Isolation, Identification, and Characterization of Microorganisms which Possess the Flocculating Activity to Food Wastewater (음식물류폐수에 특이적 응집제를 생성하는 미생물의 분리, 동정 및 응집특성)

  • Six bacteria, which showed the flocculating activity to food wastewater, were isolated from various environment. These strains were identified as Bacillus pumilus, Enterobacter sp., Pantotea agglomerans, Bacillus licheniformis, and two Bacillus sps. Among them, the flocculating activities of three strains, such as Enterobacter sp.(YK102), Bacillus sp.(YK103), and Pantotea agglomerans (YK104), were eight times or more higher than that of the control strain, Zoogloea ramigera. in the test with 0.5% kaolin. In the experiment with food wastewater, Enterobacter sp.(YK102) showed the highest flocculating activity which was 2.5 times higher than that of a control strain, Pseudomonas fluorescens.

An Implementation of YK2 Cipher System for Electronic Commerce Security (전자상거래 보안을 위한 YK2 암호시스템의 구현)

  • EC(Electronic Commerce) which is done on the virtual space through Internet has strong point like independence from time and space. On the contrary, it also has weak point like security problem because anybody can access easily to the system due to open network attribute of Internet, Therefore, we need the solutions that protect the EC security problem for safe and useful EC activity. One of these solutions is the implementation of strong cipher system. YK2(Young Ku King) cipher system proposed in this paper is good solution for the EC security and it overcome the limit of current block cipher system using 128 bits key length for input, output, encryption key and 32 rounds. Moreover, it is designed for the increase of time complexity by adapting more complex design for key scheduling algorithm regarded as one of important element effected to encryption.

Isolation and Characterization of Plant Growth Promoting Rhizobacterium Bacillus subtilis YK-5 from Soil (토양에서 식물생육촉진 활성을 가진 균주 Bacillus subtilis YK-5의 분리 및 특성)

  • For the isolation of a plant growth-promoting rhizobacteria, strain YK-5 was selected from approximately 400 thermostable strains isolated from special soil samples. Strain YK-5 produced an antifungal compound, and optimum carbon and nitrogen sources for the production of the antifungal compound were investigated against Aspergillus flavus as a test strain. Modified LB medium containing 1% peptone, 1% yeast extract and 5% black sugar was determined to be the optimal medium for growth and antifungal compound production. Culture broth of strain YK-5 potently inhibited growth of the phytopathogenic fungus Fusarium oxysporum KACC 40052 for 7 days. The plant growth-promotion function of strain YK-5 was tested against radish and rice in pot trials. Leaf number, plant height and root length in YK-5-treated radish markedly exceeded (> 60%) those of untreated radish. Leaf length and white rootlet development were markedly more prominent than in commercially-treated rice plants. Strain YK-5 was determined to be Bacillus subtilis YK-5 by physiological, chemotaxonomical, and phylogenetical analyses.

  • The purpose of this work was to investigate the induction of stress shock proteins (SSPs) in Burkholderia sp. YK-2 in response to 2,4-dichlorophenoxyacetic acid (2,4-D), and Pseudomonas sp. DJ-12 to benzoate, 4-chlorobenzoate (4-CBA), 4-hydroxybenzoate, and biphenyl. The SSPs, which contribute to the resistance of the cytotoxic effect of the toxic aromatic compounds including 2,4-D and 4-CBA, were induced at different concentrations of the compounds in exponentially growing cultures of Burkholderia sp. YK-2 or Pseudomonas sp. DJ-12. This response involved the induction of a 43 kDa DnaK and 41 kDa GroEL proteins in Burkholderia sp. YK-2, characterized by SDS-PAGE and Western blot using the anti-DnaK and anti-GroEL monoclonal antibodies. In Pseudomonas sp. DJ-12, 70 kDa DnaK and 60 kDa GroEL proteins was induced as SSPs, respectively. The total SSPs were analyzed by 2-D PAGE. Survival of Burkholderia sp. YK-2 or Pseudomonas sp. DJ-12 with time in the presence of different concentrations of the compounds was monitored, and viable counts paralleled the induction of the SSPs in these strains. Cells treated with the increased concentrations of toxic compounds showed some destructive openings on the cell envelopes.

  • The eleven strains, which could be used lactic and acetic acids as carbon sources, were isolated from kimchi and identified; the strains were facultative microorganisms which could be grown at low temperature (10$\circ$C) and around pH 3.2. As results of morphological, biochemical and physiological tests, 5 species of 3 genera were identified as Debaryomyces coudertii, Pichia edia, Pichia chambardii, Pichia haplophilia and Saccharomyces fermentati. Each strain was grown in basal media. In acidic resistance and acid utilization test, Saccharomyces sp. YK- 17 and Saccharomyces fermentati YK-19 were grown well in basal and YM media containing 0.3% lactic acid. And two strains were grown in basal and YM media containing O.3% lactic acid and 0.6% acetic acid. Since strain YK-19 was grown better at 10$\circ$C than that in 25$\circ$C, strain YK-19 was known to be a psychrophilic strain.

