Prevalence of Cryptosporidium sp among calves and pigs in Jeonbuk province (전북지역의 소와 돼지에서 Cryptosporidium sp의 감염실태 조사)

  • Cryposporidium species have a wide host range. These coccidian parasites are found in close association with epithelial cells of many species of animals including mm. The gastrointestinal tract is most commonly affected in young ruminants and this parasite is thought to be considerable importance in calf diarrhea complex. Major outbreaks of cryptosporidiosis have been reported in calves, lambs, pigs and others including avian species. Cryptosporidiosis is transmitted by oocysts of Cryptosporidium species. Because cryptosporidiosis is common infection among animals, early literature considered it a zoonosis. Human infections contracted from calves, cats, and horse feces. However, the resrvoir host is longer considered the major source of infection. Mild cases of disease have been reported in farm workers. Immunosuppressed, very young and very old persons should avoid contact with this parasite because it may cause severe diarrhea. In order to detect of Crytosporidium sp infection from feces of cattle and pigs at Chonbuk Iksan and Kunsan area, sedimentation and modified acid fast stain were applied. The positive rate of Cryptosporidium sp infection from 1, 176 of cattle and 267 of pigs were 0.5 % and 16.8%, respectively. According to area in Iksan and Kunsan, the positive rates were 0.4% and 0.9% from cattle, and 18.9% and 12.1% from pigs, respectively. In cattle, positive detection rate was 0.6% in milking cows but not in Korean cattle.

Endoparasites of Rats Caught at Jeollabuk-do in Korea (전라북도 지역 집쥐의 체내 기생충 감염 조사)

  • 집쥐가 인체 기생충 전파에 어느 정도의 역할을 하는지 그 양상을 밝히고자 전라북도 남원, 익산지역에서 포획한 집쥐의 체내 기생충 감염상을 조사하였다. 섬진강 상류 남원지역에서 32마리, 만경강 중류 익산지역에서 53마리를 채집하여 총 85마리였고, 종별로는 곰쥐(Rattus rattus) 28마리, 시궁쥐(Rattus norvegicus) 57마리였다. 결과를 요약하면 다음과 같다. 1. 남원, 익산지역의 집쥐 85마리 중 71마리(83.5%)에서 체내 기생충이 검출되었다. 2. 폐장이 회수되었던 집쥐 74마리 중 35마리(47.3%)에서 폐장에 기생하는 조직 기생충인 폐포자충(Pneumocystis carinii)이 검출되었다. 3. 전체 85마리의 가로막에서 선모충(Trichinella spiralis)을 조사하였고, 폐심장 혈관계에서 광동주혈선충(Angiostrongylus cantonensis)을 조사하였으나 한 예도 검출하지 못하였다. 4. 간을 조사한 바 85마리 중 간모세선충(Capillaria hepatica) 22례(25.9%), Taenia taeniaeformis의 유충(Cysticercus fasciolaris) 9례(10.6%), 간흡충(Clonorchis sinensis) 1례(1.2%)가 검출되었다. 5. 장내용물을 조사 한 바 85 마리 중 50례(58.9%)에서 윤충 및 원충이 검출되었다. 윤충은 쥐조충(Hymenolepis diminuta), 극구흡충류(Echinostoma sp.), 쥐요충류(Syphacia sp.), 분신충류(Strongyloides sp.) 등이었고 원충은 대장아메바(Entamoeba coli) 등이었다. 이상의 결과에서 인수공통 질환을 일으킬 수 있는 폐포자충, 쥐조충, 간흡충 등이 집쥐에 감염되어 있어 철저한 집쥐 관리가 요망된다.

Molecular Identification of Diphyllobothrium nihonkaiense from 3 Human Cases in Heilongjiang Province with a Brief Literature Review in China

  • Human diphyllobothriasis is a widespread fish-borne zoonosis caused by the infection with broad tapeworms belonging to the genus Diphyllobothrium. In mainland China, so far 20 human cases of Diphyllobothrium infections have been reported, and the etiologic species were identified as D. latum and D. nihonkaiense based on morphological characteristics or molecular analysis. In the present study, proglottids of diphyllobothriid tapeworms from 3 human cases that occurred in Heilongjiang Province, China were identified as D. nihonkaiense by sequencing mitochondrial cytochrome c oxidase subunit I (cox1) and NADH dehydrogenase subunit 5 (nad5) genes. Two different cox1 gene sequences were obtained. One sequence showed 100% homology with those from humans in Japan. The remaining cox1 gene sequence and 2 different nad5 gene sequences obtained were not described previously, and might reflect endemic genetic characterizations. D. nihonkaiense might also be a major causative species of human diphyllobothriasis in China. Meanwhile, the finding of the first pediatric case of D. nihonkaiense infection in China suggests that infants infected with D. nihonkaiense should not be ignored.

