Baculovirus transfer and expression vectors with Hyphantria cunea nuclear polyhedrosis virus (HcNPV) were constructed. An initial transfer vector, pHcEV, constructed using HcNPV was previously reported (Park et al. 1993. J. Kor. Soc. Viral. 23: 141-151). Herein, the size of the vector was properly reduced, and a functionally perfect vector was constructed and named pHcEV-IV (6.7 kb). The vector has a 2.2-kb HcNPV DNA sequence in the 5'-flanking region of the vector's polyhedrin gene promoter. The 1.8-kb HcNPV DNA sequence, poly A signal sequence, T3 primer sequence, and 13 multicloning site sequences, in order, were ligated in front of the translation start codon of the polyhedrin gene. The cloning indicating marker lacZ gene was inserted into the pHcEV-IV, named pHcEV-IV-lacZ, and transferred into the wild-type virus. Recombinant expression virus, lacZ-HcNPV, was constructed by replacing the lacZ gene in the pHcEV-IV-lacZ with the polyhedrin gene of the wild-type virus. The recombinant virus was isolated from blue plaques that produce $\beta$-galactosidase without polyhedra. The lacZ gene insertion was confirmed by Southern hybridization analysis. The expression of the lacZ gene in Spodoptera frugiperda cells infected with the lacZ-HcNPV was examined by SDS-PAGE and colorimetric assay. One 116-kDa LacZ protein band appeared on the PAGE. The production rate of the $\beta$-galactosidase was approximately 50 international units (IU) per min per ml between 2 to 5 days postinfection (p.i.). The highest activity occurred at five days p.i. was 170 IU/min/$m\ell$. The enzyme activity first appeared about 20 h p.i. as measured by colorimetric assay.
The geometrical structures, atomic charges, and relative energies of tetracoordinated Pd complexes [PdCl3Z (Z = Cl-, Br-, OH?-, H2O, NH3, PH3), PdCl2Z2 (Z = Br-, OH?-, H2O, NH3, PH3), PdZ?2X (Z = Cl-, OH?-, H2O, NH3, PH3; X = oxalate, O2-?CCO2-), and PdZ2Y (Z = Cl?-, OH?-, H2O, NH3, PH3; Y = succinate, CO2-?CHCHCO2-?)] and the ligand exchange reactions of the Pd complexes were investigated using the ab initio second order Mller-Plesset perturbation (MP2) and Density Functional Theory (DFT) methods. The geometrical characteristics of the tetracoordinated Pd(II) complexes with mono- and bidentate ligands, the effects of the atomic charges for the charged and uncharged ligands, the (dz2-p ) interactions between the dz2-orbital of Pd(II) and the p -orbital of bidentates, and the relative stabilities between the isomers of PdCl2Z2 and PdZ2Y were investigated in detail. The potential energy surfaces for the ligand exchange reactions used for the conversions of {[PdCl2(NH3)2] + H2O} to {[PdCl(NH3)2(H2O)]+ + Cl?-?} and {[PdCl2(PH3)2] + H2O} to {[PdCl(PH3)2(H2O)]+ + Cl?-?]} were investigated. The geometrical structure variations, molecular orbital variations (HOMO and LUMO), and relative stabilities for the ligand exchange processes were also examined quantitatively.
Purpose: The problem for the traditional control chart is that it is unable to monitor the very small fraction of nonconforming and the underlying distribution is the normal distribution. $Z_p$ control chart is useful where it controls the vert small fraction on nonconforming. In this study, we will design the $Z_p$ control chart in order to use under non-normal process. Methods: $Z_p$ is calculated not by failure rate based on attribute data but using variable data. Control limit for non-normal $Z_p$ control chart is designed based on ${\alpha}$-risk calculated by cumulative distribution function of Burr distribution. ${\beta}$-risk, which is for performance evaluation, obtains in the Burr distribution's cumulative distribution function and control limit. Results: The control limit for non-normal $Z_p$ control chart is designed based on Burr distribution. The sensitivity can be checked through ARL table and OC curve. Conclusion: Non-normal $Z_p$ control chart is able to control not only the very small fraction of nonconforming, but it is also useful when $Z_p$ distribution is non-normal distribution.
