벼 흰잎마름병균 파지의 생태학적 특성 및 이를 이용한 생물방제 (Ecological Characteristics of Bacteriophages Infecting Xanthomonas oryzae pv. oryzae and Their Use as Biocontrol Agents)

  • Xanthomonas oryzae pv. oryzae에 의해서 발생하는 벼 흰잎마름병은 세계적으로 벼 재배지역에서 큰 피해를 주고 있다. 그러나, 이 병의 효과적인 방제 수단은 적은 실정이다. 본 연구에서는 국내 벼 재배지의 관개수에서 분리한 Xoo 파지를 이용하여 흰잎마름병을 생물적으로 방제하고자 하였다. 먼저 포장에서의 사용가능성을 확인하기 위하여 pH, 온도, 자외선에 대한 Xoo 파지의 안정성을 조사하였다. Xoo 파지는 pH 5-pH 10 사이에서는 안정적이었고, $50^{\circ}C$에서 1시간 이상 처리시 대부분 불활성화 되었다. 자외선을 조사하였을 때는 약 2분간의 노출에서도 불활성화 되었다. 벼의 유묘에 살포한 파지는 세균이 없이도 7일간 벼 잎에서 활성을 유지하였고, 스킴 밀크에 혼합하여 살포하면 PBS 완충액에 넣어 살포한 것 보다 10배 이상의 생존율을 보였다. 그러나 이러한 생존율의 증가에도 불구하고 포장에서의 방제효과는 저조하였다. 하지만 파지 혼합액을 Tecloftalam WP, ASM 등과 혼합하여 동시에 살포하면 방제효과가 월등히 증가하였다. 이상의 결과는 Xoo 파지가 흰잎마름병의 방제에 사용될 수 있으며, 약제의 사용 빈도나 농도를 줄여 줄 수 있을 것임을 의미한다.

  • Xanthomonas oryzae pv. oryzae (Xoo) causes bacterial leaf blight (BLB) disease in rice (Oryza sativa L.) and it is among the most destructive pathogen responsible for severe yield losses. Potential bacterial biocontrol agents (BCAs) with plant growth promotion (PGP) abilities can be applied to better manage the BLB disease and increase crop yield, compared to current conventional practices. Thus, this study aimed to isolate, screen, and identify potential BCAs with PGP abilities. Isolation of the BCAs was performed from internal plant tissues and rhizosphere soil of healthy and Xoo-infected rice. A total of 18 bacterial strains were successfully screened for in vitro antagonistic ability against Xoo, siderophore production and PGP potentials. Among the bacterial strains, 3 endophytes, Bacillus sp. strain USML8, Bacillus sp. strain USML9, and Bacillus sp. strain USMR1 which were isolated from diseased plants harbored the BCA traits and significantly reduced leaf blight severity of rice. Simultaneously, the endophytic BCAs also possessed plant growth promoting traits and were able to enhance rice growth. Application of the selected endophytes (BCAs-PGP) at the early growth stage of rice exhibited potential in suppressing BLB disease and promoting rice growth.

Screening and Identification of Antimicrobial Compounds from Streptomyces bottropensis Suppressing Rice Bacterial Blight

  • Xanthomonas oryzae pv. oryzae (Xoo) is the most devastating pathogen to Oryza sativa and has been shown to cause bacterial blight. Two bioactive compounds showing antimicrobial activities against Xoo strain KACC 10331 were isolated from a Streptomyces bottropensis strain. The ethyl acetate extract was fractionated on a Sephadex LH-20 column, and then purified by preparative HPLC. The purified compounds were identified as bottromycin A2 and dunaimycin D3S by HR/MS and $^1H$ NMR analyses. The MIC value against Xoo and the lowest concentration still capable of suppressing rice bacterial blight were 2 ${\mu}g$/ml and 16 ${\mu}g$/ml for bottromycin A2, and 64 ${\mu}g$/ml and 0.06 ${\mu}g$/ml for dunaimycin D3S, respectively. These two compounds were shown to exert different bioactivities in vitro and in rice leaf explants.

  • The expression of HR-like lesion inducing gene of Oryza sativa (OsHRL) was slightly increased by Xanthomonas oryzae pv. oryzae (Xoo) infection. Transgenic rice plants over-expressing OsHRL gene were challenged with Xoo and the development of disease symptoms were examined to investigate the effect of OsHRL gene expression on plant defense responses. The over-expression of OsHRL increased disease resistance against Xoo compared with wild type plants.

Deciphering the Role of Tyrosine Sulfation in Xanthomonas oryzae pv. oryzae Using Shotgun Proteomic Analysis

  • A bacterial tyrosine sulfotransferase, RaxST, is required for activation of rice XA21-mediated immunity, and it catalyzes sulfation of tyrosine residues of Omp1X and RaxX in Xanthomonas oryzae pv. oryzae, a causal agent of bacterial blight in rice. Although RaxST is biochemically well-characterized, biological functions of tyrosine sulfation have not been fully elucidated. We compared protein expression patterns between the wildtype and a raxST knockout mutant using shotgun proteomic analysis. Forty nine proteins displayed a more than 1.5-fold difference in their expression between the wildtype and the mutant strains. Clusters of orthologous groups analysis revealed that proteins involved in cell motility were most abundant, and phenotypic observation also showed that the twitching motility of the mutant was dramatically changed. These results indicate that tyrosine sulfation by RaxST is essential for Xoo movement, and they provide new insights into the biological roles of RaxST in cellular processes.