Whole genome sequencing of foot-and-mouth disease virus using benchtop next generation sequencing (NGS) system

  • In countries with FMD vaccination, as in Korea, typical clinical signs do not appear, and even in FMD positive cases, it is difficult to isolate the FMDV or obtain whole genome sequence. To overcome this problem, more rapid and simple NGS system is required to control FMD in Korea. FMDV (O/Boeun/ SKR/2017) RNA was extracted and sequenced using Ion Torrent's bench-top sequencer with amplicon panel with optimized bioinformatics pipelines. The whole genome sequencing of raw data generated data of 1,839,864 (mean read length 283 bp) reads comprising a total of 521,641,058 (≥Q20 475,327,721). Compared with FMDV (GenBank accession No. MG983730), the FMDV sequences in this study showed 99.83% nucleotide identity. Further study is needed to identify these differences. In this study, fast and robust methods for benchtop next generation sequencing (NGS) system was developed for analysis of Foot-and-mouth disease virus (FMDV) whole genome sequences.

Chemogenomics Profiling of Drug Targets of Peptidoglycan Biosynthesis Pathway in Leptospira interrogans by Virtual Screening Approaches

  • Leptospirosis is a worldwide zoonosis of global concern caused by Leptospira interrogans. The availability of ligand libraries has facilitated the search for novel drug targets using chemogenomics approaches, compared with the traditional method of drug discovery, which is time consuming and yields few leads with little intracellular information for guiding target selection. Recent subtractive genomics studies have revealed the putative drug targets in peptidoglycan biosynthesis pathways in Leptospira interrogans. Aligand library for the murD ligase enzyme in the peptidoglycan pathway has also been identified. Our approach in this research involves screening of the pre-existing ligand library of murD with related protein family members in the putative drug target assembly in the peptidoglycan biosynthesis pathway. A chemogenomics approach has been implemented here, which involves screening of known ligands of a protein family having analogous domain architecture for identification of leads for existing druggable protein family members. By means of this approach, one murC and one murF inhibitor were identified, providing a platform for developing an anti-leptospirosis drug targeting the peptidoglycan biosynthesis pathway. Given that the peptidoglycan biosynthesis pathway is exclusive to bacteria, the in silico identified mur ligase inhibitors are expected to be broad-spectrum Gram-negative inhibitors if synthesized and tested in in vitro and in vivo assays.

An Overview of Fish-borne Nematodiases among Returned Travelers for Recent 25 Years- Unexpected Diseases Sometimes Far Away from the Origin

  • Along with globalization of traveling and trading, fish-borne nematodiases seems to be increasing in number. However, apart from occasional and sporadic case reports or mini-reviews of particular diseases in particular countries, an overview of fish-borne nematodiasis among travelers have never been performed. In this review, we gathered fish-borne nematodiasis among travelers for recent 25 years by an extensive global literature survey using appropriate keywords, e.g. travelers diseases, human infection, anisakiasis, gnathostomiasis, capillariasis, sushi, sashimi, ceviche, Gnathostoma, Pseudoterranova, Anisakis, Capillaria, etc., as well as various combinations of these key words. The Internet search engines PubMed, Medline, Google and Googler Scholar were used as much as possible, and the references of every paper were checked in order to identify useful and reliable publications. The results showed unexpectedly high incidence of gnathostomiasis and low incidence of anisakidosis. The different incidence values of the infection with several fish-borne zoonotic nematode species are discussed, as well as some epidemiological aspects of the infections. The difficulties of differential diagnosis in non-endemic countries are emphasized. It is concluded that travelers must avoid risky behaviors which can lead to infection and that physicians and health authorities must advice travelers on the risks of eating behaviors during travel.