During the last decade, several papers have focused on linear q-difference equations of the form ${\sum}^n_{j=0}a_j(z)f(q^jz)=a_{n+1}(z)$ with entire or meromorphic coefficients. A tool for studying these equations is a q-difference analogue of the lemma on the logarithmic derivative, valid for meromorphic functions of finite logarithmic order ${\rho}_{log}$. It is shown, under certain assumptions, that ${\rho}_{log}(f)$ = max${{\rho}_{log}(a_j)}$ + 1. Moreover, it is illustrated that a q-Casorati determinant plays a similar role in the theory of linear q-difference equations as a Wronskian determinant in the theory of linear differential equations. As a consequence of the main results, it follows that the q-gamma function and the q-exponential functions all have logarithmic order two.
This study evaluates the adsorption properties of Sr ions in an aqueous solution of the synthetic zeolite (Z-Y1) prepared using coal fly ash generated from a thermal power plant. In order to investigate the adsorption characteristics, the effects of various parameters such as the initial concentrations of Sr ion, contact time, and solution pH were investigated in a batch mode. The Langmuir and Redlich-Peterson model fitted the adsorption isotherm data better than the Freundlich model. The maximum adsorption capacity of Sr ions, as determined the Langmuir model, was 181.68 mg/g. It was found that by varying the Sr ion concentration, pH, and temperature, the pseudo-second-order kinetic model describes the adsorption kinetics of the Sr ion better than the pseudo-first-order kinetic model. The calculated thermodynamic parameters of ${\Delta}H^0$ and ${\Delta}G^0$ showed that the adsorption of Sr ions on Z-Y1 was occurred through a spontaneous and an endothermic reaction. We found that the adsorption of Sr ions by Z-Y1 was more affected by pH than by temperature and Sr ion concentration.
Statement of problem: The esthetic component of dental care has become increasingly more important, while new tooth-colored materials are continually marketed. Various new indirect composite materials have been developed with required advantages. The most recent development in the indirect composites has been the introduction of the second-generation laboratory composite or poly-glass materials. They are processed by different laboratory techniques based on combinations of heat, pressure, vacuum and light polymerization. Although, second generation products became available in 1995, their characteristics and clinical performance have not been adequately investigated. Purpose: The aim of this study was to measure the mechanical properties of the second generation indirect resin system and compare these with an existing universal direct composite resin. Material and method: In this study four indirect composite material (Adoro LC, BelleGlass HP, Tescera, Synfony) were tested for flexural strength, wear resistance, hardness and their degree of conversion against Z250, a light cure direct composite. Results: Within the limitations of this study, the following conclusions were drawn: 1. From the abrasion wear result, Adoro showed the least volume loss while Synfony showed the greatest volume loss. Z250 and BelleGlass HP didn't show significant difference (p>0.05), but they showed significant difference with other groups (p<0.05). From the attrition wear, BelleGlass HP showed the least volume loss and it didn’t show significant difference with Tescera (p>0.05). While Synfony showed the greatest volume loss that it showed significant difference with other groups (p>0.05). 2. Mean values of flexural strength by means of three point bending test was in the order of Z250, Adoro, Belleglass HP, Tescera and Synfony. Mean elastic modulus was in the order of Z250, BelleGlass HP, Tescera, Adoro and Synfony. 3. The result of Vicker‘s microhardness value showed that significantly higher value in Z250 (p<0.05), and is in the order of BelleGlass HP, Tescera, Adoro and Synfony. 4. The degree of conversion measured by FT-IR showed significantly higher value in BelleGlass HP (p<0.05), and is in the order of Adoro, Synfony, Tescera and Z250. Conclusion: Significant differences were found in the flexural strength, wear resistance, hardness and their degree of conversion.
과거 Z. cao는 Relation matrix를 사용한 정밀한 추론이 가능한 NFRM(New fuzzy reasoning method)을 제안하였다. 이는 추론의 규칙 수가 적음에도 불구하고 Mamdani의 퍼지추론방식에 비하여 좋은 성능을 보였다. 그러나 정밀한 추론을 위하여 relation matrix는 시행착오법을 사용하여 구하고, 이는 많은 시간과 노력이 필요하다. 본 연구에서는 이러한 relation matrix를 구하기 위하여 시행착오법에 의해 소요되는 많은 시간과 노력을 줄이고, 더욱 정밀한 추론 성능의 개선을 위하여 경사감소학습법을 사유한 학습기능을 갖는 Z. Cao의 퍼지추론 방식을 제안하고자 한다.