Purification and Characterization of Transglutaminase from a Newly Isolated Streptomyces platensis YK-2 (토양 방선균 Streptomyces platensis YK-2가 생산하는 Transglutaminase의 정제 및 효소학적 특성)

  • A species producing transglutaminase (EC 2.3.2.13) was isolated from forest soil and identified as Streptomyces platensis YK-2. The transglutaminase was purified from culture broth by 50% methanol precipitation, followed by successive chromatography on DEAE-Sephadex. The yield and purification-fold was 63.4% and 2.2-fold, respectively. The purified microbial transglutaminase (MTG) migrated as a single band of approximately 45 kDa upon sodium dodecyl sulfate polyacrylamide gel eletrophoresis. The isoelectric point determined by multichambered electrofocusing was pH $6.0{\sim}7.0$. The enzyme was strongly inhibited by $Hg^{++}$, but was activated by $Cd^{++}$, $Mg^{++}$, $Mn^{++}$, $Pb^{++}$ and reducing agents such as dithiothreitol and mercaptoethanol.

  • Using Mu dJ(Km lac) operon fusion, several oxygen-regulated genetic loci were identified in Salmonella typhimurium. Nine aerobically inducible(oxi) and thirteen anaerobically inducible(ani) operon fusions were newly identified. Based on the control by oxrA regulatory locus, the ani-lacZ fusions were grouped into two classes: class I loci were regulated by oxrA regulatory locus; class II genes were not affected by this locus. Some of the ani-lacZ fusions had required growth in CAA and LB before they exhibited the inducible phinotype. Most of all ani-lacZ fusions were repressed by nitrate and fumarate. Three of the ani loci were mapped into $59{\pm}0.14$ map unit (YK114), $64{\pm}0.5$ map unit(YK120), and $93{\pm}0.29$ map unit(YK112) by testing the cotransduction frequency, respectively.

  • Cytosine deaminase (cytosine aminohydrolase, EC 3.5.4.1) stoichiometrically catalyzes the hydrolytic deamination of cytosine and 5-fluorocytosine to uracil and 5-fluorouracil, respectively. Optimal medium compositions for production of cytosine deaminase from Chromobacterium violaceum YK 391 were 0.75% soluble starch, 1.5% peptone, 0.1% meat extract, 0.1% yeast extract, 0.01% NaCl, 0.01% $MgCl_2{\cdot}7H_2O$ and 0.05% $K_2HPO_4$. The optimal pH of medium and incubation temperature were 7.0 and $30^{\circ}C$, respectively. C. violaceum reached stationary phase after 30 hr, and produced a maximum cytosine deaminase (120 units/ml) after 72 h in batch culture.

Bioactivity of an Extract of Lespedezea Cuneata G. Don to Rabbit Corpus Cavernosum Smooth Muscle Tone (야관문(Lespedezea cuneata G. Don) 추출물이 토끼 음경해면체 평활근에 미치는 생리활성)

  • The extracts in n-hexane layer from Lespedezea cuneata G. Don exelled at sexual activity according to in vitro experiment using biopsy of rabbit corpus cavernosum to investigate effects on sexual function. The extracts were injected into 9 samples at $10{\mu}l$. The lowest relaxation was showed at sample C, $4.8{\pm}1.4%$ and the highest relaxation was showed at sample A, $20.2{\pm}6.0%$. The relaxation by every sample, except sample C and G-1, were higher than by $10^{-7}M$ ACh, $7.8{\pm}5.1%$, and their effects were above 10%. Also, the extracts were injected into 9 samples at $15{\mu}l$. The lowest relaxation was showed at sample C, $15.4{\pm}9.7%$ and the highest relaxation was showed at sample J, $54.8{\pm}9.7%$. The relaxation by sample A and D was as much as by $10^{-6}M$ ACh, $28.0{\pm}20.1%$. The relaxation by sample H was $41.9{\pm}7.3%$. The relaxation by sample J was $54.8{\pm}9.7%$ and it was higher than by $10^{-5}M$ ACh, $53.9{\pm}25.9%$. Also, the extracts were injected into 9 samples at $20{\mu}l$. The lowest relaxation was showed at sample E, $28.9{\pm}0.6%$ and it was a little higher than by $10^{-6}M$ ACh. The relaxation by sample G was as much as by $10^{-5}M$ ACh. The two higher relaxation were showed at sample H, $99.4{\pm}16.0%$ and J, $95.7{\pm}7.2%$, and their relaxation against contraction reaction by PhE were near 100%. Experiment by sample I was not performed for lack of sample amount.

Design, Characterization, and Antimicrobial Activity of a Novel Antimicrobial Peptide Derived from Bovine Lactophoricin

  • Lactophoricin (LPcin), which is a part of proteose peptone isolated from bovine milk, is a cationic amphipathic ${\alpha}-helical$ antimicrobial peptide. Its truncated variants and mutated analogs were designed and their antimicrobial activities were evaluated by using various assays, like broth dilution methods and disk diffusion methods as well as hemolysis assay. Three analogs, LPcin-C8 (LPcin-YK1), LPcin-T2&6W (LPcin-YK2), and LPcin-T2&6W-C8 (LPcin-YK3), which showed better antibiotic activities than LPcin, were selected. Their secondary structures were also characterized by using CD spectropolarimetry. These three analogs of LPcin could be used as an alternative source of powerful antibacterial agents.
