsed Plasma Thruster

  • An assessment of a novel laser-electric hybrid propulsion system was conducted, in which a laser-induced plasma was induced through laser beam irradiation onto a solid target and accelerated by electrical means instead of direct acceleration only by using a laser beam. A fundamental study of newly developed rectangular laser-assisted pulsed-plasma thruster (PPT) was conducted. On discharge characteristics and thrust performances with increased peak current compared to our previous study to increase effects of electromagnetic forces on plasma acceleration. Maximum peak current increased for our early study by increasing electromagnetic effects in a laser assisted PPT. At 8.65 J discharge energy, the maximum current reached about 8000 A. Plasma behaviors emitted from a thruster in various cases were observed with an ICCD camera. It was shown that the plasma behaviors were almost identical between low and high voltage cases in initial several hundred nanoseconds, however, plasma emission with longer duration was observed in higher voltage cases. Canted current sheet structures were also observed in the higher voltage cases using a larger capacitor. With a newly developed torsion-balance type thrust stand, thrust performances of laser assisted PPT could be estimated. The impulse bit and specific impulse linearly increased. On the other hand, coupling coefficient and the thrust efficiency did not increase linearly. The coupling coefficient decreased with energy showing maximum value (20.8 ?Nsec/J) at 0 J, or in a pure laser ablation cases. Thrust efficiency first decreased with energy from 0 to 1.4 J and then increased linearly with energy from 1.4 J to 8.6 J. At 8.65 J operation, impulse bit of 38.1 ?Nsec, specific impulse of 3791 sec, thrust efficiency of 8 %, and coupling coefficient of 4.3 ?Nsec/J were obtained.