In a grid-connected photovoltaic (PV) system, the traditional Z-source inverter uses a low frequency transformer to ensure galvanic isolation between the grid and the PV system. In order to combine the advantages of both Z-source inverters and transformerless PV inverters, this paper presents a modified single-phase transformerless Z-source PV grid-connected inverter and a corresponding PWM strategy to eliminate the ground leakage current. By utilizing two reversed-biased diodes, the path for the leakage current is blocked during the shoot-through state. Meanwhile, by turning off an additional switch, the PV array is decoupled from the grid during the freewheeling state. In this paper, the operation principle, PWM strategy and common-mode (CM) characteristic of the modified transformerless Z-source inverter are illustrated. Furthermore, the influence of the junction capacitances of the power switches is analyzed in detail. The total losses of the main electrical components are evaluated and compared. Finally, a theoretical analysis is presented and corroborated by experimental results from a 1-kW laboratory prototype.
Objectives: $^{32}P$-postlabeling assay is the most sensitive method of detecting DNA adduct formation. However, it is limited by a low sample throughput and use of radioisotopes (RI). In this study, we modified it to minimize these limitations and applied it to Z. platypus exposed to Benzo(a)pyrene (BaP) in order to investigate DNA adduct formation (effect biomarker for pollutants) in Z. platypus for assessing risk of waterborne BaP exposure. Methods: DNA hydrolysis was performed only with Micrococcal nuclease (MNase), RI reduction test was performed and the overlapping steps between thin layer chromatography (TLC) and radioisotope high-performance liquid chromatography (RI-HPLC) were omitted. The application of a modified method to Z. platypus exposed to BaP was performed. Results: The results revealed that the amount of RIs used can be reduced roughly 10-fold. Because the analysis time was shortened by 8.5 hours, the sample throughput per hour was increased compared with the previous method. The results of applying modified $^{32}P$-postlabeling assay to Z. platypus, DNA adduct formation in Z. platypus showed dose-dependency with the BaP concentration. Only BPDE-dGMP was detected as a DNA adduct. Conclusion: These results demonstrate that the modified $^{32}P$-postlabeling assay is a suitable method for detecting DNA adduct formation in Z. platypus exposed to waterborne BaP and will be useful in risk assessment of carcinogenic effect in aquatic environment due to BaP.
In order to develop oral cephalosporin having a new substituent at 3 position, the synthesis of cephalosporins modified at C-3 and the effect of the substituents on the oral absorption is studied. 7-[(Z)-2-(2-Aminothiazole- 4-yl)-2-methoxyiminoacetamidol-3-[4-(2-pyridyl )piperazinyl] thiocarbonylthiomethyl-3-cephem-4-carboxylic acid (CEN1) and 7-[(Z)-2-(2-aminothiazole-4-yl)-2-methoxyiminoacetamido]-3-[4-(2-pyrimid yl)piperazinylthiocarbonylthiomethyl-3-cephem-4-carboxylic acid (CEN2) were synthesized from 4-(2-piridyl)piperazinyl dithiocarbamate potassium salt or 4-(2-pirimidyl)piperazinyl dithiocarbamate potassium salt and cefotaxime. Also pivaloyloxymethyl esters of CEN1 and CEN2, pivaloyloxymethyl 7-[(Z)-2-(2-aminothiazole-4-yl)-2-methoxyiminoacetamido]-3-[4-(2-pyridyl )piperazinyllthiocarbonylthiomethyl-3-cephem-4-carboxylate (CENIP) and pivaloyloxymethyl 7-[(Z)-2-(2-aminothiazole-4-yl)-2-methoxyiminoacetamidol-3- [4-(2-pyrimid yl)piperazinyllthiocarbonylthiomethyl-3-cephem-4-carboxylate (CEN2P) were synthesized. The in vitro activities of two new oral cephalosporins, CEN1 and CEN2, were compared with the in vitro activities of cefaclor and cefotaxime against a variety of bacterial species. CEN2 has a broad antibacterial spectrum covering Gram-positive and Gram-negative bacteria, similar to that exhibited by CEN1 and cefotaxime. CEN1 and CEN2 were more active in vitro than cefaclor against Streptococcus pyogenes, Klebsiella aerogenes and Enterobacter cloacae.