Xanthomonas oryzae pv. oryzae triggers complex transcriptomic defense network in rice

  • High throughput transcriptome investigations of immunity in plants highlight the complexity of gene networks leading to incompatible interaction. To identify genes crucial to resistance against Xanthomonas oryzae pv oryzae, functional genetic analysis of selected differentially expressed genes from our microarray data set was carried out. A total of 13 overexpression vector constructs were made using 35S CaMV promoter which drive constitutive expression in rice. Most of the genes are developmentally expressed especially during maximum tillering stage and are commonly highly expressed in the leaves. When screened against Xoo strain K2, the transgenic plants displayed shorter lesion length compared with wild type Dongjin which indicates partial resistance. The levels of ROS continuously magnified after inoculation which indicates robust cellular sensing necessary to initiate cell death. Elevated transcripts levels of several defense-related genes at the downstream of defense signal network also corroborate the phenotype reaction of the transgenic plants. Moreover, expression assays revealed regulation of these genes by cross-communicating signal-transductions pathways mediated by salicylic and jasmonic acid. These collective findings revealed the key immune signaling conduits critical to mount full defense against Xoo.

Endophytic Bacillus subtilis MJMP2 from Kimchi inhibits Xanthomonas oryzae pv. oryzae, the pathogen of Rice bacterial blight disease

  • An endophytic bacterial strain was isolated from kimchi, a Korean traditional fermented Brassica campestris and identified as Bacillus subtilis MJMP2 based on the 16S rRNA sequence. This strain showed strong antagonistic activity against Xanthomonas oryzae pv. oryzae (Xoo) KACC10331, the pathogen of bacterial rice blight disease, as well as activity against some other rice phytopathogenic fungi. The active compound was purified through size-exclusion chromatography and preparative High-performance liquid chromatography. The molecular weight was determined as m/z 1043 by mass spectroscopy, which is identical to that of iturin A. Furthermore, a crude extract from the culture supernatant of Bacillus subtilis MJMP2 showed inhibitory activity against rice blight disease in both a rice leaf explant assay and a pot assay. The crude extract also enhanced the length of roots of Arabidopsis thaliana. These results suggest that the strain Bacillus subtilis MJMP2 could be used as a biological agent to control rice blight disease.

Virulence Reduction and Differing Regulation of Virulence Genes in rpf Mutants of Xanthomonas oryzae pv. oryzae

  • To define the functions of the rpf genes in Xanthomonas oryzae pv. oryzae (Xoo), which regulates pathogenicity factors in Xanthomonas campestris pv. campestris (Xcc), marker-exchange mutants of each rpf gene were generated. When the mutants were inoculated on a susceptible cultivar, the lesion lengths caused by the rpfB, rpfC, rpfF, and rpfG mutants were significantly smaller than those caused by the wild type, whereas those caused by the rpfA, rpfD, and rpfI mutants were not. Several virulence determinants, including extracellular polysaccharide (EPS) production, xylanase production, and motility, were significantly decreased in the four mutants. However, the cellulase activity in the mutants was unchanged. Complementation of the rpfB and rpfC mutations restored the virulence and the expression of the virulence determinants. Expression analysis of 14 virulence genes revealed that the expression of genes related to EPS production (gumG and gumM), LPS (xanA, xanB, wxoD, and wxoC), phytase (phyA), xylanase (xynB), lipase (lipA), and motility (pitA) were reduced significantly in the mutants rpfB, rpfC, rpfF, and rpfG. In contrast, the expression of genes related to cellulase (eglxob, clsA), cellobiosidase (cbsA), and iron metabolism (fur) was unchanged. The results of this study clearly show that rpfB, rpfC, rpfF, and rpfG are important for the virulence of Xoo KACC10859, and that virulence genes are regulated differently by the Rpfs.

Xanthomonas oryzae pv. oryzae RpfE Regulates Virulence and Carbon Source Utilization without Change of the DSF Production

  • It has been known that most regulation of pathogenicity factor (rpf ) genes in xanthomonads regulates virulence in response to the diffusible signal factor, DSF. Although many rpf genes have been functionally characterized, the function of rpfE is still unknown. We cloned the rpfE gene from a Xanthomonas oryzae pv. oryzae (Xoo) Korean race KACC10859 and generated mutant strains to elucidate the role of RpfE with respect to the rpf system. Through experiments using the rpfE-deficient mutant strain, we found that mutation in rpfE gene in Xoo reduced virulence, swarm motility, and production of virulence factors such as cellulase and extracellular polysaccharide. Disease progress by the rpfE-deficient mutant strain was significantly slowed compared to disease progress by the wild type and the number of the rpfE-deficient mutant strain was lower than that of the wild type in the early phase of infection in the inoculated rice leaf. The rpfE mutant strain was unable to utilize sucrose or xylose as carbon sources efficiently in culture. The mutation in rpfE, however, did not affect DSF synthesis. Our results suggest that the rpfE gene regulates the virulence of Xoo under different nutrient conditions without change of DSF production.