Biochemical characteristics of Brucella abortus isolated from cattle in Gyungbuk province (경북지방 소에서 분리한 Brucella abortus의 생화학적 특성)

  • Bovine brucellosis is a zoonosis, long incubation period and chronic infectious disease, usually caused by Brucella abortus. This study was carried out to investigate the biotyping and biochemical characterization of B. abortus isolated from 208 farm 871 korean cattle and holstein diagnosed brucellosis by serological positive in Gyeongbuk province during the period from 2002 to 2006. B. abortus was isolated from 124 (14.2%) of 871 cattle, and isolated 110 (13.4%) of 820 Korean cattle and 14 (27.5%) of 51 holstein in breed. The uterus of korean cattle was isolated in 8 (17.8%) of 45 cattle and supramammary lymph none of holstein was isolated 11 (68.8%) of 16 cattle. 101 (12.5%) of 810 serological positive blood samples were isolated B. abortus. The isolation rate of B. abortus was correlated with antibody titers. The biochemical characterization of isolates was non-hemolytic, production of H$_2$S, oxidase-positive, catalase-positive, hydrolyzation of urea and growth of basic fuchsin dye medium. As a result, all of isolates was identified B. abortus bv 1. 124 isolates were susceptible to ampicillin, lincospectin, amikacin, gentamicin, kanamycin, neomycin, streptomycin, tetracycline, ciprofloxacin, norfloxacin and enrofloxacin.

Awareness on Zoonoses among Livestock(Korean Native Cattle) Farmers in Gyeongsangbuk-do (경북 일부지역 축산(한우) 농업인의 인수공통감염병 인지도)

  • Objectives: General livestock farmers are known as a high riskgroup for zoonoses, but studies of general livestock farmers in regard tozoonoses have been rare in Korea. We surveyed awareness of zoonoses amonggeneral livestock farmers to suggest directions for education. Methods: A questionnaire was developed examining the workbehaviors and risk factors of general livestock farmers. We conducted a questionnairesurvey on awareness on zoonoses among 265 general livestock farmers. Results: The awareness rates for zoonosis itself,brucellosis, and q-fever were 13.0%, 65.0%, and 2.3%, respectively. The awarenessrate of zoonoses and brucellosis tended to increase with sex. Conclusions: Livestock is the principal reservoir ofzoonoses. Therefore, effective working guidelines for preventing zoonoses amonggeneral livestock farmers must be developed and an educational program onzoonoses is needed for general livestock farmers. Furthermore, publicityactivities on the prevention of zoonoses are needed for high-risk groups.

Anticancer and Antimicrobial Activities of 13(E)-labd-13-ene-8α,15-diol from Brachyglottis monroi

  • In a previous study, we reported that 13(E)-labd-13-ene-$8{\alpha}$,15-diol (13E) possesses antiviral and anticancer activities. In this study, the anticancer and antimicrobial activities of 13(E) were evaluated against 4 cancer cell lines and 6 bacteria. 13(E) showed inhibitory effect on a variety of cancer cell lines. The $IC_{50}$ values was 8.3-21.3 ${\mu}g/mL$. 13(E) was the most effective growth inhibitor of murine leukaemia cell lines P388, producing approximately 8.3 ${\mu}g/mL$ of $IC_{50}$ in the cytopathic effect (CPE) method. 13(E) also inhibited the growth of the gram-positive bacteria (Staphylococcus aureus, Bacillus cereus and Listeria monocytogenes) and gram-negative bacteria (Vibrio parahaemolyticus, Escherichia coli and Salmonella enteritidis) with a range of minimum inhibitory concentration (MIC) values from 0.092 to 0.598 mg/mL and gram-negative bacteria were more sensitive to the compound (MIC, 0.092 mg/mL).

The dynamics of proteinase activity at different stages of toxocariasis in mice (개회충에 감염된 생쥐에 있어서 시기별 단백분해효소 활성의 변동)

  • Toxocariasis is a parasitic zoonosis caused by infestation of humans with larvae of Toxocara canis, the common roundworm in dogs. Two syndromes have been identified : visceral larva migrans and ocular toxocariasis. In this study we were characterized proteinase activity in crude extracts from liver, lung, kidney and heart of mice infected with Toxocara canis and the dynamics of their changes in different stages of disease. The optimal pH was 5.5. In liver of mice infected with Toxocara canis, the maximun activity of proteinase was observed in 5 day post infection. In lung, the activity reached its maximun on 5th day in A group (infected with 100 embryonated eggs), and on 5th week in B group (infected with 50 embryonated eggs). In kidney, the maximum activity was shown at 6th week in A group, and in B group was shown at 10th day. In early infection, the activity reached its maximun in heart of mice infected with Toxocara canis. As we could see, the dynamics of the changes of proteinase activity in mice is similar in the case of the disease with other biochemical and immunological indices observed in toxocariasis.
